Occurrence of Ergasilus megaceros Wilson, 1916, in the sea lamprey and other fishes from North America.

Ergasilus megaceros (Copepoda: Ergasilidae) was recovered from the nasal fossae (lamellae) of the olfactory sac in 1 (1.8%) of 56 sea lampreys, Petromyzon marinus Linne, 1758, collected in May 2002 from the Cheboygan River, Michigan. Although the sea lamprey is a new host record for E. megaceros, this fish species may not be a preferred host because of its low prevalence. Ergasilus megaceros is the second ergasilid species reported from the sea lamprey in North America. This is the third report of an ergasilid species infecting the nasal fossae of fishes in North America, with E. rhinos being

Fine structure of the liver in the larval lamprey, Petromyzon marinus L.; bile ducts and gall bladder.

The purpose of the present investigation was to describe ultrastructural characteristics of hepatic bile ducts and the gall bladder in larvae of the sea lamprey, Petromyzon marinus, using freeze--fracture replicas as well as ultrathin sections. Comparison of these structures with those of other vertebrates was necessary to provide a basis in future studies for characterization of biliary degeneration during metamorphosis. The bile ducts were composed of a simple cuboidal to columnar epithelium with the cells separated by wide lateral intercellular spaces and containing a prominent brush border

The hypothalamo-hypophysial system of the lamprey, Lampetra fluviatilis L. II. The proximal neurosecretory contact region.

The proximal neurosecretory contact region (PNCR) of the lamprey, a homologue of the median eminence of tetrapods, was studied by light, fluorescence and electron microscopy. Paraldehyde fuchsin-positive neurosecretory fibers are seen mainly in the central part of the rostral subdivision of the PNCR. The Falck-Hillarp technique reveals a weak, mainly diffuse yellow-green fluorescence in the PNCR. The ultrastructure of the tanycyte layer of the PNCR is very similar to that in the neurohypophysis of the same species, although the funnel-shaped protrusions of the third ventricle in the rostral pa

Cholinergic and GABAergic neuronal elements in the pineal organ of lampreys, and tract-tracing observations of differential connections of pinealofugal neurons.

The putative cholinergic and GABAergic elements of the pineal organ of lampreys were investigated with immunocytochemistry to choline acetyltransferase (ChAT) and gamma-aminobutyric acid (GABA), and by acetylcholinesterase (AChE) histochemistry. For comparison we also carried out immunocytochemistry to serotonin (5-HT) and a tract-tracing investigation of the two types of projecting cells, i. e., ganglion cells and long-axon photoreceptors. Most photoreceptors were ChAT-immunoreactive (ChAT-ir) and AChE-positive, while ganglion cells and the pineal tract were ChAT-negative and AChE-negative or

A comparison of endocrine pancreatic tissue in adults of four species of lampreys in British Columbia: a morphological and immunohistochemical study.

Histological techniques and immunohistochemistry with antisera against insulin and somatostatin were used in a study of the identification and the distribution of endocrine pancreatic tissue in adults of four species of lampreys in British Columbia. The fine structure of the pancreatic cells of one of the species was also examined. The four species (Lampetra ayresi, Lampetra tridentata, Lampetra macrostoma, and Lampetra richardsoni) all possess cranial and caudal pancreatic masses and an intervening intermediate cord, but there are variations in amount and deposition of the pancreatic tissue w

Adult sea lamprey tolerates biliary atresia by altering bile salt composition and renal excretion.

The sea lamprey (Petromyzon marinus) is a genetically programmed animal model for biliary atresia, as it loses its bile ducts and gallbladder during metamorphosis. However, in contrast to patients with biliary atresia or other forms of cholestasis who develop progressive disease, the postmetamorphosis lampreys grow normally to adult size. To understand how the adult lamprey thrives without the ability to secrete bile, we examined bile salt homeostasis in larval and adult lampreys. Adult livers were severely cholestatic, with levels of bile salts >1 mM, but no evidence of necrosis, fibrosis, or

Anatomical and molecular reinvestigation of lamprey endostyle development provides new insight into thyroid gland evolution.

