Transcriptomic analysis reveals mechanism of light-sensitive albinism in tea plant Camellia sinensis 'Huangjinju'.

BACKGROUND: Camellia sinensis 'Huangjinju' is an albino tea variety developed recently in China. Young leaves of 'Huangjinju' demonstrate bright yellow when cultivated under natural sunlight, but regreens under reduced light intensity. To elucidate the physiological and molecular mechanisms of this light-sensitive albinism, we compared leaf pigmentation, metabolites, cellular ultrastructure and transcriptome between plants cultured under natural sunlight and shade. RESULTS: Shading treatment doubled the chlorophyll concentration and regreened albino leaves; carotenoid also increased by 30%. E

Proteome Analysis of the ROF-FKBP Mutants Reveals Functional Relations among Heat Stress Responses, Plant Development, and Protein Quality Control during Heat Acclimation in Arabidopsis thaliana.

In the present study, a differential screening following heat stress acclimation was performed in Arabidopsis thaliana WT and ROF-FKBP mutated plants using mass spectrometry, and the results were used to understand and analyze the effect of the ROF PPIases during thermotolerance acquisition in plants. Our data highlight the central role of these two PPIases in heat stress and point to their direct or indirect effect on other proteins participating in cellular functions such as protein folding and quality control, cell division, photosynthesis, and other metabolic and signaling processes. A quantum chemical study on the effects of varying the central metal in extended photosynthetic pigments.

In a certain period of Earth's history, chlorophylls with Mg as their central metal would have been selected as the major photosynthetic pigments, reflecting the radiation in habitats. Assuming evolution in different light and material environments, different photosynthetic pigments would occur. This study is the first attempt to evaluate the physical and chemical properties of model photosynthetic pigments and their potential to function in a variety of light environments using quantum chemistry calculations. Specifically, bacteriochlorophyll b (Bchl b), phthalocyanine (Pht) and meso-dibenzop

Identification of chloroplast transit peptides inDunaliella salina and their application for heterologous astaxanthin production.

Dunaliella salinais a promising green cell factory, but efficient chloroplast targeting of exogenous proteins requires suitable chloroplast transit peptides (cTPs). In this study, six endogenous cTPs fromD. salinawere shown to direct mCherry import into the chloroplast, and downstream extensions beyond the cleavage site enhanced import efficiency and accuracy. To validate their utility, DAT-2 and DAT-3 were engineered to expressHaematococcus lacustrisβ-carotene hydroxylase and ketolase fused to the cTPs derived from light-harvesting complex II chlorophyll a/b binding protein 1 and tocopherol c

Non-photochemical quenching in cryptophyte alga Rhodomonas salina is located in chlorophyll a/c antennae.

Photosynthesis uses light as a source of energy but its excess can result in production of harmful oxygen radicals. To avoid any resulting damage, phototrophic organisms can employ a process known as non-photochemical quenching (NPQ), where excess light energy is safely dissipated as heat. The mechanism(s) of NPQ vary among different phototrophs. Here, we describe a new type of NPQ in the organism Rhodomonas salina, an alga belonging to the cryptophytes, part of the chromalveolate supergroup. Cryptophytes are exceptional among photosynthetic chromalveolates as they use both chlorophyll a/c pro

Planar arrangement of bacteriochlorophyll c aggregates in chlorosomes of Chloroflexus aurantiacus.

Chlorosomes are main light-harvesting complexes of green photosynthetic bacteria. Pigments are organized in self-assembling aggregates within chlorosomes rather than being coordinated by proteins as seen for other photosynthetic complexes. Chlorosomes isolated from Chloroflexus aurantiacus were subjected to cryoelectron tomography, which revealed the long-range organization of bacteriochlorophyll c within the chlorosome interior. The resulting electrostatic scattering potential map is well described by a planar lamellar model. Molecular dynamics simulations substantiated an important role of v

Cholera toxin elevates pathogen resistance and induces pathogenesis-related gene expression in tobacco.

