Hydrogen Sulfide Mitigates Kidney Injury in High Fat Diet-Induced Obese Mice.

Obesity is prevalent worldwide and is a major risk factor for the development and progression of kidney disease. Hydrogen sulfide (H2S) plays an important role in renal physiological and pathophysiological processes. However, whether H2S is able to mitigate kidney injury induced by obesity in mice remains unclear. In this study, we demonstrated that H2S significantly reduced the accumulation of lipids in the kidneys of high fat diet- (HFD-) induced obese mice. The results of hematoxylin and eosin, periodic acid-Schiff, and Masson's trichrome staining showed that H2S ameliorated the kidney stru

Glucagon receptor antagonist and GIP agonist combination for diet-induced obese mice.

Ablation of glucagon receptor signaling represents a potential treatment option for type 2 diabetes (T2DM). Additionally, activation of glucose-dependent insulinotropic polypeptide (GIP) receptor signaling also holds therapeutic promise for T2DM. Therefore, this study examined both independent and combined metabolic actions of desHis(1)Pro(4)Glu(9)(Lys(12)PAL)-glucagon (glucagon receptor antagonist) and d-Ala(2)GIP (GIP receptor agonist) in diet-induced obese mice. Glucagon receptor binding has been linked to alpha-helical structure and desHis(1)Pro(4)Glu(9)(Lys(12)PAL)-glucagon displayed enha

Morin attenuates hepatic insulin resistance in high-fat-diet-induced obese mice.

Morin is a natural bioflavonoid that exhibits antioxidant and anti-inflammatory properties. The present study was designed to evaluate the effect of morin on insulin resistance, oxidative stress, and inflammation in a high-fat-diet (HFD)-induced obese mice. Obesity was induced in ICR mice by feeding a HFD (60 % kcal from fat) for 12 weeks. After the first 6 weeks, obese mice were treated with morin (50 or 100 mg/kg/day) once daily for further 6 weeks. Blood glucose, lipid profile, insulin, leptin, adiponectin, and markers of oxidative stress and inflammation were then measured. Liver was excis

Moderate physical activity promotes basal hepatic autophagy in diet-induced obese mice.

Obesity is a known risk factor for the development of hepatic disease; obesity-induced fatty liver can lead to inflammation, steatosis, and cirrhosis and is associated with degeneration of the mitochondria. Lifestyle interventions such as physical activity may ameliorate this condition. The purpose of this study was to investigate regulation of mitochondrial and autophagy quality control in liver following Western diet-induced obesity and voluntary physical activity. Eight-week-old C57BL/6J mice were fed a Western diet (WD) or normal chow (NC, control) for 4 weeks; afterwards, groups were divi

Rapamycin-induced metabolic defects are reversible in both lean and obese mice.

A poor uterine environment causes changes in fetal development that affect the health of offspring long-term. Although there are multiple pathways that contribute to the development of cardiovascular and neurological disease, low birth weight or fetal growth restriction (FGR) predisposes offspring to these diseases. There is a link between fetal exposure to adverse influences and hypertension later in life. Many epidemiological studies support the link between fetal life and the risk of disease later in life. Experimental models have sought to provide mechanistic proof of this link while simul

Losartan and ozagrel reverse retinal arteriolar constriction in non-obese diabetic mice.

OBJECTIVE: Reductions in retinal blood flow are observed early in diabetes. Venules may influence arteriolar constriction and flow; therefore, we hypothesized that diabetes would induce the constriction of arterioles that are in close proximity to venules, with the constriction mediated by thromboxane and angiotensin II. METHODS: Using nonobese diabetic (NOD) mice, retinal measurements were performed three weeks following the age at which glucose levels exceeded 200 mg/dL, with accompanying experiments on age-matched normoglycemic NOD mice. The measurements included retinal arteriolar diamete

The concept of selective leptin resistance: evidence from agouti yellow obese mice.

