"Ears of the lynx" sign in a marchiafava-bignami patient: structural basis and fiber-tracking DTI contribution to the understanding of this imaging abnormality.

The "ears of the lynx" sign was previously reported as a neuroimaging finding observed in patients with autosomal recessive hereditary spastic paraplegia in association with a thin corpus callosum (ARHSP-TCC). We report a patient with a chronic form of Marchiafava-Bignami disease (MBD) that presented with this imaging feature. Diffusion tensor imaging (DTI) and fiber-tracking data support that this finding is a consequence of the structural derangement, which enlarges a preexisting border zone of the bundles of fibers from the corpus callosum (CC) genu to the forceps minor and anterior corona

Antibody Detection and Molecular Characterization of Toxoplasma gondii from Bobcats (Lynx rufus), Domestic Cats (Felis catus), and Wildlife from Minnesota, USA.

Little is known of the epidemiology of toxoplasmosis in Minnesota. Here, we evaluated Toxoplasma gondii infection in 50 wild bobcats (Lynx rufus) and 75 other animals on/near 10 cattle farms. Antibodies to T. gondii were assayed in serum samples or tissue fluids by the modified agglutination test (MAT, cut-off 1:25). Twenty nine of 50 bobcats and 15 of 41 wildlife trapped on the vicinity of 10 farms and nine of 16 adult domestic cats (Felis catus) and six of 14 domestic dogs resident on farms were seropositive. Toxoplasma gondii oocysts were not found in feces of any felid. Tissues of all sero

Spider toxins comprising disulfide-rich and linear amphipathic domains: a new class of molecules identified in the lynx spider Oxyopes takobius.

In addition to the conventional neurotoxins and cytotoxins, venom of the lynx spider Oxyopes takobius was found to contain two-domain modular toxins named spiderines: OtTx1a, 1b, 2a and 2b. These toxins show both insecticidal activity (a median lethal dose against flesh fly larvae of 75 μg·g(-1)) and potent antimicrobial effects (minimal inhibitory concentrations in the range 0.1-10 μm). Full sequences of the purified spiderines were established by a combination of Edman degradation, mass spectrometry and cDNA cloning. They are relatively large molecules (~ 110 residues, 12.0-12.5 kDa) and con

Developing biological resource banks as a supporting tool for wildlife reproduction and conservation The Iberian lynx bank as a model for other endangered species.

This work presents a Biological Resource Bank generated as a complementary supporting tool for the reproduction and the in situ and ex situ conservation of the Iberian lynx. In its design we prioritized the preservation of a maximum of the current genetic and biological diversity of the population, and the harmless collection of the samples. To provide future reproductive opportunities through any possible technique, we processed and cryopreserved germinal cells and tissues from dead animals, 7 males and 6 females, as well as somatic cells and tissues from 69 different individuals. This somati

Isolation and Genetic Characterization of Toxoplasma gondii from Black Bears (Ursus americanus), Bobcats (Lynx rufus), and Feral Cats (Felis catus) from Pennsylvania.

Toxoplasma gondii infects virtually all warm-blooded hosts worldwide. Recently, attention has been focused on the genetic diversity of the parasite to explain its pathogenicity in different hosts. It has been hypothesized that interaction between feral and domestic cycles of T. gondii may increase unusual genotypes in domestic cats and facilitate transmission of potentially more pathogenic genotypes to humans, domestic animals, and wildlife. In the present study, we tested black bear (Ursus americanus), bobcat (Lynx rufus), and feral cat (Felis catus) from the state of Pennsylvania for T. gond

Development and validation of a fecal PCR assay for Notoedres cati and application to notoedric mange cases in bobcats (Lynx rufus) in Northern California, USA.

