The Janus face of CaMKII: from memory consolidation to neurotoxic switch in Alzheimer's disease.

Alzheimer's disease (AD), a neurodegenerative "memory killer" demanding urgent global intervention, has long been shrouded in mystery regarding its core pathological mechanisms. Although the traditional amyloid-β (Aβ) hypothesis remains dominant, recent groundbreaking research has revealed that early activation of aberrant calcium (Ca2⁺) signaling pathways serves as the "initiating trigger" of AD pathogenesis-preceding even the formation of classical Aβ plaques-a discovery that fundamentally overturns the existing cognitive framework. This study systematically deconstructs, for the first time,

Consolidation deficits in episodic memory define distinct clinical and neurodegenerative profiles in Huntington's disease.

BACKGROUND: Huntington's disease (HD) is primarily associated with executive dysfunction, but episodic memory impairment is also present. Traditionally, these memory deficits have been attributed to retrieval difficulties linked to fronto-striatal dysfunction, rather than to disruptions in encoding or consolidation processes. However, the specific nature and diversity of memory impairments in HD remain underexplored. OBJECTIVE: To characterize the profile of episodic memory impairment in HD, identify distinct cognitive phenotypes, and examine their clinical, neuroanatomical, and biomarker cor

Sleep-dependent engram reactivation during hippocampal memory consolidation associated with subregion-specific biosynthetic changes.

Post-learning sleep is essential for hippocampal memory processing, including contextual fear memory consolidation. We labeled context-encoding engram neurons in the hippocampal dentate gyrus (DG) and assessed reactivation of these neurons after fear learning. Post-learning sleep deprivation (SD) selectively disrupted reactivation of inferior blade DG engram neurons, linked to SD-induced suppression of neuronal activity in the inferior, but not superior DG blade. Subregion-specific spatial profiling of transcripts revealed that transcriptomic responses to SD differed greatly between hippocampa

Randomly fluctuating neural connections may implement a consolidation mechanism that explains classic memory laws.

How can we reconcile the massive fluctuations in neural connections with a stable long-term memory? Two-photon microscopy studies have revealed that large portions of neural connections (spines, synapses) are unexpectedly active, changing unpredictably over time. This appears to invalidate the main assumption underlying the majority of memory models in cognitive neuroscience, which rely on stable connections that retain information over time. Here, we show that such random fluctuations may in fact implement a type of memory consolidation mechanism with a stable very long-term memory that offer

Transcranial direct current stimulation applied after encoding facilitates episodic memory consolidation in older adults.

Episodic memory shows the largest degree of age-related memory decline. There is evidence that consolidation, the process that stabilizes memories after encoding, is reduced in older adults. Previous studies have shown that transcranial direct current stimulation (tDCS) applied during intentional encoding or immediately after a contextual reminder enhanced delayed episodic memory performance, suggesting a potential interaction between tDCS and consolidation or reconsolidation processes. The present randomized, double-blind, sham-controlled study addressed the question whether tDCS applied imme

Transient inactivation of perirhinal cortex disrupts encoding, retrieval, and consolidation of object recognition memory.

Damage to perirhinal cortex (PRh) impairs object recognition memory in humans, monkeys, and rats when tested in tasks such as delayed nonmatching to sample, visual paired comparison, and its rodent analog, the spontaneous object recognition task. In the present study, we have capitalized on the discrete one-trial nature of the spontaneous object recognition task to investigate the role of PRh in several distinct stages of object recognition memory. In a series of experiments, transient inactivation of PRh was accomplished with bilateral infusions of lidocaine directly into PRh immediately befo

Impaired sleep-related consolidation of declarative memories in idiopathic focal epilepsies of childhood.

OBJECTIVE: Declarative memory is consolidated during sleep in healthy children. We tested the hypothesis that consolidation processes are impaired in idiopathic focal epilepsies (IFE) of childhood in association with frequent interictal epileptiform discharges (IEDs) during sleep. METHODS: A verbal (word-pair association) and a nonverbal (2D object location) declarative memory task were administrated to 15 children with IFEs and 8 control children 6-12 years of age. Patients had either centrotemporal (11 patients) or occipital (4 patients) IEDs. All but 3 patients had a history of unprovoked

Imaging systems level consolidation of novel associate memories: a longitudinal neuroimaging study.

Previously, a standard theory of systems level memory consolidation was developed to describe how memory recall becomes independent of the medial temporal memory system. More recently, an extended consolidation theory was proposed that predicts seven changes in regional neural activity and inter-regional functional connectivity. Using longitudinal event-related functional magnetic resonance imaging of an associate memory task, we simultaneously tested all predictions and additionally tested for consolidation-related changes in recall of associate memories at a sub-trial temporal resolution, an

Effect of naloxone and amphetamine on acquisition and memory consolidation of active avoidance responses in rats.

Pretraining IP injection of naloxone (0.3 mg/kg) or amphetamine (2 mg/kg) enhanced performance during acquisition, but did not improve retention of active avoidance responses in rats. Naloxone (0.1 or 3 mg/kg) had no effect on acquisition or on retention. The combination of naloxone (0.3 mg/kg) plus amphetamine (2 mg/kg) did not produce the facilitation observed when each of the two drugs was administered alone. Pretreatment with the higher dose of naloxone (3 mg/kg) blocked the facilitative effect of amphetamine on acquisition. Post-training administration of naloxone (0.3 mg/kg) or amphetami

Retrograde amnesia produced by electroconvulsive shock after reactivation of a consolidated memory trace.

