The mechanism of hyperlipidaemia in the nephrotic syndrome.

The mechanism of hyperlipidaemia in the nephrotic syndrome has not been fully established. We propose that it results from hypoalbuminaemia due to inhibition of the reaction catalysed by lecithin cholesterol acyltransferase converting cholesterol of high density lipoproteins to cholesterol esters and to an inhibition of high density lipoprotein particle formation from very low density lipoproteins due to reduced activity of lipoprotein lipase.

[Acid lipase deficiency: Wolman disease and cholesteryl ester storage disease].

Wolman disease and cholesteryl ester storage disease (CESD) are caused by a deficiency of lysosomal acid lipase activity, resulting in massive accumulation of cholesteryl ester and triglycerides. Wolman disease occurs in infancy, with hepatosplenomegaly, steatorrhea and adrenal calcification. It is fatal before the age of 1 year. In CESD, hepatomegaly may be the only clinical abnormality, although lipid deposition is widespread. Lysosomal acid lipase hydrolyzes both triaclyglycerols and cholesteryl esters, and the enzyme plays an important role in the cellular processing of plasma lipoproteins

Review on anti-lipoprotein lipase antibodies.

Lipoprotein lipase (LPL) is a member of the lipase family, and LPL is known to hydrolyze triglyceride molecules found in lipoprotein particles. Understanding the pathogenesis of atherosclerosis in autoimmune diseases, particularly systemic lupus erythematosus (SLE), is vital to reduce morbidity and mortality of this disease. It is now recognized that SLE has a particular pattern of dyslipoproteinemia characterized by low HDL levels and increased triglycerides, which is aggravated by flare. Antilipoprotein lipase (anti-LPL) antibodies have been recently described in rheumatic diseases, mainly i

Post-heparin plasma lipase activity in hypopituitary adults.

Twenty-six hypopituitary patients on conventional replacement therapy were compared with 31 matched normal controls. Plasma lipoprotein lipase (LPL) and hepatic lipase (HL) activities were measured 15 min after a bolus of intravenous heparin (100 IU/kg body weight). Fasting plasma lipids, lipoproteins, glucose and insulin and triceps to subscapular skin fold ratio and waist to hip circumference ratio were also measured. Total and low density lipoprotein cholesterol were significantly higher in patients than in controls. Plasma activity of both LPL and HL was significantly higher in hypopituita

Lipolysis of host triacylglyceride-rich lipoproteins creates a toxic microenvironment for Staphylococcus aureus.

Staphylococcus aureus secretes lipases that hydrolyze glycerol esters to mitigate host detection and scavenge fatty acids for membrane synthesis, but exposure to host-derived polyunsaturated fatty acids (PUFA) can be toxic. Here, we show that host hyperlipidemia protects against S. aureus osteomyelitis by engaging this vulnerability. Using a mouse model of severe hyperlipidemia, we show that PUFA-enriched plasma triacylglycerides are associated with reduced bacterial burden and bone destruction from injury-associated osteomyelitis. In vitro , human triacylglyceride-rich lipoproteins (TRL) exhi

Dual roles for lipolysis and oxidation in peroxisome proliferation-activator receptor responses to electronegative low density lipoprotein.

Low density lipoprotein (LDL) exists in various forms that possess unique characteristics, including particle content and metabolism. One circulating subfraction, electronegative LDL (LDL(-)), which is increased in familial hypercholesterolemia and diabetes, is implicated in accelerated atherosclerosis. Cellular responses to LDL(-) remain poorly described. Here we demonstrate that LDL(-) increases tumor necrosis factor alpha (TNFalpha)-induced inflammatory responses through NF kappa B and AP-1 activation with corresponding increases in vascular cell adhesion molecule-1 (VCAM1) expression. LDL

Chylomicron and very-low-density lipoprotein levels in type I hyperlipoproteinaemia. The role of the liver in determining biochemical phenotype.

