Melanin photosensitization and the effect of visible light on epithelial cells.

Protecting human skin from sun exposure is a complex issue that involves unclear aspects of the interaction between light and tissue. A persistent misconception is that visible light is safe for the skin, although several lines of evidence suggest otherwise. Here, we show that visible light can damage melanocytes through melanin photosensitization and singlet oxygen (1O2) generation, thus decreasing cell viability, increasing membrane permeability, and causing both DNA photo-oxidation and necro-apoptotic cell death. UVA (355 nm) and visible (532 nm) light photosensitize 1O2 with similar yields

Extracellular and intracellular melanin in inflammatory middle ear disease.

OBJECTIVES/HYPOTHESIS: Melanin is a pigmented polymer with a known role in dermal solar protection. In vertebrates, melanogenesis has been reported in leukocyte populations, suggesting a potential role in innate immunity. In this study, we report the novel finding of melanin associated with chronic inflammation and speculate on its potential role in the middle ear and mastoid. STUDY DESIGN: Retrospective review of case series. METHODS: Medical records of six patients who demonstrated melanin in the ear were reviewed. RESULTS: Six patients from 1 to 63 years of age were identified with extra

Melanin, a promising radioprotector: mechanisms of actions in a mice model.

The radioprotective effect of extracellular melanin, a naturally occurring pigment, isolated from the fungus Gliocephalotrichum simplex was examined in BALB/C mice, and the probable mechanism of action was established. At an effective dose of 50mg/kg body weight, melanin exhibited both prophylactic and mitigative activities, increasing the 30-day survival of mice by 100% and 60%, respectively, after exposure to radiation (7Gy, whole body irradiation (WBI)). The protective activity of melanin was primarily due to inhibition of radiation-induced hematopoietic damages as evidenced by improvement

Magnetic beads method for determination of binding of drugs to melanin.

Binding to melanin is considered to be a reason for several adverse effects of drugs and should be known to reduce the failure rate due to inappropriate pharmacokinetics in search for better pharmaceuticals. A new, reliable and convenient method of determination of affinity of drugs and drug candidates to melanin has been proposed employing magnetic beads. For that aim the reaction conditions to effectively covalently immobilize melanin on surface of superparamagnetic beads have been determined. Binding efficiency of melanin towards antipsychotic and other basic drugs has been determined and c

The regulation of alcohol intake by melanin-concentrating hormone in rats.

Given into the brain, melanin-concentrating hormone (MCH) increases alcohol consumption, but the mechanism and physiological relevance of this effect are unclear. We hypothesized that endogenous MCH will enhance alcohol drinking and that MCH increases alcohol's reinforcing properties. An MCH receptor 1 (MCHR1) antagonist, or saline was administered centrally alone, or preceding MCH or saline to rats trained to drink 10% alcohol using sucrose fading. Blocking MCHR1 neither reduced alcohol intake (saline=0.4+/-0.1 g, 30 microg MCHR1 antagonist=0.4+/-0.1 g/kg alcohol), nor attenuated MCH-induced

Binding affinity of bunazosin, dorzolamide, and timolol to synthetic melanin.

PURPOSE: The purpose of this study was to compare the binding affinity of bunazosin and dorzolamide to synthetic melanin relative to that of timolol. METHODS: Synthetic melanin was prepared from dopa by the action of tyrosinase. Timolol, dorzolamide, and bunazosin were incubated separately at a concentration of 10(-4) M in 2 ml of 0.066 M phosphate buffer containing 5 mg of synthetic melanin. After centrifugation, the absorbance of each free drug in the supernatant was measured at its optimum wavelength. The percentage of each drug bound to melanin was calculated directly from the change in a

Blastomyces dermatitidis produces melanin in vitro and during infection.

Melanin is made by several important pathogenic fungi and is implicated in the pathogenesis of a number of mycoses. This study investigates whether the thermally dimorphic fungal pathogen Blastomyces dermatitidis produces melanin. Using techniques developed to study melanization in other fungi, we demonstrate that B. dermatitidis conidia and yeast produce melanin in vitro and that yeast cells synthesize melanin or melanin-like pigment in vivo. Melanization reduced susceptibility to amphotericin B, but not to itraconazole or voriconazole. Since melanin is an important virulence factor in other

The function of melanin or six blind people examine an elephant.

The pigment melanin is found in all living kingdoms and in many different structures and forms. When its various functions are examined separately, its behaviors seem disparate and conflicting. It has a clear role in camouflage and sexual display. Other major roles are examined critically. It can act as a sun screen but is not a very effective one. It can also scavenge active chemical species, but this, too, is not done very effectively. It produces active radicals that can damage DNA. It binds to drugs in ways that are either beneficial or deleterious. Aside from camouflage, its other roles c

The influence of inner ear melanin on susceptibility to TTS in humans.

