Mansonella ozzardi and its vectors in the New World: an update with emphasis on the current situation in Haiti.

Mansonella ozzardi (Nematoda: Onchocercidae) is a little studied filarial nematode. This human parasite, transmitted by two families of dipteran vectors, biting midges (most of them members of the genus Culicoides) and blackflies (genus Simulium), is endemic to the Neotropical regions of the New World. With a patchy geographical distribution from southern Mexico to north-western Argentina, human infection with M. ozzardi is highly prevalent in some of the Caribbean islands, along riverine communities in the Amazon Basin, and on both sides of the border between Bolivia and Argentina. Mansonella ozzardi in Brazil: prevalence of infection in riverine communities in the Purus region, in the state of Amazonas.

This study was undertaken to investigate the prevalence of Mansonella ozzardi infection and to estimate the parasitic infection rate (PIR) in simuliid black flies in the municipality of Pauini, Amazonas, Brazil. We used thick blood films to examine 921 individuals in 35 riverine communities along the Pauini and Purus Rivers. Simuliids were caught in several communities. Flies were identified, stained with haematoxylin and dissected. Overall, 44 (24.86%) of 177 riverines were infected in communities on the Pauini River and 183 (24.19%) of 744 on the Purus. The prevalence was higher in men (31.8

Mansonella perstans filariasis in Uganda: patterns of microfilaraemia and clinical manifestations in two endemic communities.

Surveys for Mansonella perstans infection and potentially related clinical manifestations were undertaken in two endemic communities in Mukono and Luwero districts of Uganda where no other human filarial infections are transmitted. A sensitive and accurate counting chamber method was used for quantifying microfilaraemia in 100microl of finger-prick blood. Among 575 and 991 examined individuals aged >or=1 year in the two communities, the overall microfilariae (mf) prevalence was significantly higher in Mukono (76.5%) than in Luwero (57.7%). As early as age 1-4 years, 40.6% and 20.5% of the chil

Rapid assessment of the geographical distribution of Mansonella perstans infections in Uganda, by screening schoolchildren for microfilariae.

The geographical distribution of Mansonella perstans infections in Uganda was assessed by day-time examination of school-aged children for microfilariae. Overall, 12,207 children from 76 sites representing the various topographical and ecological zones in the country were examined. Children with M. perstans microfilaraemia were detected at 47 (61.8%) of the study sites, with prevalences ranging from 0.4% to 72.8%. A broad, east-west-oriented belt of high endemicity was identified, stretching across the central part of the country from the southern end of Lake Albert to the north-western shores

Prevalence of Mansonella perstans in western Uganda and its detection using the QBC-fluorescence method.

The prevalence of the apathogenic filaria Mansonella perstans was studied in four parishes in western Uganda as part of an onchocerciasis control programme to avoid futile treatment. Blood samples from 1543 persons aged over 14 years from 19 villages were examined for the presence of microfilariae using a modified Knott method. The prevalence of microfilaraemic persons ranged between the parishes from 39% (95% CI 35.9-42.0%) to 81% (95% CI 76.2-84.8%). With exception of single microfilariae of Onchocerca volvulus no other filaria species was detected. Onchocerciasis mass treatment campaigns di

Development of Mansonella ozzardi in Simulium amazonicum, S. argentiscutum, and Culicoides insinuatus from Amazonas, Colombia.

Simulium amazonicum, S. argentiscutum and Culicoides insinuatus were collected from the Amazon region of Colombia after having blood-fed on volunteers naturally infected with Mansonella ozzardi. Dissection of the specimens revealed that these species supported the development of the microfilaria of M. ozzardi to the infective stage. Infective larvae were obtained from Simulium within 7 days at temperatures ranging from 23-30 degrees C. S. argentiscutum ingested more microfilariae and had a correspondingly higher experimental infection rate. Three of 31 C. insinuatus dissected beyond day 5 post

Mansonella perstans, Onchocerca volvulus and Strongyloides stercoralis infections in rural populations in central and southern Togo.

BACKGROUND: Mansonella perstans, Onchocerca volvulus and Strongyloides stercoralis are widespread helminth parasites in the tropics. Their distribution remains difficult to determine as it may change during national disease control programs and with regional mass drug administration (MDA). Epidemiological surveys are of importance to evaluate the geographical distribution of these helminth parasites and the diseases they may cause, however, up to date epidemiological evaluations on M. perstans and S. stercoralis in Togo are rare, and surveys on O. volvulus are important especially under the as

Ecological Drivers of Mansonella perstans Infection in Uganda and Patterns of Co-endemicity with Lymphatic Filariasis and Malaria.

BACKGROUND: Mansonella perstans is a widespread, but relatively unknown human filarial parasite transmitted by Culicoides biting midges. Although it is found in many parts of sub-Saharan Africa, only few studies have been carried out to deepen the understanding of its ecology, epidemiology, and health consequences. Hence, knowledge about ecological drivers of the vector and parasite distribution, integral to develop spatially explicit models for disease prevention, control, and elimination strategies, is limited. METHODOLOGY: We analyzed data from a comprehensive nationwide survey of M. perst

Lack of evidence for transplacental transfer of microfilarial antigens in filariasis due to Loa loa and Mansonella perstans.

