Effects of a non-selective TRPC channel blocker, SKF-96365, on melittin-induced spontaneous persistent nociception and inflammatory pain hypersensitivity.

OBJECTIVE: Melittin is the main peptide in bee venom and causes both persistent spontaneous nociception and pain hypersensitivity. Our recent studies indicated that both transient receptor potential (TRP) vanilloid receptor 1 (TRPV1) and canonical TRPs (TRPCs) are involved in mediating the melittin-induced activation of different subpopulations of primary nociceptive cells. Here, we further determined whether TRPC channels are involved in melittin-induced inflammatory nociceptive responses in behavioral assays. METHODS: The anti-nociceptive and anti-hyperalgesic effects of localized periphera

Respiratory syncytial virus reverses airway hyperresponsiveness to methacholine in ovalbumin-sensitized mice.

Each year, approximately 20% of asthmatics in the United States experience acute symptom exacerbations, which commonly result from pulmonary viral infections. The majority of asthma exacerbations in very young children follow infection with respiratory syncytial virus (RSV). However, pathogenic mechanisms underlying induction of asthma exacerbations by RSV are not well understood. We therefore investigated the effect of post-sensitization RSV infection on lung function in ovalbumin (OVA)-sensitized BALB/c mice as a model of RSV asthma exacerbations. OVA sensitization of uninfected female BALB/

Simultaneous binding of drugs with different chemical structures to Ca2+-calmodulin: crystallographic and spectroscopic studies.

The modulatory action of Ca2+-calmodulin on multiple targets is inhibited by trifluoperazine, which competes with target proteins for calmodulin binding. The structure of calmodulin crystallized with two trifluoperazine molecules is determined by X-ray crystallography at 2.74 A resolution. The X-ray data together with the characteristic and distinct signals obtained by circular dichroism in solution allowed us to identify the binding domains as well as the order of the binding of two trifluoperazine molecules to calmodulin. Accordingly, the binding of trifluperazine to the C-terminal hydrophob

Snake venom cardiotoxins and bee venom melittin activate phospholipase C activity in primary cultures of skeletal muscle.

The effects of cardiotoxin fractions from Naja naja kaouthia and Naja naja atra snake venoms and synthetic melittin peptide were examined on lipolytic activity in red blood cells and primary skeletal muscle cultures. Both native cardiotoxin fractions caused considerable production of free fatty acids in red blood cells. This production was abolished when the fractions were first treated with p-bromophenacyl bromide to reduce the venom phospholipase A2 activity contamination. In equine and human primary cultures of skeletal muscle, the N. n. kaouthia cardiotoxin (10 microM) and melittin (2 micr

Evaluation of the role of calcium in cytotoxic injury in isolated rat pancreatic acini.

The role of extracellular Ca2+ in pancreatic acinar membrane damage (cellular injury) by nicotine, membrane-active agents (mellitin, snake venom and Ca2+ ionophore A23187) and secretagogues (CCK-8 and secretin) was investigated. Freshly isolated dispersed pancreatic acini from 18 h fasted adult rats were incubated with one of the aforementioned agents, in the absence and presence of Ca2+. Cellular injury was assessed by measuring the release of pulse-labeled 51Cr and LDH. In addition, release of amylase, trypsinogen and chymotrypsinogen was also determined. In the absence of Ca2+ nicotine (6 m

Intracellular Ca2+ sequestration in cultured mouse fibroblasts: ATP-dependent Ca2+ uptake by saponin-permeabilized Swiss 3T3 cells.

Intracellular Ca2+ sequestration was studied by determining 45Ca2+ uptake by saponin-permeabilized Swiss 3T3 mouse fibroblasts. Uptake was enhanced by addition of ATP and was dependent on the concentrations of ATP and saponin. ADP and UTP also enhanced uptake, but less than did ATP. Mitochondrial Ca2+ uptake was distinguished from nonmitochondrial uptake by the use of antimycin A plus oligomycin at various Ca2+ concentrations. Major sites for sequestration at 10(-5) M Ca2+ are probably mitochondrial and those at 10(-7) M Ca2+ nonmitochondrial, because these drugs inhibited ATP-dependent Ca2+ u

A Comprehensive Review of the Antimicrobial Peptide CM11: Structure, Biological Properties, and Therapeutic Prospects.

