Impact of encapsulating a probiotic (Pediococcus pentosaceus Li05) within gastro-responsive microgels on Clostridium difficile infections.

Antibiotic treatment is often followed by Clostridium difficile infection (CDI), which causes severe diarrhea and other health issues. Oral administration of Pediococcus pentosaceus Li05 (Li05) has been shown to have great potential in preventing CDI. However, the viability of Li05 is greatly reduced during storage and passage through the gastrointestinal (GI) tract, which limits its biological activity. In this study, a gastro-responsive microgel was designed to encapsulate and protect Li05 to enhance its efficacy against CDI. The viability of Li05 encapsulated within the microgels was signif

Bioprinted dECM particles-laden microgels enhance the mesenchymal stromal cell paracrine effect for myocardial infarction treatment.

Despite guideline-directed reperfusion and pharmacological therapies, myocardial infarction (MI) continues to pose substantial clinical challenges, with high incidence and mortality rates. Mesenchymal stromal cell (MSC) transplantation represents a promising therapeutic strategy, yet its efficacy remains limited by poor survival and unstable paracrine function of transplanted cells within the hostile infarct microenvironment. Here, we developed an integrated microcarrier system via digital light processing, which encapsulates MSCs within microgels functionalized with cardiac-derived decellular

Microgels Containing Protease Resistant CXCL12 and FasL Enable Long-term Allo-Islet Function Without Systemic Immunosuppression.

Cotransplantation of allogeneic islets with microgels coated with streptavidin fused with Fas ligand and protease-resistant CXCL12 results in robust and sustained diabetes reversal for over 6 months in streptozotocin-induced diabetic mice and induces local immunomodulation, enhances Treg recruitment within the graft, and improves graft vascularization. This study addresses a critical challenge in cell transplantation, namely the need to develop technologies that enable local immunoprotection of the graft while eliminating the need for systemic immunosuppression. This combination microgel techn

Engineering Resatorvid-Loaded Sub-Microgels of Epigallocatechin-3-gallate/Hyaluronic Acid to Treat Acute Lung Injury.

Acute lung injury (ALI) and acute respiratory distress syndrome (ARDS) remain life-threatening conditions with high morbidity and mortality, largely due to excessive reactive oxygen species (ROS) generation and uncontrolled inflammatory cascades. Despite extensive pharmacological investigations, effective and targeted therapies are still lacking. This study develops an innovative hyaluronic acid (HA)-epigallocatechin gallate (EGCG)-amino phenylboronic acid (PBA) based sub-microgel (HAEP sub-microgel) platform capable of integrating multiple dynamic interactions (esterification, boronic ester b

ROS-triggered transformation of microgels to bioadhesive patch for orally administrated treatment of inflammatory bowel disease.

Inflammatory bowel diseases (IBD) are driven by excessive reactive oxygen species (ROS) and progressive disruption of the intestinal mucus barrier, yet sustained mucosal protection remains insufficiently addressed by current therapies. Here, we develop an orally administered, inflammation-activated microgel system (iPatch) that undergoes a ROS-triggered microgel-to-patch transformation specifically at inflamed intestinal sites. Unlike pre-formed hydrogels or enema-based formulations, this in situ transformation enables prolonged mucosal retention, adaptive barrier reconstruction, and sequentia

Combinatorial Microgels for 3D ECM Screening and Heterogeneous Microenvironmental Culture of Primary Human Hepatic Stellate Cells.

Nonalcoholic fatty liver disease affects 30% of the United States population and its progression can lead to nonalcoholic steatohepatitis (NASH), and increased risks for cirrhosis and hepatocellular carcinoma. NASH is characterized by a highly heterogeneous liver microenvironment created by the fibrotic activity of hepatic stellate cells (HSCs). While HSCs have been widely studied in 2D, further advancements in physiologically relevant 3D culture platforms for the in vitro modeling of these heterogeneous environments are needed. In this study, the use of stiffness-variable, extracellular matri

Advances in the development of cyclodextrin-based nanogels/microgels for biomedical applications: Drug delivery and beyond.

