From mechanical triggering to metabolic-inflammatory driving: a new paradigm of knee osteoarthritis pathogenesis.

BACKGROUND: Knee osteoarthritis (KOA) is a global public health crisis and a leading cause of disability among middle-aged and elderly populations. Historically characterized as a passive "wear-and-tear" process, the understanding of KOA has undergone a fundamental paradigm shift. While aberrant biomechanical loading remains a primary initiator of joint damage, it is increasingly recognized that systemic metabolic dysfunction and chronic low-grade inflammation act as the critical forces driving sustained disease progression. MAIN BODY: Abnormal mechanical loading is the primary initiator of K

Periodic Fracture of Active Tissues.

Fracture typically signifies mechanical failure in engineering materials, whereas controlled cracking may actively sculpt tissues through precise biological regulation. Here, we establish a multiscale nonlinear peridynamic theory that accounts for cellular mechanosensing to decipher the spontaneous fracture of active tissues. We show that tissues cultured in a ring-shaped domain can undergo periodic fracture to generate multicellular aggregates with regular spacing, recapitulating prior morphogenetic experiments on avian dermal cell collectives. It is found that the number of cracks varies non

Mechanical sensing of fluid shear stress biphasically regulates chondrocyte metabolism and osteoarthritis progression: Implications for therapeutics.

Articular cartilage homeostasis relies on chondrocytes to maintain extracellular matrix (ECM) integrity, with fluid shear stress (FSS) emerging as a critical regulator of chondrocyte function. Osteoarthritis (OA), a leading cause of global disability, is driven by abnormal mechanical loading, yet the dual role of FSS in joint health and disease remains incompletely defined, and existing reviews have not fully integrated its biphasic dose-effect boundaries, mechanotransduction networks, and translational pathways within a unified framework. To address this gap, we synthesize evidence that FSS e

The Frontiers of Orthodontically Induced Inflammatory Root Resorption: Mechanism, Treatment, and Future Prospects.

INTRODUCTION: Orthodontically induced inflammatory root resorption (OIIRR) is a frequent complication of orthodontic treatment, featured by sterile inflammatory destruction of root hard tissues, which arises from the crosstalk between biomechanical forces and molecular pathways in the periodontal microenvironment. METHODS: A literature review was conducted to analyze the key mechanisms, biomarkers, and potential interventions involved in OIIRR. RESULTS: Multiple factors contribute to OIIRR, including inflammatory signaling (interleukin [IL]-6, IL-1β, IL-18), mechanotransduction (Piezo1, sphi

Myosin 7a is required for maintaining the transducing stereocilia and for force transmission to the MET channel during cochlear hair cell development.

Shaker-1 mice carry a spontaneous missense mutation in Myo7a (Myo7aSh1) that interferes with the motor function of MYO7A. Mutation in the orthologous gene in humans causes syndromic (Usher 1B) or non-syndromic forms of deafness. In hair cells, MYO7A is expressed throughout the stereocilia, where it has been implicated in tip-link tensioning required for gating the mechanoelectrical transducer (MET) channel and setting its resting open probability (Po). The Myo7aSh1 mutation progressively dysregulated the height of shorter stereocilia rows from the end of the first postnatal week onwards, assoc

The Mechano-Immune-Vascular Activation Loop: An Integrative Conceptual Framework for Positive Feedback in Hypertension.

Hypertension is increasingly recognized as involving chronic inflammation beyond its hemodynamic origins. This review proposes the Mechano-Immune-Vascular Activation Loop as an integrative conceptual framework-a positive feedback circuit in which aberrant mechanical forces in the hypertensive vascular wall may activate immune cells through mechanosensitive sensors, potentially triggering inflammation that drives vascular stiffening and amplifies mechanical stress. We examine molecular force sensors including piezo-type mechanosensitive ion channel component 1 (Piezo1), transient receptor poten

Primary cilia as mechanosensors in musculoskeletal homeostasis and disease.

The musculoskeletal system relies on precise mechanical signal transduction to maintain physiological homeostasis, yet the underlying mechanisms remain incompletely characterized. Primary cilia (PC), membrane-bound organelles that protrude into the extracellular matrix, acting as cellular sensors to integrate biomechanical forces into cellular responses. Moreover, their dynamic assembly and disassembly are tightly coupled to mechanical stress perception, which also suggests a pivotal role in regulating musculoskeletal tissue function. This review systematically synthesizes current evidence on

Mechanosensory and command contributions to the Drosophila grooming sequence.

Flies groom in response to competing mechanosensory cues in an anterior-to-posterior order using specific legs. From behavior screens, we identified a pair of cholinergic command-like neurons, Mago-no-Te (MGT), whose optogenetic activation elicits thoracic grooming by the back legs. Thoracic grooming is typically composed of body sweeps and leg rubs in alternation, but clonal analysis coupled with amputation experiments revealed that MGT activation only commands the body sweeps: initiation of leg rubbing requires contact between the leg and thorax. With new electron microscopy (EM) connectome

Squishy matters - Corneal mechanobiology in health and disease.

The cornea, as a dynamic and responsive tissue, constantly interacts with mechanical forces in order to maintain its structural integrity, barrier function, transparency and refractive power. Cells within the cornea sense and respond to various mechanical forces that fundamentally regulate their morphology and fate in development, homeostasis and pathophysiology. Corneal cells also dynamically regulate their extracellular matrix (ECM) with ensuing cell-ECM crosstalk as the matrix serves as a dynamic signaling reservoir providing biophysical and biochemical cues to corneal cells. Here we provid

Biomechanical properties of ovarian tissue and their impact on the activation of follicular growth: a narrative review.