Notoedric mange in felids is a devastating disease caused by a hypersensitivity reaction to the mite Notoedres cati. The burrowing of the mite causes intense pruritis resulting in self-mutilation, secondary bacterial infection, and often death of affected felids if left untreated. Our understanding of how notoedric mange is maintained in felid populations, and the true geographic extent of infestations, has been hampered because wild felids are elusive and, thus, traditional diagnostic methods are difficult to implement. To create a noninvasive diagnostic test, we developed and validated a nov

LYmphoid NeXt-Generation Sequencing (LYNX) Panel: A Comprehensive Capture-Based Sequencing Tool for the Analysis of Prognostic and Predictive Markers in Lymphoid Malignancies.

B-cell neoplasms represent a clinically heterogeneous group of hematologic malignancies with considerably diverse genomic architecture recently endorsed by next-generation sequencing (NGS) studies. Because multiple genetic defects have a potential or confirmed clinical impact, a tendency toward more comprehensive testing of diagnostic, prognostic, and predictive markers is desired. This study introduces the design, validation, and implementation of an integrative, custom-designed, capture-based NGS panel titled LYmphoid NeXt-generation sequencing (LYNX) for the analysis of standard and novel m

Pathology and Tissue Distribution of Highly Pathogenic Avian Influenza Virus Clade 2.3.4.4b in Free-Ranging Mountain Lions (Puma concolor) and Bobcats (Lynx rufus) from Colorado, USA.

During a 1-mo period in early 2023, four adult free-ranging mountain lions (Puma concolor) and two adult free-ranging bobcats (Lynx rufus) died or were euthanized during an outbreak of H5N1 highly pathogenic avian influenza virus (HPAIV) in Colorado, USA. All six animals tested positive for HPAIV clade 2.3.4.4b, and necropsy findings resembled those described by others for domestic cats and wild felids infected with HPAIV clade 2.3.4.4b in North America. The most consistent findings in the Colorado wild felids were necrotizing and lymphoplasmacytic meningoencephalitis (6/6) and interstitial to

Subcutaneous daratumumab plus carfilzomib and dexamethasone (D-Kd) versus carfilzomib and dexamethasone (Kd) in patients with relapsed/refractory multiple myeloma who received previous daratumumab treatment: LYNX study.

Daratumumab-based regimens demonstrate clinical efficacy in relapsed/refractory multiple myeloma (RRMM). As more patients receive frontline daratumumab-based therapy, evaluation of daratumumab retreatment is needed. In the phase 2 LYNX study (ClinicalTrials.gov Identifier: NCT03871829), 88 patients with RRMM who received 1-3 prior lines of therapy, one of which contained daratumumab, were randomized to receive subcutaneous daratumumab plus carfilzomib/dexamethasone (D-Kd; n = 44) or carfilzomib/dexamethasone (Kd; n = 44). The primary endpoint was the very good partial response or better (≥VGPR

Identification of mcr-13 in the Endangered Lynx pardinus, and mcr-1 in Sus scrofa, Expands the Diversity of Plasmid-Mediated Colistin Resistance Determinants of Escherichia coli from Wildlife.

Background: Plasmid-mediated colistin resistance threatens the effectiveness of one of the last-line antibiotics for treating multidrug-resistant Gram-negative infections. Although wildlife is increasingly recognized as a reservoir of mobile antimicrobial resistance genes, its role in the emergence and dissemination of novel plasmid-mediated colistin resistance (mcr) genes remains poorly understood. Objectives: This study aimed to characterize two distinct mcr-carrying E. coli isolates identified in Spanish wildlife. Methods: Among 807 faecal samples from healthy wild animals screened, two mcr

Forceps minor region signal abnormality "ears of the lynx": an early MRI finding in spastic paraparesis with thin corpus callosum and mutations in the spatacsin gene (SPG11) on chromosome 15.