Leptin, a hormone secreted by adipose tissue, acts to inhibit appetite and promote metabolism, thereby reducing body weight. Leptin also increases sympathetic activity and arterial pressure. Several murine models of obesity, including agouti obese mice, exhibit resistance to the anorexic and weight-reducing effects of leptin. Hypertension in agouti mice has been attributed to hyperleptinemia. These observations pose a seeming paradox. If these mice are leptin-resistant, then how can leptin contribute to hypertension? We tested the novel hypothesis that these mice have selective leptin resistan

Involvement of COX inhibitor and arachidonic acid in manipulating obesity and obesity-induced bone resorption markers in obese mice.

Obesity and bone-loss have remained a focus of research. Obesity stimulates adipose tissue expansion and adipocyte hypertrophy, resulting in chronic low-grade inflammation in the adipocytes. This enlarged adipocyte secretes a variety of pro-inflammatory chemicals. Because of their endocrine signaling, these substances indirectly promote osteoclast activity and bone-loss. However, the role of COX-2 signaling in obesity-induced bone resorption gene expression has yet to be investigated. Thus, we conducted this study in the context of obesity, employing a high-fat diet-induced obese mouse model.

Pre-obese and obese agouti mice are sensitive to the anorectic effects of peptide YY(3-36) but resistant to ghrelin.

OBJECTIVE: The role of the melanocortin system in the feeding effects of peripheral peptide YY(3-36) (PYY(3-36)) and ghrelin was investigated using the agouti (A(y)/a) mouse as a model of abnormal melanocortin signalling. Furthermore, we examined whether the ectopic expression of agouti protein in A(y)/a mice results in complete MC4-R inhibition, by studying the effects of peripheral alpha-melanocyte-stimulating hormone (alpha-MSH) and leptin on food intake. DESIGN: Adult A(y)/a mice were studied in the pre-obese state (7-8 weeks) and obese state (14-15 weeks). Animals received PYY(3-36) (0.0

Intermittent peripheral exposure to lipopolysaccharide induces exploratory behavior in mice and regulates brain glial activity in obese mice.

BACKGROUND: Consecutive peripheral immune challenges can modulate the responses of brain resident microglia to stimuli. High-fat diet (HFD) intake has been reported to stimulate the activation of astrocytes and microglia in the arcuate nucleus (ARC) of the hypothalamus in obese rodents and humans. However, it is unknown whether intermittent exposure to additional peripheral immune challenge can modify HFD-induced hypothalamic glial activation in obese individuals. METHODS: In this study, we administered 1 mg/kg LPS (or saline) by intraperitoneal (i.p.) injection to 8-week-old male mice after

Quantitative modeling of insulin signaling reveals mechanisms of insulin resistance in obese mice.

Insulin resistance is a deficiency in insulin-mediated regulation of glucose metabolism and is considered a cause of multiple diseases such as diabetes and hypertension. Because the insulin signaling pathway plays a central role in insulin action, it has been extensively studied in the context of insulin resistance. However, the insulin signaling pathway's contribution to insulin resistance remains incompletely understood. Insulin receptor (IR) mutations can cause insulin resistance; however, reduced IR expression in obesity may still permit sufficient signaling due to the characteristics of I

Myofiber stearoyl-CoA desaturase 1 moderates MAFLD and metabolic syndrome in obese mice.

INTRODUCTION: Obesity-induced ectopic lipid accumulation inside skeletal muscle (SKM) (myosteatosis) and the liver (hepatosteatosis) contributes to local metabolic dysfunction and serves as a launchpad for the progression of diabetes. While stearoyl-CoA desaturase 1 (SCD1), an ER-bound enzyme that catalyzes the rate-limiting step in monounsaturated fatty acid (MUFA) synthesis, is widely recognized as a lynchpin facilitator of steatosis, much controversy exists on its efficacy as a therapeutic target. In particular, a total lack of SKM-specific SCD1 KO models has greatly limited our understandi

Liraglutide modulates ALCAT1-Mediated cardiolipin remodeling to improve cardiac function in obese mice.