In order to investigate the function of the inner ear melanin, the relationship between skin pigmentation and noise-induced temporary hearing loss (TTS) was studied. Forty-four normal-hearing Caucasian subjects were divided into three groups according to their sun sensitivity. Hearing thresholds before and after exposure were ascertained with a computerized sweep frequency audiometer in the frequency range 2-8 kHz. The noise exposure consisted of a 1/3-octave band-filtered noise with a centre frequency of 2 kHz at 105 dB SPL for 10 min. The mean TTS in the frequency range 2-8 kHz showed statis

Further evidence that the pigment in the Dubin-Johnson syndrome is not melanin.

The pigment in the Dubin-Johnson syndrome (DJS) is shown unequivocally not to be a typical melanin or closely related polymer. In electron spin resonance (ESR) studies of DJS pigment from a hepatoma, it is shown that, unlike true melanins, the pigment associated with the DJS syndrome has no free radical in the absence of light. Exposure to even low levels of visible light over a broad frequency range induces a free radical in the DJS pigment. Previous studies did not appreciate the sensitivity to light of this pigment and therefore erroneously concluded that the DJS pigment had a permanent fre

Pharmacological treatment of tinnitus with special reference to the role of melanin.

Local anaesthetics have hitherto been thought to exert their effects on tinnitus through a centrally operative mechanism, analogously to their effects in epilepsy and central pain. Evidences for a peripheral mechanism, probably related to the affinity of these drugs to inner ear melanin, are reviewed. The physico-chemical properties of melanin and findings indicating that this substance plays a role in hearing are also discussed.

Effect of macrophages on elimination of dermal melanin from the dermis.

To facilitate the mobilization of dermal melanin from the skin, we studied the role of macrophages in the elimination of dermal melanin in incontinentia pigmenti. The protein and melanin moieties of mouse-melanoma melanosomes were labeled with 14C and 3H, respectively. Labeled melanosomes were injected intracutaneously into the backs of guinea pigs. Carrageenan, a macrophage toxic agent, showed an inhibitory effect on the degradation and elimination of protein moiety of melanosomes from the dermis, when carrageenan was injected i.p. at the time of inoculation of melanosomes. The melanin pigmentary disorder in a family with Hermansky-Pudlak syndrome.

The albinotic skin and hair of 2 patients with Hermansky-Pudlak syndrome were investigated by light and electron microscopy. Incubation of hairbulbs and epidermis in 1-dopa revealed a weak tyrosinase activity. The epidermal melanocyte population was of normal density. The most striking feature was the presence of numerous giant melanosomes resembling those mainly reported in various hyperpigmented skin lesions. The association of this melanosomal disorder with the platelet dysfunction and ceroid storage typical of the autosomal recessive Hermansky-Pudlak syndrome might provide new insights int

Photothermal Release by Melanin-like Nanoparticles: Biomedical Applications.

Melanin-like nanoparticles (NPs) exhibit a remarkable ability to absorb light across a wide range of wavelengths, from the ultraviolet (UV) to the near-infrared (NIR) spectrum. This characteristic enables them to serve as effective photothermal agents (PTAs). Upon irradiation, especially within the NIR window, a region where biological tissues are highly transparent, these NPs efficiently convert light energy into heat. This phenomenon, known as the photothermal effect, leads to localized temperature increases. The resulting heat can be strategically employed to induce selective cell death in

Mutations in SLC45A2 lead to loss of melanin in parrot feathers.

Bird plumage coloration is a complex and multifactorial process that involves both genetic and environmental factors. Diverse pigment groups contribute to plumage variation in different birds. In parrots, the predominant green color results from the combination of 2 different primary colors: yellow and blue. Psittacofulvin, a pigment uniquely found in parrots, is responsible for the yellow coloration, while blue is suggested to be the result of light scattering by feather nanostructures and melanin granules. So far, genetic control of melanin-mediated blue coloration has been elusive. In this

Measuring Laccase Activity and Melanin Production in Cryptococcus neoformans.

Melanin is a complex dark pigment synthetized by the phenoloxidase enzyme laccase in Cryptococcus neoformans. In vitro, this enzyme oxidizes exogenous catecholamines to produce melanin that may be secreted or incorporated into the fungal cell wall. This pigment has multiple roles in C. neoformans virulence during its interaction with different hosts and probably also in protecting fungal cells in the environment against predation and oxidative and radiation stresses, among others. However, it is important to note that laccase also has melanin-independent roles in C. neoformans interactions wit