In animals, plants and fungi, cholera toxin (CTX) can activate signalling pathways dependent on heterotrimeric GTP binding proteins (G-proteins). We transformed tobacco plants with a chimeric gene encoding the A1 subunit of CTX regulated by a light-inducible wheat Cab-1 promoter. Tissues of transgenic plants expressing CTX showed greatly reduced susceptibility to the bacterial pathogen Pseudomonas tabaci, accumulated high levels of salicylic acid (SA) and constitutively expressed pathogenesis-related (PR) protein genes encoding PR-1 and the class II isoforms of PR-2 and PR-3. In contrast, the

Barley Cultivar Sarab 1 Has a Characteristic Region on the Thylakoid Membrane That Protects Photosystem I under Iron-Deficient Conditions.

The barley cultivar Sarab 1 (SRB1) can continue photosynthesis despite its low Fe acquisition potential via roots and dramatically reduced amounts of photosystem I (PSI) reaction-center proteins under Fe-deficient conditions. We compared the characteristics of photosynthetic electron transfer (ET), thylakoid ultrastructure, and Fe and protein distribution on thylakoid membranes among barley cultivars. The Fe-deficient SRB1 had a large proportion of functional PSI proteins by avoiding P700 over-reduction. An analysis of the thylakoid ultrastructure clarified that SRB1 had a larger proportion of

Evidence for Protein Dynamics-Induced Energy Transfer Pathway Changes in Photosynthetic Complexes.

Photosystem I (PSI) is a chlorophyll-binding protein complex that plays a central role in photosynthesis. PSI isolated from cyanobacteria, Thermosynechococcus vestitus, contains a light-harvesting system that consists of approximately 100 chlorophyll a molecules. The effect of the protein fluctuation dynamics on the excitation energy migration within the pigment array is fundamentally important. In 2009, Brecht and co-workers proposed that protein dynamics can induce variations in the excitation energy transfer (EET) pathways within PSI ( Brecht et al. Proc. Natl. Acad. Sci. U. S. A.2009, 106,

Mechanistic Insights into Dinitrogen Reduction to Ammonia in Light-Controlled Nanocrystal:Nitrogenase Complexes.

Developing systems that can efficiently capture photon energy and convert this energy into fuels and chemicals requires understanding how to assemble molecular components with diverse functions into complete systems possessing selectivity and efficiency in directing charge carriers to catalytic reactions. There are many challenges to achieving this goal. One promising approach is the development of hybrid systems that combine semiconductor nanocrystals (NCs) for light capture and enzymes as efficient catalysts. Such biohybrid systems capitalize on the tunable electronic and optical properties

Carbonic anhydrase activity under low zinc in Phaeodactylum tricornutum largely depends on metal-free iota-carbonic anhydrase.

Carbonic anhydrases (CAs) typically possess a Zn2+ cofactor and are involved in a wide range of adaptive metabolisms involving pH and dissolved inorganic carbon homeostasis. In photosynthetic microorganisms, they are involved in CO2 concentrating mechanisms (CCM) that supply CO2 to ribulose-1,5-bisphosphate carboxylase-oxygenase. Here we show that the model diatom Phaeodactylum tricornutum possesses a CA from the newly identified iota class, P. tricornutum ι-carbonic anhydrase (Pt-ιCA), which exhibited catalytic activity without a metal cofactor, in contrast to most CAs. Metal-free Pt-ιCA acti

Quantum dots and fluorescent protein FRET-based biosensors.

There has been considerable recent interest in the creation of nanoparticle-biomolecule hybrid materials for uses such as in vitro and in vivo biosensing, biological imaging, and drug -delivery. Nanoparticles have a high surface to volume ratio, making them capable of being decorated with -various biomolecules on their surface which retain their biological activity. Techniques to bind these biomolecules to nanoparticle surfaces are also advancing rapidly. Here we demonstrate hybrid materials assembled around CdSe/ZnS core/shell semiconductor quantum dots (QDs). These intrinsically fluorescent

Non-Markovianity between Site Pairs in FMO Complex Using Discrete-Time Quantum Jump Model.

The Fenna-Mathews-Olson (FMO) complex present in green sulfur bacteria is known to mediate the transfer of excitation energy between light-harvesting chlorosomes and membrane-embedded bacterial reaction centers. Due to the high efficiency of this transport process, it is an extensively studied pigment-protein complex system with the eventual aim of modeling and engineering similar dynamics in other systems and using it for real-time application. Some studies have attributed the enhancement of transport efficiency to wavelike behavior and non-Markovian quantum jumps resulting in long-lived and

A calcium-binding protein in bile and gallstones.