The growing prevalence of drug-resistant infections and the constraints of existing cancer treatment have increased efforts to find new therapeutic agents. CM11 is a synthetic hybrid peptide derived from Cecropin and Melittin, which is a promising candidate because it has a broadspectrum of antimicrobial and anticancer effects. Preclinical research indicates that it has a rapid mechanism of action, interfering with bacterial membranes, preventing biofilm formation, and inducing apoptosis in various cancer cell lines, including leukemia and hepatocellular carcinoma. To address the inherent limi

Adjuvant action of melittin following intranasal immunisation with tetanus and diphtheria toxoids.

Melittin, a 26-amino acid peptide and the major active component of the venom of the honey bee--Apis mellifera--has recently been shown to have absorption enhancing properties in Caco-2 cells at levels well below the level required for the generation of cytotoxicity. Given the potential of absorption enhancing agents to act as adjuvants when administered nasally [Alpar, H.O., Eyles, J.E., Williamson, E.D. and Somavarapu, S. (2001) "Intranasal vaccination against plague, tetanus and diphtheria", Adv. Drug Delivery Rev. 51, 173-201] we hypothesized that melittin may have potential as a mucosal a

Increase in filtration coefficient from actions of melittin on neutrophils in isolated rabbit lungs.

Activation of neutrophils may contribute to lung injury in the adult respiratory distress syndrome. We added rabbit neutrophils to the pulmonary circulation of salt-perfused and ventilated isolated rabbit lungs. These neutrophils were activated by adding synthetically pure melittin to the perfusate. This led to lung injury as measured by filtration coefficient under no-flow conditions. We also activated neutrophils in vitro before addition to the pulmonary circulation. These preactivated neutrophils also produced lung injury, indicating a primary action of melittin on neutrophils rather than o

Sequence and specificity of two antibacterial proteins involved in insect immunity.

Immune responses have been described for many different insect species. However, it is generally acknowledged that immune systems must therefore differ from those of vertebrates. An effective humoral immune response has been found in pupae of the cecropia moth, Hyalophora cecropia. The expression of this multicomponent system requires de novo synthesis of RNA and proteins and its broad antibacterial activity is due to at least three independent mechanisms, the most well known of which is the insect lysozyme. However, this enzyme is bactericidal for only a limited number of Gram-positive bacter

Melittin- and Apamin-Standardized Bee Venom: UHPLC-ESI-MS Characterization, Antibacterial Activity, and Apoptosis Induction in Human Keratinocytes.

Bee venom (BV) is valued therapeutically mainly for its peptides, melittin and apamin. This study determined the dose-dependent biological activity of bee venom standardized to its melittin and apamin content by UHPLC-ESI-MS, as well as that of its principal peptides, melittin and apamin, tested at matched concentrations. Antibacterial activity was assessed as showing the minimum inhibitory and minimum bactericidal concentrations (MIC, MBC) against Staphylococcus aureus (MSSA and MRSA) and Staphylococcus epidermidis (MRSE). Cytotoxicity toward primary human keratinocytes (HEKa) was determined

Efficient synthesis, stability-guided optimization and anti-ulcerative colitis evaluation of bee venom peptide melittin.

Ulcerative colitis (UC) is considered as one of the most prevalent inflammatory bowel diseases (IBDs), which increases risks for colectomy and colorectal cancer. However, due to moderate efficiency and potential side effects of first-line drugs, it is desirable to develop more efficient anti-UC agents. The natural peptide Melittin, which is the main active ingredient of bee venom, is considered as a potential scaffold for the development of anti-UC drugs. Nevertheless, as a linear amphipathic peptide, Melittin could be degraded by various proteases, suffering from poor stability and short half

Melittin-loaded solid lipid nanoparticles inhibit pulmonary fibrosis via the NOVA2-TGF-β/Smads signaling pathway.

