Measles virus persistence in human lymphocytes: a role for virus-induced interferon.

Because of the association of measles virus with persistent infections such as subacute sclerosing panencephalitis, and its possible involvement in multiple sclerosis, we have investigated the capacity of this virus to establish chronic infections in human peripheral blood lymphocytes (PBLs). We have demonstrated that a latent, persistent infection of human PBLs with measles virus results in low levels of infectious virus production in which large amounts of virus-induced interferon could be detected. Further, treatment of these silently infected cells with an anti-human leukocyte interferon s

Detection of Live Attenuated Measles Virus in the Respiratory Tract Following Subcutaneous Measles-Mumps-Rubella Vaccination.

The live attenuated measles vaccine is extremely effective in preventing measles and induces mucosal immunity in the respiratory tract; however, the mechanism is not known. We show that live attenuated measles virus (LAMV) RNA is frequently detected in the respiratory tract 7-21 days after subcutaneous measles-mumps-rubella (MMR) vaccination in healthy children (n = 5/20) and macaques (n = 6/8). Replicating LAMV was isolated from the lungs of 2 macaques, with no evidence of transmission to unvaccinated individuals. These observations suggest that LAMV in the respiratory tract may play a role i

Comparison of laboratory diagnostic methods for measles infection and identification of measles virus genotypes in Hong Kong.

The sensitivities of IgM detection, virus isolation, and RT-PCR for the diagnosis of measles infection were assessed using samples collected from confirmed measles cases from 2006 to 2009. The optimal timing of specimen collection and the preferred specimen type(s) for these tests were also determined. IgM detection showed highest sensitivity when serum samples were collected >or=5 days after rash onset. Virus isolation gave the highest sensitivity when samples were collected <or=3 days after rash onset, with nasopharyngeal aspirate being the best specimen type, followed by urine and throat/co

Variations in measles vaccine-specific humoral immunity by polymorphisms in SLAM and CD46 measles virus receptors.

BACKGROUND: Measles infection requires 2 cellular receptors, signaling lymphocyte activation molecule (SLAM) and CD46. Known and novel single nucleotide polymorphisms (SNPs) in SLAM and CD46 genes might influence the immune response to measles vaccine. OBJECTIVE: We sought to identify SNP associations in SLAM and CD46 genes with variations in measles antibody response. METHODS: We genotyped known SNPs in SLAM and CD46 genes in 339 subjects vaccinated with 2 doses of measles-mumps-rubella vaccine. We also sequenced the measles virus-binding domains of SLAM and CD46 to identify novel SNPs. RE

Antibody response to structural proteins of measles virus in patients with natural measles and subacute sclerosing panencephalitis.

By immunoprecipitation and SDS-polyacrylamide gel electrophoresis, antibody responses to the structural proteins of measles virus were examined on patients with various forms of natural measles, atypical measles, and subacute sclerosing panencephalitis (SSPE). The serum of atypical measles most strongly reacted with four structural proteins, i.e., hemagglutinin (H), nucleocapsid (NC), fusion (F), and matrix (M) proteins. In natural measles, antibodies to the four structural proteins were detected at such an early convalescent stage as one month after the onset of disease. In late convalescent

Growth of measles virus in nervous tissues. IV. Neurovirulence of wild measles and SSPE viruses in monkeys.

Neurovirulence of in vivo-passed wild measles virus and that of the cell-associated SSPE virus were compared by intracerebral inoculation into monkeys. The wild measles virus was found to lack neutrovirulence without producing neurological signs or significant histological changes in the brains, whereas the virus was confirmed to preserve the properties characteristic of the wild virus. In contrast, inoculation of the SSPE virus induced rapid onset of neurological signs with mild but definite histological changes including degeneration of nerve cells. The fact that SSPE virus exhibited neurovi

MeV-Stealth: A CD46-specific oncolytic measles virus resistant to neutralization by measles-immune human serum.

The frequent overexpression of CD46 in malignant tumors has provided a basis to use vaccine-lineage measles virus (MeV) as an oncolytic virotherapy platform. However, widespread measles seropositivity limits the systemic deployment of oncolytic MeV for the treatment of metastatic neoplasia. Here, we report the development of MeV-Stealth, a modified vaccine MeV strain that exhibits oncolytic properties and escapes antimeasles antibodies in vivo. We engineered this virus using homologous envelope glycoproteins from the closely-related but serologically non-cross reactive canine distemper virus (

Detection of measles antigen in conjunctival epithelial lesions staining by lissamine green during measles virus infection.

In a study in Kenya of corneal lesions occurring in children with measles virus infection, conjunctival lesions of a characteristic appearance were seen concomitant with the rash. The lesions became visible after staining by Lissamine green or Rose Bengal. In order to investigate whether these lesions were virus-induced, paired biopsies were performed on the conjunctiva of five measles patients. One biopsy was taken of a staining lesion; the other one, taken of nonstaining conjunctiva of the same eye, served as control. Measles-specific immunofluorescence on cryostate sections of the biopsies

Protective effect of measles virus inoculation on subacute sclerosing panencephalitis virus-infected mice.