Nanogels/microgels are swollen cross-linked polymer networks with tunable physicochemical properties and are commonly employed for the effective delivery of hydrophilic drugs. By structural engineering, they can be adapted for the delivery of hydrophobic drugs. Likewise, the use of cyclodextrins (CDs) as pharmaceutical excipients in nanogels drastically boosts the loading capacity of lipophilic drugs while enhancing their stability, bioavailability, and permeability owing to their capability of hosting drugs in their somewhat lipophilic cavity. Here, the synthesis and biomedical applications o

Antimicrobial Activity of Microgels with an Immobilized Copper(II) Complex Linked to Cross-Linking and Composition.

The resistance of many bacteria against currently available antimicrobial agents is increasing worldwide at an alarming pace. The described structure-activity relationship study was prompted by the extraordinary ability of water-dispersed microgels to hydrolyze glycosidic bonds similar to building blocks of the peptidoglycan layer of Gram-positive bacteria. The results establish polyacrylate microgels with embedded copper(II) complex as antimicrobial agents. The systematic study reveals that Staphylococcus aureus is susceptible to the microgels, while common commercial agents are found interme

Walnut and peanut protein microgel particles: production, characterization, and application in stabilizing emulsions and gels.

The functional differences between peanut protein (PPI) and walnut protein (WPC) in constructing microgel particles and the structure-property regulation mechanism in stabilizing emulsions and forming gels were investigated in this study. The results revealed that superior stability was exhibited by the high internal phase emulsions (HIPPEs) prepared therefrom due to the stronger hydrophobicity possessed by walnut protein microgel particles (WPC-MPs). In contrast, enhanced gel-forming ability and higher gel strength were observed in HIPPEs prepared from peanut protein microgel particles (PPI-M

An all-in-one microgel-derived cellulosic triboelectric foam for robust and sensitive CO2 monitoring.

The reliance of conventional carbon dioxide (CO₂) sensors on external power limits their application scope. Self-powered triboelectric nanogenerators offer a promising alternative, yet the development of high-performance triboelectric sensing materials remains a key challenge. This study presents an ingenious design using microgels from polyethyleneimine (PEI)-crosslinked nanocellulose as dual-functional building blocks to fabricate a mechanically robust and CO2-sensitive triboelectric foam. The microgels (optimized with 25,000 Da PEI) formed an optimal pore-forming template, facilitating the

Beetle-inspired responsive photonic microgel assemblies for multi-sensing enhanced by machine learning.

Bioinspired photonic hydrogels hold promise as sensors; however, their use in triple-analyte sensing optical devices has been minimally explored. Temperature, serum Fe3+ levels, and X-ray doses are critical factors for predicting and monitoring medical and health issues. For the first time, poly(N-isopropylacrylamide-co-vinyl ferrocene) microgel-based assemblies, also known as interferometers, integrated with machine learning (ML) are introduced for detecting these analysts of interest. The interferometers demonstrate the exceptional capability for sensing with superior sensitivity, reversibil

Microgel-encapsulated tetrandrine nanoparticles promote spinal cord repair by sustaining neuroinflammation inhibition.

Traumatic spinal cord injury (SCI) initiates an intricate secondary injury cascade, characterized by persistent inflammatory responses with neurotoxic microglia and astrocyte activation. Inhibition of neuroinflammation would significantly benefit SCI treatment. Here, tetrandrine with anti-neuroinflammatory activity was delivered into the intrathecal space for SCI treatment. The tetrandrine was encapsulated in MPEG-PDLLA nanoparticles and further loaded into GelMA microgels via a fast 3D printing process based on digital light. After intrathecal injection, the drug-loaded microgels could sustai

Cartilage tissue engineering by extrusion bioprinting utilizing porous hyaluronic acid microgel bioinks.