Patients with type 1 hyperlipoproteinaemia present with relatively normal plasma levels of very-low-density lipoprotein (VLDL) together with marked hyperchylomicronaemia. Since the aetiological basis for the type 1 phenotype is a severe reduction in peripheral lipoprotein lipase (LPL) activity, the discrepancy between the two triglyceride-rich lipoprotein fractions is paradoxical. In order to account for these observations we have previously proposed (G.M.B) that the hepatic secretion of lipoproteins in these patients is characterized by the production of chylomicron-like particles rather than

Letting lipids go: hormone-sensitive lipase.

PURPOSE OF REVIEW: Despite their pathophysiological importance, the molecular mechanisms and enzymatic components of lipid mobilization from intracellular storage compartments are insufficiently understood. The aim of this review is to evaluate the role of hormone-sensitive lipase in this process. RECENT FINDINGS: Hormone-sensitive lipase exhibits a broad specificity for lipid substrates such as triglycerides, diglycerides, cholesteryl esters, and retinyl esters and the enzyme is in a wide variety of tissues. The high enzyme activity in adipose tissue was considered rate-limiting in the degra

Secondary type V hyperlipoproteinemia in an acromegalic patient without overt diabetes.

A rare case of acromegaly developing gross hyperlipidemia not associated with overt diabetes was presented. Gross hyperlipidemia revealed type V hyperlipoproteininemia, according to agarose gel electrophoretic pattern of serum lipoprotein. Before pituitary surgery, the serum levels of cholesterol and triglyceride were 320 mg/dl and 1830 mg/dl respectively, and the basal level of growth hormone was markedly elevated, ranging from 129 to 231 ng/ml with the mean +/- SD of 168 +/- 39 ng/ml. The enzymatic activities of hepatic triglyceride lipase (H-TGL) and lipoprotein lipase (LPL) were 14.2 and 1

Carnitine palmitoyltransferase II deficiency with normal carnitine palmitoyltransferase I in skeletal muscle and leucocytes.

Deficiency of carnitine palmitoyltransferase II (CPT II), was found to be the cause of the syndrome of muscle pain and myoglobinuria following strenuous exercise in an otherwise healthy young man. During fasting, serum creatine kinase remained low and ketogenesis was normal. The clearance of a fat emulsion and the activity of extrahepatic lipoprotein lipase was lowered, while the hepatic lipoprotein lipase was normal. A skeletal muscle biopsy did not show abnormal lipid storage. CPT II was deficient in skeletal muscle and leucocytes, while CPT I activity was normal and exhibited normal kinetic

Circulating lipids and lipoproteins in glycogen storage disease type I with nocturnal intragastric feeding.

With the advent of nocturnal intragastric feeding which protects against acute metabolic complications and promotes growth, patients with glycogen storage disease type I are attracting less attention. However, several biochemical alterations persist and suggest that the long-term risk of atherosclerotic heart disease remains high. Persisting hypertriglyceridemia and hypercholesterolemia were found in seven glycogen storage disease type I subjects, six of them following 5-6 yr of nocturnal intragastric feeding. When compared to ten age-matched controls, the patients showed significantly (P less

Androgens and women's health: genetic and epidemiologic aspects of lipid metabolism.

Lipoprotein metabolism may be viewed as a process whereby large, triglyceride-carrying particles from the intestine and liver are broken down into smaller cholesterol-enriched lipoprotein particles. In the process, triglyceride is transported from the intestine and liver to adipose and other storage tissues. Androgen appears to affect lipoprotein metabolism in a number of ways. These include: increasing the activity of lipoprotein lipase and hepatic triglyceride lipase, resulting in higher levels of triglyceride in adipose tissue and a drop in total circulating high-density lipoprotein levels,

Relationship between insulin-like growth factor-I and low-density lipoprotein cholesterol levels in primary hypothyroidism in women.

