Chromosome 16p13.11 microduplication is a risk factor associated with various neurodevelopmental disorders such as attention-deficit/hyperactivity disorder, intellectual disabilities, developmental delay and autistic spectrum disorder. The underlying molecular mechanism of this genetic variation remained unknown, but its core genetic locus-conserved across mice and humans-contains seven genes. Here, we generated bacterial artificial chromosome-transgenic mice carrying a human 16p13.11 locus, and these mice showed the behavioral hyperactivity phenotype. We identified miR-484 as the responsible

We describe a Saccharomyces cerevisiae bioluminescence assay for UV and arsenate in which bacterial luciferase genes are regulated by the promoter of the yeast gene, UFO1. UFO1 encodes the F-box subunit of the Skp1–Cdc53–F-box protein ubiquitin ligase complex and is induced by DNA damage and by arsenate. We engineered the UFO1 promoter into an existing yeast bioreporter that employs human genes for detection of steroid hormone-disrupting compounds in water bodies. Our analysis indicates that use of an endogenous yeast promoter in different mutant backgrounds allows discrimination between diffe

Alanine/glyoxylate aminotransferase 1 (AGT) is peroxisomal in most normal humans, but in some patients with the hereditary disease primary hyperoxaluria type 1 (PH1), AGT is mislocalized to the mitochondria. In an attempt to identify the sequences in AGT that mediate its targeting to peroxisomes, and to determine the mechanism by which AGT is mistargeted in PH1, we have studied the intracellular compartmentalization of various normal and mutant AGT polypeptides in normal human fibroblasts and cell lines with selective deficiencies of peroxisomal protein import, using immunofluorescence microsc

One limitation of employing lux bioreporters to monitor in situ microbial gene expression in dynamic, laboratory-scale systems is the confounding variability in the luminescent responses. For example, despite careful control of oxygen tension, growth stage, and cell number, luminescence from Pseudomonas putida RB1353, a naphthalene-degrading lux bioreporter, varied by more than sevenfold during saturated flow column experiments in our laboratory. Therefore, this study was conducted to determine what additional factors influence the luminescent response. Specifically, this study investigated th

Yersinia pestis is a tier 1 agent due to its contagious pneumopathogenicity, extremely rapid progression, and high mortality rate. As the disease is usually fatal without appropriate therapy, rapid detection from clinical matrices is critical to patient outcomes. We previously engineered the diagnostic phage ΦA1122 with luxAB to create a "light-tagged" reporter phage. ΦA1122::luxAB rapidly detects Y. pestis in pure culture and human serum by transducing a bioluminescent signal response. In this report, we assessed the analytical specificity of the reporter phage and investigated diagnostic uti

Toll-like receptors (TLRs) are one of the most important innate immune receptors, which recognize various pathogen-associated molecular patterns and activate the downstream immune response. Mouse TLR13 has been found to recognize a highly conserved sequence from bacterial or viral RNA and activate the myeloid differentiation primary response gene 88-dependent signaling response. The function of teleost tlr13 is still not fully understood, especially its relationship with bacterial RNA. In our study, we identified and characterized a tlr13 from Epinephelus coioides (orange-spotted grouper). The

INTRODUCTION: Listeria monocytogenes (L. monocytogenes) is an intracellular bacterial pathogen that requires coordinated innate and adaptive immune responses for effective clearance. However, the epigenetic mechanisms governing host defense against L. monocytogenes remain poorly understood.
METHODS: JMJD2D-deficient mice and wild-type littermates were infected with L. monocytogenes to assess host susceptibility, bacterial burdens, tissue damage, and survival. Bone marrow-derived macrophages (BMDMs) and peritoneal macrophages (PMs) were isolated for in vitro infection assays. Molecular mechani

The gold standard in microbiology for monitoring bacterial dissemination in infected animals has always been viable plate counts. This method, despite being quantitative, requires sacrificing the infected animals. Recently, however, an alternative method of in vivo imaging of bioluminescent bacteria (IVIBB) for monitoring microbial dissemination within the host has been employed. Yersinia pestis is a Gram-negative bacterium capable of causing bubonic, septicemic, and pneumonic plague. In this study, we compared the conventional counting of bacterial colony forming units (cfu) in the various in

Live, attenuated Vibrio cholerae vaccines can induce potent immune responses after only a single oral dose. The strategy of harnessing these strains to present antigens from heterologous pathogens to the mucosal immune system shows great promise. To fully realize this possibility, V. cholerae strains must be created that stably express antigens in vivo in sufficient quantity to generate an immune response. In vivo-induced promoters have been shown to increase the stability and immunogenicity of foreign antigens expressed from multicopy plasmids. We report the construction of a series of geneti

