Diagnosis of essential thrombocythemia (ET) is controversial and remains mainly an exclusion diagnosis. Endogenous megakaryocyte colony (EMC) formation have been largely evaluated to identify specific criteria for ET, but results are impeded by the lack of medium standardization. We evaluated megakaryocyte (MK) colony formation in a serum-free collagen-based medium, without cytokine and in the presence of various concentrations of thrombopoietin (TPO). Thirty-six bone marrows from patients diagnosed with ET (n = 11), polycythemia vera (PV; n = 12), reactive thrombocytosis (RT; n = 6) and healt

Circulating megakaryocytic cells abound in chronic myeloid leukemia (CML) seen in India and uniquely provide a setting for observing megakaryocytic maturation in the peripheral blood, a milieu not native to megakaryocytes. Peripheral blood megakaryocytic cells were studied in 324 cases of CML (235 chronic, 65 accelerated and 24 blastic phases). Two maturation themes were evident. Megakaryocytic blasts, especially in some cases of blast crisis, precociously make a foray into platelet formation and end up producing huge agranular or poorly granular cytoplasmic lobulated masses, that break off an

The generation of megakaryocytes (MKs) from human somatic cells through chemical reprogramming represents a promising strategy for developing alternative platelet sources. Building on our prior chemical reprogramming protocol for converting erythroblasts to MKs, we established a robust method that successfully generated induced MKs (iMKs) from human cord blood-derived CD3+ T cells, which is a more abundant source. This method used a five-small molecule cocktail containing a reprogramming booster, AZD4205, to promote erasure of T cell identity and facilitate fate transition toward MKs. T cell-d

Induced pluripotent stem cell-derived immortalized megakaryocyte cell lines (imMKCLs) exhibit substantial heterogeneity, which poses a major barrier to efficient ex vivo production of platelet-like particles (PLPs). In this study, we combined proteomic profiling with principal component analysis to uncover regulatory factors influencing imMKCL maturation. Through correlation-based omics screening, prohibitin 2 (PHB2) was identified as a putative inhibitory node, and JI051, a small molecule previously reported to bind PHB2, was found to enhance PLP production. JI051 accelerated proplatelet-like

Despite significant progress in the cultivation of hematopoietic stem cells (HSCs), the establishment of lineage-specific cell culture systems remains inadequately developed. This study compares the effects of recombinant human serum albumin (r-HSA) and polyvinyl alcohol (PVA) in serum-free culture systems on the differentiation of HSCs into multiple lineages. Both single-cell and multi-cellular culture systems are used, and differentiation is evaluated by flow cytometry and slides-based techniques under various cytokine conditions. Our results show that r-HSA strongly promotes differentiation

Megakaryocytes (MKs) are specialized hematopoietic cells long recognized for their ability to produce platelets. Increasing evidence now highlights MKs as multifunctional immune effectors that bridge hematopoiesis with host immunity. In the bone marrow (BM), MKs arise through thrombopoietin (TPO)-mediated differentiation of hematopoietic stem cells (HSCs) and show substantial heterogeneity, with discrete subsets specialized for platelet production (thrombopoiesis), HSC niche maintenance, or immune modulation. Outside the BM, MKs in the lungs and spleen perform tissue-specific immune functions,

Autoantibodies targeting platelet surface glycoproteins (GP) are the primary cause of immune thrombocytopenia (ITP), however testing for these autoantibodies is not routinely employed mainly because current diagnostic tests lack sufficient sensitivity. To overcome this and other diagnostic limitations, we generated HLA class I-negative, blood group O, induced pluripotent stem cell lines, and gene-edited them to produce a novel panel of GP-deficient, in vitro-derived megakaryocytes (MK). Using GPIIb-, GPIbα- and GPIX-deficient MK allowed sensitive and specific identification and characterizatio

BACKGROUND: Adenoviruses, adenoviral vector vaccines, and gene delivery systems have been implicated in thrombosis/thrombocytopenia syndrome. The underlying cause remains unknown. We have previously hypothesized that infection of megakaryocytes (MKs), the progenitor cells of platelets, plays a central role in adenovirus- or adenoviral vector-induced thrombosis/thrombocytopenia. Further, recent work has highlighted that the MK population comprises multiple subsets, including cells with immune function, and we propose that this MK subset is involved in adenoviral infection and the subsequent res

