Determination of the new psychoactive substance dichloropane in saliva by microextraction by packed sorbent - Ion mobility spectrometry.

A simple procedure based on microextraction by packed sorbent (MEPS) has been proposed for the extraction of dichloropane in oral fluids and its determination by ion mobility spectrometry (IMS). Extraction conditions such as type of sorbent (octyl and octadecyl silica), sample pH, number of sample loadings, and elution volume were evaluated to obtain the most appropriate values. Dichloropane was extracted from saliva samples using C8 MEPS, loading with 100 μL sample (adjusted to pH 7) in 4 cycles, washing with 100 μL deionized water, and eluting with 50 μL 2-propanol in 10 cycles. The proposed

Computational methods for metabolomic data analysis of ion mobility spectrometry data-reviewing the state of the art.

Ion mobility spectrometry combined with multi-capillary columns (MCC/IMS) is a well known technology for detecting volatile organic compounds (VOCs). We may utilize MCC/IMS for scanning human exhaled air, bacterial colonies or cell lines, for example. Thereby we gain information about the human health status or infection threats. We may further study the metabolic response of living cells to external perturbations. The instrument is comparably cheap, robust and easy to use in every day practice. However, the potential of the MCC/IMS methodology depends on the successful application of computat

Characterization of TATP gas phase product ion chemistry via isotope labeling experiments using ion mobility spectrometry interfaced with a triple quadrupole mass spectrometer.

Identification of the fragment ion species associated with the ion reaction mechanism of triacetone triperoxide (TATP), a homemade peroxide-based explosive, is presented. Ion mobility spectrometry (IMS) has proven to be a key analytical technique in the detection of trace explosive material. Unfortunately, IMS alone does not provide chemical identification of the ions detected; therefore, it is unknown what ion species are actually formed and separated by the IMS. In IMS, ions are primarily characterized by their drift time, which is dependent on the ion׳s mass and molecular cross-section; thu

Probing Gas-Phase-Clustering Thermodynamics with Ion Mobility-Mass Spectrometry: Association Energies of Phenylalanine Ions with Gas-Phase Alcohols.

Vapor assisted mobility shift measurements were made with atmospheric pressure drift-tube ion mobility-mass spectrometry (IM-MS) to determine the thermodynamic properties of weakly bound ion-molecule clusters formed from protonated phenylalanine and neutral vapor molecules with hydroxyl functional groups. Relative binding energies and gas-phase association energies of amino acid ions clustered with small organic molecules have been established previously using high-pressure mass spectrometry. However, the issue of volatility largely prohibits the use of high-pressure mass spectrometry for the

Rapid Recognition of Matrine Enantiomers Using Ion Mobility-Mass Spectrometry and Theoretical Modeling with Macrolide as Chiral Selectors.

The precise analysis of chirality is crucial for elucidating their stereoselective effects. Ion mobility-mass spectrometry (IM-MS) is a promising tool for chirality analysis, yet it is constrained by limited resolving power and the correlation between collision cross-section (CCS) values and accurate structures. In this work, a rapid method for recognizing matrine (MAT) and its enantiomer sophoridine (SPD) was established using IM-MS and theoretical modeling. With Natamycin (Nat) and Amphotericin B (Amp) as chiral selectors, binary noncovalent complexes were formed with target analytes to enha

Overexpression of GM3 and Ganglioside Pattern Remodeling in Lung Adenocarcinoma Brain Metastases Identified by Ion Mobility Mass Spectrometry.

Lung adenocarcinoma (LUAD), the most prevalent subtype of non-small cell lung carcinoma (NSCLC), commonly metastasizes to the brain, particularly in advanced stages. Since brain metastases (BMs) are a leading cause of morbidity and mortality in LUAD patients, their early detection is critical, necessitating the identification of reliable biomarkers. Gangliosides (GGs), a class of bioactive glycosphingolipids involved in cell signaling, adhesion, and immune regulation, have emerged as promising candidates for diagnostic and therapeutic targeting in LUAD-associated brain metastases (BMLA). [Development of Enantioselective Imaging Method for Chiral Molecules by on-Tissue Chiral Derivatization and Ion Mobility Mass Spectrometry].

Bioactive molecules are mostly chiral, and their enantiomers often exhibit different biological properties, including pharmacological effects and toxicity. Recent biochemical studies have revealed that certain trace-level chiral metabolites are associated with specific pathological conditions, including chronic kidney disease and metabolic disorders. This finding highlights the importance of enantioselective imaging techniques that can help visualize the spatial distribution and dynamic behavior of individual enantiomers. Although enantioselective biochemical analyses, such as those based on c

Characterization and Discrimination of Volatile Compounds of Donkey and Horse Meat Based on Gas Chromatography-Ion Mobility Spectrometry.