Inducible transcription and position-independent expression are critical issues after gene transfer. To gain insight into the amount of variability of transcriptional regulation due to random proviral integration, we analyzed a total of 200 C6 glioma and rat-1 fibroblast clones retrovirally infected with the conventional and reverse tet systems where a luciferase reporter gene was placed under control of a tetracycline-responsive promoter. Repressed luciferase activities differed by up to 81000-fold among individual clones. Repressed activities close to baseline levels were observed in eight c

We present a method for constructing a Kunjin virus (KUNV) infectious clone capable of stable proliferation in bacterial systems. This KUNV infectious clone successfully produces infectious viral particles within host cells. In vitro, the recombinant virus demonstrates the ability to replicate in primary duck cells and induces 100 % mortality in duck embryos. In vivo, the recombinant virus exhibits virulence in our established duckling model, leading to clinical signs such as hepatic haemorrhage, splenomegaly, and pancreatic haemorrhage. Furthermore, the recombinant rKUNV induced a mortality r

The determination of the chronic toxicity is time-consumed and costly, so it's of great interest to predict the chronic toxicity based on acute data. Current methods include the acute to chronic ratios (ACRs) and the QSTR models, both of which have some usage limitations. In this paper, the acute and chronic mixture toxicity of three types of antibiotics, namely sulfonamides, sulfonamide potentiators and tetracyclines, were determined by a bioluminescence inhibition test. A novel QSTR model was developed for predicting the chronic mixture toxicity using the acute data and docking-based descrip

Anti-restriction proteins ArdA and Ocr are specific inhibitors of type I restriction-modification enzymes. The IncI1 transmissible plasmid ColIb-P9 ardA and bacteriophage T7 0.3(ocr) genes were cloned in pUC18 vector. Both ArdA (ColIb-P9) and Ocr (T7) proteins inhibit both restriction and modification activities of the type I restriction-modification enzyme (EcoKI) in Escherichia coli K12 cells. ColIb-P9 ardA, T7 0.3(ocr), and the Photorhabdus luminescens luxCDABE genes were cloned in pZ-series vectors with the P(ltetO-1) promoter, which is tightly repressible by the TetR repressor. The Staphylococcus aureus SarA global regulator controls the expression of numerous virulence genes, often in conjunction with the agr quorum-sensing system and its effector RNA, RNAIII. In the present study, we have examined the role of both SarA and RNAIII on the regulation of the promoter of tst, encoding staphylococcal superantigen toxic shock syndrome toxin 1 (TSST-1). In vitro DNA-protein interaction studies with purified SarA using gel shift and DNase I protection assays revealed one strong SarA binding site and evidence for a weaker site nearby within the minimal 400-bp promoter region

Dominant selectable markers, reporter genes and regulatable systems remain powerful molecular tools for genetic and cell biology studies in fungi. Among Saccharomycotina, it is currently accepted that most species belonging to the genus Candida have adopted a specific codon usage, whereby the CTG codon encodes serine instead of leucine. This group is now widely referred to as the CTG clade. For a long time, this uncommon genetic code has precluded the use of the available Saccharomyces or bacterial markers and reporter systems for genetic studies in Candida species. Over the last 15 years, inc

BACKGROUND: Leprosy is a chronic infectious disease caused by Mycobacterium leprae (M. leprae). Despite significant progress in disease control, the molecular mechanisms regulating host responses to M. leprae infection remain incompletely understood. MicroRNAs (miRNAs) are important post-transcriptional regulators that modulate immune responses and host-pathogen interactions in many infectious diseases. However, the specific miRNA-mediated regulatory networks involved in leprosy and their potential implications for monitoring therapeutic responses remain poorly defined.

Suppressor of cytokine signaling 1 (SOCS1) is a pivotal negative regulator of cytokine-mediated signaling in mammals, yet its role in teleost fish immunity remains poorly understood. In this study, we cloned and characterized the full-length SOCS1 cDNA from Japanese eel (Anguilla japonica), which encodes a 203-amino acid protein containing a conserved SH2 domain and SOCS box. Phylogenetic analysis placed AjSOCS1 within the teleost clade, showing high conservation with other vertebrate SOCS1 homologs. AjSOCS1 was ubiquitously expressed across tissues, with the highest levels in intestine and im

BACKGROUND AND OBJECTIVE: Connective tissue growth factor (CTGF) is up-regulated in the lungs of patients with chronic obstructive pulmonary disease (COPD). Cigarette smoke and repeated bacterial infections, both of which are rich sources of LPS, are major causes of COPD. The high levels of LPS in lung epithelial lining fluid also suggest that it may have a considerable impact on the airway epithelium, in terms of cytokine and growth factor production. The aim of this study was to clarify the mechanism of LPS-induced CTGF expression in bronchial epithelial cells.
METHODS: The expression and t