Follicular recruitment is tightly regulated to ensure long-term balance between the pools of dormant and growing follicles. While the growth of secondary to antral follicles is well understood, the initiation of folliculogenesis remains elusive. Several processes have been described, and a new approach is mechanotransduction. The aim of this review is to present the latest findings on the biomechanical properties of the ovary, and their role during the initiation of folliculogenesis. A search of PubMed using keywords related to the biomechanical properties of ovarian tissue and ovarian mechano

Asymmetric mechanotransduction by hair cells of the zebrafish lateral line.

In the lateral line system, water motion is detected by neuromast organs, fundamental units that are arrayed on a fish's surface. Each neuromast contains hair cells, specialized mechanoreceptors that convert mechanical stimuli, in the form of water movement, into electrical signals. The orientation of hair cells' mechanosensitive structures ensures that the opening of mechanically gated channels is maximal when deflected in a single direction. In each neuromast organ, hair cells have two opposing orientations, enabling bi-directional detection of water movement. Interestingly, Tmc2b and Tmc2a

Prolonged Dexamethasone Exposure Enhances Zebrafish Lateral-Line Regeneration But Disrupts Mitochondrial Homeostasis and Hair Cell Function.

The synthetic glucocorticoid dexamethasone is commonly used to treat inner ear disorders. Previous work in larval zebrafish has shown that dexamethasone treatment enhances hair cell regeneration, yet dexamethasone has also been shown to inhibit regeneration of peripheral nerves after lesion. We therefore used the zebrafish model to determine the impact of dexamethasone treatment on lateral-line hair cells and primary afferents. To explore dexamethasone in the context of regeneration, we used copper sulfate (CuSO4) to induce hair cell loss and retraction of nerve terminals, and then allowed ani

Targeting cell-matrix interface mechanobiology by integrating AFM with fluorescence microscopy.

Mechanosensing at the interface of a cell and its surrounding microenvironment is an essential driving force of physiological processes. Understanding molecular activities at the cell-matrix interface has the potential to provide novel targets for improving tissue regeneration and early disease intervention. In the past few decades, the advancement of atomic force microscopy (AFM) has offered a unique platform for probing mechanobiology at this crucial microdomain. In this review, we describe key advances under this topic through the use of an integrated system of AFM (as a biomechanical testi

Long-range mechanical signaling in biological systems.

Cells can respond to signals generated by other cells that are remarkably far away. Studies from at least the 1920's showed that cells move toward each other when the distance between them is on the order of a millimeter, which is many times the cell diameter. Chemical signals generated by molecules diffusing from the cell surface would move too slowly and dissipate too fast to account for these effects, suggesting that they might be physical rather than biochemical. The non-linear elastic responses of sparsely connected networks of stiff or semiflexible filament such as those that form the ex

Sensory Innervation of the Larynx and the Search for Mucosal Mechanoreceptors.

Purpose The larynx is a uniquely situated organ, juxtaposed between the gastrointestinal and respiratory tracts, and endures considerable immunological challenges while providing reflexogenic responses via putative mucosal mechanoreceptor afferents. Laryngeal afferents mediate precise monitoring of sensory events by relay to the internal branch of the superior laryngeal nerve (iSLN). Exposure to a variety of stimuli (e.g., mechanical, chemical, thermal) at the mucosa-airway interface has likely evolved a diverse array of specialized sensory afferents for rapid laryngeal control. Accordingly, m

Disease-associated keratin mutations reduce traction forces and compromise adhesion and collective migration.

Keratin intermediate filament (IF) proteins constitute the major cytoskeletal components in epithelial cells. Missense mutations in keratin 5 (K5; also known as KRT5) or keratin 14 (K14; also known as KRT14), highly expressed in the basal epidermis, cause the severe skin blistering disease epidermolysis bullosa simplex (EBS). EBS-associated mutations disrupt keratin networks and change keratinocyte mechanics; however, molecular mechanisms by which mutations shape EBS pathology remain incompletely understood. Here, we demonstrate that, in contrast to keratin-deficient keratinocytes, cells expre

Visceral pain from colon and rectum: the mechanotransduction and biomechanics.

Visceral pain is the cardinal symptom of functional gastrointestinal (GI) disorders such as the irritable bowel syndrome (IBS) and the leading cause of patients' visit to gastroenterologists. IBS-related visceral pain usually arises from the distal colon and rectum (colorectum), an intraluminal environment that differs greatly from environment outside the body in chemical, biological, thermal, and mechanical conditions. Accordingly, visceral pain is different from cutaneous pain in several key psychophysical characteristics, which likely underlies the unsatisfactory management of visceral pain

Mechanosensing and Mechanoregulation of Endothelial Cell Functions.

Vascular endothelial cells (ECs) form a semiselective barrier for macromolecules and cell elements regulated by dynamic interactions between cytoskeletal elements and cell adhesion complexes. ECs also participate in many other vital processes including innate immune reactions, vascular repair, secretion, and metabolism of bioactive molecules. Moreover, vascular ECs represent a unique cell type exposed to continuous, time-dependent mechanical forces: different patterns of shear stress imposed by blood flow in macrovasculature and by rolling blood cells in the microvasculature; circumferential c

Transient receptor potential channels and mechanosensation.

Transient receptor potential (TRP) channels act as sensors for a range of stimuli as diverse as light, sound, touch, pheromones, and tissue damage. Their role in mechanosensation in the animal kingdom, identified by gene ablation studies, has raised questions about whether they are directly mechanically gated, whether they act alone or in concert with other channels to transduce mechanical stimuli, and their relative importance in various functions and disease states in humans. The ability of these channels to form heteromultimers and interact with other ion channels underlies a range of cell-