The production of high-quality specialty meats has emerged as a prominent research focus within the livestock industry, under the broader concept of big food. However, the composition and variances of volatile compounds (VOCs) in donkey meat (DM) and horse meat (HM) remain unclear, which complicates their effective identification. In the present study, the VOCs of DM and HM were analyzed using gas chromatography-ion mobility spectrometry (GC-IMS) in combination with a multivariate analysis. Our results indicate that a total of 39 VOCs were identified in both DM and HM. These VOCs were categori

Understanding the structural dynamics of human islet amyloid polypeptide: Advancements in and applications of ion-mobility mass spectrometry.

Human islet amyloid polypeptide (hIAPP) forms amyloid deposits that contribute to β-cell death in pancreatic islets and are considered a hallmark of Type II diabetes Mellitus (T2DM). Evidence suggests that the early oligomers of hIAPP formed during the aggregation process are the primary pathological agent in islet amyloid induced β-cell death. The self-assembly mechanism of hIAPP, however, remains elusive, largely due to limitations in conventional biophysical techniques for probing the distribution or capturing detailed structures of the early, structurally dynamic oligomers. The advent of I

Characterization of Volatile Compounds in Donkey Meat by Gas Chromatography-Ion Mobility Spectrometry (GC-IMS) Combined with Chemometrics.

Volatile compounds (VOCs) are an important factor affecting meat quality. However, the characteristic VOCs in different parts of donkey meat remain unknown. Accordingly, this study represents a preliminary investigation of VOCs to differentiate between different cuts of donkey meat by using headspace-gas chromatography-ion mobility spectrometry (HS-GC-IMS) combined with chemometrics analysis. The results showed that the 31 VOCs identified in donkey meat, ketones, alcohols, aldehydes, and esters were the predominant categories. A total of 10 VOCs with relative odor activity values ≥1 were found

Exploring the potential of using ion mobility-mass spectrometry to separate matrix interferences from analytes in food control.

During residue analysis in complex matrices for food safety purposes, interfering signals can sometimes overlap with those of the analyte of interest. Access to an additional separation dimension besides chromatographic and mass separation, such as ion mobility, can aid in removing interfering signals, allowing for correct analyte identification in these cases. In our laboratory, during routine LC-MS/MS analysis of liver samples for growth promoter residues, an interfering signal was found that matches the retention time and m/z values for stanozolol, a synthetic anabolic steroid. In the prese

Screening apo-SOD1 conformation stabilizers from natural flavanones using native ion mobility mass spectrometry and fluorescence spectroscopy methods.

RATIONALE: A large number of studies have shown that the production of aberrant and deleterious copper zinc superoxide dismutase (SOD1) species is closely related to amyotrophic lateral sclerosis (ALS). Therefore, it is of great significance to screen effective inhibitors of misfolding and aggregation of SOD1 for treating ALS disease. METHODS: The interaction between flavanone compounds with apo-SOD1was investigated using native electrospray ion mobility mass spectrometry (native ESI-IM-MS). Binding affinities of ligands were compared using native MS, ESI-MS/MS, collision-induced unfolding, a

Analyzing the Photo-oxidation of 2-propanol at Indoor Air Level Concentrations Using Field Asymmetric Ion Mobility Spectrometry.

We demonstrate a versatile protocol to be used for determining the effectiveness of photocatalysts in degrading indoor air concentration (ppb) volatile organic carbons (VOCs), illustrating this with a titanium dioxide based catalyst, and the VOC 2-propanol. The protocol takes advantage of field asymmetric ion mobility spectroscopy (FAIMS), an analysis tool that is capable of continuously identifying and monitoring the concentration of VOCs such as 2-propanol and acetone at the ppb level. The continuous nature of FAIMS allows detailed kinetic analysis, and long-term reactions, offering a signif

Evaluation of high-field asymmetric waveform ion mobility spectrometry coupled to nanoelectrospray ionization for bioanalysis in drug discovery.

The potential of high-field asymmetric waveform ion mobility spectrometry (FAIMS) coupled to nanoelectrospray ionization (nanoESI) as a method to improve sample throughput for bioanalysis in a discovery pharmaceutical setting was explored in this work. The ability of FAIMS to separate gas-phase ions in the millisecond timescale was exploited to eliminate the need for liquid chromatography. Samples were introduced into the FAIMS electrodes/mass spectrometer using offline nanoESI at 20 nL/min and 1.5 kV. Signals were averaged for 30 s after which the next sample could be analyzed. The separation

Investigation of the reaction of thiomolybdate and copper by electrospray-trapped ion mobility-mass spectrometry (ESI-TIMS-MS).

Molecular mass spectrometry was utilized for the first time to gather information of formed compounds consisting of tetrathiomolybdate (TTM) and Cu in the context of Wilson's disease (WD). Electrospray ionization (ESI)-mass spectrometry was used to elucidate four in vitro-formed TTM-Cu compounds, including MoS4Cu-, (MoS4)2Cu3-, (MoS4)3Cu5-, and (MoS4)4Cu7-. Based on the ions' net charges, it could be concluded that Cu(II) had been reduced to Cu(I), which then binds to TTM. By increasing the potential applied to the ESI source, it was observed that the heavier compounds fragment into MoS4Cu-, w