3D bioprinting offers an excellent opportunity to provide tissue-engineered cartilage to microtia patients. However, hydrogel-based bioinks are hindered by their dense and cell-restrictive environment, impairing tissue development and ultimately leading to mechanical failure of large scaffoldsin vivo. Granular hydrogels, made of annealed microgels, offer a superior alternative to conventional bioinks, with their improved porosity and modularity. We have evaluated the ability of enzymatically crosslinked hyaluronic acid (HA) microgel bioinks to form mature cartilagein vivo. Microgel bioinks wer

In situ formation of osteochondral interfaces through "bone-ink" printing in tailored microgel suspensions.

Osteochondral tissue has a complex hierarchical structure spanning subchondral bone to articular cartilage. Biomaterials approaches to mimic and repair these interfaces have had limited success, largely due to challenges in fabricating composite hard-soft interfaces with living cells. Biofabrication approaches have emerged as attractive methods to form osteochondral analogues through additive assembly of hard and soft components. We have developed a unique printing platform that is able to integrate soft and hard materials concurrently through freeform printing of mineralized constructs within

Cosolvent-Mediated Carbon Dots Aggregation in Microgel Microenvironments for Multicolor Dynamic Fluorescence Modulation.

Dynamic fluorescent materials are attractive for tunable emission colors, but most multicolor systems are limited by a few switching states and complex modulation. To address these challenges, a composite system (MG-CDs) was designed by embedding aggregation-induced color-tuning carbon dots (CDs) into microgels. When respectively swollen in water (H2O), ethanol (EtOH), or ethylene glycol (EG), MG-CDs formed distinct internal hydrogen-bonding networks, yielding varied CDs aggregation states and fluorescence emissions. In an H2O-EtOH-EG cosolvent, MG-CDs established more complex hydrogen-bonding

Optical Detection of Proteins Using Microgel-Stabilized Pickering Liquid Crystal-in-Water Emulsions.

Herein, we present a novel liquid crystal (LC)-based sensing platform utilizing microgel-stabilized Pickering LC droplets dispersed in water for simple and label-free detection of proteins in an aqueous environment. This could be achieved by tailoring the surface of 4-cyano-4'-pentylbiphenyl (5CB) LC droplets dispersed in aqueous medium through the interfacial adsorption of poly(N-isopropylacrylamide) (PNIPAM) microgel particles, followed by the introduction of model surfactants, such as anionic sodium dodecyl sulfate and cationic dodecyltrimethylammonium bromide. Sprayable proteinic adhesive microgel-based immunosuppressive therapeutic coating for effective xenograft transplantation.

Tissue transplantation is an essential therapeutic approach for restoring function in patients with tissue damage caused by aging or accidents. However, immune rejection remains a significant challenge because the host immune system recognizes transplanted xenografts as foreign, triggering systemic inflammatory responses and graft failures. Although conventional oral or intravenous immunosuppressive therapies can mitigate immune rejection, they are often associated with severe systemic side effects and increased risk of infection. To address these challenges, a sprayable proteinic adhesive mic

3D Printable Poly(N-isopropylacrylamide) Microgel Suspensions with Temperature-Dependent Rheological Responses.

Microgel suspensions have garnered significant interest in fundamental research due to their phase transition between liquid-like to paste-like behaviors stemming from tunable interparticle and particle-solvent interactions. Particularly, stimuli-responsive microgels undergo faster volume changes in response to external stimuli in comparison to their bulk counterparts, while maintaining their structural integrity. Here, concentrated and diluted suspensions of poly(N-isopropylacrylamide) (PNIPAm) microgels are dispersed to different packing fractions in water for the characterizations of temper

Intravitreal long-term sustained ranibizumab delivery using injectable microgel-embedded hydrogel.

Retinal vascular disease is the leading cause of visual impairment. Although intravitreal drug injections are the most suitable approach for addressing retinal disorders, existing clinical treatments necessitate repeated administration, imposing a substantial burden on patients with various intraocular complications. This study introduces an injectable and biodegradable hyaluronan microgel (Hm)-embedded gelatin-poly(ethylene glycol)-tyramine hydrogel (HmGh) designed for sustained intravitreal ranibizumab (RBZ) delivery to reduce patient burden and minimize the side effects associated with freq
