Cerebrospinal fluid pressure (CSFP) by lumbar puncture, systemic blood pressure (BP) and pulse rate (PR) were measured for 15 minutes during the induction phase of general anaesthesia in seven groups of six healthy female patients each. Intravenous drugs, thiopentopne 5 mg . kg-1, alfathesin 50 microliters . kg-1 and diazepam 0.5 mg . kg-1 given in 10 to 20 seconds caused a fall of CSFP and BP, whereas ketamine 2 mg . kg-1 and a three-minute induction with halothane three per cent, trichloroethylene one per cent, or methoxyflurane 0.75 per cent caused a sharp highly significant but short-lived

Chinese hamster fibroblast cells in stirred suspension cultures were exposed to volatile anaesthetic agents for 3 hr or for 24 hr. Cell viability was measured as colony forming ability and was found to be reduced, in a dose related manner, by halothane, chloroform, methoxyflurane, trichloroetheylene and diethyl ether.

A simple heated precolumn device constructed from standard gas chromatographic components is described and evaluated. The device permits the analysis of concentrations of anaesthetic agents by direct injection of whole blood containing a suitable internal standard. A method is described for the analysis of halothane, chloroform, trichloroethylene, and methoxyflurane. The procedure is rapid and of acceptable accuracy.

Nephrotoxicity can be grouped by the xenobiotics place of action, by the clinical presentation or by the generic toxic effect. The latter can be dose related, indirect, idiosyncratic or allergic. Nephrotoxicity of lithium, demeclocycline, aminoglycosides, cyclosporine, mercuric ion, nonsteroidal anti-inflammatory drugs, methoxyflurane, ethylene glycol, D-penicillamine and methicillin is reviewed in light of all these three viewpoints, but emphasis is on toxic mechanisms.

The hydro-osmotic response of the toad bladder to antidiuretic hormone and cyclic AMP was inhibited by the methoxyflurane metabolite, fluoride. The osmotic transfer of water in the absence of hormone was unaffected by fluoride as was the hydroosmotic response due to hypertonicity of the serosal bathing media. Fifteen related dogs were studied for susceptibility to malignant hyperthermia using halothane challenge and caffeine contracture tests. These dogs had hypertrophied muscles, were of a nervous temperament and had rectal temperatures at the upper limit of the normal range. Anesthetic agents are used in laboratory animals to prevent pain or distress due to an experimental procedure or for restraint to facilitate a technically difficult procedure. This unit provides three basic protocols: injectable anesthesia for mouse, rat, and hamster; inhalant anesthesia using methoxyflurane for mouse, rat, and hamster; and injectable anesthesia using ketamine/xylazine for rabbit. An Alternate Protocol describes sedation using butorphanol/acetylpromazine in the rabbit. The Commentary further describes and compares these methods of anesthesia for various applications.

The TM3 Gas differentiator, a simple device introduced to differentiate between nitrous oxide and oxygen, was found to be capable of positively distinguishing between these gases when only they were known to be present. The differentiator gave nearly equal responses to nitrous oxide and carbon dioxide. Cyclopropane, halothane, methoxyflurane, trichloroethylene and nitrogen all produced smaller responses on the differentiator's built-in meter. This apparatus has proved in use to be a practicable and simple method of checking the composition of gases emerging from pipelines.

Two cases are reported of combined renal and hepatic failure following exposure to halothane anesthesia. Both patients presented with postoperative fever and rapidly deteriorating liver and kidney function. Both required peritoneal dialysis.
