Porcine Leydig cells: central regulators of boar reproductive development and fertility.

Leydig cells within the interstitium of the testis produce the sex steroids (e.g., testosterone and 17β-estradiol), which are essential to male fertility. Unlike most species, pigs have three distinct phases of Leydig cell development and function. The first transient phase occurs during early fetal life, leading to the production of testosterone, which is critical for sex differentiation and early testicular development. The second transient phase in Leydig cell development occurs during the perinatal/neonatal period; during this developmental window, gonadal steroids further masculinize the

The Pregnane-X receptor regulates steroid synthesis in mouse Leydig cells.

INTRODUCTION: Pregnane X Receptor (PXR, NR1I2) is a ligand-dependent transcription factor belonging to the nuclear receptor superfamily, that can be activated by a wide variety of endogenous and exogenous ligands. It is a major actor of the endo- and xeno-biotic detoxification process. It also regulates biological processes such as lipid metabolism in large number of tissues. Pxr was shown to be expressed in human, mouse, rat and pig testis, however its roles in the regulation of testicular functions have been little explored so far. METHODS: To determine the potential involvement of PXR in t

Molecular Mechanisms Elicited by d-Aspartate in Leydig Cells and Spermatogonia.

A bulk of evidence suggests that d-aspartate (d-Asp) regulates steroidogenesis and spermatogenesis in vertebrate testes. This review article focuses on intracellular signaling mechanisms elicited by d-Asp possibly via binding to the N-methyl-d-aspartate receptor (NMDAR) in both Leydig cells, and spermatogonia. In Leydig cells, the amino acid upregulates androgen production by eliciting the adenylate cyclase-cAMP and/or mitogen-activated protein kinase (MAPK) pathways. d-Asp treatment enhances gene and protein expression of enzymes involved in the steroidogenic cascade. d-Asp also directly affe

Ectopic Leydig cells of testis An immunohistochemical study on tissue microarray.

The incidence of ectopic Leydig cells (LCs) has been reported to be 40% to 90%. Several theories have been proposed to account for ectopic LCs, including in situ differentiation, migration from testicular interstitium, and trapping of peritubular LCs in the tunica propria of the seminiferous tubule during its thickening. To document the nature of ectopic LCs and to compare them with interstitial LCs, Sertoli cells, epididymal cells, mesothelial cells, and epithelial cells in rete testis, immunostainings with calretinin, CD10, inhibin, CK7, and CK20 were performed in 40 cases of orchiectomy spe

Phthalate ester toxicity in Leydig cells: developmental timing and dosage considerations.

Humans have significant exposures to phthalates, as these chemical plasticizers are ubiquitously present in flexible plastics. Recent epidemiological evidence indicates that boys born to women exposed to phthalates during pregnancy have an increased incidence of congenital genital malformations and spermatogenic dysfunction, signs of a condition referred to as testicular dysgenesis syndrome (TDS). TDS is thought to develop as a result of environmental factors that cause a testicular disturbance at an early fetal stage with a resultant spectrum of clinical testicular dysfunction, ranging from i

Characterization of mouse glandular kallikrein 24 expressed in testicular Leydig cells.

Mouse kallikrein 24 is thought to encode a functional serine protease belonging to the mouse glandular kallikrein gene family. Preliminary results suggest that this kallikrein may play a role in testis function in adult mice. In order to obtain insights into its physiological functions, we undertook molecular and biochemical analyses of this enzyme. We cloned a cDNA for kallikrein 24 from the adult mouse testis cDNA library. Kallikrein 24 was expressed in the kidney, submandibular glands, ovary, epididymis, and testis of the mouse. In the testis, kallikrein 24 mRNA was detectable at 4 weeks of

[Evaluation of FSH and Leydig cells function in patients with varicocele].

The endocrine alterations in patients affected with varicocele are heterogenous in relation to some clinical aspects: orchidometry, bilateral lesion, decreased spermatozoa count and disease history. The decreased function of Leydig cells in patients affected with varicocele is related to alterations of the serotonin metabolism and to the impairment of testosterone biosynthesis. An exaggerated increase of FSH and LH after GnRH stimulation can be observed. The response to GnRH stimulation can predict a positive outcome of surgical treatment of infertile patients with varicocele. Finally it was t

Sport improves function of roosters testicular Leydig cells by mediating FUNDC1.

Roosters play a crucial role in breeder chicken production. A decline in reproductive performance during the late breeding stage is a major factor limiting economic returns. Testosterone, a key hormone for maintaining secondary sexual characteristics and supporting spermatogenesis, is primarily synthesized and processed within mitochondria. With advancing age, mitochondrial function deteriorates in roosters, leading to reduced testosterone synthesis and impaired reproductive capacity. This study aimed to elucidate the mechanisms through which rest and sport affect the reproductive performance

DAPK3 is Essential for DBP-Induced Autophagy of Mouse Leydig Cells.

