The effects of water sample treatment, preparation, and storage prior to cyanotoxin analysis for cylindrospermopsin, microcystin and saxitoxin.

Cyanobacterial harmful algal blooms occur in freshwater lakes, ponds, rivers, and reservoirs, and in brackish waters throughout the world. The wide variety of cyanotoxins and their congeners can lead to frequent exposure of humans through consumption of meat, fish, seafood, blue-green algal products and water, accidental ingestion of contaminated water and cyanobacterial scum during recreational activities, and inhalation of cyanobacterial aerosols. Cyanotoxins can also occur in the drinking water supply. In order to monitor human exposure, sensitive analytical methods such as enzyme linked im

Protective efficacy and the recovery profile of certain chemoprotectants against lethal poisoning by microcystin-LR in mice.

The cyclic peptide toxins microcystins and nodularins are the most common and abundant cyanotoxins present in diverse water systems. They have been the cause of human and animal health hazards and even death. Development of suitable chemoprotectants against microcystin is essential considering the human health importance. In the present study, three agents cyclosporin-A (10mg/kg), rifampin (25mg/kg) and silymarin (400mg/kg) pre-treatment gave 100% protection against lethal dose of microcystin-LR (100 microg/kg). Various biochemical parameters were evaluated to study the recovery profile of pro

Pivotal role of mitochondrial Ca(2+) in microcystin-induced mitochondrial permeability transition in rat hepatocytes.

We have shown earlier that microcystin-LR (MLR), a specific hepatotoxin, induced onset of mitochondrial permeability transition (MPT) and apoptosis in rat hepatocytes. Here we attempted to investigate the role of mitochondrial Ca(2+) in MLR-induced onset of MPT and cell death. Using confocal microscopy, we found that MLR caused an early surge of mitochondrial Ca(2+) prior to the onset of MPT and cell death. Pretreatment with 1,2-bis(O-aminophenoxyl)ethane-N,N,N',N'-tetracetic acid tetra(acetoxymethyl)ester (an intracellular Ca(2+) chelator) or ruthenium red (an inhibitor of mitochondrial Ca(2+

Identification of an enzymatically formed glutathione conjugate of the cyanobacterial hepatotoxin microcystin-LR: the first step of detoxication.

Cyanobacterial toxins have adverse effects on mammals, birds and fish and are being increasingly recognised as a potent stress factor and health hazard factor in aquatic ecosystems. Microcystins, cyclic heptapeptides and a main group of the cyanotoxins are mainly retained within the producer cells during cyanobacterial bloom development. However, these toxins are released into the surrounding medium by senescence and lysis of the blooms. Any toxin present could then come into contact with a wide range of aquatic organisms including phytoplankton grazers, invertebrates, fish and aquatic plants.

Engineering a nanobody-alkaline phosphatase bifunctional probe for enhanced immunoassay performance toward microcystin-LR.

Eutrophication caused by excessive nutrient discharge has led to the frequent occurrence of cyanobacterial blooms, during which toxic metabolites such as microcystins (MCs) are released into aquatic environments. Among these, microcystin-LR (MC-LR) is the most widespread and toxic congener, posing serious threats to both ecological safety and human health. In this study, a novel bifunctional detection probe (A2.3-AP) was developed by genetically fusing a MC-LR-specific nanobody (A2.3) with alkaline phosphatase (AP) via a flexible (GGGGS)3 linker. The A2.3-AP bifunctional detection probe integr

Transporter-Mediated Uptake of Microcystin-LR in Human Trophoblasts: Regulation By Oxygen Concentration and Cell Fusion.

BACKGROUND: Rising global temperatures and eutrophication are increasing the intensity and frequency of cyanobacterial harmful algal blooms that release toxins including microcystin-LR (MC-LR). MC-LR inhibits protein phosphatases in the human liver and brain, but its accumulation in the placenta is unclear. Placental transporter expression varies across pregnancy and is influenced by physiological cues, such as low oxygen concentrations which activate HIF1A, and trophoblast cell fusion forming syncytiotrophoblasts that engage CREB-driven transcription. This study examined whether MC-LR accumul

Assessing the risk of cyanobacterial toxins in freshwater ecosystems: microcystin-LR exposure in Chironomus riparius larvae.

