Genetic polymorphism and plant growth promotion traits of potent fungal entomopathogens of rice leaf folder.

Entomopathogenic fungal biocides are preferred for environment friendly sustainable management of insect pests due to their host specificity and harmlessness to non-target insects. Plant growth promotion (PGP) functions of the entomofungi are also important attributes but hitherto insignificantly explored. Therefore, virulence of 17 natural fungal entomocides (Cordyceps, Beauveria, Metarhizium, Nomuraea, Fusarium, Verticillium, Trichoderma and Paecilomyces spp.) were evaluated for pathogenicity against five rice pests (brown plant hopper (Nilaparvata lugens) and green leaf hopper (Nephotettix

From soil to target: Investigating the role of native entomopathogenic fungi as biocontrol agents against Scaphoideus titanus ball (Hemiptera: Cicadellidae) in grapevines.

Entomopathogenic fungi (EPF) are promising biological control agents for the integrated pest management of Scaphoideus titanus Ball, the principal vector of Flavescence dorée phytoplasma in European vineyards. This study assessed the diversity, abundance, pathogenicity, and endophytic colonization ability of native EPF isolated from vineyard, woodland, and fallow field soils in the Piedmont Region (northwestern Italy). A total of 151 EPF isolates were recovered using selective media plating and the Galleria bait method and identified by ITS sequencing. Of the 14 species identified, eight belon

Rice recycling: a simple strategy to improve conidia production in solid-state cultures.

Here we studied at a laboratory scale a potential strategy to revalorize the residual rice remaining at the end of a conventional conidia production process in solid-state culture. The conidia production of Trichoderma asperellum Th-T4 (3) and Metarhizium robertsii Xoch-8.1 started with the use of fresh rice (unrecycled rice) as the substrate (cycle one), and continued with the use of recycled rice in successive cycles of conidia production. The rice remaining at the end of the first cycle was reused without any further sterilization or reinoculation. As a result, it was observed that the coni

A scorpion neurotoxin increases the potency of a fungal insecticide.

The low virulence of the insecticidal fungus Metarhizium anisopliae has stymied its widespread use in controlling insect pests. We show that high-level expression of an insect-specific neurotoxin from the scorpion Androctonus australis in hemolymph by M. anisopliae increases fungal toxicity 22-fold against tobacco hornworm (Manduca sexta) caterpillars and ninefold against adult yellow fever mosquitoes (Aedes aegypti) without compromising host specificity. Prelethal effects include reduced mobility and feeding of the insects targeted.

Nine new species of cordycipitoid fungi (Hypocreales, Sordariomycetes) from China.

Cordycipitoid fungi, encompassing the four families Clavicipitaceae, Cordycipitaceae, Ophiocordycipitaceae, and Polycephalomycetaceae, have been the focus of intensive molecular phylogenetic and taxonomic research over the past twenty years. Utilizing a six-locus dataset (LSU, ITS, SSU, tef1-α, rpb1, and rpb2) with expanded taxon sampling, this study establishes a robust phylogenetic framework for this group, with particular emphasis on the Ophiocordycipitaceae. Our work significantly expands the documented diversity of cordycipitoid fungi in China through the description of nine new species:

Mechanism of destruxin a inhibits juvenile hormone binding protein transporting juvenile hormone to affect insect growth.

Destruxin A, a non-ribosomal peptide toxin produced by Metarhizium, exhibits potent insecticidal activity by targeting various tissues, organs, and cells of insects. Our previous research has revealed that DA possesses the ability to bind to multiple proteins. In this study, we aimed to identify the most sensitive binding proteins of DA and investigate the physiological processes in which DA regulated. Through RNAi technology, we screened 22 binding proteins of DA in silkworm hemolymph. Among them, the juvenile hormone binding protein (JHBP), a hormone transport protein crucial for growth and

Unveiling thermotolerant and entomopathogenic fungi potential for solid-state fermentation: Multiscale evaluation in packed-bed bioreactor.