The Biken strain of subacute sclerosing panencephalitis (SSPE) virus caused a fatal neurologic disease in adult mice after intracerebral inoculation. However, the mice were completely protected from the disease when a high dose of measles virus was given intracerebrally after the SSPE virus infection. The measles virus inoculation induced interferon production and immune responses. An experiment with athymic nude mice showed that interferon and anti-measles antibody were able to prolong the incubation period of the disease but not to protect the SSPE virus-infected nude mice from death. For co

Measles Virus Genotypes Causing Outbreaks in Tanzania, 2022-2024.

Globally, 24 measles virus genotypes have been detected, and these genotypes have been classified into eight clades based on 450 nucleotides of the C-terminal region of the nucleoprotein gene. Genotype B3 is predominant in Africa, but there are limited data from Tanzania since the introduction of the second dose of measles-containing vaccine in 2014. A total of 129 nasopharyngeal samples and corresponding sera were collected during measles outbreaks between 2022 and 2024. Viral RNA was extracted from nasopharyngeal swabs prior to RT-qPCR and sequencing of a 450-nucleotide segment of the nucleo

Slow clearance of measles virus RNA after acute infection.

BACKGROUND: Measles virus (MV) RNA was detected 1 month after hospitalization with measles in more than half of Zambian children but the duration of detectable RNA was not determined. OBJECTIVES: To characterize the time course of MV clearance and identify factors associated with presence of viral RNA at late times after clinical recovery from infection. STUDY DESIGN: Blood, urine and nasopharyngeal specimens from 49 Zambian children with laboratory-confirmed measles were collected a median of 100 days (range 65-118) after rash onset. Samples were assayed for MV nucleocapsid and hemagglutini

Immunosuppression caused by measles virus: role of viral proteins.

Measles virus (MV) causes transient but profound immunosuppression resulting in increased susceptibility to secondary bacterial and viral infections. Due to the development of these opportunistic infections, measles remains the leading vaccine-preventable cause of child death worldwide. Different immune abnormalities have been associated with measles, including disappearance of delayed-type hypersensitivity reactions, impaired lymphocyte and antigen-presenting cell functions, down-regulation of pro-inflammatory interleukin 12 production and altered interferon alpha/beta signalling pathways. [Bronchiolitis obliterans with organized pneumonia associated with measles virus].

Bronchiolitis obliterans with organizing pneumonia (BOOP) is a nonspecific form of pulmonary injury related to different etiologic agents. The disorder is not only limited to the alveoli but also affects to the alveolar ducts and distal bronchioles. This fact may differentiate this disorder of others with chronic inflammatory diffuse alveolar lesion as chronic eosinophilic pneumonia (CEP), extrinsic allergic alveolitis (EAA) or diffuse alveolar damage (DAD). It has been described an idiopathic form and other associated with connective tissue diseases, infections, organ's transplantation, drug

Defective measles virus in human subacute sclerosing panencephalitis brain.

Evidence is presented showing that the brain cells of patients with subacute sclerosing panencephalitis (SSPE) contain mutant measles (MV) genomes having the characteristics of 5' copy-back defective interfering (DI) RNAs. Using a polymerase chain reaction-based amplification specific for copy-back DIs, abundant, discrete cDNAs representing different-sized MV defective RNA species were generated from each SSPE brain. The defective genomes were cloned in two portions. The most common of these defective species were sequenced, confirming their MV genome origin and 5' copy-back nature. We deduced

Immunohistochemical evidence of measles virus antigens in active otosclerosis.

Despite intensive investigation, the cause of otosclerosis remains uncertain. Recent studies of Paget's disease of bone have revealed a possible viral origin. Because of similarities between otosclerosis and Paget's disease, we have pursued investigation of a possible viral cause of otosclerosis. Four temporal bone specimens from patients with otosclerosis, processed for immunohistochemistry, demonstrated positive specific reactivity with monoclonal antibodies to measles virus antigens using the indirect immunofluorescent and immunoperoxidase techniques. Reactivity was most intense in active f

Chronic, relapsing encephalomyelitis associated with experimental measles virus infection.

An experimental central nervous system (CNS) disease that bears some similarity to human multiple sclerosis and that is associated with infection by measles virus has been induced in hamsters. This disease has been described previously (Carrigan and Johnson: Proceedings of the National Academy of Sciences 77:4297-4300, 1980). At that time, it was believed that the disease was restricted to the spinal cords of affected animals. The present work demonstrates that the disease can also affect the brain, describes in more complete detail the types of clinical disease that occur, illustrates the his

Measles virus nucleotide sequences: detection by hybridization in situ.

A tritium-labeled probe that detects measles virus nucleotide sequences was hybridized in situ to cells infected with measles virus and to sections of brain tissue from patients with subacute sclerosing panencephalitis and from patients with multiple sclerosis. The measles virus genome was detected in many cells in subacute sclerosing panencephalitis where this virus would have been missed by methods such as immunofluorescence. Measles virus sequences were also found in two foci in one of four cases of multiple sclerosis. This refined method of hybridization in situ, which can be useful in the

Antibodies against measles virus polypeptides in different disease conditions.

The occurrence of antibodies to the nucleoprotein and matrix (M) antigens of measles virus was determined in early and late measles convalescent sera and in sera from patients with multiple sclerosis, subacute sclerosing panencephalitis, chronic active hepatitis, and atypical measles. Antibodies to the two components were identified separately in serially diluted samples both by radioimmune precipitation assays and by complement fixation tests employing purified nucleoprotein and M components as antigens. The antibody response to M antigen in connection with acute infections was weak, and with
