Clinical autonomic research : official journal of the Clinical Autonomic Research Society2006eng
In this study we evaluated by indirect immunohistochemistry the prevalence of cerebrospinal fluid (CSF) antibodies reacting with structures of rat pons/medulla in patients with multiple system atrophy (MSA) (n = 29), Parkinson disease with neurogenic orthostatic hypotension (n = 13), or pure autonomic failure (n = 11) and in control subjects without autonomic failure (n = 33). About 10-20% of CSF samples had positive immunoreactivity to rat locus coeruleus (LC), regardless of clinical diagnosis. The results failed to confirm the previously reported high prevalence of immune binding to rat LC i

Somatostatin (SS14) binding sites within locus coeruleus (LC) were localized at the light microscope level by [125I][Tyr0,D-Trp8]SS14 radioautography combined with an immunohistochemical/neurotoxic lesioning approach. In intact rats, the dense accumulation of SS14 binding sites of LC conspicuously overlapped with the cluster of tyrosine hydroxylase (TH) immunoreactive neurons; SS14 specific binding was directly proportional to the number of TH immunostained (TH+) cell bodies per mg of tissue throughout LC. Complete lesion of catecholaminergic nerve cell bodies of LC by intracerebroventricular

Young, 4-week-old spontaneously hypertensive rats (SHR) had significantly higher concentrations of dopamine and 3,4-dihydroxyphenylacetic acid and a faster rate of accumulation of L-dihydroxyphenylalanine after decarboxylase inhibition in the locus coeruleus than age-matched Wistar-Kyoto rats, whereas the norepinephrine levels were similar to each other. In the brainstem, these changes were restricted to the locus coeruleus and were not present in adult, 14-week-old rats with established hypertension. These results are suggestive of a noradrenergic overactivity in the locus coeruleus of young

Neuromelanin-(NM) containing organelles are sub-cellular auto-lysosomal structures composed of three main compartments: NM pigment, protein matrix, and lipid bodies. These organelles accumulate during aging and are found predominantly in the catecholaminergic neurons of Substantia Nigra (SN) and Locus Coeruleus (LC), the main regions affected in Parkinson's disease (PD). NM serves a protective function by sequestering potentially toxic metals like Cu, Fe, and Al. However, NM released from degenerating neurons may lead to a cascade of events resulting in neuroinflammation and neurodegeneration.

BACKGROUND: Fibromyalgia is a chronic pain condition without an established aetiology. However, noradrenergic dysfunction is a possible mechanism to explain the constellation of symptoms associated with fibromyalgia. Noradrenaline synthesis in the locus coeruleus (LC) results in a paramagnetic by-product, neuromelanin. Recently, a magnetic resonance imaging sequence sensitive to neuromelanin has been used to assay LC signal intensity, a proxy for noradrenergic system function. Here, we use MR imaging to investigate the noradrenergic-locus coeruleus system in participants with fibromyalgia and

Alzheimer's disease (AD) often begins with non-cognitive symptoms such as olfactory deficits, which can predict later cognitive decline, though the mechanisms remain unclear. Pathologically, the brainstem locus coeruleus (LC), the main source of the neurotransmitter noradrenalin (NA) modulating olfactory information processing is affected early. Here we show early and distinct loss of noradrenergic input to the olfactory bulb (OB) coinciding with impaired olfaction in an AD mouse model, before appearance of amyloid plaques. Mechanistically, OB microglia recognize and phagocytose LC axons. Redu

Dysregulation in contextual processing is believed to affect several forms of psychopathology, such as post-traumatic stress disorder (PTSD). The dentate gyrus (DG), a subregion of the hippocampus, is thought to be an important brain region for disambiguating new experiences from prior experiences. Noradrenergic (NE) neurons in the locus coeruleus (LC) are more tonically active during stressful events and send dense projections to the DG, yet an understanding of their function in DG-dependent contextual discrimination has not been established. Here, we isolate a key function of the LC-NE-DG ci

Sleep and brain health are closely coupled, yet the mechanisms underlying this relationship remain incompletely understood. The locus coeruleus (LC) is a key modulator of sleep architecture, and LC dysfunction (LCd) is implicated in a wide range of neurodegenerative and psychiatric disorders, including Alzheimer's disease, Parkinson's disease, depression, and post-traumatic stress disorder. The isolated effects of LCd on sleep and brain health cannot be studied in humans, but they can be modeled in animals using the noradrenergic neurotoxin DSP4, which produces chronic and selective LC degener

BACKGROUND: The locus coeruleus (LC) is a key noradrenergic nucleus of the brain. Its dysfunction is implicated in neurodegenerative diseases like Alzheimer's disease and Parkinson's disease, as well as in psychiatric disorders. However, imaging the LC with sufficient contrast-to-noise ratio (CNR) is challenging due to its small size and deep location in the brainstem. This study optimizes a 3D gradient echo (GRE) sequence with magnetization transfer contrast (MTC) to enable rapid, high-resolution LC imaging in under five minutes.
METHODS: A high-resolution 3D-GRE-MTC sequence was optimized o