This work presents the production of fungal conidia from the thermotolerant and entomopathogenic fungi Metarhizium robertsii, by means of solid-state fermentation (SSF) in packed-bed bioreactors (PBR) up to a pilot scale of 100 L. The M. robertsii strain used (EAMa 01/158-su) presents both thermotolerant and entomopathogenic properties. Its thermotolerance is an unusual trait as entomopathogenic strain, and can help to surpass heat accumulation, the main limitation when increasing scale in SSF. Using a mixture of beer spent grains (BSG) and wood chips as substrates, work is performed at 3 diff

Is the developmental stage important for the plasticity of transgenerational immune priming?

Parental transfer of immune memory can increase offspring survival, although its benefits may vary across developmental stages. We investigated transgenerational immune priming in the fall armyworm (Spodoptera frugiperda) infected with the fungus Metarhizium brunneum. Using a split design, fathers and mothers were exposed to a low-dose fungal challenge at larval stages L5 or L6, and their offspring were subsequently challenged with a high dose at L5 or L6. We assessed survival, development, immune responses (phenoloxidase, lytic activity), and oxidative stress (total antioxidant capacity, cata

Pulmonary fungal granulomas and fibrinous pneumonia caused by different hypocrealean fungi in reptiles.

In contrast to fungal dermatitis, fungal glossitis and disseminated visceral mycosis, fungal infection of the lung has so far rarely been described in reptiles. Pulmonary fungal granulomas were diagnosed histopathologically within the scope of post mortem examinations. Fragments of the 18S-internal transcribed spacer1-5.8S rDNA (SSU-ITS1-5.8S) and 28S rDNA (LSU), including domains (D)1 and D2 as well as the protein coding gene translation elongation factor 1 alpha (TEF) were used for phylogenetical analysis after isolation of the fungal pathogen by culturing. Ten reptiles, including lizards (n

Beyond toxicity: Systems-level virulence of entomopathogenic fungi in termite social immunity.

Termites live in dense colonies where frequent social contact can facilitate pathogen transmission, but coordinated social immunity can also limit disease spread. Entomopathogenic fungi (EPF) are particularly relevant to termite pathology because they infect primarily through the cuticle, using adhesion, enzymatic degradation, penetration, and internal proliferation to overcome host barriers. This review synthesizes current knowledge on EPF virulence in termites and argues that fungal pathogenicity should be understood not only as direct toxicity (lethal effects of pathogen-derived compounds)

Ethanol tolerance of fungal strains underlies biocontrol of invasive ambrosia beetles.

Entomopathogenic fungi are widely recognized as biological control agents showing potential against insect pests that are difficult to manage with chemical pesticides. Invasive ambrosia beetles threaten a wide variety of tree crops and prove challenging to manage with chemical pesticides due to their unique life history: entrenched in the xylem of tree hosts, feeding on symbiotic fungi. Many ambrosia beetle species attack trees that are flood-stressed and producing ethanol, a key component of growth for their fungal symbiont. Ethanol causes osmotic and chaotropic stress in microbes and could p

Efficacy and environmental persistence of nootkatone for the control of the blacklegged tick (Acari: Ixodidae) in residential landscapes.

The ability of the plant-derived compound nootkatone to control nymphs of the blacklegged tick, Ixodes scapularis Say, was evaluated at lawn perimeter plots at homes in Lyme disease endemic areas of Connecticut. Three formulations of nootkatone ranging from 0.05 to 0.84% (0.06 - 1.03 g AI/m2) were applied by a hydraulic sprayer from 2008 to 2010. In 2008, the 0.84% emulsifiable nootkatone formulation provided 100% control of I. scapularis through week 1, but declined to 49 and 0% by 2 and 3 wk posttreatment, respectively. A combination of 0.05% nootkatone and entomopathogenic fungus, Metarhizi

Assessing pesticide compatibility with entomopathogenic fungi for biological control: evidence from in vivo and in vitro assays.

Integrating entomopathogenic fungi (EPF) into crop protection programs requires ensuring their compatibility with insecticides, herbicides, and fungicides to enable simultaneous applications and reduce operational costs. Compatibility is typically assessed by incorporating pesticides into growth media and measuring toxicity through germination, growth, and sporulation (Biological Index). However, field applications mainly expose conidia directly to pesticides, potentially leading to mismatches between laboratory and real-world outcomes. Here, we evaluated the compatibility of 5 fungicides (cop

An entomopathogen-based small-RNA delivery system promotes the biocontrol efficacy against termites.