The thyroid gland of vertebrates is considered to be homologous to the endostyle of non-vertebrate chordates (cephalochordates, urochordates), a key character for understanding the origin and evolution of the chordate body plan. In lampreys, the larval endostyle transforms into an adult thyroid gland during metamorphosis, reflecting evolutionary changes that occurred in the vertebrate lineage. Focussing on thyroid-like cells in the endostyle, we here relate morphologically visible steps of lamprey (Lampetra fluviatilis) endostyle differentiation to embryonic stages and determine the onset of t

Proliferating cell nuclear antigen (PCNA) immunoreactivity and development of the pineal complex and habenula of the sea lamprey.

The development of the pineal complex and the habenula of the sea lamprey was studied with proliferating cell nuclear antigen (PCNA) immunocytochemistry. The pineal organ and the habenula primordia appeared in late embryos, and neuron differentiation began in prolarvae, as indicated by the presence of PCNA-negative cells. The parapineal primordium could not be distinguished until early prolarval stages, and cell differentiation was delayed to the larval period. Although the number of cycling (PCNA-immunoreactive) cells gradually decreased during the larval period in the three organs studied, t

Three-dimensional reconstruction of serotonin-immunoreactive photoreceptors in the pineal organ of the river lamprey, Lampetra japonica.

Serotonin-immunoreactive (5-HT IR) photoreceptors are present in the pineal complex (pineal and parapineal organ) of the river lamprey, Lampetra japonica. They are so-called modified pineal photoreceptors and have been regarded as photoneuroendocrine cells which secrete melatonin. We reconstructed 5-HT IR cells with a computer to demonstrate their three-dimensional structures from optical sections taken by a confocal laser scanning microscope. The 5-HT IR cell possesses a basal process, and it appears that the process does not branch out. These processes contact each other at the basal region

Time course of locomotor recovery and functional regeneration in spinal cord-transected lamprey: in vitro preparations.

1. Previous studies indicate that after transection of the rostral spinal cord, larval lamprey begin to recover locomotor behavior 2 wk posttransection and recovery is complete at approximately 8 wk. To examine the mechanisms underlying behavioral recovery after spinal cord transection, in the present study the time course and extent of recovery of locomotor function was examined in in vitro brain/spinal cord preparations. With these preparations the contributions of functional regeneration of descending brain stem projections to recovery of spinal locomotor function can be examined in the abs

Calcium-dependent potassium channels play a critical role for burst termination in the locomotor network in lamprey.

1. The possible involvement of calcium-dependent potassium channels (KCa) in the termination of locomotor bursts was investigated by administration of a specific blocker, apamin, in the lamprey spinal cord in vitro. The effects were examined by recording the efferent activity in ventral roots and by intracellular recording from interneurons and motoneurons. During fictive locomotion induced by N-methyl-D-aspartate (NMDA), apamin was found to affect both the frequency of bursting and the regularity of the locomotor pattern. 2. At the single cell level, NMDA can induce pacemaker-like membrane po

Signal transmission from pineal photoreceptors to luminosity-type ganglion cells in the lamprey, Lampetra japonica.

In order to study the signal transmission from pineal photoreceptors to luminosity (achromatic)-type ganglion cells of the lamprey, Lampetra japonica, the electrical activity of these cell groups was investigated using intra- and extracellular electrodes. By intracellular recording, it was shown that the photoreceptor cells responded to flashes of light with hyperpolarizations, and the ganglion cells also hyperpolarized with concurrent suppression of spike discharges. Concerning the slow membrane potentials, the light intensity-response relationships of both cell groups followed the Naka-Rusht

A light- and electron-microscopic study of the pineal complex of the ammocoete larva of the southern lamprey Geotria australis.

The pineal complex of larval Geotria australis lampreys has been examined by light- and electron-microscopy. The complex consists of a pineal organ and a smaller parapineal organ, the former being situated dorsal to the latter. It is concluded that the pineal organ is a functioning photoreceptor, with neural and endocrine output. The parapineal appears to be a more basic neuroendocrine organ without photosensory capability. Day-night comparisons of the pineal complex show no obvious differences in ultrastructure; this could be related to the chronobiological role of the complex as a constant m

[Optical spectroscopy of methemoglobin from the Lampetra fluviatilis river lamprey and its complexes reduced at 77K].

