Additional evidence for the cyclic GMP signaling pathway resulting in the photophobic behavior of Stentor coeruleus.

We report that exo- and endogenous guanosine 3',5'-cyclic monophosphate (cGMP) specifically influenced the photophobic response. In behavioral experiments the slowly hydrolyzable and membrane-permeable analogs of cGMP (8-bromo-cGMP [Br-cGMP] and N6,2'-o-dibutyryl-cGMP) dramatically prolonged the time for ciliary stop response and decreased the duration of ciliary reversal in a dose-dependent manner. When analogs of adenosine 3',5'-cyclic monophosphate (cAMP) (8-bromo-cAMP or N6,2'-o-dibutyryl-cAMP) were used, no essential effects were detected on the kinetics of the photophobic response. Both

Retinal venous blood carbon monoxide response to bright light in male pigs: A preliminary study.

The physical mechanism by which light is absorbed in the eye and has antidepressant and energizing effects in Seasonal Affective Disorder and other forms of psychiatric major depression is of scientific interest. This study was designed to explore one specific aspect of a proposed humoral phototransduction mechanism, namely that carbon monoxide (CO) levels increase in retinal venous blood in response to bright light. Eleven mature male pigs approximately six months of age were kept for 7days in darkness and fasted for 12h prior to surgery. Following mild sedation, anesthesia was induced. Silas

TaUSP_6D-5 integrates light signalling and heat stress to regulate thermotolerance and developmental transitions in plants.

Universal Stress Proteins (USPs) constitute a conserved class of stress-responsive proteins that play important roles in helping plants adapt to adverse environmental conditions, including heat, oxidative stress, and endoplasmic reticulum (ER) stress. However, the molecular mechanisms through which USPs integrate environmental and developmental signalling pathways remain poorly understood. In this study, we characterized the wheat Universal Stress Protein TaUSP_6D-5 and identified it as a potential link between light signalling and thermotolerance. Our results demonstrated that TaUSP_6D-5 is l

Cancer chemopreventive effects of oral feeding alpha-tocopherol on ultraviolet light B induced photocarcinogenesis of hairless mouse.

Ultraviolet light is the most common cause of skin cancers in humans and several effects of ultraviolet light B (UVB: 290-320 nm) are thought to contribute to skin photocarcinogenesis. The generation of free radicals and related oxidants produced by UVB exposure, result in photocarcinogenesis by directly damaging DNA. On the other side, activating of transcription factor, activator protein 1 (AP-1) induced by UVB exposure causes tumor promotion. alpha-tocopherol has two principal physiological activities and one is an antioxidant activity through which alpha-tocopherol protects unsaturated fat

Analysis of combinatorial loss-of-function mutants in the Arabidopsis ethylene receptors reveals that the ers1 etr1 double mutant has severe developmental defects that are EIN2 dependent.

Ethylene responses in Arabidopsis are controlled by the ETR receptor family. The receptors function as negative regulators of downstream signal transduction components and fall into two distinct subfamilies based on sequence similarity. To clarify the levels of functional redundancy between receptor isoforms, combinatorial mutant lines were generated that included the newly isolated ers1-2 allele. Based on the etiolated seedling growth response, all mutant combinations tested exhibited some constitutive ethylene responsiveness but also remained responsive to exogenous ethylene, indicating that

Controllable gene delivery via masked adeno-associated viral vectors.

Controllable gene delivery to specific tissue by adeno-associated viruses (AAVs) is a challenge in gene therapy. Here we develop masked AAVs, whose transduction is blocked and restored only on pathological activation or by activation triggered by exogenous signals. The masking effect is accomplished by genetic encoding of non-canonical amino acid with tetrazine groups into AAV capsid proteins, allowing efficient reaction with trans-cyclooctene-modified truncated AAV receptor or polyethylene glycol to completely block viral infectivity in mice. With three different cleavable linkers, masking gr

Regulatory role of E3 ubiquitin ligases in multiple myeloma: from molecular mechanisms to therapeutic strategies.

Multiple myeloma (MM) is a hematological tumor characterized by the malignant proliferation of plasma cells in bone marrow (BM). Despite the prolonged survival of MM patients, a significant amount of patients relapse or become drug resistant. This underlines the importance of the development and investigation of novel targets to improve MM therapy. Increasing evidences have shed light on the emerging roles of E3 ubiquitin ligases in MM. E3 ubiquitin ligases play an essential role in protein ubiquitination, which is involved in the regulation of protein degradation, protein-protein interactions

The role of E3 ubiquitin ligase HECTD3 in cancer and beyond.

Ubiquitin modification plays significant roles in protein fate determination, signaling transduction, and cellular processes. Over the past 2 decades, the number of studies on ubiquitination has demonstrated explosive growth. E3 ubiquitin ligases are the key enzymes that determine the substrate specificity and are involved in cancer. Several recent studies shed light on the functions and mechanisms of HECTD3 E3 ubiquitin ligase. This review describes the progress in the recent studies of HECTD3 in cancer and other diseases. We propose that HECTD3 is a potential biomarker and a therapeutic targ

[Research status and prospect of orthodontically induced inflammatory root resorption].

Orthodontically induced inflammatory root resorption (OIIRR) is one of the most common and troublesome complications in orthodontic treatment, which not only affects the long-term stability of teeth, but also can cause irreversible damage to the patient's oral health. In light of the rising demand for orthodontic treatment, the prevention and treatment of OIIRR has become a primary focus for clinicians and researchers. Recent multidisciplinary studies have revealed the complex regulatory network of OIIRR, including mechanical signal transduction, immune microenvironment remodelling, cell diffe

Transcription Regulation and Membrane Stress Management in Enterobacterial Pathogens.

