Keratins Regulate p38MAPK-Dependent Desmoglein Binding Properties in Pemphigus.

Keratins are crucial for the anchorage of desmosomes. Severe alterations of keratin organization and detachment of filaments from the desmosomal plaque occur in the autoimmune dermatoses pemphigus vulgaris and pemphigus foliaceus (PF), which are mainly caused by autoantibodies against desmoglein (Dsg) 1 and 3. Keratin alterations are a structural hallmark in pemphigus pathogenesis and correlate with loss of intercellular adhesion. However, the significance for autoantibody-induced loss of intercellular adhesion is largely unknown. In wild-type (wt) murine keratinocytes, pemphigus autoantibodie

Ultrastructural identification of basic abnormalities as clues to genetic disorders of the epidermis.

The present article discusses specific, directly gene-dependent ultrastructural markers of dominantly inherited epidermal disorders that serve as clues to their underlying molecular genetic abnormalities. These are epidermolysis bullosa simplex Koebner and Weber-Cockayne with rupture or non-assembly of basal cell keratins and point mutations in keratins 5 and 14. Clumping of basal cell keratins is pathognomonic of EB Dowling-Meara and caused by mutations in hot spots of the rod domain of K5 and K 14. Clumps and aggregates of basal keratins occur side by side in the same cell and thus do not in

Dual-specificity phosphatases in the hypo-osmotic stress response of keratin-defective epithelial cell lines.

Although mutations in intermediate filament proteins cause many human disorders, the detailed pathogenic mechanisms and the way these mutations affect cell metabolism are unclear. In this study, selected keratin mutations were analysed for their effect on the epidermal stress response. Expression profiles of two keratin-mutant cell lines from epidermolysis bullosa simplex patients (one severe and one mild) were compared to a control keratinocyte line before and after challenge with hypo-osmotic shock, a common physiological stress that transiently distorts cell shape. Fewer changes in gene exp

Bullous ichthyosiform erythroderma, developmental delay, aortic and pulmonary stenosis in association with a FRA12A.

We present an 11-year-old female with bullous ichthyosiform erythroderma (BIE), learning disability, patent ductus arteriosus and mild stenosis of the aortic and pulmonary arteries. Chromosome analysis showed the expression of the rare folate-sensitive fragile site FRA12A at 12q13 in 8/20 (40%) of blood lymphocytes cultured in folate-deficient medium in the presence of trimethoprim. Her mother and maternal grandmother are phenotypically normal, but her mother shows expression of the same fragile site in 4/20 (20%) of cells cultured under the same conditions. Lymphocytes from the grandmother on

Elastic globes: electron microscopic and immunohistochemical observations.

Specimens from a patient with epidermolysis bullosa contained many elastic globes in the dermis. Ultrastructurally they were composed of (i) medium electron-dense amorphous substances, (ii) electron-dense round structures, and (iii) fine filaments. These various elements were seldom organized into typical normal elastic fiber and, therefore, it was difficult ultrastructurally to recognize them as such or components thereof. Immunohistochemically, elastic globes were strongly reactive with NKH-1, which stains elastic microfibrils, and antibody to serum amyloid P component (anti-SAP), which bind

Clinical outcomes of a peri-implant keratinized mucosa augmentation procedure using concentrated growth factor: a prospective case series.

BACKGROUND: Adequate keratinized mucosa around implants is crucial for maintaining peri-implant hard and soft tissue health. Concentrated growth factor (CGF), a novel platelet concentrate, has been widely applied to promote tissue regeneration and wound healing, which could also be effective in peri-implant keratinized mucosa augmentation. This prospective case series aimed to present the clinical outcomes of apically repositioned flap (ARF) combined with CGF and to provide histological evidence. METHODS: Patients with insufficient keratinized mucosa (< 2 mm) were included. ARF combined with

Mucogingival problems, prevalence and therapy in children.

The clinical impressions of the authors and a survey of mucogingival problems in children lead to the following conclusions: 1. Mucogingival problems occur in children. 2. The prevalence of such problems is 12 to 19% in sample of 100 patients examined. 3. Insufficient keratinized tissue is developmentally related to: a. eruption pattern of the permanent incisors. b. buccolingual width of the alveolar process. 4. The autogenous free gingival graft is recommended as an acceptable procedure to prevent incipient mucogingival problems from progressing. 5. Where orthodontic therapy is anticipated an

Mesothelioma with signet-ring cell features: report of 23 cases.

Signet-ring cell mesothelioma is uncommon and only two case reports have been published on this mesothelioma variant, both of which were initially misdiagnosed as signet-ring cell carcinoma. Herein are reported 23 signet-ring cell mesotheliomas that were investigated by immunohistochemistry, 12 of which were also studied by electron microscopy. Twenty-one of the cases originated in the pleura and two in the peritoneum. For comparison purposes and in order to determine the value of these techniques in the differential diagnosis of these tumors, seven cases of signet-ring cell lung adenocarcinom

Solution blow spun keratin - PVP nanofibers as a green platform for high-efficiency curcumin encapsulation and controlled release.