The absorption spectra from met-haemoglobin from lamprey Lampetra fluviatilis and its complexes with F-, CN-, N3- and imidazole were studied in non-equilibrium states arising in low-temperature (77K) reduction of water-glycerol of oxidized forms of protein with thermolyzed electrons. The Soret band was shown to be not split in low-temperature absorption spectra of reduced met-haemoglobin and its complexes unlike that of the equilibrium deoxyhaemoglobin. It was concluded that met-haemoglobin has a higher symmetry of the active centre as compared to deoxyhaemoglobin.

Periductal fibrosis and lipocytes (fat-storing cells or Ito cells) during biliary atresia in the lamprey.

Transmission and scanning electron microscopy were utilized to follow the degeneration of bile ducts of lampreys (Petromyzon marinus L.) during metamorphosis. The convoluted bile ducts of larval lampreys are surrounded by rich sinusoids, but this intimate biliovascular relationship is lost during metamorphosis because the bile duct degeneration is accompanied by the development of thick periductal fibrosis. Lipocytes, which are present not only in the parenchyma but also in the interstitial tissue of the liver, increase in number in the periductal fibrous tissue, and their processes are direct

Thermostable lamprey variable lymphocyte receptor antibody for detection of Plasmodium falciparum histidine rich protein-2.

Lampreys use variable lymphocyte receptors (VLR) comprising leucine-rich-repeat (LRR) segments for antigen recognition, distinct from immunoglobulin-based receptors of jawed vertebrates. Lamprey VLRs are as diverse and antigen-specific as immunoglobulin-based antibodies, with unique advantages including high avidity, pH stability, and recognition of novel antigen epitopes. Here we describe the generation of VLR monoclonal antibody against histidine rich protein-2 (HRP-2) of Plasmodium falciparum, a causative agent of malaria. HRP-2, expressed by all parasite stages and secreted into plasma, se

Lamprey FOXN1 rescues the block of thymic epithelial cell development in the mouse Foxn1-deficient thymic rudiment.

All vertebrate adaptive immune systems exhibit distinct lymphocyte lineages. In jawless vertebrates, such as lampreys, T-like cells are thought to develop in lympho-epithelial structures at the tips of gill filaments, termed thymoids. However, it is unclear whether thymoids are functionally equivalent to the thymus of jawed vertebrates. Indeed, because the structural modules that are somatically assembled to form the antigen receptors of jawless and jawed vertebrates differ, development and selection of T cells may be governed by clade-specific genetic networks. To address this question, we ha

A new clarification method to visualize biliary degeneration during liver metamorphosis in Sea Lamprey (Petromyzon marinus).

Biliary atresia is a rare disease of infancy, with an estimated 1 in 15,000 frequency in the southeast United States, but more common in East Asian countries, with a reported frequency of 1 in 5,000 in Taiwan. Although much is known about the management of biliary atresia, its pathogenesis is still elusive. The sea lamprey (Petromyzon marinus) provides a unique opportunity to examine the mechanism and progression of biliary degeneration. Sea lamprey develop through three distinct life stages: larval, parasitic, and adult. During the transition from larvae to parasitic juvenile, sea lamprey und

Functional characterization of two melanocortin (MC) receptors in lamprey showing orthology to the MC1 and MC4 receptor subtypes.

BACKGROUND: The melanocortin (MC) receptors have a key role in regulating body weight and pigmentation. They belong to the rhodopsin family of G protein-coupled receptors (GPCRs). The purpose of this study was to identify ancestral MC receptors in agnathan, river lamprey. RESULTS: We report cloning of two MC receptors from river lamprey. The lamprey receptors, designated MCa and MCb, showed orthology to the MC1 and MC4 receptor subtypes, respectively. The molecular clock analysis suggested that lamprey MC receptor genes were not duplicated recently and diverged from each other more than 400 M

Chloride transport in red blood cells of lamprey Lampetra fluviatilis: evidence for a novel anion-exchange system.

The existence of a furosemide-sensitive Cl- transport pathway activated by external Ca2+ and Mg2+ has been demonstrated previously in studies of Cl- influx across the lamprey erythrocyte membrane. The aim of the present study was to characterize further specific Cl- transport pathways, especially those involved in Cl- efflux, in the red blood cell membrane of Lampetra fluviatilis. Cl- efflux was inhibited by 0.05 mmol l-1 dihydroindenyloxyalkanoic acid (DIOA) (81%), 1 mmol l-1 furosemide (76%) and 0.1 mmol l-1 niflumic acid (54%).