INTRODUCTION: Locus coeruleus (LC) is one of the earliest structures altered in Alzheimer's disease (AD). Inflammation is also now considered critical in AD pathology, early stage included. However, no association between LC degeneration and the peripheral inflammation has been reported yet.
METHODS: A cohort of 102 patients was studied for which both magnetic resonance imaging (MRI) scans and blood samples were available. LC integrity was assessed by MRI, and plasma soluble TAMs (Tyro3, Axl, and MerTK) receptor levels were measured by enzyme-linked immunosorbent assay (ELISA).
RESULTS: We f

INTRODUCTION: In murine models of Alzheimer's disease (AD), lesioning the locus coeruleus-norepinephrine (LC-NE) system with DSP-4 exacerbates AD-like neuropathology and cognitive impairment. However, the impact of LC lesions during prodromal stages is poorly characterized.
METHODS: TgF344-AD and wild-type rats received monthly injections of DSP-4 or saline from 1-5 months of age, a time point preceding forebrain plaque or tangle deposition in TgF344-AD rats, after which behavior and pathology were assessed.
RESULTS: DSP-4 compromised LC cell bodies, fibers, and NE content. LC lesion and the

Chronic exposure to stress throughout the lifespan has been the focus of many studies on Alzheimer's disease (AD) because of the similarities between the biological mechanisms involved in chronic stress and the pathophysiology of AD. In fact, the earliest abnormality associated with the disease is the presence of phosphorylated tau protein in locus coeruleus neurons, a brain structure highly responsive to stress and perceived threat. Here, we introduce allostatic load as a useful concept for understanding many of the complex, interacting neuropathological changes involved in the AD degenerativ

This study aimed to explore the underlying molecular mechanisms of idiopathic Parkinson's disease (IPD) by bioinformatics analysis. Gene expression profile GSE34516 was downloaded from the Gene Expression Omnibus. Eight locus coeruleus post-mortem tissue samples derived from four IPD patients and four neurological healthy controls were used to identify the differentially expressed genes (DEGs) by paired t test. Based on the DEGs, principal components were analyzed. The Gene Ontology functional and Kyoto Encyclopedia of Genes and Genomes pathway analysis of the genome microarray data were then

The locus coeruleus (LC) is a noradrenergic brainstem structure that is considered to play a role in promoting arousal. To further clarify the role of LC noradrenergic neurons, we performed an optogenetic assay by injecting AAV-channelrhodopsin-2 (ChR2) into the LC of cre-tyrosine hydrolase (TH) mice. We found here that the specific activation of LC noradrenergic neurons produced a significant increase in wakefulness and a significant decrease in non-rapid eye movement (NREM) sleep during photostimulation. On the other hand, neuropathic pain is believed to significantly interfere with sleep, a

Parkinson's disease (PD) is characterized not only by a progressive loss of dopaminergic neurons in the substantia nigra pars compacta (SNc) but also by a degeneration of locus coeruleus (LC) noradrenergic neurons. It has been suggested that deficient LC noradrenergic mechanisms might play a critical role in symptomatology and in the progression of PD. However, the effect of LC depletion on levodopa-induced motor complications, such as the motor fluctuations, is still unknown. Male Sprague-Dawley rats received 50 mg/kg intraperitoneal (i.p.) of [N-(2-chloroethyl)-N-ethyl-2-bromobenzylamine] (D

Previous studies have demonstrated a widespread stimulation of regional cerebral metabolic rate(s) for glucose (rCMRglc) in morphine-dependent rats subjected to opioid withdrawal precipitated by systemic injection of naloxone. Nonetheless, many of the behavioral signs of opioid withdrawal are produced by intracerebral injections of an opioid antagonist, methylnaloxonium (MN), into the locus coeruleus (LC). The purpose of the present work was to determine the extent to which cerebral metabolic alterations in opioid withdrawal could be initiated by a local action in LC. Intracerebral injections

Plastic changes in axon terminals of NA LC neurons following repeated stress and antidepressant treatments were examined using electrophysiological or morphological methods. For stress treatment, rats restrained in a small cage were immersed up to the neck in warm water for 10 min daily. Electrophysiological experiments were performed under urethane anesthesia on the day following the termination of stress treatment. To quantify the density of cortical axon terminals arising in the LC, the percentage of LC neurons activated antidromically from the cerebral cortex was assessed. The percentage o

INTRODUCTION: Sleep disruption, particularly loss of slow-wave sleep (SWS), is common in Alzheimer's disease (AD), but its neurobiological underpinnings remain unclear. We investigated whether locus coeruleus (LC) integrity relates to SWS across the AD continuum and whether sex and perivascular spaces (PVSs) modify these associations.
METHODS: In a cohort (11 controls, 30 mild cognitive impairment, 17 AD dementia) we combined overnight polysomnography with LC-sensitive magnetic resonance imaging of the LC, basal ganglia and centrum semiovale PVS ratings, and cerebrospinal fluid noradrenaline.