Calcium salts are often present in the center of all types of gallstones. Matrix proteins are known to be essential for biomineralization and may therefore also be important in the formation and growth of gallstones. Other researchers have described an anionic peptide fraction of a biliary lipoprotein complex in bile and a low-molecular weight acidic glycoprotein present in gallstones. Our goal was to determine whether such a protein was present in bile and whether this protein has any calcium-binding properties. We identified a pigment-associated, highly acidic protein that precipitates from

Transcript profiling of salinity stress responses by large-scale expressed sequence tag analysis in Mesembryanthemum crystallinum.

The common ice plant, Mesembryanthemum crystallinum, is a halophytic (salt-loving) member of the Aizoaceae, which switches from C3 photosynthesis to Crassulacean acid metabolism (CAM) when exposed to salinity or water-deficit stress. CAM is a metabolic adaptation of photosynthetic carbon fixation that improves water use efficiency by shifting net CO2 uptake to the night, thereby reducing transpirational water loss. To improve our understanding of the molecular genetic underpinnings and control mechanisms for Crassulacean acid metabolism (CAM) and other salinity stress response adaptations, a t

Cyclic lipopeptide antibiotics bind to the N-terminal domain of the prokaryotic Hsp90 to inhibit the chaperone activity.

Chemical arrays were employed to screen ligands for HtpG, the prokaryotic homologue of Hsp (heat-shock protein) 90. We found that colistins and the closely related polymyxin B interact physically with HtpG. They bind to the N-terminal domain of HtpG specifically without affecting its ATPase activity. The interaction caused inhibition of chaperone function of HtpG that suppresses thermal aggregation of substrate proteins. Further studies were performed with one of these cyclic lipopeptide antibiotics, colistin sulfate salt. It inhibited the chaperone function of the N-terminal domain of HtpG. H

De novo sequencing and analysis of the Ulva linza transcriptome to discover putative mechanisms associated with its successful colonization of coastal ecosystems.

BACKGROUND: The green algal genus Ulva Linnaeus (Ulvaceae, Ulvales, Chlorophyta) is well known for its wide distribution in marine, freshwater, and brackish environments throughout the world. The Ulva species are also highly tolerant of variations in salinity, temperature, and irradiance and are the main cause of green tides, which can have deleterious ecological effects. However, limited genomic information is currently available in this non-model and ecologically important species. Ulva linza is a species that inhabits bedrock in the mid to low intertidal zone, and it is a major contributor

A Natural qE- and qZ-Deficient Alga Retains Functional Plant-Like and Chlorophycean Violaxanthin De-Epoxidases.

Photosynthetic organisms protect themselves from excess light through non-photochemical quenching (NPQ), a process that safely dissipates excess light energy as heat. Current understanding of NPQ is largely drawn from a few model systems, leaving it unclear how universal NPQ mechanisms are across the green lineage. We characterize the slow, sustained NPQ of the green alga, Auxenochlorella protothecoides x symbiontica (UTEX 250-A). The Auxenochlorella genome lacks Light-Harvesting Complex Stress Related (LHCSR), one of the two proteins required for the fast energy-dependent quenching (qE) compo

The rapid yellowing of spruce at a mountain site in the Central Black Forest (Germany). Combined effects of Mg deficiency and ozone on biochemical, physiological and structural properties of the chloroplasts.

Biochemical, physiological and ultrastructural changes of the chloroplasts were examined in the course of the rapid yellowing process of spruce (Picea abies (L.) Karst.) at a Mg-deficient and ozone polluted mountain site (Schöllkopf mountain, Central Black Forest, Germany, 840 m a.s.l.). While at an early stage of yellowing the chlorophyll (Chl) content of the needles decreased slowly, significant changes occurred in the chloroplasts: The lability of the light-harvesting Chl a/b protein complex LHC II increased; the thylakoid cross-sectional area of chloroplasts in the outer mesophyll of the n