BACKGROUND AND PURPOSE: A thin corpus callosum on magnetic resonance imaging (MRI) characterizes a type of autosomal recessive disorder with progressive spastic paraparesis and cognitive impairment. Known as Hereditary Spastic Paraparesis with Thin Corpus Callosum (HSP-TCC), it has been associated with mutations of the SPG11 gene. No other specific MRI findings have been reported. METHODS: We studied with MRI four patients from three families with HSP-TCC who had identified causal mutations in the SPG11 gene. RESULTS: In all individuals studied the region of the forceps minor of the corpus c

Detection of Trichinella spp. in free-ranging carnivores and wild boars in Switzerland.

Wolves (Canis lupus lupus) and lynx (Lynx lynx) are officially monitored species in Switzerland. Deceased individuals are subjected to post-mortem examination and collection of baseline health data. The procedure includes an assessment of different infectious agents, including Trichinella spp., the cause of a notifiable zoonotic infection. Between May 2009 and May 2023, a total of 100 wolves and 250 lynx were tested at the National Reference Laboratory for Trichinellosis by the artificial digestion method. Additionally, muscle samples from 8838 wild boars (Sus scrofa), 27 red foxes (Vulpes vul

Prevalence and molecular characterization of Strongyloides spp. in domestic and wild felids from Switzerland.

Strongyloides spp. infecting felids remain largely understudied. To date, four Strongyloides species have been reported in felids worldwide: S. felis, S. planiceps, S. stercoralis, and S. tumefaciens. Most epidemiological studies performed in felids identified the parasites only to genus level. This study aimed to determine the prevalence of Strongyloides spp. in domestic cats (Felis catus), free-ranging Eurasian lynx (Lynx lynx), and European wildcats (Felis silvestris) from Switzerland, while providing detailed morphological and molecular characterization of the parasites. A total of 490 int

Translocation effects on regional and local population viability and connectivity.

Translocations and reintroductions aim to improve the viability of isolated populations and promote connectivity for large carnivores. However, there is no established framework for assessing their success. We used the Eurasian lynx (Lynx lynx) in western and central Europe to assess the impact of translocations on the viability of six populations and their interconnectivity. Lynx populations are small and isolated and have low genetic diversity. Population reinforcements have been carried out, but their impact has not been determined. We devised a workflow to evaluate how releasing new indivi

New insights into the distribution of cardio-pulmonary nematodes in road-killed wild felids from Romania.

BACKGROUND: The population of wild felids is large and stable in Romania with many carnivore habitats being protected. Felids can be infected with a wide variety of cardio-pulmonary nematodes and can act as reservoirs of infection for domestic cats. The aim of this study was to evaluate the distribution and species diversity of cardio-pulmonary nematodes in wild felids from Romania. METHODS: A total of 54 wild felids (7 Lynx lynx and 47 Felis silvestris) were legally collected from different locations in Romania and analysed by complete parasitological necropsy. The entire respiratory tract w

Cortical parvalbumin and somatostatin GABA neurons express distinct endogenous modulators of nicotinic acetylcholine receptors.

BACKGROUND: Inhibition from GABAergic interneurons in brain circuits is a critical component of cognitive function. This inhibition is regulated through a diverse network of neuromodulation. A number of recent studies suggest that one of the major regulators of interneuron function is nicotinic acetylcholinergic transmission and dysregulation of both systems is common in psychiatric conditions. However, how nicotinic modulation impacts specific subpopulations of diverse GABAergic interneurons remains in question. One potential way of conferring specificity to the convergence of GABAergic and n

Point-of-care p24 antigen detection for early infant diagnosis of HIV infection: cross-sectional and longitudinal studies in Zambia.

BACKGROUND: Early infant diagnosis of HIV infection is challenging in sub-Saharan Africa, particularly in rural areas, leading to delays in diagnosis and treatment. Use of a point-of-care test would overcome many challenges. This study evaluated the validity of a novel point-of-care p24 antigen detection test (LYNX) in rural and urban settings in southern Zambia. METHODS: Two studies were conducted: a cross-sectional study from 2014 to 2015 at Macha Hospital (LYNX Hospital study) and a longitudinal study from 2016 to 2018 at 12 health facilities in Southern Province, Zambia (NSEBA study).
