Centrally-administered AMPA antagonists increase locomotion in parkinsonian rats.

It was shown in the present study that three antagonists of the alpha-amino-3-hydroxy-5-methylisoxazole-4-propionic acid (AMPA) glutamate receptor, including 2,3-dihydroxy-6-nitro-7-sulfamoylbenzo(f)quinoxaline (NBQX), 1-(4-aminophenyl)-4-methyl-7,8-methylenedioxy-5H-2,3-benzodiazepine (GYKI 52466) and 6-(1H-imidazole-1-yl)-7-nitro-2,3-(1H, 4H)-quinoxalinedione (YM90K), caused marked reversal of akinesia when administered into the entopeduncular nucleus of rats rendered parkinsonian by bilateral substantia nigra pars compacta lesion. These data suggest that centrally active AMPA antagonists ma

Variable gearing during locomotion in the human musculoskeletal system.

Human feet and toes provide a mechanism for changing the gear ratio of the ankle extensor muscles during a running step. A variable gear ratio could enhance muscle performance during constant-speed running by applying a more effective prestretch during landing, while maintaining the muscles near the high-efficiency or high-power portion of the force-velocity curve during takeoff. Furthermore, during acceleration, variable gearing may allow muscle contractile properties to remain optimized despite rapid changes in running speed. Forceplate and kinematic analyses of running steps show low gear r

Head stabilization during locomotion. Perturbations induced by vestibular disorders.

Head kinematics was studied in 10 normal subjects (NS) and 7 patients (P) with bilateral vestibular deficit while they executed various locomotor tasks. The movement of the body was recorded with a video system which allowed a computer reconstruction of the motion of joint articulations and other selected points on the body in three dimensions. Analyses focus on head translation along the vertical axis and rotation in the sagittal plane. Two conditions were studied: free walking (W) and hopping (H). The subjects were tested in light and in darkness. In NS, while walking in darkness, mean head

Locomotion-specific metabolic demands in 30-s anaerobic efforts.

PURPOSE: This study aimed to examine whether energy system contributions differ across locomotion modalities during 30-s all-out exercise. METHODS: Sixteen trained male team-sport athletes completed a Wingate Anaerobic Test (WAnT), a 30-s continuous jumping test (CJ30), and a 30-s all-out running test (RUN30) in randomized order. Aerobic, glycolytic, and phosphagen energy contributions were estimated using the PCr-La-O2 method based on oxygen uptake, blood lactate accumulation, and excess post-exercise oxygen consumption. RESULTS: Total energy expenditure was higher in RUN30 (130.8 ± 18.0 kJ

Nanocollision promotes locomotion of dendritic cells for tumor therapy.

Mechanical stimuli originating from extracellular matrix have been proved to affect antitumor immunity by regulating dendritic cell (DC) locomotion. However, the immunological consequences of ultrasmall bioparticle-induced nanocollision remain largely an unexplored realm despite its ubiquitous and incessant occurrence within the in vivo milieu. Herein, we disclose that nanocollisions caused by endogenous bioparticles, such as extracellular vesicles, can induce localized membrane deformations. This spatially confined mechanical input activates Piezo1 at collision sites and promotes myosin IIA p

Tube feet dynamics drive adaptation in sea star locomotion.

Sea stars use hundreds of tube feet on their oral surface to crawl, climb, and navigate complex environments, despite lacking a central brain. While tube foot morphology and function as muscular hydrostats are well described, the mechanisms that coordinate their collective dynamics remain poorly understood. To investigate these dynamics, we employed an optical imaging method based on frustrated total internal reflection (FTIR) to visualize and quantify tube foot adhesive contacts in real time in the species Asterias rubens across individuals spanning a wide size range. Our results reveal an in

Shape-Dependent Locomotion of DNA-Linked Magnetic Nanoparticle Films.

The shape-dependent aero- and hydro-dynamics found in nature have been adopted in a wide range of areas spanning from daily transportation to forefront biomedical research. Here, we report DNA-linked nanoparticle films exhibiting shape-dependent magnetic locomotion, controlled by DNA sequences. Fabricated through a DNA-directed layer-by-layer assembly of iron oxide and gold nanoparticles, the multifunctional films exhibit rotational and translational motions under magnetic fields, along with reversible shape morphing via DNA strand exchange reactions. Notably, the shape of the film significant

[Cortical Control of Steps During Locomotion to Avoid Obstacles].

Although it has long been known that cortical contribution is undoubtedly necessary for visually guided modification of steps during locomotion, most of the physiological evidence for cortical contribution has been accumulated since the 1980s. This article reviews evidence obtained from experiments on cats that have demonstrated the involvement of the primary motor cortex (MI), posterior parietal cortex (PPC), and premotor areas (PM) in visually guided locomotion. Activity in the MI, which is tightly coupled with muscle activity of the contralateral limbs, has been thought to control muscle sy

REST overexpression in mice causes deficits in spontaneous locomotion.

Overexpression of REST has been implicated in brain tumors, ischemic insults, epilepsy, and movement disorders such as Huntington's disease. However, owing to the lack of a conditional REST overexpression animal model, the mechanism of action of REST overexpression in these disorders has not been established in vivo. We created a REST overexpression mouse model using the human REST (hREST) gene. Our results using these mice confirm that hREST expression parallels endogenous REST expression in embryonic mouse brains. Further analyses indicate that REST represses the dopamine receptor 2 (Drd2) g

Whisker touch sensing guides locomotion in small, quadrupedal mammals.

