Copper, an essential trace element for the human body, plays a key role in energy metabolism, mitochondrial respiration, redox reactions, and neural signal transmission. The recently proposed concept of "cuproptosis" has further revealed the unique status of copper in cellular regulation: when copper abnormally accumulates within cells, it can directly bind to the lipoylated proteins of the mitochondrial TCA cycle, triggering protein aggregation and metabolic disorders, ultimately leading to cell death. This form of cell death plays an important role in various neurodegenerative diseases of th

Copper is an essential trace element, and plays a vital role in numerous physiological processes within the human body. During normal metabolism, the human body maintains copper homeostasis. Copper deficiency or excess can adversely affect cellular function. Therefore, copper homeostasis is stringently regulated. Recent studies suggest that copper can trigger a specific form of cell death, namely, cuproptosis, which is triggered by excessive levels of intracellular copper. Cuproptosis induces the aggregation of mitochondrial lipoylated proteins, and the loss of iron-sulfur cluster proteins. In

Obsessive-compulsive disorder (OCD) affects 1-3% of the global population and ranks among the top ten most disabling medical conditions. While abnormalities in cortico-striato-thalamo-cortical circuits have been implicated in OCD pathophysiology, the molecular mechanisms underlying these neural aberrations remain incompletely understood. Protein palmitoylation, a reversible post-translational modification, is essential for neuronal development and synaptic function, potentially affecting neurotransmitter systems linked to OCD. We employed Mendelian randomization (MR) to explore causal relation

Despite its prevalence and the severity of symptoms, little is known about the pathogenesis and etiology of adenomyosis. In our previous study, Scribble localization has been found to be partially translocated to cytoplasm; however, its regulatory mechanism is known. In consideration of the important role of supraphysiologic estrogen production in the endometrium in the development of adenomyosis, we analyzed the effect and mechanism of estrogen on Scribble localization in vivo and in vitro. Firstly, we found Scribble translocation from the basolateral membrane to the cytoplasm was easily to b

The spike proteins of enveloped viruses are transmembrane glycoproteins that typically undergo post-translational attachment of palmitate on cysteine residues on the cytoplasmic facing tail of the protein. The role of spike protein palmitoylation in virus biogenesis and infectivity is being actively studied as a potential target of novel antivirals. Here, we report that palmitoylation of the first five cysteine residues of the C-terminal cysteine-rich domain of the SARS-CoV-2 S protein are indispensable for infection, and palmitoylation-deficient spike mutants are defective in membrane fusion.

Post-translational modification of proteins by the lipid palmitate is critical for protein localization and function. Palmitoylation is regulated by the opposing enzymes palmitoyl acyltransferases (PATs) and acyl protein thioesterases, which add and remove palmitate from proteins, respectively. Palmitoylation is particularly important for a number of processes including neuronal development and synaptic activity in the central nervous system. Dysregulated palmitoylation contributes to neuropsychiatric disease. In total six PATs (HIP14, HIP14L, ZDHHC8, ZDHHC9, ZDHHC12, and ZDHHC15) and one thio

OBJECTIVE: The objective of this study was to comprehensively characterize the expression profiles and prognostic significance of palmitoylation-related genes (PRGs) in bladder cancer (BLCA). METHODS: Transcriptomic, clinical, and somatic mutation data of BLCA patients were obtained from The Cancer Genome Atlas (TCGA) and the GEO dataset (GSE13507), and batch effects were corrected using the ComBat method. Consensus clustering was performed to identify PRG-related molecular subtypes. Prognostic differentially expressed genes (DEGs) were further used for gene subtyping, and a PRG-based prognost

The tegument protein UL51 is conserved among herpesviruses, but the mechanisms underlying its role in viral pathogenesis remain unclear. In this study, using Anatid herpesvirus 1 (AnHV-1) as a model, we show that deletion of UL51 markedly attenuates viral replication and virulence in ducks. Immunoprecipitation coupled with LC-MS/MS identified interactions between pUL51 and multiple viral proteins, among which the association with pUL10 depends on pUL51 palmitoylation. This modification promotes pUL10 stability and its localization to the Golgi apparatus. Further analysis revealed that the palm

The palmitoylation system is intricate, multidimensional, and plays a crucial role in various inflammatory and immune-related disorders. Palmitoylation controls protein stability, cargo sorting, signal transmission, as well as cell differentiation and death. Notably, a growing body of studies has highlighted its participation in inflammatory processes, either directly or indirectly, indicating its broad and complex involvement in the development of sepsis. Understanding the mechanisms underlying palmitoylation is essential for advancing research on sepsis. We began this review with a brief sum

Loss-of-function mutations in the depalmitoylating enzyme palmitoyl protein thioesterase 1 (PPT1) cause neuronal ceroid lipofuscinosis (NCL), a devastating neurodegenerative disease. The substrates of PPT1 are largely undescribed, posing a limitation on molecular dissection of disease mechanisms and therapeutic development. Here, we provide a resource identifying >100 novel PPT1 substrates. We utilized Acyl Resin-Assisted Capture (Acyl RAC) and mass spectrometry to identify proteins with increased in vivo palmitoylation in PPT1 knockout (KO) mouse brains. We then validated putative substrates

