The unilateral 6-hydroxydopamine mouse lesion models of Parkinson's disease have received increasing attention in recent years, but comparison of the different lesion models was largely focused at a histological level. An extensive behavioural comparison between different mouse models on tests of motor function has yet to be carried out, to pin point tests that accurately discriminate between different extents of dopaminergic depletion. In the present study we examine the consequences of injection of the toxin at three sites along the nigrostriatal tract (substantia nigra, medial forebrain bun

The short-term effects of bilateral electrical stimulation of the lateral component of the lateral hypothalamic/medial forebrain bundle area (LHA) on peripheral plasma levels of glucose (G), immunoreactive insulin (IRI) and glucagon (IRG) were measured in food-derived, anesthetized rats in the presence of continuous low i.v. glucose infusion. In normal rats LHA stimulation induced a rapid but moderate rise in IRI in the presence of a simultaneous rapid rise in G. At the end of the 5 min stimulation period the glucose-induced IRI response was clearly attenuated. In rats receiving a continuous p

Stimulation of carotid body chemoreceptors activates putative vasopressin neurons in the supraoptic nucleus, an effect which has been abolished by lesions in the caudal ventrolateral medulla. Stimulation within the A1 catecholamine cell group in the ventrolateral medulla also activates supraoptic neurons and releases vasopressin. Therefore the A1 catecholamine neurons may be the means by which carotid body chemoreceptors influence the supraoptic nucleus and other parts of the forebrain. To test this possibility the influence of carotid body chemoreceptors on the discharge of rostrally-projecti

The thalamic reticular nucleus (TRN) is a critical structure influencing information transfer to the forebrain. In crocodilians, the TRN shares many features with its mammalian counterpart. One area that has not been explored is how individual neurons in the crocodilian TRN compare with those found in mammals. In mammals, TRN neurons are aligned parallel to the external border of the dorsal thalamus, have their dendrites oriented perpendicular to the fibers in the internal capsule, have fine, filamentous dendritic appendages, are either bipolar or multipolar, and are commonly considered to be

Studies on the genetic basis of addiction indicate that the tendency to develop alcoholism is inherited. In addition, alcoholism appears to be associated with a specific neurochemical disorder. Research has focused on the mesolimbic system, which is associated with the ability to feel pleasure (i.e. hypothalamic control centres are related to daily survival activities, and the medial forebrain bundle is involved in the positive reinforcement of addictive drugs). Current findings support the hypothesis that a neurochemical deficiency causes alcohol-dependent individuals to drink. Thus, pharmaco

All sectors of the human cerebral cortex receive dense cholinergic input. The origin of this projection is located in the Ch4 cell group of the nucleus basalis of Meynert. However, very little is known about the location of the pathways which link the cholinergic neurons of the nucleus basalis to the human cerebral cortex. This question was addressed in whole-hemisphere sections processed for the visualization of multiple cholinergic markers. Two highly organized and discrete bundles of cholinergic fibres extended from the nucleus basalis to the cerebral cortex and amygdala and were designated

The normal morphology of the rat preoptic region has been briefly described on the basis of Nissl- and silver-stained preparations and its efferent connections have been studied autoradiographically in over 50 rat brains with single small injections of 3H-proline, or various mixtures of 3H-proline, 3H-leucine, and 3H-lysine. Injections in the anteroventral part of the lateral preoptic area labeled fibers projecting through, and perhaps to, the anterior and lateral hypothalamic areas and ending in the supramammillary region, and ventral fiber lamina of the mammillary complex; other labeled fibe

The use of differential pulse voltammetry, performed with electrochemically treated carbon fiber electrodes, enables us to detect in vitro or in vivo, in the striatum of anaesthetized rats, an oxidation peak (3) at a potential of + 300 mV. Electrolytic or 5-7-dihydroxytryptamine lesions of the medial forebrain bundle are followed by a decrease of 59 and 62% respectively of this peak. Biochemical measurements are significantly correlated with the measured peak (3) and decreases. Thus, peak (3) increases obtained after injection of L-tryptophan and/or Reserpine, as well as the decreases observed

Intrastriatal implantation with dopaminergic of nondopaminergic tissue can elicit behavioral recovery in parkinsonian animals. Because in these animals, especially in 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine-lesioned monkeys, there are still considerable numbers of dopaminergic neurons left in the mesencephalon, implantation-induced trophic effects on host residual dopaminergic neurons have been suggested as a mechanism underlying the behavioral recovery. Gliosis around the graft is a universal finding in any implantation procedure and is probably mediated by interleukin-1 (IL-1); in addit

Deep Brain Stimulation (DBS) is a therapeutic option for treatment resistant (TR) obsessive-compulsive disorder (OCD). The OCD network comprises different sub-networks with homeostatic functions, altered under disease and modifiable with DBS. Connectomic analyses of DBS data sets have defined fiber selections explaining anti-OCD efficacy. This is a retrospective stimulation and outcome derived anatomical overlay analysis of 26 TR-OCD patients who received DBS at two academic centers. Using a reinstatement procedure, it has been shown that intermittent footshock stress reliably reinstates extinguished drug-taking behaviour in rats. Here we studied the role of noradrenaline (NE), one of the main brain neurotransmitters involved in responses to stress, in reinstatement of heroin seeking. We first determined the effect of clonidine, an alpha-2 adrenergic receptor agonist that decreases NE cell firing and release, on stress-induced reinstatement of heroin seeking. Rats were trained to self-administer heroin (0.1 mg/kg per infusion, IV, three 3-h sessions per day) for 9-10 days.