Despite its wide range of beneficial pharmacological effects, curcumin (Cur) has limited clinical applicability due to its extremely low water solubility. In this study, a novel curcumin-loaded nanofiber system was fabricated by combining polyvinylpyrrolidone (PVP) and keratin (Ker) using the solution blowing spinning (SBS) method. The nanofibers (Cur-loaded PxKyF) were produced using water as the solvent, without the use of any crosslinking agents. The incorporation of keratin resulted in thinner (∼ 349 nm) and more uniform nanofibers, with enhanced elongation at break and toughness. Among th

Reduced Keratin-Based Bioadhesives for Peri-Implant Soft Tissue Sealing around Transcutaneous Metallic Prostheses.

When a percutaneous metallic implant breaches the skin barrier, the lack of peri-implant soft tissue sealing arising from a foreign body reaction and epithelial downgrowth results in delayed wound healing and closure. This facilitates microbial pathogen entry, leading to infection, inflammation, and ultimately, peri-implantitis. Toward this end, bioglue formulations of -human hair keratin (HHK) and chicken feather keratin coupled with polyphenols, dopamine (DOPA), quercetin (Q), and tannic acid (TA) were developed for peri-implant soft tissue sealing. A freeze-thaw gelation mediated by thiol r

Mapping of epidermolysis bullosa simplex mutation to chromosome 12.

Epidermolysis bullosa simplex (EBS) is a dominantly inherited genodermatosis characterized by intraepidermal blister formation. Recent reports have suggested that EBS mutations may relate to keratin abnormalities. In this study, we conducted RFLP analyses to test the hypothesis that EBS is linked to one of the keratin gene clusters on chromosome 12 or chromosome 17. Although these keratin gene loci are not defined by RFLPs, several mapped RFLPs in the same chromosomal regions could be tested for linkage. A large EBS family with 14 affected and 12 unaffected individuals in three generations was

Role of miR-182 in response to oxidative stress in the cell fate of human fallopian tube epithelial cells.

BACKGROUND: Epidermolysis bullosa simplex (EBS) is an inherited skin fragility disorder caused by mutations in keratin intermediate filament proteins. While discoveries of these mutations have increased understanding of the role of keratins and other intermediate filaments in epithelial tissues, progress towards the development of therapy for these disorders is much slower. OBJECTIVES: Cell culture model systems that display these structural defects are needed for analysis of the cellular consequences of the mutations and to enable possible therapeutic strategies to be developed. Under Pressure: A unique mechanoresponsive mechanism of body site-specific keratin regulation in palmoplantar epidermis.

UNLABELLED: Palmoplantar skin is structurally and molecularly distinct from other body sites. Notably, the type 1 keratin 9 ( KRT9 /K9) is exclusively expressed in palmoplantar epidermis. Mutations in KRT9 /K9 are causative for epidermolytic palmoplantar keratoderma (EPPK), a genetic disorder typified by palmoplantar keratoderma. Surprisingly little is known about the ontogeny, regulation and significance of Krt9 /K9. Here we characterize the regulation of Krt9 /K9 in postnatal palmoplantar epidermis and uncover a novel role of K9 in modulating YAP1 signaling. Expression of Krt9/ K9 rises dram

Keratin utilizes H2S to promote HMGB1 sulfhydration for inflammation resolution.

Despite the established importance of hydrogen sulfide (H2S) in redox biology, strategies to modulate its endogenous levels remain largely unexplored. Here, we show that a keratin fraction (KF) obtained from natural sources modulates H2S homeostasis and protein redox state through sulfhydration. Chemoproteomic analysis identified keratin 2 (K2; UniProtKB P25691, keratin, type II microfibrillar, component 5) as the most hyperreactive keratin toward sulfhydration. Oral administration of K2 to mice alleviated lipopolysaccharide (LPS)-induced inflammatory fever and led to the generation of K2-deri

Bisphenol pollutants bind with human hair keratin: Combining evidence from fluorescence spectroscopy and molecular docking.

Bisphenols, including bisphenol A (BPA) and its analogs such as bisphenol F (BPF), bisphenol S (BPS), tetrabromobisphenol A (TBBPA), tetrachlorobisphenol A (TCBPA) and tetrabromobisphenol S (TBBPS), are typical endocrine disruptors widely used in plastic production. However, until now, the occurrence mechanisms of these bisphenols in hair, a non-invasive material for human biomonitoring, have been inadequately explored. This study employed fluorescence spectroscopy and molecular docking to investigate the interactions between these 6 bisphenols and hair keratin. The findings revealed that thes

Deleterious mutations of a claw keratin in multiple taxa of reptiles.

We have recently shown that homologs of mammalian hair keratins are expressed in the claws of the green anole lizard, Anolis carolinensis. To test whether reptilian hair keratin homologs are functionally associated with claws, we investigated the conservation of the prototypical reptilian hair keratin homolog, hard acidic keratin 1 (HA1), in representative species from all main clades of reptiles. A complete cDNA of HA1 was cloned from the claw-forming epidermis of the lacertid lizard Podarcis sicula, and partial HA1 gene sequences could be amplified from genomic DNA of tuatara, lizards, gekko