The transplacental transfer of microfilarial antigens could induce prenatal tolerance, or conversely resistance, which would influence the development of filariasis after birth. The passage across the placenta of immunogens constitutive of a crude microfilarial Loa loa extract has been investigated by dosing specific IgG, IgM and IgE antibodies in 92 maternal and corresponding cord blood samples, IgG levels were shown to be identical in about half of the mother-cord paired samples while in the other half the cord serum had a lower titre. Specific IgM or IgE could not be evidenced in any of the

Comparative development of human filariae Loa loa, Onchocerca volvulus and Mansonella perstans in immunocompromised mouse strains.

INTRODUCTION: Mouse models of human filarial infections are not only urgently needed to investigate the biology of the nematodes and their modulation of the host's immunity, but will also provide a platform to screen and test novel anti-filarial drugs. Recently, murine Loa loa infection models have been stablished using immunocompromised mouse strains, whereas murine Mansonella perstans infections have not been implemented until now. METHODS: Therefore, we aim to establish experimental M. perstans infections using the immunocompromised mouse strains RAG2IL-2Rγ-/- (lack B, T and natural killer

Culicoides Species of the Rain Forest Belt of the Littoral Region of Cameroon: Their Incrimination in the Transmission of Mansonella perstans.

Biting midges belonging to the genus Culicoides are tiny stout-shaped hematophagous insects and are thought to transmit the filarial nematode Mansonella perstans. Little is known about the Culicoides fauna in the rain forest belt of the Littoral Region of Cameroon. This study was designed to investigate the diversity, abundance and distribution of Culicoides spp. and their role as the purported vector(s) of M. perstans. Overnight light trap collections and human landing catches (HLCs) revealed eight species of Culicoides with C. grahamii being the most abundant species followed by C. milnei. F

Phase III Clinical Trial to Evaluate Ivermectin in the Reduction of Mansonella ozzardi infection in the Brazilian Amazon.

The treatment of mansonelliasis is still a challenge because there are few clinical trials for the treatment of the disease. This double-blind, randomized, placebo-controlled study (phase III clinical trial) was conducted to evaluate the effectiveness of a single oral dose of ivermectin (0.15 mg/kg) in the reduction of the Mansonella ozzardi microfilaraemia and the occurrence of adverse effects in infected people compared with the control group treated with placebo. A total of 49 microfilaraemic patients were randomly selected from the municipality of Lábrea, State of Amazonas, in the Brazilia

Historic accounts of Mansonella parasitaemias in the South Pacific and their relevance to lymphatic filariasis elimination efforts today.

There are two species of filarial parasites with sheathless microfilariae known to commonly cause parasitaemias in humans: Mansonella perstans and Mansonella ozzardi. In most contemporary accounts of the distribution of these parasites, neither is usually considered to occur anywhere in the Eastern Hemisphere. However, Sir Patrick Manson, who first described both parasite species, recorded the existence of sheathless sharp-tailed Mansonella ozzardi-like parasites occurring in the blood of natives from New Guinea in each and every version of his manual for tropical disease that he wrote before

Population dynamics of Loa loa and Mansonella perstans infections in individuals living in an endemic area of the Congo.

A follow-up of Loa loa and Mansonella perstans microfilaremia was carried out in an adult population living in a highly endemic area of the Congo. Infection rates and parasite loads were found to be stable in the general population, both in the short-term (two months) and long-term (3-4 years) followup. The microfilarial status of most of the subjects examined did not change between tests. At the individual level, the microfilarial densities of L. loa and M. perstans also remained remarkably constant over time. This results in a qualitative and quantitative stability of the parasitic material

Comparison of thick blood smear and saponin haemolysis for the detection of Loa loa and Mansonella perstans infections.

In an endemic area, Loa loa and Mansonella perstans microfilariae were detected by the examination of 5 ml blood in respectively 7.4 and 26.2% of subjects who would have been erroneously considered amicrofilaraemic by the conventional method of two thick blood smears (40 microliters blood). Correction factors to be applied to the results obtained with thick blood films in order to approach the true parasitological prevalences were 1.5 for L. loa and 1.6 for M. perstans. In addition, the analysis of a large volume of blood provides a better estimation of microfilarial load of the parasitized po

Filariasis in Amerindians of western Guyana with observations on transmission of Mansonella ozzardi by a Simulium species of the amazonicum group.

In order to conduct experimental infection studies of Mansonella ozzardi in local haematophagous Diptera, volunteers infected with this parasite were identified during a microfilaria survey in four Amerindian villages in the Pakaraima Mountains of western Guyana, near the Brazilian border. M. ozzardi microfilariae were detected in thick blood smears from 8-21% of persons examined. They were also found in skin snips from 8/73 persons all of whom were positive by blood smear examination. No Onchocerca volvulus microfilariae were detected. Dipetalonema perstans infections were found in three of t
