[Protective effect of cholestyramine in the hepatocarcinogenesis of the rat (author's transl)].

The influence of cholestyramine on hepatocarcinogenesis was investigated in the Wistar rat. After 14 weekly gastric instillation of 50 mg diethylnitrosamine/kg and simultaneous administration of diet supplement with 1.3% cholestyramine, half of the animals developed hepatic tumors. In contrast, nearly all the animals in the control group under similar experimental conditions, but without the cholestyramine supplement in their diet, developed hepatic tumors. The sequestration of bile acids and the reduction of the noxious lithocholic acid could be considered as an explanation for the modificati

Altered gut microbiota and metabolite profiles in community-acquired pneumonia: a metagenomic and metabolomic study.

UNLABELLED: Emerging evidence suggests that altered gut microbiota is linked to community-acquired pneumonia (CAP), but the potential mechanisms by which gut microbiota and its metabolites contribute to the development of CAP remain unclear. Fecal samples from 32 CAP patients and 36 healthy controls were analyzed through metagenomic sequencing and metabolomic profiling. The gut microbiota composition in CAP patients showed significant differences and lower diversity compared to healthy controls. Genera involved in short-chain fatty acid (SCFA) production, such as Faecalibacterium, Ruminococcus

Targeting the Eph-ephrin System with Protein-Protein Interaction (PPI) Inhibitors.

Eph-ephrin system is emerging as a new potential target in several diseases including cancer, diabetes, neurodegenerative diseases and inflammation. In the last decade, several efforts have been made to develop small molecule antagonists of Eph receptors. Both natural and synthetic compounds were discovered with (poly) phenol and steroidal derivatives on one side and the α1 agonist doxazosin, 2,5-dimethylpyrrol- 1-yl-benzoic acids and amino acid conjugates of lithocholic acid on the other. In the present paper we critically present available data for these compounds and discuss their potential

Synthesis of dimeric steroid-glycylglycine conjugates by Ugi four-component reaction. NMR and X-ray diffraction characterization.

Ugi-four component reaction of bile acids, formaldehyde, ethyl isocyanoacetate and 1,4-phenylenediamine produced dimeric steroid-glycylglycine conjugates in low yields. The structure of the new compounds consists of two steroid-dipeptide conjugate units linked by a 1,4-phenylene bridge. Detailed NMR characterization provided sufficient evidence on the obtained structures that were confirmed by single crystal X-ray diffraction. Interestingly X-ray crystallography revealed that the dimeric lithocholic acid-glycylglycine conjugate crystalizes in a conformation having the tertiary and secondary am

A novel role for the retinoic acid-catabolizing enzyme CYP26A1 in Barrett's associated adenocarcinoma.

Vitamin A deficiency is associated with carcinogenesis, and upregulation of CYP26A1, a major retinoic acid (RA)-catabolizing enzyme, has recently been shown in cancer. We have previously demonstrated alterations of RA biosynthesis in Barrett's oesophagus, the precursor lesion to oesophageal adenocarcinoma. The aims of this study were to determine CYP26A1 expression levels and functional effects in Barrett's associated carcinogenesis. Retinoic acid response element reporter cells were used to determine RA levels in non-dysplastic and dysplastic Barrett's cell lines and endoscopic biopsies. CYP2

Diversity of bile acids in the fetus and newborn infant.

The bile acids found in the fetus and newborn are more numerous and diverse than has generally been appreciated. The four conventional bile acids, cholic, chenodeoxycholic, deoxycholic, and lithocholic acids, are found. In addition, however, stereoisomers of the conventional bile acids, bile acids with functional groups at different positions or in greater number than found in conventional bile acids, and "short-chain" and "long-chain" bile acids are found. The site of origin, pathways of synthesis, metabolism, and excretory routes of these unconventional bile acids are largely unknown. Their

The cytotoxicity of plasma from patients with acute hepatic failure to isolated rabbit hepatocytes.

The cytotoxicity of plasma from patients with various types of liver disease to rabbit hepatocytes maintained in vitro has been investigated using a microcytotoxicity assay system. Plasma from patients with fulminant hepatic failure and uncomplicated viral hepatitis showed significant cytotoxicity compared to controls. The cytotoxic effect of plasma from patients with fulminant hepatic failure was reduced by charcoal haemoperfusion, or heating and dialysis. The bile acids chenodeoxycholic acid and lithocholic acid were cytotoxic when added to control human plasma. Such toxic factors may be res

Serum and urinary bile acids in patients with primary biliary cirrhosis.

Serum concentrations and daily urinary excretions of unsulphated and total cholic (C) and chenodeoxycholic (CDC) acid were determined by radioimmunoassay in 15 patients with primary biliary cirrhosis. Thirteen patients had increased fasting serum bile acid concentrations; two of them had an increase of C only. An increase of C and CDC in serum was always followed by an increased urinary excretion of C and CDC. The individual serum bile acids were separated by gas-liquid chromatography. 3 beta-hydroxy-5-cholenoic acid was increased in nine patients, and its serum concentration was correlated to

Obeticholic acid accelerates portal vein embolization-induced liver hypertrophy in rabbits by improving bile salt homeostasis.

Obeticholic acid (OCA), a potent agonist of the bile salt-activated nuclear receptor farnesoid X receptor (FXR), accelerates liver hypertrophy after portal vein embolization (PVE), but the mechanisms underlying this effect are incompletely understood. In a rabbit model of PVE, New Zealand White rabbits received OCA (10 mg/kg/day) or vehicle by oral gavage before and after embolization. We quantified systemic and hepatic bile salt pools, profiled FXR-regulated genes in ileum and liver, and related these parameters to volumetric and histological indices of regeneration. OCA shifted bile salt com

Bile acid synthesis by long-term cultured cell line established from human hepatoblastoma.

