Structural elements required for coupling ion and substrate transport in the neurotransmitter transporter homolog LeuT.

The coupled transport of ions and substrates allows transporters to accumulate substrates using the energy of transmembrane ion gradients and electrical potentials. During transport, conformational changes that switch accessibility of substrate and ion binding sites from one side of the membrane to the other must be controlled so as to prevent uncoupled movement of ions or substrates. In the neurotransmitter:sodium symporter (NSS) family, Na+ stabilizes the transporter in an outward-open state, thus decreasing the likelihood of uncoupled Na+ transport. Substrate binding, in a step essential fo

Coupled transport of water and ions in intercalated graphene nanochannels.

Understanding the coupled transport of water and ions under nanoscale confinement is essential for advancing nanofluidic applications. In this work, we employed molecular dynamics simulations to investigate the electrohydrodynamic transport behavior of aqueous NaCl solution within graphene nanochannels containing an intercalated graphene layer. The results show that in a narrow channel, the intercalated layer induces strong confinement effects, leading to complete suppression of anion migration and nearly 100% unidirectional water transport, resembling the function of biological nanoscale pump

Preeclampsia and transport of ions and small molecules: A literature review.

Preeclampsia (PE) is a prevalent obstetric complication affecting approximately 3-5% of pregnancies worldwide and is a major cause of maternal and perinatal morbidity and mortality. Preeclampsia is considered a disease of the endothelial system that can progress to eclampsia, characterized by seizures. Early diagnosis and appropriate management are crucial to improving maternal and fetal outcomes, as preeclampsia can lead to severe complications such as placental abruption, fetal growth restriction, and stroke. The pathophysiology of PE is complex, involving a combination of genetic, acquired,

Progress and prospects toward supramolecular bioactive ion transporters.

The majority of cellular physiological processes depend on natural ion channels, which are pore-forming membrane-embedded proteins that let ions flow across the cell membranes selectively. This selective movement of ions across the membranes balances the osmolality within and outside the cell. However, mutations in the genes that encode essential membrane transport proteins or structural reorganisation of these proteins can cause life-threatening diseases like cystic fibrosis. Artificial ion transport systems have opened up a way to replace dysfunctional natural ion channels to cure such disea

Membrane transport proteins in melanosomes: Regulation of ions for pigmentation.

Melanosomes are unique organelles in melanocytes that produce melanin, the pigment for skin, hair, and eye color. Tyrosinase is the essential and rate-limiting enzyme for melanin production, that strictly requires neutral pH for activity. pH maintenance is a result of the combinational function of multiple ion transport proteins. Thus, ion homeostasis in melanosomes is crucial for melanin synthesis. Defect of the ion transport system causes various pigmentation phenotypes, from mild effect to severe disorders such as albinism. In this review, we summarize the up-to-date knowledge of the ion tr

Analysis of changes in sodium and chloride ion transport in the skin.

The measurement of electric potential and resistance reflect the transport of sodium and chloride ions which take place in keratinocytes and is associated with skin response to stimuli arising from external and internal environment. The aim of the study was to assess changes in electrical resistance and the transport of chloride and sodium ions, under iso-osmotic conditions and following the use of inhibitors affecting these ions' transport, namely amiloride (A) and bumetanide (B). The experiment was performed on 104 fragments of rabbit skin, divided into three groups: control (n = 35), A-inhi

Indispensable Role of Ion Channels and Transporters in the Auditory System.

Ear is a complex system where appropriate ionic composition is essential for maintaining the tissue homeostasis and hearing function. Ion transporters and channels present in the auditory system plays a crucial role in maintaining proper ionic composition in the ear. The extracellular fluid, called endolymph, found in the cochlea of the mammalian inner ear is particularly unique due to its electrochemical properties. At an endocochlear potential of about +80 mV, signaling initiated by acoustic stimuli at the level of the hair cells is dependent on the unusually high potassium (K+ ) concentrati

The family of SMF metal ion transporters in yeast cells.

Metal ions are vital for all organisms, and metal ion transporters play a crucial role in maintaining their homeostasis. The yeast (Saccharomyces cerevisiae) Smf transporters and their homologs in other organisms have a central role in the accumulation of metal ions and their distribution in different tissues and cellular organelles. In this work we generated null mutations in each individual SMF gene in yeast as well as in all combinations of the genes. Each null mutation exhibited sensitivity to metal ion chelators at different concentrations. The combination of null mutants DeltaSMF1 + Delt

Molecular cloning of ion transporters: potential clinical implications.

The ion transporters are a group of integral membrane proteins which, like enzymes, can be regulated at multiple levels. Understanding of function and regulation of these proteins has been greatly facilitated by the knowledge of their molecular structures. In this article, we discuss the various approaches used in the cloning of these proteins. We then focus on genetic disorders known to be caused by abnormal structure of the transporter protein including cystic fibrosis, generalized myotonia and myotonia congenita, hyperkalemic periodic paralysis, hereditary ovalocytosis, hereditary spherocyt

Transport of organic ions through lipid bilayers. The barbiturates.

In a previous article [Naunyn-Schmiedeberg's Arch Pharmacol (1981) 316:266-272] transport of a number of organic ions across lipid bilayers was investigated using single bilayer liposomes. In this investigation the translocation of structurally closely related barbiturates across lipid bilayers is studied. The anionic form of the barbiturates can pass lipid bilayers, provided that the lipophilicity of the 5,5-disubstituted side chain is high enough. This permeability of barbiturates in the ionized form explains their uncoupling action of the oxidative phosphorylation and the deviations from th

Programmable Inter-Droplet Communication by Polymer-Gated Ion Transport Through Nanopore Lattices.