Cyanobacteria produce a variety of secondary metabolites, including the potent and widespread toxin microcystin-LR (MC-LR). While the toxic effects of MC-LR on aquatic species are well documented, its impact on Chironomid larvae, key organisms in freshwater food chains, remains insufficiently understood. This study investigated the acute toxicity of MC-LR at environmentally relevant concentrations (5, 10, 15, and 30 µg/L) on Chironomus riparius (Diptera, Chironomidae). The effects of the tested concentrations were evaluated using a multi-biomarker approach, including hemoglobin levels, oxidati

Nanozyme mediated Raman-NLISA dual-modal immunosensor for accurate and sensitive detection of microcystin-LR.

A Raman scattering and nanozyme-linked immunosorbent assay (NLISA) dual modal immunosensor, was constructed by mesoporous SiO2/Au-Pt nanozymes (m-SAP) and nanobodies (A2.3-SBP). Oxidized TMB served as Raman and ELISA signals in a competitive binding assay. Under optimized conditions, an inverse correlation was established between the Microcystin-LR (MC-LR) concentration and the signals, spanning Raman and ELISA ranges of 0.1-100 μg L-1 and 1.0-500 μg L-1, with limit of detections (LODs, 3σ/S) of 0.015 μg L-1 and 0.12 μg L-1, respectively. The LODs showed over 90 times and 11 times higher sensi

Gold nanobipyramid-based colorimetric sensor for rapid on-site detection of microcystin-LR in drinking water.

In recent years, algal blooms have occurred frequently. Microcystin-LR (MC-LR), a harmful metabolite produced by cyanobacterial cells, is widely distributed in water environments worldwide. MC-LR can enter the human body through drinking water. Humans may suffer from its severe hepatotoxicity, which in extreme situations may result in fatalities and incidents involving public health and safety. Therefore, a detection method based on chicken yolk antibody, immunomagnetic separation technology and gold nanobipyramids (AuNBPs) colorimetric technology was developed for visual qualitative and semi-

Unraveling the mechanisms underlying the fluorescent probe detection of microcystin-LR and its binding with CT-DNA.

Cyanobacteria blooms are concerning due to algal toxins like microcystin-leucine arginine (MC-LR). Despite progress in detecting MC-LR and understanding its toxic effects, including calf thymus DNA (CT-DNA) damage, the mechanisms for fluorescent probe detection of MC-LR and its binding to CT-DNA are poorly understood. In this study, we designed three fluorescent probes for MC-LR detection. Probe 1, with an acidic recognition site, is effective but influenced by solution pH. Probe 2, featuring a benzene ring structure, shows stable detection regardless of pH. Probe 3 offers the best performance

Oxidative stress and mitochondrial dysfunctions induced by cyanobacterial microcystin-LR in primary grass carp hepatocytes.

Microcystin-LR (MC-LR), a cyclic heptapeptide produced by freshwater cyanobacteria, induces a range of liver injuries. However, the mechanisms underlying MC-LR toxicity in primary hepatocytes of aquatic organisms remains poorly understood. In this study, we investigated the effects of MC-LR on oxidative stress and mitochondrial function using primarily cultured grass carp hepatocytes. The results revealed that IC50 of MC-LR on grass carp primary liver cells for 24 hours was 2.40 μmol/L. Based on 24h-IC50, concentrations of 0, 0.30, 0.60, and 1.20 μmol/L were used in subsequent experiments. MC-

Environmental factors driving microcystin contamination of estuarine bivalve populations downstream of freshwater cyanobacterial blooms.

Estuaries are dynamic environments that provide vital habitat to ecologically and commercially important bivalves. In some cases, freshwater tributaries can introduce cyanobacteria and associated cyanotoxins into estuaries that may subsequently accumulate in estuarine bivalves. Temporarily open/closed estuaries (TOCEs), which only experience tidal input for limited periods of time, may be particularly vulnerable to the accumulation of cyanotoxins in bivalves as they can be subject to freshwater input without tidal flushing and may experience lower salinities and cyanobacterial blooms. This stu

Microplastics as vectors for microcystin-LR: Enhanced bioavailability, oxidative damage, and immune dysregulation in zebrafish spleen.