Entomopathogenic fungus-derived microRNA-like RNAs (milRNAs) have been determined as novel virulence factors that exhibit cross-kingdom RNAi effects on insect hosts. However, which factors and how they function in social insects are still largely unknown. Using the pathogen Metarhizium anisopliae and its social host Reticulitermes aculabialis, we investigated how pathogen-derived small RNAs mediate cross-kingdom RNAi to promote infection in social insect colonies. By using sRNA-seq, we identified 27 milRNAs outside the fungal cells. Among them, Ma-milR-4 was significantly increased in the resp

Efficacy of biological control agents against Popillia japonica: A strategy to reduce the risk of pest spread via potted plant substrates.

The international trade of potted plants represents a pathway for the unintentional spread of harmful organisms, leading to new infestations in pest-free areas. The Japanese beetle Popillia japonica, which is considered one of the most threatening insect pests in Europe, can evade detection at the larval stage by hiding within the substrates of potted plants. To reduce the risk of P. japonica dispersal via potted plants, we tested the efficacy of entomopathogenic nematodes (EPNs) and fungi (EPF) as biological control agents (BCAs). Field trials with potted cherry trees were conducted in an Ita

Potential of adjuvants and an entomopathogenic fungus for the management of Helicoverpa armigera.

Helicoverpa armigera (Hübner) (Lepidoptera: Noctuidae) is a polyphagous species that feeds on crops of high economic importance in Brazil. Biological control, a component of Integrated Pest Management (IPM), employs parasitoids, predators, and entomopathogenic fungi to control agricultural pests as a strategy to reduce insecticide use and promote more sustainable practices. Adjuvants and activators, including vegetable oils, mineral oils, and fusel oil, can enhance the efficacy and toxicity of bioinsecticides. Thus, the objective of this study was to evaluate the effects of different adjuvants

Double trouble? Quantifying the risk from co-exposure to multiple pathogens in Tenebrio molitor at different CO2 concentrations.

The insect mass-rearing industry to produce feed and food is expanding rapidly. Insects in production frequently encounter multiple pathogens and environmental stressors simultaneously, which can lead to significant economic losses. Our understanding of the interactions between different stressors remains limited, and existing methods primarily focus on determining overall patterns of additivity, synergism, or antagonism. However, the interactions between different stressors may exhibit more intricate response patterns, such as time or dose dependency. With the expanding industry of insect pro

Encapsulation of entomopathogenic fungal conidia: evaluation of stability and control potential of Rhipicephalus microplus.

The use of chemical acaricides is the primary strategy to control tick infestations. Nonetheless, chemical resistance in ticks has been reported. Thus, complementary methods such as biological control using entomopathogenic fungi (EPF) have been investigated. EPF, although efficient, have their viability compromised when applied under natural conditions, which indicates that formulation development is essential. Some researchers have demonstrated the efficacy of ionic gelation in protecting EPF against deleterious abiotic factors. In the present study, we conducted the ionic gelation technique

Discovery of a fungal Tc toxin complex and functional mycoserpin through a novel two-by-two comparative genomics approach.

INTRODUCTION: Fungi have been a rich source of pharmaceuticals such as antibiotics, immunosuppressants, and cholesterol-lowering drugs; however, their therapeutic potential remains largely untapped due to difficulties in culturing and elucidating the genetic basis of beneficial traits. Fungi contain 'cryptic' genes that are expressed under certain, and often obscure, growth conditions and can produce complex compounds that are difficult to synthesize economically. Developments in genome sequencing and DNA-synthesis technologies offer new opportunities to produce such compounds using biotechnol

Auto-dissemination of commercially available fungal pathogens in a laboratory assay for management of the brown dog tick Rhipicephalus sanguineus.

Rhipicephalus sanguineus is a canine tick that infests dogs throughout the world and is frequently found in homes and dog kennels. Management of this tick species is complicated by the presence of resistance to commonly utilized acaricides. Fungal formulations could provide a valuable alternative tool for management and are especially relevant indoors where detrimental environmental effects on fungal spores are of less concern. Two commercially available fungal formulations, one containing Metarhizium anisopliae and the other containing Beauveria bassiana, were compared for time to death and s
