Biodegradable microspheres containing adriamycin in the treatment of proliferative vitreoretinopathy.

The use of biodegradable polymer microspheres containing adriamycin for the treatment of proliferative vitreoretinopathy (PVR) in an experimental rabbit model was investigated. A single injection of microspheres containing 10 micrograms of adriamycin effectively decreased traction retinal detachment to 10% (n = 10), whereas 50% of eyes injected with blank microspheres (n = 10) developed retinal detachment (P < 0.05). A single injection of microspheres, containing 3 micrograms of adriamycin, did not suppress retinal detachment. Electroretinographic and histologic studies confirmed that the 10 m

Temporary chemoembolization of colorectal liver metastases with degradable starch microspheres.

Increasing drug delivery to the tumor should induce improved tumor response. To study this the effect of degradable starch microspheres (DSM) and mitomycin was evaluated in 11 patients with chemoembolization of colorectal liver metastases (CRLM) and previous floxuridine (FUDR) treatment. In 10 patients access to the hepatic artery was obtained either by infusaid pump or infusion chambers. Indications for chemoembolization were: Failure of continuous FUDR treatment (n = 7), biliary sclerosis (n = 2), incomplete liver perfusion (n = 2), extensive disease (n = 2). Preliminary observations showed

Submicron Perovskite Quantum Dot Glass Microspheres for Micro-LED Displays.

Quantum-dot (QD) color-conversion technology is considered a promising strategy for constructing a full-color micro-LED display. Perovskite quantum dots (PQDs) are the preferred luminescent materials for constructing color-conversion micro-LED pixels, but their poor environmental stability severely limits their practical application in micro-LED displays. Here, we design a novel submicron-sized PQD glass microspheres (PQDGMS) with high quantum yield and excellent stability for color conversion micro-LED displays. Kilogram scale (batch 2 kg) submicron-sized PQDGMS was prepared by a top-down str

Ordered compartmentalization in colloidal microspheres: Design strategies and biomedical applications.

The rational design of colloidal microspheres with ordered internal compartments represents a frontier challenge in colloid and interface science, offering unique opportunities to mimic the structural and functional complexity of biological cells. Orange-flap shaped multicompartmental microspheres (OFMM) exemplify this platform by featuring radially separated, geometrically uniform compartments that enable precise spatial control over multi-enzyme cascade reactions, directional inter-compartmental signal transduction, and programmed payload release. Unlike conventional core-shell or randomly d

Organic di-selenide hydrogel microspheres for multimodal treatment of osteoarthritis.

Osteoarthritis (OA) involves multiple pathological processes and presents significant clinical challenges in treatment. Traditional therapies focus on individual factors in cartilage, synovium, or subchondral bone, limiting their ability to comprehensively address OA pathogenesis. In this study, a ROS/MMP13 dual-responsive organic selenium hydrogel microsphere (HSPHR) is developed to trigger a localized microenvironmental response specific to early OA by exploiting the disease's pathological features. Simultaneously, the organic selenium component effectively enhances selenoprotein levels in c

Dual antibiotic PLGA microspheres for the treatment of traumatic osteomyelitis.

Traumatic osteomyelitis (TO) treatment remains challenging due to biofilm formation and poor antibiotic penetration. We developed poly(lactic-co-glycolic acid) (PLGA) microspheres co-loaded with moxifloxacin/rifampicin (M/R-P) to address these limitations. Key findings: In vitro: The microspheres showed (1) no cytotoxicity (CCK-8 assay; live/dead staining; cytoskeletal staining; SEM visualization), and (2) no inhibition of osteogenic potential (ALP activity). In vivo: M/R-P microspheres significantly reduced MRSA burden ( 8 ± 3 × 10³ CFU/g) compared to debridement-only controls (3143 ± 727 × 10

Self-Replenishable Metabolically Augmented Synbiotic Microspheres Remodel Gut-Bone Homeostasis.

Gut microbiota dysbiosis in postmenopausal osteoporosis (PMO) is frequently accompanied by aberrant metabolism and absorption of short-chain fatty acids (SCFAs). However, current oral probiotic therapies neglect the crucial role of probiotic-driven SCFAs metabolism in restoring gut-bone homeostasis. In this study, commencing with the sequencing of fecal samples from clinical patients with PMO, a self-replenishable metabolically augmented synbiotic microsphere (SMASM) is fabricated via a thiol-ene click reaction to restore gut-bone homeostasis using Lactobacillus rhamnosus GG (LGG) as a viable

Porous Chitosan/Hydroxyapatite Composite Microspheres for Vancomycin Loading and Releasing.

Porous chitosan/hydroxyapatite (Chi-HAp) composite microspheres were prepared in an aqueous solution containing chitosan, calcium nitrate, and ammonium dihydrogen phosphate by using a hydrothermal method at various temperatures. The investigation indicated that temperature significantly impacted the final product's appearance. Hydroxyapatite (HAp) coupled with dicalcium phosphate dihydrate (DCPD) flakes were obviously found at 65 and 70 °C, while the latter gradually disappeared at higher temperatures. Conversely, synthesis at 90 °C led to smaller particle sizes due to the broken chitosan chai

Visualizable and Lubricating Hydrogel Microspheres Via NanoPOSS for Cartilage Regeneration.