Obesity, a significant risk factor for cardiovascular diseases, induces cardiolipin (CL) remodeling. Acyl-CoA:lysocardiolipin acyltransferase-1 (ALCAT1), a key enzyme in CL metabolism, drives mitochondrial impairment and cardiac dysfunction in obesity. Although glucagon-like peptide-1 receptor agonists (GLP-1RAs) exhibit cardioprotective properties, their effects on ALCAT1-mediated CL remodeling in obesity-induced myocardial injury remain unclear. Male C57BL/6 mice fed a high-fat diet (HFD) or standard diet (STD) for 12 weeks received liraglutide (200 μg/kg/day) or saline during the last 4 wee

Diminazene aceturate, an ACE2 activator, ameliorates testicular injury in diet-induced obese mice.

IN BRIEF: Obesity is a prevalent health concern globally, often associated with testicular damage that can lead to male infertility. This study investigates the improvement of obesity-induced testicular damage by ACE2 agonist diminazene aceturate (DIZE) and explores the role of the ACE2/Ang 1-7/MasR axis in this process. ABSTRACT: DIZE improved obesity and metabolic disturbances in diet-induced obesity (DIO) mice. An increase in sperm count and motility, along with improved morphology and increased male fertility, was observed after DIZE treatment. Both serum and intratesticular testosterone

Tirzepatide improves neutrophilic inflammation in obese asthmatic mice by downregulating Th17 cell differentiation.

BACKGROUND: Asthma is a chronic respiratory disease affecting more than 330 million people worldwide. Obesity not only increases the risk of asthma attacks but also worsens symptoms and diminishes responsiveness to standard drug treatments. Tirzepatide, a dual GIP and GLP-1 receptor agonist, has demonstrated superior glycemic control and weight loss effects compared to selective GLP-1 receptor agonists in clinical trials. However, its impact on neutrophilic inflammation in obese asthmatic mice and the underlying molecular mechanisms remain unclear. METHODS: This study first established an obes

Obese-diabetic female Ksr2 knockout mice develop brittle bones near end of life.

The influence of obesity and type 2 diabetes (T2D) on the skeleton is complex, with affected individuals having higher fracture risk despite having higher BMD. To evaluate how obesity and T2D affect skeletal health, we studied mice with disruption of a gene that regulates energy intake and expenditure, Ksr2, which results in reduced metabolic rate and severe insulin resistance in both mice and a subpopulation of humans. Relative to 28-wk-old littermate sibling controls, Ksr2 mutants weighed more than double the body and fat weight. Moreover, leptin and insulin were elevated by 20- and 10-fold

Positive Regulation of Acetate in Adipocyte Differentiation and Lipid Deposition in Obese Mice.

Acetate is associated with adipocyte differentiation and lipid deposition. To further develop this scientific point, obese mice on a high-fat diet were given an intragastric administration of acetate for 8 weeks and mouse adipose mesenchymal stem cells (mAMSCs) were treated with acetate for 24 h. The results showed that the body weight, food intake, Lee's index, adipose tissue coefficient, liver index, blood lipid levels, insulin resistance, pro-inflammatory factors levels and fatty lesions in liver and adipose tissue in obese mice treated with acetate increased markedly, while anti-inflammato

Brown adipocyte mineralocorticoid receptor deficiency impairs metabolic regulation in diet-induced obese mice.

Activation of brown adipose tissue (BAT) contributes to energy dissipation and metabolic health. Although mineralocorticoid receptor (MR) antagonists have been demonstrated to improve metabolism under obesity, the underlying mechanisms remain incompletely understood. We aimed to evaluate the role of BAT MR in metabolic regulation. After 8 weeks of high-fat diet (HFD) feeding, BAT MR KO (BMRKO) mice manifested significantly increased bodyweight, fat mass, serum fasting glucose, and impaired glucose homeostasis compared with littermate control (LC) mice, although insulin resistance and fasting s
