Short-Term Impact of Betaine Supplementation on Ruminal Microbial Relative Abundance, Nutrient Digestibility, Serum Metabolites, and Milk Composition in Heat-Stressed Dairy Cows.

This short-term study evaluated the effects of dietary betaine supplementation over 4 weeks on feed intake, milk production, nutrient digestibility, blood metabolites, and ruminal microbial communities in heat-stressed Holstein cows. Thirty lactating cows were randomly divided into a control group and a treatment group supplemented with 80 g/day of natural betaine extract for 4 weeks during summer. Betaine supplementation significantly increased dry matter intake (21.0 vs. 18.7 kg/day; p ≤ 0.05) and milk yield (35.4 vs. 31.2 kg/day; p ≤ 0.05) compared to the control. Milk composition, includin

Pre-Diagnostic Saliva Microbiota of School-Aged Children Who Developed Type 1 Diabetes or Inflammatory Bowel Diseases.

Altered commensal microbiota composition has been associated with pediatric type 1 diabetes mellitus (T1D) and inflammatory bowel diseases (IBD), but the causal relationship is still unclear. To search for potential pre-diagnostic biomarkers for pediatric T1D or IBD, we compared microbiota in saliva samples in a nested case-control design comprising children developing T1D (nchildren = 52) or IBD (nchildren = 21) and controls with a similar age, sex, and residential area (nchildren = 79). The pre-diagnostic saliva microbiota alpha- and beta-diversity of children who would develop T1D (nsamples

Perturbations of gut microbiome and metabolome of pigs infected with Mycoplasma hyorhinis.

BACKGROUND: Mycoplasma hyorhinis is a prevalent respiratory pathogen in swine, causing significant economic loss to pig producers. There is growing evidence that respiratory pathogen infections have a large impact on intestinal microecology. To study the effect of M. hyorhinis infection on gut microbial composition and metabolome profile, pigs were infected with M. hyorhinis. Metagenomic sequencing analysis was performed of fecal samples and a liquid chromatography/tandem mass spectrometry (LC-MS/MS) analysis of gut digesta was made. RESULTS: Pigs infected with M. hyorhinis had enriched Sutte

Preterm labor and bacterial vaginosis-associated bacteria among urban women.

AIMS: Bacterial vaginosis (BV) affects millions of women, is extremely prevalent and is frequently chronic. We recognize numerous microbiologic variations among women with BV and this variability may explain the limited effectiveness of metronidazole in curing BV and/or reducing the risk of spontaneous preterm birth (SPTB) among BV-positive pregnant women. We assessed the independent role of seven common BV-associated bacteria on the risk of spontaneous preterm birth (SPTB) among urban pregnant women. METHODS: This prospective cohort study was conducted within an urban obstetrics practice at

Distinct Gut Microbiome Signatures in Hemodialysis and Kidney Transplant Populations.

Background: Gut microbiota plays a critical role in host metabolism, immunity, and intestinal barrier integrity. Both chronic kidney disease (CKD) and kidney transplantation (KTR) are associated with gut dysbiosis, driven by uremic toxins, comorbidities, and immunosuppressive therapy. However, direct comparisons between hemodialysis (HD), KTR, and healthy controls (HC), while accounting for dietary factors, remain limited. Methods: We conducted a cross-sectional study including 48 HD patients, 75 KTR patients, and 13 HC. Stool patient samples were analyzed using 16S rRNA amplicon sequencing ta

Comparative Analysis of Rumen Microbiota Composition in Dairy Cows with Simple Indigestion and Healthy Cows.

Simple indigestion in cows leads to substantial economic losses in the dairy industry. Despite ongoing efforts, an effective treatment for this issue remains elusive. Previous studies have emphasized the vital role of rumen microbes in maintaining ruminant health. To deepen our comprehension of the intricate interplay between rumen microbiota and simple indigestion, we undertook a study involving the analysis of rumen fluid from eight cows with simple indigestion and ten healthy cows. Additionally, we collected data pertaining to milk production, rumination behavior, and rumen characteristics.

Differences in gut microbiome between autosomal dominant polycystic kidney disease with and without intracranial aneurysms.

Autosomal dominant polycystic kidney disease (ADPKD) is a genetic disorder characterized by cyst formation in the kidneys, and is associated with an elevated risk of intracranial aneurysms (IAs). Although a family history is a recognized risk factor for IAs in patients with ADPKD, emerging research suggests that gut microbiome composition may influence IA development. We investigated the relationship between the gut microbiome and the development of IA in patients with ADPKD. We recruited patients with ADPKD with (IA group) and without (non-IA group) IA from Osaka University between October 20

Molecular characterization of vaginal microbiota using a new 22-species qRT-PCR test to achieve a relative-abundance and species-based diagnosis of bacterial vaginosis.

BACKGROUND: Numerous bacteria are involved in the etiology of bacterial vaginosis (BV). Yet, current tests only focus on a select few. We therefore designed a new test targeting 22 BV-relevant species. METHODS: Using 946 stored vaginal samples, a new qPCR test that quantitatively identifies 22 bacterial species was designed. The distribution and relative abundance of each species, α- and β-diversities, correlation, and species co-existence were determined per sample. A diagnostic index was modeled from the data, trained, and tested to classify samples into BV-positive, BV-negative, or transit

Oral microbiome and health.

The oral microbiome is diverse in its composition due to continuous contact of oral cavity with the external environment. Temperatures, diet, pH, feeding habits are important factors that contribute in the establishment of oral microbiome. Both culture dependent and culture independent approaches have been employed in the analysis of oral microbiome. Gene-based methods like PCR amplification techniques, random amplicon cloning, PCR-RELP, T-RELP, DGGE and DNA microarray analysis have been applied to increase oral microbiome related knowledge. Studies revealed that microbes from the phyla Firmic

The role of lactate metabolism in shaping intestinal microbiota and health.

Lactate, particularly the D-isomer, which is produced exclusively by microbial fermentation, is usually low or undetectable in the healthy colon but can accumulate when there is a microbial imbalance, contributing to low pH and inflammation. Intestinal lactate-utilizing bacteria, such as Anaerobutyricum soehngenii, Anaerostipes species, and Coprococcus catus, convert lactate to short-chain fatty acids, including butyrate, potentially important for health. Here, we mined genomes of lactate utilizers for genes involved in propionate formation and conducted metabolic network modeling to investiga

Advances of Polymerase Chain Reaction Sperm Testing in Men With Chronic Prostatitis and Pelvic Pain Syndrome.

Prostate inflammation is a common condition in men characterized by swelling of the prostate gland, often associated with other prostate diseases. Understanding the role of chronic inflammation in prostatic diseases is important due to the changes in prostatic cells and the persistence when undiagnosed. The evaluation and management of chronic prostatitis (CP) and chronic pelvic pain (CPP) involve specific diagnostic tests. This study aimed to evaluate the efficiency of the polymerase chain reaction (PCR) detection kits that use multiplex real-time PCR in comparison to standard microbiology sp