Bile acids in the spent medium for the cell culture were analyzed by gas-liquid chromatography and gas-liquid chromatography-mass spectrometry to determine whether human hepatoblastoma cell line could synthesize bile acids. Cholic, chenodeoxycholic, and lithocolic acids were found in the culture medium, and a portion of chenodeoxycholic acid and all of lithocholic acid were sulfated. Since the cells had been cultured in serum-free medium, it is clear that the bile acids were newly synthesized and sulfated by the cultured cells. Chenodeoxycholic acid was the main bile acid in the medium, sugges

Vitamin D receptor as an intestinal bile acid sensor.

The vitamin D receptor (VDR) mediates the effects of the calcemic hormone 1alpha,25-dihydroxyvitamin D3 [1,25(OH)2D3]. We show that VDR also functions as a receptor for the secondary bile acid lithocholic acid (LCA), which is hepatotoxic and a potential enteric carcinogen. VDR is an order of magnitude more sensitive to LCA and its metabolites than are other nuclear receptors. Activation of VDR by LCA or vitamin D induced expression in vivo of CYP3A, a cytochrome P450 enzyme that detoxifies LCA in the liver and intestine. These studies offer a mechanism that may explain the proposed protective

Bilirubin and bile acids in osteocytes and bone tissue. Potential role in the cholestatic-induced osteoporosis.

BACKGROUND AND AIMS: Osteoporosis is a common complication in patients with primary biliary cholangitis. Both bilirubin and lithocholic acid (LCA) result in detrimental effects on osteoblastic cells, and ursodeoxycholic acid (UDCA) counteracts these outcomes. However, there is no information on the consequences of these retained substances of cholestasis and sera from cholestatic patients in osteocytes. METHODS: The impact of bilirubin, LCA, UDCA and serum from jaundiced patients on viability, differentiation, mineralization and apoptosis has been assessed in MLO-Y4 and MLO-A5 osteocyte cell

Cellular Accumulation and Toxic Effects of Bile Acids in Cyclosporine A-Treated HepaRG Hepatocytes.

Alteration of bile acid (BA) profiles and secretion by cholestatic drugs represents a major clinical issue. Species differences exist in BA composition, synthesis, and regulation; however presently, there is no in vitro reproducible cell model to perform studies on BAs in humans. We have evaluated the capacity of the human HepaRG cell line to synthesize, conjugate, and secrete BAs, and analyzed changes in BA content and profile after cyclosporine A (CsA) treatment. Our data show that HepaRG cells produced normal BAs at daily levels comparable, though in different proportions, to those measured

Rational Design of pH-Switchable 3α-Amino Lithocholate-Modified Lipids for Efficient siRNA Delivery.

Lipid nanoparticles (LNPs) represent a state-of-the-art platform for RNA-based therapeutics, yet inefficient endosomal escape remains a critical barrier to cytosolic RNA delivery. We designed novel pH-switchable 3α-amino lithocholate-modified lipids (LMLs) with a unique mechanism of action. Under weakly basic conditions, the 3α-amino lithocholate moieties sequester within LNP membranes, while acidic environments (e.g., endosomal pH 5.0) trigger their reorientation to the lipid-water interface. The pH-driven flipping behavior amplified the surface charge of LNPs, yielding a much higher zeta pot

A Computational Workflow for Prioritizing Microbial Metabolite-Associated Host Genes in Constipation-Predominant Irritable Bowel Syndrome.

No standardized computational pipeline exists for systematically prioritizing microbial metabolite-associated host genes and protein-ligand complexes from publicly available chemical, genomic, and structural databases. This article describes an eight-stage workflow that accepts a user-defined set of gut microbiota-derived metabolites and produces a ranked shortlist of candidate metabolite-associated host genes, enriched biological pathways, and structurally prioritized protein-ligand complexes for experimental follow-up. The pipeline integrates (i) chemoinformatic metabolite profiling; (ii) mu

A microbiota-derived bile acid modulates biofilm formation by the probiotic strain Escherichia coli Nissle 1917.

Bacteria that colonize the human gut must withstand a variety of stressors, including detergent-like compounds known as bile acids. Here, we investigated how bile acids found in the human cecum and colon impact the behavior of the probiotic strain Escherichia coli Nissle 1917 (EcN). We found that lithocholic acid (LCA), which is a microbiota-derived secondary bile acid, promotes the formation of a distinctive surface-coating biofilm by EcN, including on an organoid-derived model of the human colonic epithelium. Mechanistic investigations revealed that LCA upregulates the production of several

The effect of bile salts on human vascular endothelial cells.

The uptake and release of radiochromium from adult human vascular endothelial cells in culture was employed to determine the relative toxicity of different bile salts. Endothelial cells after pre-incubation with 51Cr for 18 h were incubated with bile salts for 24 h and percentage chromium release was taken as a measure of toxicity to cells. Lithocholic acid (LC) (potassium salt) was cytotoxic at concentrations greater than 50 microM. However, LC glucuronide, sulfate and the beta-epimer were progressively less toxic with toxicity seen at concentrations of 60, 110 and 180 microM, respectively. T

Taurocholic acid metabolism by gut microbes and colon cancer.

Colorectal cancer (CRC) is one of the most frequent causes of cancer death worldwide and is associated with adoption of a diet high in animal protein and saturated fat. Saturated fat induces increased bile secretion into the intestine. Increased bile secretion selects for populations of gut microbes capable of altering the bile acid pool, generating tumor-promoting secondary bile acids such as deoxycholic acid and lithocholic acid. Epidemiological evidence suggests CRC is associated with increased levels of DCA in serum, bile, and stool. Mechanisms by which secondary bile acids promote CRC are
