Evaluating effects of penicillin treatment on the metabolome of rats.

Penicillin (PEN) V, a well-known antibiotic widely used in the treatment of Gram-positive bacterial infections, was evaluated in this study. LC/MS- and NMR-based metabolic profiling were employed to examine the effects of PEN on the host's metabolic phenotype. Male Sprague Dawley rats were randomly divided into groups that were orally administered either 0.5% methylcellulose vehicle, 100 or 2400mg PEN/kg body weight once daily for up to 14 consecutive days. Urine, plasma and tissue were collected from groups sacrificed at 6h, 24h or 14d. The body fluids were subjected to clinical chemistry and

Uncovering the metabolomic signatures in adolescent depression using a targeted metabolomics approach.

BACKGROUND: Depression is a leading cause of mortality worldwide and remains a growing public health concern. Most research has focused on adults, despite evidence that adolescence is a common period of onset of mental health problems. Metabolomics, which reflects the ongoing bodily biochemical processes through metabolite profiling, has emerged as a promising avenue to identify possible indicators of disease state. To contribute to this growing field, this study aims to identify early occurring metabolomic signatures associated with adolescent depression using a targeted metabolomics approach

Untargeted Metabolomics Revealed Metabolomic Profile in Patients with Primary Systemic Sclerosis.

Systemic sclerosis (SSc) is a systemic autoimmune disease characterized by increased morbidity and mortality. The intestinal microbiome and serum metabolites had been implicated in SSc, but the connection between the gut microbiome and serum metabolites remains poorly understood. In this study, we aimed to investigate the relationship between the gut microbiome and serum metabolome in SSc patients. Untargeted metabolomics was employed to examine the metabolic profiles of SSc patients. The results revealed significant alterations in metabolic pathways, particularly beta-alanine metabolism and p

A Metabolomics Approach to Screening for Autism Risk in the Children's Autism Metabolome Project.

Autism spectrum disorder (ASD) is biologically and behaviorally heterogeneous. Delayed diagnosis of ASD is common and problematic. The complexity of ASD and the low sensitivity of available screening tools are key factors in delayed diagnosis. Identification of biomarkers that reduce complexity through stratification into reliable subpopulations can assist in earlier diagnosis, provide insight into the biology of ASD, and potentially suggest targeted interventions. Quantitative metabolomic analysis was performed on plasma samples from 708 fasting children, aged 18 to 48 months, enrolled in the

Metabolomic Characterization of Acute Ischemic Stroke Facilitates Metabolomic Biomarker Discovery.

Acute ischemic stroke (AIS) is characterized by a sudden blockage of one of the main arteries supplying blood to the brain, leading to insufficient oxygen and nutrients for brain cells to function properly. Unfortunately, metabolic alterations in the biofluids with AIS are still not well understood. In this study, we performed high-throughput target metabolic analysis on 44 serum samples, including 22 from AIS patients and 22 from healthy controls. Multiple-reaction monitoring analysis of 180 common metabolites revealed a total of 29 metabolites that changed significantly (VIP > 1, p < 0.05).

When the proteome meets the metabolome observational and Mendelian randomization analyses.

OBJECTIVE: The basis for protein synthesis is the genetic code. Many of these proteins will affect intermediary metabolites by acting as enzymes, hormones, or by other actions. The aim of the present study was to assess the relationships of a large number of proteins with endogenous metabolites. METHODS: Plasma protein levels were measured by the proximity extension assay (PEA) and metabolites by mass spectrometry. Cross-sectional relationships of 242 proteins and 790 metabolites were evaluated in the EpiHealth and POEM studies using a discovery/validation approach. Genetic instruments identi

Metabolome profiling reveals the resistance mechanisms of rice against brown planthopper.

Rice (Oryza sativa L.) is a crucial global cereal crop, severely threatened by the brown planthopper (BPH), which causes extensive damage through direct feeding and virus transmission. To elucidate the resistance mechanism mediated by the BPH-resistance gene Bph6, metabolic responses to BPH infestation were compared between the resistant rice line G6 and the susceptible cultivar Nipponbare using LC-MS-based metabolomics combined with quantitative real-time PCR analysis. The Bph6 gene conferred both antixenotic and antibiotic resistance to BPH. A total of 673 metabolites were identified across

Natural selection shapes the evolution of the metabolome in Saccharomyces cerevisiae.