OBJECTIVE: Melittin (MLT) is a cationic hemolytic peptide that suppresses inflammation and fibrosis in various animal models. However, the application of MLT in clinical practice is limited owing to its side effects. Solid lipid nanoparticles have the advantages of cell-specific delivery and sustained release of hydrophobic drugs, which can be exploited against many diseases. This study aimed to evaluate the effects of MLT-loaded solid lipid nanoparticles (MLT@NPs) on pulmonary fibrosis. METHODS: In this study, the MLT-loaded solid lipid nanoparticles (MLT@NPs) were designed and their antifib

Increased lethality of calmodulin antagonists and bleomycin to human bone marrow and bleomycin-resistant malignant cells.

The effect of bleomycin and calmodulin antagonists on human cells was studied using a clonogenic assay. A 1-h exposure to nontoxic concentrations of the calmodulin antagonists melittin (0.5 microM), pimozide (5 microM), and chlorpromazine (20 microM) increased the lethality of bleomycin to human ovarian carcinoma cells (SK-OV). No increase was seen with chlorpromazine sulfoxide, which lacks calmodulin antagonistic activity. Maximum enhancement of bleomycin lethality by calmodulin antagonists was seen when the antagonist was present simultaneously with bleomycin rather than before or after bleo

Isolation and characterization of a new Mr 18,000 protein with calcium vector properties in amphioxus muscle and identification of its endogenous target protein.

A new Ca2+-binding protein, called CaVP, has been detected in muscle of the cephalochordate amphioxus and purified to electrophoretic homogeneity. The Mr 18,000 protein (pI = 4.9) binds 2 Ca2+ atoms in a noncooperative way with an intrinsic binding constant of 8.2 X 10(6) M-1. Ca2+, but not Mg2+, induces a 10% increase in alpha-helical content in the metal-free protein. CaVP does not interact with chlorpromazine, but forms a Ca2+-dependent complex with melittin. In situ, CaVP forms a high affinity Ca2+-dependent complex with an Mr 36,000 protein present in muscle extracts of amphioxus. This co

pH-responsive polymeric-peptide complex enhances tumor immunogenic cell death by membrane disruption.

Melittin, the principal bioactive component of bee venom, exhibits potent anticancer activity. However, its clinical application is hindered by nonspecific hemolytic toxicity. In this study, we developed a pH-responsive polymeric-peptide complex (PCM) to improve anti-tumor activity of melittin while minimizing off-target effects. PCM remained structurally stable under physiological conditions (pH 7.4) but underwent rapid disassembly in the mildly acidic tumor microenvironment (pH 6.8) to expose melittin. The exposed melittin bound to tumor cell membranes, compromised membrane integrity through

Neuroprotective Effects of Melittin Against Cerebral Ischemia and Inflammatory Injury via Upregulation of MCPIP1 to Suppress NF-κB Activation In Vivo and In Vitro.

Melittin, a principal constituent of honeybee venom, exhibits diverse biological effects, encompassing anti-inflammatory capabilities and neuroprotective actions against an array of neurological diseases. In this study, we probed the prospective protective influence of melittin on cerebral ischemia, focusing on its anti-inflammatory activity. Mechanistically, we explored whether monocyte chemotactic protein-induced protein 1 (MCPIP1, also known as ZC3H12A), a recently identified zinc-finger protein, played a role in melittin-mediated anti-inflammation and neuroprotection. Male C57/BL6 mice wer

Eradication of vancomycin-resistant Staphylococcus aureus on a mouse model of third-degree burn infection by melittin: An antimicrobial peptide from bee venom.

Third-degree burn infections caused by antibiotic-resistant bacteria are of high clinical concern. Chemical antibiotics are not promising in eradication of bacterial infections. In this challenging condition, antimicrobial peptides (AMPs) are recently introduced as novel promising agents to overcome the issue. Accordingly, our study aimed to evaluate the efficiency of 'melittin' as natural peptide in bee venom, in eradicating vancomycin resistant Staphylococcus aureus (VRSA) on a mouse model of third-degree burn infection. In vitro pharmacological value of melittin was determined by examining
