In this report, we show that a novel capillary-based photopolymerized monolith offering unprecedented efficiency (approximately 80%) for DNA extraction from submicroliter volumes of whole blood (Wen, J.; Guillo, C.; Ferrance, J. P.; Landers, J. P. Anal. Chem. 2006, 78, 1673-1681) can be translated to microfluidic devices. However, owing to the large mass of protein present in blood, both DNA binding capacity and extraction efficiency were significantly decreased when extraction of DNA was carried out directly from whole blood (38+/-1%). To circumvent this, a novel two-stage microdevice was dev

Articular cartilage is a load-bearing connective tissue whose structural integrity depends on a highly specialized extracellular matrix and the activity of resident chondrocytes. In vivo, chondrocytes are continuously exposed to mechanical stimuli, including fluid-induced shear stress associated with synovial fluid movement, which plays a critical role in regulating cell phenotype and matrix homeostasis. Osteoarthritis (OA) is a degenerative joint disease marked by extracellular matrix breakdown, chondrocyte dysfunction, and inflammation. Despite its high prevalence, effective disease-modifyin

As the population ages, the worldwide prevalence of Alzheimer's disease (AD) as the most common dementia in the elderly is increasing dramatically. However, a long-term challenge is to achieve rapid and accurate early diagnosis of AD by detecting hallmarks such as amyloid beta (Aβ42). Here, a multi-channel microfluidic-based plasmonic fiber-optic biosensing platform is established for simultaneous detection and differentiation of multiple AD biomarkers. The platform is based on a gold-coated, highly-tilted fiber Bragg grating (TFBG) and a custom-developed microfluidics. TFBG excites a high-den

We report on a microfluidic platform for culture of whole organs or tissue slices with the capability of point access reagent delivery to probe the transport of signaling events. Whole mice retina were maintained for multiple days with negative pressure applied to tightly but gently bind the bottom of the retina to a thin poly-(dimethylsiloxane) membrane, through which twelve 100 μm diameter through-holes served as fluidic access points. Staining with toluidine blue, transport of locally applied cholera toxin beta, and transient response to lipopolysaccharide in the retina demonstrated the cap

The information regarding tumor cells, the immune environment and microorganisms can provide insights into the formation and progression of gastric cancer (GC) from various perspectives. However, there remains a need for a diagnostic strategy capable of integrating and analyzing these multiple dimensions to accurately assess the progression of GC. Extracellular vesicles (EVs), which carry multidimensional information, represent a promising non-invasive biomarker for the diagnosis and monitoring of GC. Despite their potential, the development of an efficient method for the enrichment and precis

Towards addressing the need for detecting and eliminating biothreats, we describe a micromotor-based approach for screening, capturing, isolating and destroying anthrax simulant spores in a simple and rapid manner with minimal sample processing. The B. globilli antibody-functionalized micromotors can recognize, capture and transport B. globigii spores in environmental matrices, while showing non-interactions with excess of non-target bacteria. Efficient destruction of the anthrax simulant spores is demonstrated via the micromotor-induced mixing of a mild oxidizing solution. The new micromotor-

Quantifying CD11b in whole blood from patients with sepsis is analytically challenging because conventional preprocessing can perturb activation state and bias the measured surface-marker abundance. To address this limitation, we report a biomimetic-acoustic cooperative platform for rapid CD11b-based neutrophil activation readout. CD11b expression is transduced into a quantitative microsphere-depletion signal using a competitive microsphere format, and residual microspheres are identified and counted by an object-detection model assisted by frequency-domain image-quality features to improve ro

Quantifying trace amounts of magnetic iron in living cells remains a major challenge in cellular biology and nanomedicine. Existing approaches such as magnetometry or elemental analysis either require large cell populations or cannot discriminate endogenous iron from magnetic phases, preventing measurements at the femtogram-per-cell scale. Here, we introduce a high-gradient magnetophoresis approach combined with multicellular spheroids to achieve femtogram-level sensitivity in intact 3D cellular assemblies. By exploiting the scaling of magnetic force with spheroid volume and hydrodynamic drag

Microfluidic platforms produce RNA-loaded lipid nanoparticles (RNA-LNPs) with superior uniformity, encapsulation efficiency, and control over size compared to bulk methods, and unlike bulk approaches, their throughput can be scaled over orders of magnitude via parallelization without altering the formulation. Despite these advantages, continuous manufacturing of RNA-LNPs by microfluidics has been hindered by rapid fouling of microchannels exposed to the complex mixtures of lipids, cholesterol, and RNA in mixed aqueous-organic solvents used for LNP formulation, degrading chip output quality ove