Injections of kainic acid (KA) into the lateral hypothalamus (LH) produce neuronal loss in this region without apparent damage to medial forebrain bundle fibers passing through the area. Cellular destruction is not limited to the LH; the neuronal loss in the thalamic reticular nucleus, the subthalamic nucleus and zona incerta is more extensive than that in the LH. Since all of the nuclei of ventral thalamic origin except the ventral lateral geniculate nucleus (VLGN) were destroyed by LH injections of KA, we sought to determine whether this nucleus also is sensitive to KA. Injections directly i

The prototype 5-HT2A receptor agonist hallucinogens LSD, mescaline, and psilocybin are classified as Schedule 1 drugs of abuse by the U.S. Drug Enforcement Administration. Accumulating clinical evidence has also suggested that acute or repeated "microdosing" with these drugs may have utility for treatment of some mental health disorders, including drug abuse and depression. The goal of the present study was to evaluate LSD, mescaline, and psilocybin effects on intracranial self-stimulation (ICSS), a procedure that has been used to evaluate abuse-related effects of other classes of abused drugs

Agenesis of the corpus callosum (ACC) is a relatively common brain abnormality resulting from developmental defects either limited to the structures leading to the proper formation of the corpus callosum or involving the embryo forebrain more generally. ACC is genetically heterogeneous with autosomal dominant, autosomal recessive, and X linked inheritance and has also been reported in subjects with aneuploidies involving several chromosomes. Among them, distal 6q deletions have been consistently reported in association with ACC, suggesting that there is a gene in the deleted region whose haplo

Heart rate (HR) can be voluntarily regulated when individuals receive real-time feedback. In a rat model of HR biofeedback, the neocortex and medial forebrain bundle were stimulated as feedback and reward, respectively. The rats reduced their HR within 30 minutes, achieving a reduction of approximately 50% after 5 days of 3-hour feedback. The reduced HR persisted for at least 10 days after training while the rats exhibited anxiolytic behavior and an elevation in blood erythrocyte count. This bradycardia was prevented by inactivating anterior cingulate cortical (ACC) neurons projecting to the v

Growth/differentiation factor 5 (GDF5), a novel member of the transforming growth factor beta superfamily, promotes the survival of dopaminergic neurones in vitro. We present here the first evidence for a neuroprotective action of GDF5 in vivo. We investigated the effects of intracerebral administration of GDF5 on a rat model of Parkinson's disease. GDF5 was administered just above the substantia nigra and into the lateral ventricle immediately before ipsilateral injection of 6-hydroxydopamine into the medial forebrain bundle. GDF5 prevented the development of amphetamine-induced rotations and

1. Male Sprague-Dawley rats received either a unilateral injection of 6-hydroxy-dopamine or vehicle injection into the medial forebrain bundle. 2. Two weeks post surgery, all rats received a pemoline challenge (250 mg/kg s.c.), and rotational and stereotyped behaviors were videotaped and analyzed. 3. All rats regardless of injection expressed stereotyped behaviors and hyper-locomotion after pemoline challenge. 4. High performance liquid chromatography (HPLC) with electrochemical detection was used to evaluate changes in the levels of dopamine, serotonin and their metabolites in neostriata. 5.

Twenty-six embryos (6-11 mm) of stage 15 (approximately 33 days) were studied in detail and graphic reconstructions of three of them were prepared. Characteristic features of this stage include closed lens vesicles, presence of nasal pits, and retinal pigment. The neuromeric pattern is still visible. Each cerebral hemisphere is limited by the torus hemisphericus internally and by the di-telencephalic sulcus externally. The medial (diencephalic) eminence of the basal nuclei (previously misinterpreted by others as the lateral) had appeared in stage 14, and the lateral eminence, which is telencep

Deep brain stimulation (DBS) is a promising therapeutic modality for managing treatment-resistant depression. Most DBS research has focused on single brain regions resulting in unclear optimal stimulation targets and vague mechanisms. Here, we introduce an experimental paradigm in which multiple graphene fiber stimulating electrodes were implanted in various brain regions of the same depressive animal for behavioral testing and DBS-functional MRI studies. We observed an instantaneous alleviation of depressive-like symptoms with a high response rate in Wistar-Kyoto rats following DBS at the med

BACKGROUND: Non-motor symptoms such as appetite loss contribute to weight and fat mass reduction in people living with Amyotrophic Lateral Sclerosis (plwALS), both of which are strong prognostic factors in the disease. Consequently, understanding the neural mechanisms underlying appetite and other non-motor disturbances in ALS is of significant clinical concern. Previous studies highlight widespread grey and white matter involvement beyond the motor system, including hypothalamic volume loss and functional changes in reward-related regions. However, it remains unclear whether structural altera