The co-occurrence of eutrophication and plastic pollution has emerged as a critical global water quality challenge. In the present study, we systematically investigated the joint effects of microcystin-LR (MC-LR) and polystyrene microplastics (PSMPs) on antioxidant defense mechanisms and innate immunity function through an orthogonal experimental design (MC-LR: 0, 0.8, 4, 20 μg/L; PSMPs: 0, 100 μg/L). After 60 d exposure, our results found that the presence of PSMPs enhanced the MC-LR bioavailability in the spleen of zebrafish. Besides, the severity of splenic injuries exhibited a concentratio

Single microcystin exposure impairs the hypothalamic-pituitary-gonadal axis at different levels in female rats.

Microcystin (MC) is most common cyanobacterial toxin. Few studies have evaluated the MC effects on the hypothalamic-pituitary-gonadal (HPG) axis and metabolic function. In this study, we assessed whether MC exposure results in HPG axis and metabolic changes. Female rats were exposed to a single dose of MC at environmentally relevant levels (5, 20 and 40 μg/kg). After 24 h, we evaluated reproductive and metabolic parameters for 15 days. MC reduced the hypothalamic GnRH protein expression, increased the pituitary protein expression of GnRHr and IL-6. MC reduced LH levels and increased FSH levels

Liver Lesions in Estuarine Dolphins in the Indian River Lagoon, Florida: Does Microcystin Play a Role?

Microcystin (MC), a hepatotoxin produced by cyanobacteria, was introduced into the Indian River Lagoon (IRL), Florida, in 2005 through freshwater outflows. Since then, MC has been detected in humans, domestic animals, and wildlife in the lagoon. Potential public health effects associated with MC exposure along the IRL include an increased risk of non-alcoholic liver disease among area residents. Yet, there are limited studies characterizing liver disease, as well as the potential role of MC, in humans and animals in this region. Thus, histopathology reports (n = 133) were reviewed in the stran

Polystyrene microplastics enhance the microcystin-LR-induced gonadal damage and reproductive endocrine disruption in zebrafish.

The coexistence of eutrophication and plastic pollution in the aquatic environment is becoming a realistic water pollution problem worldwide. To investigate the microcystin-LR (MC-LR) bioavailability and the underlying reproductive interferences in the presence of polystyrene microplastic (PSMPs), zebrafish (Danio rerio) were exposed to individual MC-LR (0, 1, 5, and 25 μg/L) and combined MC-LR + PSMPs (100 μg/L) for 60 d. Our results showed that the existence of PSMPs increased the accumulation of MC-LR in zebrafish gonads compared to the MC-LR-only group. In the MC-LR-only exposure group, se

Ginsenoside Rg3 improves microcystin-induced cardiotoxicity through the miR-128-3p/MDM4 axis.

Microcystin (MC) is the byproduct of cyanobacteria metabolism that is associated with oxidative stress and heart damage. This study aimed to investigate the effect of ginsenoside Rg3 on MC-induced cardiotoxicity. A mouse model of myocardial infarction was constructed by oral MC administration. H9C2 cells were used for in vitro analysis. Cellular oxidative stress, apoptosis, and the relationship between miR-128-3p and double minute 4 protein (MDM4) were analyzed. MiR-128-3p expression was upregulated in vitro and in vivo after MC treatment, which was downregulated after Rg3 treatment. Left vent

Microcystin - LR exposure causes cardiorespiratory impairments and tissue oxidative damage in trahira, Hoplias malabaricus.

This study investigated the effect of microcystin-LR (MC-LR) on in vivo cardiorespiratory function and on tissue biomarkers of oxidative stress in gills and liver of the trahira, a neotropical freshwater fish. Trahira were treated with an intraperitoneal injection of 100 µg MC-LR.kg-1 body mass or a saline, with the toxic effects of MC-LR then evaluated after 48 h. Rates of oxygen uptake (V̇O2) did not differ significantly between Control and the exposed group (Mcys), but exposure to MC-LR significantly reduced O2 extraction in the Mcys group at all O2 tensions. This was associated with higher