Owing to their hydrophilicity and pronounced capillary pressures, mesoporous platforms enable isotropic or multidirectional fluid transport. Based on aqueous salt solution droplets, we investigate nanopore-driven ion transport control for tailorable droplet communication via a mesoporous surface. Droplets were placed on a mesoporous silica thin film, separated by a polyelectrolyte (PEL) barrier. The PEL-barrier was incorporated into the mesoporous matrix by automated direct-laser-writing and a grafting-from SI-PET-RAFT polymerization. Permselective transport-regulated barrier breakthrough time

Phosphonium outperforms ammonium poly(ionic liquids) for fast ion transport polymer electrolytes.

Cationic poly(ionic liquids) (polyILs) are promising solid polymer electrolytes, yet the role of cationic charge-centre chemistry in regulating alkali-metal transport and interfacial behaviour remains poorly understood. Here, we computationally investigated a phosphonium-based polyIL at high lithium or sodium concentrations and compared these systems with previously reported ammonium-based analogues. The phosphonium systems demonstrated superior alkali-metal ion diffusion, which was consistent with their high ionic conductivities. Molecular-level analyses show that phosphonium cation chemistry

MOF in Polymer Electrolytes Raising Ion Transport for Breakthrough Lithium Metal Batteries.

SPEs have gained significant attention in lithium metal batteries for enhanced safety. However, low ionic conductivity and dendrite formation limit the performance. To address these limitations, a novel composite-type SPE was developed by incorporating phytic acid(PA)-functionalized ZIF-67 (ZIFP) into (N-Methyl-2-Pyrrolidone)NMP-modified (Poly(vinylidenefluoride-co-vinylchloride-co-vinylidenefluoride)_PVDF-HFP Solid polmer electrolyte(SPE). Programming Ion Transport in Layered 2D Hybrid Membranes via Coordination-Occupation Coupling.

The sustainable separation of chemically distinct yet notoriously difficult-to-separate metal ions is a critical challenge for nuclear waste management and strategic resource recovery. Layered two-dimensional (2D) membranes offer energy-efficient alternatives to conventional extraction and adsorption technologies, yet their selectivity is often constrained by fixed and poorly tunable transport pathways. Here we introduce a coordination-occupation-coupled strategy to program ion transport in layered 2D hybrid membranes combining a confined polyacrylate network between layered vermiculite. Irrev

Reprogramming Ion-Transport Dimensionality via Crystal-Channel Engineering to Stabilize Zinc Anodes.

Unstable zinc (Zn) deposition in aqueous zinc-ion batteries is intrinsically linked to the interfacial confinement of Zn2+ transport, where ion migration is dominated by lateral diffusion along the electrode surface. This quasi-two-dimensional transport amplifies local electric-field and concentration heterogeneities, leading to uneven nucleation and dendritic growth. Here, we demonstrate that Zn deposition can be fundamentally regulated by reprogramming the dimensionality of ion transport. A three-dimensionally interpenetrated covalent organic framework (COF) incorporating crown-ether moietie

Atomistic insights into water and ion transport in GO-MoS2 hybrid membranes.

Hybrid membranes composed of graphene oxide (GO) and molybdenum disulfide (MoS2) offer promising opportunities for overcoming the permeability-selectivity trade-off in membrane-based desalination. However, the influence of layer composition and stacking sequence on water and ion transport remains insufficiently understood at the molecular level. In this work, non-equilibrium molecular dynamics simulations were employed to systematically investigate pressure-driven water and ion transport through six GO-MoS2 membrane architectures, including pristine GO and MoS2 membranes and four hybrid stacki

Reversible Bond Dynamics Enable Crystallinity-Healed COF Membranes for Selective Ion Transport.

Covalent organic frameworks (COFs) offer ordered, nanometer-scale channels with programmable chemistry and topology, making them promising membrane materials for selective ion transport. However, fabricating robust COF membranes that preserve high crystallinity remains a key challenge. Here we directly address this challenge by decoupling crystallization from membrane formation. COF membranes are first made by interfacial polymerization and subsequently healed under acid-catalyzed hydrothermal conditions, which activate reversible COF linkage bond exchange and framework self-correction. From Ion Channels to Blood Pressure: Genetic Disorders of Renal Tubular Transport.

Blood pressure regulation is fundamentally dependent on renal sodium and electrolyte handling. Genetic disorders of renal tubular transport provide representative evidences that illuminate the molecular mechanisms linking ion channels to systemic hemodynamics. Monogenic conditions such as Bartter syndrome, Gitelman syndrome, Liddle syndrome, and Gordon syndrome demonstrate how specific alterations in tubular sodium, potassium, chloride, and magnesium transport translate into distinct blood pressure phenotypes. Salt-wasting disorders are characterized by hypokalemic metabolic alkalosis and low

Covalent Organic Framework Quantum Dots for Near-Frictionless Ion Transport Battery Electrolyte.

Optimization of electrolyte with enhanced energy density and durability in battery system is highly demanded yet challenging owing to the high ion transport resistance and complex by-reactions in electrolyte. In this work, we tailored the solid-liquid phase transport interface by adding covalent organic frameworks quantum dots (COFs QDs) into electrolyte. COFs QDs with abundant lipophilic ether-oxygen groups and ∼12 nm nano-size can serve as myriad ion-transport nano-engines for constantly replenishing the interface with Li+ to achieve near-frictionless ion transport (tLi + = 0.98) and excepti