S-acylation (or palmitoylation) is a reversible post-translational modification (PTM) that modulates protein activity, signalization and trafficking. Palmitoylation was found to significantly impact the activity of various membrane receptors involved in either pathogen entry, such as CCR5 (for HIV) and anthrax toxin receptors, cell proliferation (epidermal growth factor receptor), cardiac function (β-Adrenergic receptor), or synaptic function (AMPA receptor). Palmitoylation of these membrane receptors indeed affects not only their internalization, localization, and activation, but also other P

A family of zDHHC protein acyltransferase (PAT) enzymes catalyze the S-palmitoylation of target proteins via a two-step mechanism. The first step involves transfer of palmitate from the palmitoyl-CoA donor to the active site cysteine of the zDHHC PAT enzyme, releasing reduced CoA (CoASH). In the second step, the palmitoyl-PAT intermediate thioester reacts with a cysteine side chain within the target substrate to produce the palmitoylated substrate product or, in the absence of a protein substrate, the palmitoyl-PAT intermediate thioester is hydrolyzed and releases palmitate. INTRODUCTION: Cuproptosis, a novel form of cell death tied to copper homeostasis and protein lipoylation, holds significant promise for breast cancer treatment. However, its efficacy is severely hindered by the tumor microenvironment (TME) heterogeneity, such as hypoxia and elevated glutathione (GSH) levels.
METHODS: Herein, we synthesized CuNI nanoparticles via a facile hydrothermal method, which could serve as both a copper carrier and a photothermal agent, to enhance the accumulation of copper in tumor site. Following intravenous injection, CuNI accumulated and persisted in tumors via the

Renal ischemia-reperfusion injury (RIRI), a major contributor to acute kidney injury (AKI) and delayed graft function (DGF), is closely associated with dysregulation of metal ion homeostasis. Although copper and iron metabolism exhibit interconnected regulatory pathways, the temporal dynamics and functional interplay of these metal ions in RIRI pathogenesis remain poorly understood. Our study demonstrates that cuproptosis and ferroptosis, two distinct forms of cell death induced by metal ion overload, occur simultaneously within 6 h after reperfusion. Notably, ischemia-reperfusion injury induc

C opper is an essential trace element involved in mitochondrial metabolism and redox regulation, and its dysregulation has been increasingly linked to metabolic disorders. However, its specific role in diabetic kidney disease (DKD) remains unclear. In this study, we conducted an integrated analysis combining bulk RNA-seq, single-cell RNA sequencing, and spatial transcriptomics to investigate the involvement of cuproptosis-related genes in DKD. We identified consistent upregulation of FDX1 and LIAS, two key regulators of copper-induced cell death, in the kidneys of diabetic mice. These genes we

Cuproptosis, a newly identified form of cell death, is closely linked to copper homeostasis and protein lipoylation. Using a multi-omics approach, we firstly reveal that SERPINB3 confers cuproptosis resistance in pancreatic cancer and functions as a theranostic biomarker. Mechanistically, SERPINB3 inhibits FDX1 transcription by activating the MAPK signaling pathway, thereby conferring cuproptosis resistance. We further demonstrated that SERPINB3 directly interacts with MEK1, impeding its chaperone-mediated autophagic degradation, which ultimately leads to sustained activation of the MAPK signa

Acsf3, a mitochondrial malonyl-CoA synthetase, is mutated in the human inborn error Combined Malonic and Methylmalonic Aciduria. To clarify its role in mammalian metabolism, we generated Acsf3 knockout mice. We show that the loss of Acsf3 results in high concentrations of malonate and methylmalonate and exhibit expected defects in mitochondrial metabolism, but the KO mice are largely healthy similar to the human inborn error. Acsf3 KO mice exhibit redundancy in malonyl-CoA synthetase activity suggesting a genetic modifier of disease severity. Malonate and methylmalonate are metabolic poisons g

Cuproptosis is a newly identified form of cell death that relies on copper (Cu) ionophores to transport Cu into cancer cells. As a perennial herb, Patrinia scabiosaefolia Fisch (PS) has garnered significant attention owing to its analgesic, anti-inflammatory, antibacterial, and antitumor properties. Previous research has shown that the extract from PS (DEPS) can inhibit the growth of leukemia cell lines. However, the specific mechanism of its anti-leukemic effect has not been fully clarified. Therefore, this study was conducted to investigate the molecular mechanism of cuproptosis in the treat

In the last decade, the cloning and biochemical identification of mitochondrial autoantigens in primary biliary cirrhosis (PBC) as members of the 2-oxoacid dehydrogenase complex has greatly advanced the detection of antimitochondrial antibodies (AMA) and the understanding of the immunobiology of the disease. Here, we discuss the methods of detecting AMA and its isotypes, methods of epitope mapping, and using these methods in PBC liver immunohistochemistry and Ig gene usage. Increasing evidence, including the specific association of AMA with PBC, the unique similar but noncross-reactive conform

Cuproptosis is a mitochondria- and copper-dependent regulated form of cell death that has attracted growing interest as a therapeutic strategy in oncology. Its core mechanism involves the aggregation of lipoylated proteins in the tricarboxylic acid cycle to trigger proteotoxic stress and the destabilization of iron-sulfur cluster proteins, leading to mitochondrial dysfunction. These two effects synergize to initiate this regulated form of cell death. Recent studies have expanded this framework, revealing multilayered regulation through the core proteins of cuproptosis, mitochondrial respirator