The effect of insulin-like growth factor-I (IGF-I) on the disturbance of lipid metabolism during primary hypothyroidism was studied in 12 women with primary hypothyroidism. Significant increases in both low-density lipoprotein (LDL) cholesterol and intermediate-density lipoprotein cholesterol were seen. Lipoprotein concentrations reverted to normal after substitution with thyroxine (T4) until the euthyroid state was reached. A decrease in IGF-I of 65% (P less than 0.005) was seen in hypothyroid patients and this was inversely correlated (r = -0.75; P less than 0.01) with the concentration of L

Lipid nephrotoxicity: new concept for an old disease.

The prevalence of obesity in the United States remains high, exceeding 30% in most states. As this trend continues unhindered, we will continue see a persistent rise in obesity-related metabolic effects—hypertension, dyslipidemia, diabetes mellitus, and atherosclerosis. These diseases are also the leading causes of chronic kidney diseases and end-stage renal disease. The lipid nephrotoxicity hypothesis, proposed over three decades ago, suggested that proteinuria, decreased albumin levels, and the resultant hyperlipidemia may cause a glomerulosclerosis similar to atherosclerosis. More recent st

Effect of hepatoprotective ayurvedic drugs on lipolytic activities during CCl4 induced acute hepatic injury in albino rats.

Daily treatment of CCl4(3 ml/kg body wt) for 7 days induced acute hepatic necrosis in albino rats. Treatment of CCl4 caused significant alterations in the activities of acid lipase, alkaline lipase, lipoprotein lipase of liver, kidney and adipose tissue and hormone sensitive lipase of adipose tissue of albino rat. Administration of hepatoprotective ayurvedic drugs (kumari asav, kumari kalp, arogyavardhini and tamra bhasma) concomitant with CCl4 counteracted the action of CCl4 on lipolytic enzymes exhibiting hepatoprotection. The possible physiological significance of alterations in lipolytic e

Atorvastatin Improves Hepatic Lipid Metabolism and Protects Renal Damage in Adenine-Induced Chronic Kidney Disease in Sprague-Dawley Rats.

OBJECTIVE: Chronic kidney disease (CKD), including nephrotic syndrome, is a major cause of cardiovascular morbidity and mortality. The literature indicates that CKD is associated with profound lipid disorders largely due to the dysregulation of lipoprotein metabolism which further aggravates the progression of kidney disease. The present study sought to determine the efficacy of atorvastatin treatment on hepatic lipid metabolism and renal tissue damage in CKD rats. METHODS: Serum, hepatic and faecal lipid contents and the expression and enzyme activity of molecules involved in cholesterol and

Plasma lipoproteins, apolipoproteins, and triglyceride metabolism in familial hypertriglyceridemia.

Several parameters of lipoprotein metabolism were examined in 38 men with primary hypertriglyceridemia (phenotype IV). Family investigation showed that 17 men had familial combined hyperlipidemia (FCH), seven had familial hypertriglyceridemia (FHT), and 14 had unclassified hypertriglyceridemia (UNC). In all three groups, plasma high density lipoprotein (HDL) cholesterol and the concentrations of apolipoprotein A-I and A-II were decreased, and apolipoprotein B was increased, each to the same extent. These results are compatible with an increased risk of cardiovascular disease in both FCH and FH

Disorders of lipid metabolism in nephrotic syndrome: mechanisms and consequences.

Nephrotic syndrome results in hyperlipidemia and profound alterations in lipid and lipoprotein metabolism. Serum cholesterol, triglycerides, apolipoprotein B (apoB)-containing lipoproteins (very low-density lipoprotein [VLDL], immediate-density lipoprotein [IDL], and low-density lipoprotein [LDL]), lipoprotein(a) (Lp[a]), and the total cholesterol/high-density lipoprotein (HDL) cholesterol ratio are increased in nephrotic syndrome. This is accompanied by significant changes in the composition of various lipoproteins including their cholesterol-to-triglyceride, free cholesterol-to-cholesterol e