Dibutyl phthalate (DBP) has been widely used in the manufacture of various daily and industrial products. As one of the most important endocrine disruptors, DBP has male reproductive toxicity and can lead to testicular dysfunction. In view of the fact that Leydig cells are important functional and structural units in the testis, their damage will affect testicular function. However, the underlying mechanism of DBP-caused damage to mouse Leydig cells remains elusive. In the study, it is confirmed that DBP can promote the expression of death-associated protein kinase 3 (DAPK3), thereby inducing

Trps1 targets Scarb1 to regulate cholesterol acquisition in mouse Leydig cells.

Approximately 7 % of men are suffering from infertility, accounting for 40 %-50 % of all cases of all infertility, and low testosterone levels are closely associated with male infertility. Our previous study indicated that transcriptional repressor GATA binding 1 (Trps1) could inhibit testosterone synthesis in Leydig cells. In the present study, to elucidate the molecular mechanism by which Trps1 regulates testosterone synthesis in Leydig cells, differentially expressed genes (DEGs) following Trps1 knockdown was conducted by RNA-sequencing. Cyclophosphamide activates ferroptosis-induced dysfunction of Leydig cells via SMAD2 pathway†.

Cyclophosphamide (CP) is a widely used chemotherapeutic drug and immunosuppressant in the clinic, and the hypoandrogenism caused by CP is receiving more attention. Some studies found that ferroptosis is a new mechanism of cell death closely related to chemotherapeutic drugs and plays a key role in regulating reproductive injuries. The purpose of this study is to explore ferroptosis' role in testicular Leydig cell dysfunction and molecular mechanisms relating to it. In this study, the level of ferroptosis in the mouse model of testicular Leydig cell dysfunction induced by CP was significantly i

T2DM-elicited oxidative stress represses MTA3 expression in mouse Leydig cells.

Metastasis-associated protein 3 (MTA3) functions as a versatile coregulator in cancers and in physiological contexts. A predominant expression of MTA3 in interstitial Leydig cells (LCs) and its role as a local modulator of testicular steroidogenesis have recently emerged. Incubation with insulin decreased MTA3 expression in a concentration- and exposure time-dependent manner in LCs. This raises the possibility of additional endocrine actions of insulin in the direct control of MTA3 expression, which remains so far unexplored. Herein, we reported that type 2 diabetes mellitus (T2DM)-mediated in

Ochratoxin A suppresses proliferation of Sertoli and Leydig cells in mice.

Ochratoxin A (OTA) is a mycotoxin originating from Penicillium and Aspergillus. In addition to toxic effects in various tissues and cells, including neurons, immune cells, hepatocytes, and nephrons, it also causes carcinogenesis and teratogenesis. Although the negative effects of OTA with respect to the pathogenesis of diseases and the malfunction of various organs have been studied widely, the biological signaling mechanisms in testicular cells are less well known. Therefore, we determined the hazardous effect of OTA in two types of testicular cells: TM3 (mouse Leydig cells) and TM4 (mouse Se

Autophagy regulates testosterone synthesis by facilitating cholesterol uptake in Leydig cells.

Testosterone is indispensable for sexual development and maintaining male characteristics, and deficiency of this hormone results in primary or late-onset hypogonadism (LOH). Testosterone is primarily produced in Leydig cells, where autophagy has been reported to be extremely active. However, the functional role of autophagy in testosterone synthesis remains unknown. In this study, we show that steroidogenic cell-specific disruption of autophagy influenced the sexual behavior of aging male mice because of a reduction in serum testosterone, which is similar to the symptoms of LOH. The decline i

Effects of thyroid hormones on Leydig cells in the postnatal testis.

Thyroid hormones (TH) stimulate oxidative metabolism in many tissues in the body, but testis is not one of them. Therefore, in this sense, testis is not considered as a target organ for TH. However, recent findings clearly show that TH have significant functions on the testis in general, and Leydig cells in particular; this begins from the onset of their differentiation through aging. Some of these functions include triggering the Leydig stem cells to differentiate, producing increased numbers of Leydig cells during differentiation by causing proliferation of Leydig stem cells and progenitors,

Postpubertal untreated cryptorchidism. Anatomical relationship between Leydig cells and tubular sclerosis.

A histological study of the postpubertal untreated cryptorchid testis establishes a direct anatomical relationship between Leydig cells and tubular sclerosis, an age-related process ranging from mild fibrosis of the tubular tunica propria to complete tubular hyalinization. Two separate processes are involved. Mild or moderate tunical fibrosis is related to pretubular hyperplastic Leydig cell nodules, while severe tubular sclerosis and hyalinization depends on the development of intratunical Leydig cells, a finding hitherto undescribed. More probing studies are needed to define the mechanisms w

L-Cysteine Upregulates Testosterone Biosynthesis and Blood-Testis Barrier Genes in Cultured Human Leydig Cells and THP-1 Monocytes and Increases Testosterone Secretion in Human Leydig Cells.

Leydig cells are the primary source of testosterone or androgen production in male mammals. The blood-testis barrier (BTB) maintains structural integrity and safeguards germ cells from harmful substances by blocking their entry into the seminiferous tubules. L-cysteine is essential to the production of glutathione, a powerful antioxidant crucial to protecting against oxidative stress-induced damage. Animal studies have demonstrated the protective effect of L-cysteine in preventing testicular damage caused by chemicals or radiation. This study examines whether L-cysteine enhances the expression