The authors investigated the impact of antiplatelet therapy on the megakaryocyte (MK) and platelet transcriptome. RNA-sequencing was performed on MKs treated with aspirin or P2Y12 inhibitor, platelets from healthy volunteers receiving aspirin or P2Y12 inhibition, and platelets from patients with systemic lupus erythematosus (SLE). P2Y12 inhibition reduced gene expression and inflammatory pathways in MKs and platelets. In SLE, the interferon (IFN) pathway was elevated. In vitro experiments demonstrated the role of P2Y12 inhibition in reducing IFNα-induced platelet-leukocyte interactions and IFN

Myeloproliferative neoplasms (MPNs) are a heterogeneous group of chronic hematological diseases that arise from the clonal expansion of abnormal hematopoietic stem cells, of which polycythemia vera (PV), essential thrombocythemia (ET), and primary myelofibrosis (PMF) have been extensively reviewed in the context of control of clonal expansion, fibrosis, and other phenotypes. Herein, we review current knowledge on the influence of different forms of MPN on bone health. In studies, murine models and human data have implicated various degrees of effect of different forms of MPN on bone density an

Calreticulin (CALR) mutations are driver mutations in myeloproliferative neoplasms (MPNs), leading to activation of the thrombopoietin receptor and causing abnormal megakaryopoiesis. Here, we generated patient-derived CALRins5- or CALRdel52-positive induced pluripotent stem cells (iPSCs) to establish an MPN disease model for molecular and mechanistic studies. We demonstrated myeloperoxidase deficiency in granulocytic cells derived from homozygous CALR mutant iPSCs, rescued by repairing the mutation using CRISPR/Cas9. iPSC-derived megakaryocytes showed characteristics of primary megakaryocytes

Activation of innate receptors in megakaryocytes (MKs) may affect the ability to produce functional platelets. Low platelet count is one of the clinical manifestations of dengue virus (DENV) infection. In MKs, the effect of innate receptors during DENV-infection is not well studied. Here we used MEG-01 cells to investigate DENV serotype 2 induced innate receptors in these cells. DENV RNA was estimated by qRT-PCR in the culture supernatant. The expression of innate receptors was determined by western blot and qPCR. DENV infection led to increased expression of RIG-I at 24 hrs post-infection (hp

BACKGROUND/AIMS: Salicylate causes drug-induced immune thrombocytopenia. However, some clinical studies indicate the presence of additional mechanisms in the drug-induced thrombocytopenia, by which the platelet production from megakaryocytes may directly be affected. Since salicylate is amphiphilic and preferentially partitioned into the lipid bilayers of the plasma membrane, it can induce some structural changes in the megakaryocyte membrane surface and thus affect the process of thrombopoiesis.
METHODS: Employing the standard patch-clamp whole-cell recording technique, we examined the effec

Several types of large atypical epithelioid cells can mimic tumoral melanocytes and can therefore be a source of misdiagnosis of melanoma. Megakaryocytes are one of these types, and are not frequently mentioned in the literature. In the current report, we present the case of a 76-year-old man presenting with acute panmyelosis with myelofibrosis. The bone marrow biopsy contained atypical megakaryocytes that expressed S100 in the cytoplasm. We discuss how such aberrant expression could be a source of some diagnostic problems in dermatopathology, including cutaneous melanoma, metastasis of melano

An elevated Ki-67 proliferation index is a known marker of adverse prognosis in several solid and hematological tumors, but its role in Myelodysplastic Syndromes (MDS) remains underexplored. This study aimed to quantify Ki-67 expression across hematopoietic lineages in bone marrow biopsies (BMB) from MDS patients and assess its correlation with clinical features and outcomes. We performed single and double immunohistochemistry for Ki-67 with CD34, CD71, MPO, and Factor VIII on BMB samples from 73 patients diagnosed with MDS or oligoblastic acute myeloid leukemia (AML) at a single center. The c

Megakaryocytes (MKs) are polyploid cells that maintain bone marrow homeostasis by secreting cytokines, including transforming growth factor β1 (TGFβ1). During neoplastic transformation, MKs accumulate in the bone marrow, promoting fibrotic remodeling that leads to myelofibrosis. However, the therapeutic potential of targeting MK cytokine secretion remains poorly understood. Because unconventional secretion of TGFβ1 and interleukin 1β (IL1β) via secretory autophagy occurs in other cell types, we investigated whether MKs may utilize the same mechanism. Disrupting secretory autophagy, or inhibiti