All small mammals have prominent facial whiskers that they employ as tactile sensors to guide navigation and foraging in complex habitats. Nocturnal, arboreal mammals tend to have the longest and most densely packed whiskers, and semi-aquatic mammals have the most sensitive. Here we present evidence to indicate that many small mammals use their whiskers to tactually guide safe foot positioning. Specifically, in 11, small, non-flying mammal species, we demonstrate that forepaw placement always falls within the ground contact zone of the whisker field and that forepaw width is always smaller tha

Visual exploration during locomotion limited by fear of heights.

BACKGROUND: Visual exploration of the surroundings during locomotion at heights has not yet been investigated in subjects suffering from fear of heights. METHODS: Eye and head movements were recorded separately in 16 subjects susceptible to fear of heights and in 16 non-susceptible controls while walking on an emergency escape balcony 20 meters above ground level. Participants wore mobile infrared eye-tracking goggles with a head-fixed scene camera and integrated 6-degrees-of-freedom inertial sensors for recording head movements. Video recordings of the subjects were simultaneously made to co

Seizure-related automatic locomotion triggered by intracerebral electrical stimulation.

We describe the case of an eight-year-old boy, who underwent a video-stereo EEG (SEEG) investigation for the presurgical assessment of drug-resistant epilepsy, related to a right fronto-lateral cortical dysplasia and who became seizure-free after epilepsy surgery. The unexpected finding of the investigations was that intracerebral, high frequency (50 Hz) electrical stimulation (HFS) triggered the emergence of automatic and involuntary forward-backward locomotion during a focal seizure while the boy was standing. This clinical manifestation was different from the chaotic motor activity describe

Goal directed locomotion and balance control in autistic children.

This article focuses on postural anticipation and multi-joint coordination during locomotion in healthy and autistic children. Three questions were addressed. (1) Are gait parameters modified in autistic children? (2) Is equilibrium control affected in autistic children? (3) Is locomotion adjusted to the experimenter-imposed goal? Six healthy children and nine autistic children were instructed to walk to a location (a child-sized playhouse) inside the psychomotor room of the pedopsychiatric centre located approximately 5 m in front of them. A kinematic analysis of gait (ELITE system) indicates

Brainstem-mediated locomotion and myoclonic jerks. II Pharmacological effects.

Previous studies in our laboratory have demonstrated that microinjection of N-methyl-D-aspartate (NMDA) agonist into the nucleus magnocellularis (NMC) of the medial medulla increases muscle tone and/or produces locomotion, while injection of corticotropin-releasing factor (CRF) and non-NMDA agonists into the same or nearby sites suppresses muscle tone. In the first paper of this series, we report that myoclonic twitches or coordinated rhythmic leg movement (locomotion) can be induced by either NMDA or hemorrhagic bilateral lesion of the ventral mesopontine junction (vMPJ). In this paper, we re

Brainstem-mediated locomotion and myoclonic jerks. I. Neural substrates.

Eleven of 40 decerebrated cats were found to exhibit periods of spontaneous or sensory myoclonus and locomotion beginning 24 h after decerebration. Histological analysis showed that the cats generating myoclonus hemorrhagic lesions in the retrorubral nucleus (RRN) and ventral mesopontine junction (vMPJ). However, with intact RRN and vMPJ never showed myoclonus. To verify that the lesions were responsible for myoclonus, 6 additional cats received N-methyl-D-aspartate (NMDA, 0.5 M/0.5 microliter) injections in the areas of RRN and vMPJ to produce bilateral lesions. Reductions in Muscle Strength and Range of Motion Cause Locomotion Disability via Locomotion-Related Functional Limitation in Japanese Older Adults: A Cross-Sectional Study.

BACKGROUND: Functional issues (impairments, functional limitations, and disabilities) gradually occur with age. Nonetheless, maintaining physical capability may help prevent locomotion disabilities at an older age. The present study aimed to determine whether reductions in muscle strength and range of motion (ROM) cause locomotion disability via locomotion-related functional limitations among healthy older adults. METHODS: Data from a total of 144 participants (61 men, 83 women) were analyzed. To assess locomotion disability, the locomotor domain of the activities of daily living (ADLs) surve

Cancer may accelerate locomotive syndrome and deteriorate quality of life: a single-centre cross-sectional study of locomotive syndrome in cancer patients.

BACKGROUND: Thanks to recent advancement in cancer treatment, an increasing number of cancer patients are expected to live longer with cancer. The ambulatory ability is essential for cancer patients to spend their own independent lives, but locomotive syndrome (LS), a condition of reduced mobility due to impairment of locomotive organs, in cancer patients has been seldom examined. METHODS: This was a single-institutional cross-sectional study. Cancer patients receiving cancer therapy between April 2020 and March 2021 were asked to participate. LS was classified as stage 0-3, and compared with

Ground Perturbation Detection via Lower-Limb Kinematic States During Locomotion.

Falls during daily ambulation activities are a leading cause of injury in older adults due to delayed physiological responses to disturbances of balance. Lower-limb exoskeletons have the potential to mitigate fall incidents by detecting and reacting to perturbations before the user. Although commonly used, the standard metric for perturbation detection, whole-body angular momentum, is poorly suited for exoskeleton applications due to computational delays and additional tunings. To address this, we developed a novel ground perturbation detector using lower-limb kinematic states during locomotio