Transcription regulation in a temporal and conditional manner underpins the lifecycle of enterobacterial pathogens. Upon exposure to a wide array of environmental cues, these pathogens modulate their gene expression via the RNA polymerase and associated sigma factors. Different sigma factors, either involved in general 'house-keeping' or specific responses, guide the RNA polymerase to their cognate promoter DNAs. The major alternative sigma54 factor when activated helps pathogens manage stresses and proliferate in their ecological niches. In this chapter, we review the function and regulation

Negative selection at the pre-BCR checkpoint elicited by human mu heavy chains with unusual CDR3 regions.

Approximately 9% of in-frame mu heavy chain transcripts found in normal human pro-B cells encode proteins that can be expressed on the cell surface in the absence of surrogate or conventional light chains. These unusual mu heavy chains demonstrate preferential use of certain VH genes (VH3-23), frequent expression of DH regions in underrepresented reading frames, and an increased number of positively charged amino acids within the CDR3 region. Transcripts for these proteins are not found in pre-B cells or in mature B cells. When expressed in Jurkat T cells with the Ig(alpha)/Ig(beta) signal tra

Phototransduction in vertebrate rod and cone cells.

Vertebrate photoreceptor cells operate under very dim and bright illumination regimes. A protein machinery in rod and cone cells underlying the light response mediates photoexcitation, the return to the dark state, and adaptation processes. The machinery controls the homeostasis and mutual dependence of two cytoplasmic messengers, cGMP and Ca2+. The signaling pathway starts with light absorption by visual pigments (rhodopsin in rods or cone opsin in cones), which triggers an amplified signaling cascade, leading to the hydrolysis of cGMP and closure of cyclic nucleotide-gated channels in the ph

AOP2 (antioxidant protein 2): structure and function of a unique thiol-specific antioxidant.

The accumulation of reactive oxygen species (ROS) in response to extracellular signals or intracellular biochemical processes can be regulated by the coordinate action of many antioxidant proteins. Because moderate levels of ROS can act as intracellular messengers in many of these processes, this modulation is critical for the transduction of specific signals. The thiol-specific antioxidant (TSA) family is a highly conserved group of enzymes that can reduce hydroperoxides in the presence of a thiol-containing electron donor. AOP2 (antioxidant protein 2) is a newly described member that shows s

Circadian-independent light regulation of mammalian metabolism.

The daily light-dark cycle is a key zeitgeber (time cue) for entraining an organism's biological clock, whereby light sensing by retinal photoreceptors, particularly intrinsically photosensitive retinal ganglion cells, stimulates the suprachiasmatic nucleus of the hypothalamus, a central pacemaker that in turn orchestrates the rhythm of peripheral metabolic activities. Non-rhythmic effects of light on metabolism have also been long known, and their transduction mechanisms are only beginning to unfold. Here, we summarize emerging evidence that, in mammals, light exposure or deprivation profound

The small helix-loop-helix protein NsrC is a regulator of phycobilisome biosynthesis.

Nitrogen is a key nutrient that influences primary productivity in both aquatic and terrestrial ecosystems. In cyanobacteria, its deficiency affects growth and phycobiliprotein biosynthesis and leads to degradation of phycobilisomes (PBSs). While the biosynthesis genes of phycobiliprotein are regulated by strong promoters, the molecular mechanisms underlying their regulation in response to nitrogen deficiency remain largely unknown. Using nondiazotrophic cyanobacterium Synechococcus elongatus PCC 7942 as a model system, we identified a novel PBS biosynthesis regulator, a small helix-loop-helix

Ciliopathy is differentially distributed in the brain of a Bardet-Biedl syndrome mouse model.

Bardet-Biedl syndrome (BBS) is a genetically heterogeneous inherited human disorder displaying a pleotropic phenotype. Many of the symptoms characterized in the human disease have been reproduced in animal models carrying deletions or knock-in mutations of genes causal for the disorder. Thinning of the cerebral cortex, enlargement of the lateral and third ventricles, and structural changes in cilia are among the pathologies documented in these animal models. Ciliopathy is of particular interest in light of recent studies that have implicated primary neuronal cilia (PNC) in neuronal signal tran

Motility activation and second messenger signalling in spermatozoa from rat cauda epididymidis.

This study examined molecular mechanisms involved in the activation of motility in spermatozoa from the cauda epididymidis of rats. A 1.05-fold dilution of semen from the cauda epididymidis with 300 mmol sucrose l(-1) did not activate motility in spermatozoa. Addition of dibutyryl cAMP, pentoxifylline or Ca(2+) to the sucrose activated motility in the short term (<30-60 min). A fivefold dilution of semen from the cauda epididymidis with a modified Tyrode's medium (BWW) activated and sustained vigorous motility that could not be attenuated with kinase inhibitors. Molecular interactions and mutational impact upon rhodopsin (G90→D90) for hindering dark adaptation of eye: A comparative structural level outlook for signaling mechanism in night blindness.

For night blindness, a detailed structural exploration of the interactions among G-protein receptor rhodopsin, transducin and arrestin was performed. Rhodopsin is responsible for dim light vision while a point mutation (G90→D90) results in an adverse change in its photo-transduction. The validated 3D models of the three proteins were utilized, and upon mutation and interactions, rhodopsin attained higher stability (evaluated through thermodynamic energy calculations, electrostatic surface potential and solvent accessible area), thereby participating strongly with transducin.