Rats had a memory loss of a fear response when they received an electroconvulsive shock 24 hours after the fear-conditioning trial and preceded by a brief presentation of the conditioned stimulus. No such loss occurred when the conditioned stimulus was not presented. The memory loss in animals given electroconvulsive shock 24 hours after conditioning was, furthermore, as great as that displayed in animals given electroconvulsive shock immediately after conditioning. This result throws doubt on the assertion that electroconvulsive shock exerts a selective amnesic effect on recently acquired mem

Prioritizing Detailed Item Memories Through Post-Encoding Motivation Requires Consolidation.

Prioritizing memories that are motivationally salient is adaptive for goal-directed behavior, ensuring that information being retained is the most relevant to future goals. Past research demonstrates we can use value signals to adaptively modulate our memory. Crucially, however, the timing of value signals varies. Sometimes we know the significance of an upcoming event in advance, while other times, we are unaware of an event's importance until after the fact. Does the timing of a value signal relative to encoding change the cognitive mechanism by which it shapes memory? To test this question,

The effect of selective REM-sleep deprivation on the consolidation and affective evaluation of emotional memories.

Emotion boosts the consolidation of events in the declarative memory system. Rapid eye movement (REM) sleep is believed to foster the memory consolidation of emotional events. On the other hand, REM sleep is assumed to reduce the emotional tone of the memory. Here, we investigated the effect of selective REM-sleep deprivation, SWS deprivation, or wake on the affective evaluation and consolidation of emotional and neutral pictures. Prior to an 9-h retention interval, sixty-two healthy participants (23.5 ± 2.5 years, 32 female, 30 male) learned and rated their affect to 80 neutral and 80 emotion

Midazolam impairs acquisition and retrieval, but not consolidation of reference memory in the Morris water maze.

Amnesia is one of the most discussed properties of the benzodiazepine class of drugs. The effects of benzodiazepines on human memory are usually anterograde, while changes in retrograde memory functions were seldom reported. Such inconsistent findings have prompted numerous animal studies investigating the influences of these positive modulators of inhibitory neurotransmission on different stages of memory. Among the benzodiazepines, memory effects of midazolam are of special interest due to its many and varied clinical applications. The present Morris water maze study in adult male Wistar rat

Sleep to find your way: the role of sleep in the consolidation of memory for navigation in humans.

Although a large body of evidence indicates that sleep plays an important role in learning and memory processes, the actual existence of a sleep-dependent spatial memory consolidation has been not firmly established. Here, by using a computerized 3D virtual navigation tool, we were able to show that topographical orientation in humans largely benefits from sleep after learning, while 10 h of wakefulness during the daytime do not exert similar beneficial effects. In particular, navigation performance enhancement needs sleep in the first post-training night, and no further improvements were seen

An inhibitor of DNA recombination blocks memory consolidation, but not reconsolidation, in context fear conditioning.

Genomic recombination requires cutting, processing, and rejoining of DNA by endonucleases, polymerases, and ligases, among other factors. We have proposed that DNA recombination mechanisms may contribute to long-term memory (LTM) formation in the brain. Our previous studies with the nucleoside analog 1-beta-D-arabinofuranosylcytosine triphosphate (ara-CTP), a known inhibitor of DNA ligases and polymerases, showed that this agent blocked consolidation of conditioned taste aversion without interfering with short-term memory (STM). However, because polymerases and ligases are also essential for D

Effects of naloxone and naltrexone on memory consolidation in CD1 mice: involvement of GABAergic mechanisms.

The involvement of GABAergic mechanisms in the effects exerted by the opioid antagonists naloxone and naltrexone on memory consolidation was investigated in CD1 mice tested in a one-trial inhibitory avoidance task. In a first group of experiments posttraining administration of naloxone (2.0 and 4.0 but not 1.0 mg/kg) and naltrexone (0.5 and 1.0 but not 0.25 mg/kg), as well as those of the GABA-antagonists picrotoxin (0.5 and 1.0 but not 0.25 mg/kg) and bicuculline (0.25 and 0.5 but not 0.1 mg/kg) enhanced, whereas those of the GABA-agonist muscimol (1.0 and 2.0 but not 0.5 mg/kg) impaired rete

Effects of antidepressants, anesthetics and sedatives/hypnotics on sleep-associated memory consolidation - A systematic review.

Considering the wide and increasing utilization of drugs which act upon the central nervous system, a pressing, infrequently investigated question is their potential impact on sleep-associated memory consolidation. Here the influence of approved pharmaceuticals on memory consolidation during sleep in healthy humans, rats, and mice was investigated. A systematic review of the literature was conducted according to PRISMA guidelines. Criteria to be met included healthy subjects, drug application after learning and a comparable placebo condition. Exclusion criteria included but were not limited to

Does slow oscillation-spindle coupling contribute to sleep-dependent memory consolidation? A Bayesian meta-analysis.

The active system consolidation theory suggests that information transfer between the hippocampus and cortex during sleep underlies memory consolidation. Neural oscillations during sleep, including the temporal coupling between slow oscillations (SO) and sleep spindles (SP), may play a mechanistic role in memory consolidation. However, differences in analytical approaches and the presence of physiological and behavioral moderators have led to inconsistent conclusions. This metaanalysis, comprising 23 studies and 297 effect sizes, focused on four standard phase-amplitude coupling measures inclu