Precise, sheathless manipulation of blood microscale components remains a central challenge in microfluidics, particularly for Lab-on-a-Disc (LOD) platforms where inertial, rotational, and electrokinetic effects coexist. This simulation study proposes a two-phase particle-handling strategy that integrates rotation-assisted inertial focusing with high-frequency dielectrophoretic (DEP) sorting inside serpentine microchannels. In phase I, three channel geometries─an asymmetric sinusoidal, symmetric sinusoidal, and rectangular layout─were systematically evaluated to determine their ability to conf

Freshwater planarian, a model organism renowned for its remarkable regenerative capabilities, is widely used in studies of regenerative mechanisms and stem cell biology. However, current research faces challenges in automated culture, long-term tracking, and real-time in vivo imaging at the single-worm level, limiting the efficient acquisition of accurate, individual-specific data and impeding deep investigation into regeneration dynamics. To address this, we developed a planarian microfluidic chip and an integrated microdevice for the automated cultivation, individual tracking, and in vivo im

Image-activated cell sorting (IACS) enables high-speed sorting of cells based on real-time analysis of their images, providing a powerful means to link cellular morphology and function at the single-cell level on a large scale. Although IACS has been demonstrated for a variety of small-to-medium-size cells, applying it to large cells, cell clusters, and other large objects remains challenging, despite the scientific and industrial value in morphology-based sorting of such objects. The main difficulty lies in controlling large and complex cells throughout a microfluidic chip, from inlet to outl

Vector-borne diseases, including arboviral infections and malaria, pose a major global health threat and underscore the need for rapid, accurate, and multiplex nucleic acid detection technologies for clinical diagnosis and surveillance. However, previous studies on reagent-preloaded and switching valve-based microfluidic cartridge platforms have insufficiently addressed several key engineering challenges, particularly the reliable control of multichannel switching valves and the inherently critical requirement for uniform reagent distribution across multiple reaction zones. These limitations a

Bacteria motility is one of the critical bacterial behavior to exploit available resources and environments due to their biological movement toward resources supporting proliferation. Commonly used techniques investigating bacterial motility include cell staining, DNA sequencing, and fluorescent labeling, which are time consuming and challenged by limited markers and subsequent native bacterial cell high information collections. Distinguishing bacteria with high/low motility potential with simple and fast techniques are in demand. Here, a label-free DC-iDEP (Direct-current insulator based diel

Nanosatellites of CubeSat type due to, i.a., minimized costs of space missions, as well as the potential large application area, have become a significant part of the space economy sector recently. The opportunity to apply miniaturized microsystem (MEMS) tools in satellite space missions further accelerates both the space and the MEMS markets, which in the coming years are considered to become inseparable. As a response to the aforementioned perspectives, this paper presents a microfluidic mixer system for biological research to be conducted onboard CubeSat nanosatellites. As a high complexity

In the current research work, the flow behavior of a liquid-liquid extraction (LLE) process in a serpentine microchannel was analyzed. The simulation was performed using a 3D model and the results were found to be consistent with experimental data. The impact of the flow of chloroform and water on the flow model was also examined. The data indicate that once the aqua and organic phases flow rates are low and similar, a slug flow pattern is observed. However, as the overall flow rate raises, the slug flow transforms into parallel plug flow or droplet flow. An increment in the aqua flows while m

Microfluidic paper-based analytical devices (µPADs) represent one of the promising green analytical strategies for low-cost and simple determination of various analytes. The actual task is the development of such devices for quantitation of antioxidants, e.g., flavonoids. In this paper, possibilities of a novel three-reagent µPAD including silver nitrate, 4-nitrophenyldiazonium tetrafluoroborate, and iron(III) chloride as reagents are assessed with respect to the determination of dihydroquercetin. It is shown that all the three reagents produce different colorimetric responses that can be dete

Neurotransmitters play key roles in cell-to-cell communication. These chemical messengers are involved in many functional processes, including growth, reproduction, memory, and behavior. In this communication, we describe a novel microfluidic paper-based analytical device (μPAD) to detect acetylcholinesterase (AChE) activity and inhibitor screening through a colorimetric analysis. The μPAD is easily fabricated via a wax printing process whereby wax is deposited onto the surface of chromatographic paper, and heated to create a hydrophobic barrier. We report the utility of structured elastomeric membranes (SEMs) as a multifunctional microfluidic tool. These structured membranes are part of a two-layer microfluidic device, analogous to membrane valves, with the novelty that they incorporate topographical features on the roof of the fluid channel. We demonstrate that when the topographical features are recessed into the roof of the fluid channel, actuation of the membrane leads to effective confinement of fluids down to femtolitres in preformed microfluidic containers. Thus, the SEMs in this case function as fluidic traps that could be cou