Metabolite abundances represent the ultimate functional outputs of cellular processes and provide a direct link between genotype and phenotype. While the evolutionary dynamics of mRNA and protein abundances have been extensively studied, the evolutionary forces shaping metabolite abundances remain largely unexplored. Here, we present a comprehensive framework to quantify natural selection acting on metabolite abundance using Saccharomyces cerevisiae as a model system. We quantified 1,037 metabolites across 21 mutation accumulation lines, their ancestor BY4741, and 18 wild isolates using liquid

Characterizing the Elemental Metabolome in Asparagus officinalis with Multielemental Speciation Analysis.

We report a multielemental and quantitative speciation analysis study with chromatographic inductively coupled plasma-mass spectrometry (ICP-MS/MS) detection involving four elements (sulfur, selenium, arsenic, and phosphorus) in Asparagus officinalis. The samples were locally purchased and labeled to originate from different regions in two continents. Major compounds were identified and quantified with confirmation based on chemical synthesis in pure form. Nontargeted analysis revealed significant quantitative and qualitative variation for the elemental metabolomes among samples, particularly

Dynamics of the Mammalian Placental Metabolome in Placentogenesis and Embryonic Development.

Mammalian placental metabolism is crucial for both placental and embryonic development. However, the metabolic profiles of placentas and their regulatory roles in placentogenesis and embryonic development at different developmental stages remain poorly characterized. To address these questions, we collect 501 mouse placentas spanning embryonic day (E) 8.5-14.5 and construct metabolomic-transcriptomic atlases of placentogenesis. Metabolomic and transcriptomic analyses reveal that placental samples from E8.5 to E14.5 are clustered into three separated states: E8.5, E9.5-10.5, and E11.5-14.5, pin

Gut microbiome-metabolome signatures of osteosarcopenia in fracture patients in China.

BACKGROUND: Osteosarcopenia, defined as the coexistence of low bone mass and sarcopenia, is a disabling musculoskeletal condition, yet its gut microbial and metabolic characteristics in clinical populations remain incompletely understood. Integrative multi-omics approaches may help clarify species-metabolite networks associated with this condition, particularly in fracture patients. METHODS: In this single-center, prospective cross-sectional study, 69 fracture patients aged ≥50 years were classified into four phenotypes: Normal (n = 18), isolated low bone mass (Bone, n = 18), isolated sarcope

Ten mouse organs proteome and metabolome atlas from adult to aging.

BACKGROUND: Aging is a complex biological process characterized by progressive molecular alterations across multiple organ systems, significantly influencing disease susceptibility and mortality. Unraveling molecular interactions driving aging is crucial for interventions promoting healthy aging and mitigating senescence. However, the systemic mechanisms governing both inter-organ interactions and organ-specific aging trajectories remain incompletely characterized. METHODS: To investigate the molecular dynamics of aging, we conducted a systematic multi-omics analysis of 400 tissue samples col

Metabolome informs about the chemical exposome and links to brain health.

The metabolome is an intermediate phenotype, summarizing the profile of all small molecules (<1.5 kDa) in biospecimens. The metabolome provides a readout for the net influence of the chemical exposome, diet, gut microbiome, and genome on human health. Metabolic changes observed in exposome studies may thus provide clues about adverse outcome pathways related to cancer, diabetes, heart disease, cognitive impairment and other neurological conditions such as Alzheimer's disease (AD). Whilst the number of human cohort studies including both metabolomic and exposomic profiles is increasing, they ar

Changes in the metabolome, lipidomein, and gut microbiota in Behçet's disease.

BACKGROUNDS: There is growing evidence that autoimmune illnesses are associated with the metabolome and microbiota. Because Behçet's disease (BD) is not often diagnosed as a systemic disorder, the aim of this research was to investigate changes in gut flora and metabolites in BD patients. METHODS: We used 16S rRNA gut microbiota gene sequencing and UPLC-QTOF-MS analysis to gather stool and serum samples from 12 age-matched healthy controls and 17 BD patients. The correlation between changes in gut microbiota and metabolites was then further analyzed. RESULTS: In contrast to healthy controls,