The monitoring of tissue regeneration is particularly important. However, most materials do not allow direct observation of the regeneration process in the cartilage layer. Here, using sulfhydryl polyhedral oligomeric silsesquioxane (POSS-SH) as a nano-construction platform, poly(ethylene glycol) (PEG), Kartogenin (KGN), hydrogenated soya phosphatidylcholine (HSPC), and fluorescein are linked through the "click chemistry" method to construct nanomaterial with fluorescence visualization for cartilage repair: POSS linked with PEG, KGN, HSPC, and fluorescein (PPKHF). Injectable hydrogel microspheres with self-renewable hydration layers alleviate osteoarthritis.

Introducing hydration layers to hydrogel microspheres (HMs) by coating the surface with liposomes can effectively reduce friction. However, the lubrication can be inactivated when the surface coatings are damaged. To endow HMs with the ability to form self-renewable hydration layers and maintain cellular homeostasis, rapamycin-liposome-incorporating hyaluronic acid-based HMs (RAPA@Lipo@HMs) were created using microfluidic technology and photopolymerization processes. The RAPA@Lipo@HMs improve joint lubrication by using a smooth rolling mechanism and continuously exposing liposomes on the outer

Grafting MSI-78A onto chitosan microspheres enhances its antimicrobial activity.

MSI-78A (Pexiganan A) is one of the few antimicrobial peptides (AMPs) able to kill Helicobacter pylori, a pathogenic bacterium that colonizes the gastric mucosa of half of the world's population. Antibiotics fail in 20-40% of H. pylori-infected patients, reinforcing the need for alternative treatments. Herein, a bioengineered approach was developed. MSI-78A with a C-terminal cysteine was grafted onto chitosan microspheres (AMP-ChMic) by thiol-maleimide (Michael-addition) chemistry using a long heterobifunctional spacer (NHS-PEG113-MAL). Microspheres with ∼4 µm diameter (near H. pylori length)

Preparation of CaF2 Microspheres with Nanopetals for Water Vapor Adsorption.

The hierarchically structured flower-like CaF2 microspheres with nanopetals, named FL-CaF2, were synthesized via a hydrothermal method using calcium acetate Ca(Ac)2 and NaBF4 as calcium and fluorine sources, respectively, assisted by the chelating reagent trisodium citrate (Na3Cit) with the optimal pH of the synthesis solution. Meanwhile, a reference sample, named FL-CaF2-R, was reproduced using ethylenediaminetetraacetic acid disodium salt (Na2EDTA) as the chelating reagent, based on the recipe and synthesis procedure from the literature. Various techniques such as X-ray diffraction, scanning

Hydrogel microspheres evading alveolar macrophages for sustained pulmonary protein delivery.

Pulmonary delivery is a highly attractive alternative to injections for biologics such as therapeutic proteins. However, bioavailabilities generally suffer from the presence of phagocytic cells that clear particulate matter entering the lung. In this study, microgel particles were developed using an all-aqueous two-phase system approach and evaluated for their efficacy as an inhalable controlled release system. Norbornene- and thiol-modified four- and eight-armed poly (ethylene glycol) with an average molecular mass of 10,000 Da were prepared as macromonomers for microgel formation. Emulsions

Enzymatically cross-linked arabinoxylan microspheres as oral insulin delivery system.

Arabinoxylans (AX) microspheres with different insulin/AX mass ratio were prepared by formation of phenoxy radical issued from the ferulic acid by enzymatic oxidation (entrapped in situ of insulin). Phenolic acid content and FT-IR spectrum of unloaded and insulin-loaded AX microspheres revealed that the phenoxy radical issued from the ferulic acid by enzymatic oxidation did not interact covalently with insulin. The microspheres showed a spherical shape, smooth surface and an average diameter of particles of 320 μm. In vitro control release found that AX microspheres minimized the insulin loss

Antibiotic loaded microspheres as antimicrobial delivery systems for medical applications.

In this paper, we present the preparation and antibiotic loading of polymeric microspheres, composed of copolymers derived from fatty acid/amino acid components, as new polymeric platforms for antibiotic delivery systems. New polymeric materials were used to prepare microspheres with and without immobilized model antibiotics (streptomycin, chloramphenicol and amphotericin B) by a W/O/W double-emulsion/solvent evaporation method, in which chloroform and poly(vinyl alcohol) are used as the solvent and emulsifier, respectively. The antimicrobial activity of the microspheres was tested against Gra

Multiplexed Lipid Bilayers on Silica Microspheres for Analytical Screening Applications.

Most druggable targets are membrane components, including membrane proteins and soluble proteins that interact with ligands or receptors embedded in membranes. Current target-based screening and intermolecular interaction assays generally do not include the lipid membrane environment in presenting these targets, possibly altering their native structure and leading to misleading or incorrect results. To address this issue, an ideal assay involving membrane components would (1) mimic the natural membrane environment, (2) be amenable to high-throughput implementation, and (3) be easily multiplexe

Preparation and drug release properties of chitosan/organomodified palygorskite microspheres.

The novel composite microspheres, based on the hybridization of chitosan (CS) and organomodified palygorskite (OPAL), were prepared by emulsion cross-linking technique and applied as a drug carrier. Palygorskite, a kind of natural one-dimensional clay, was modified with hexadecyl betaine (BS-16) to improve the compatibility and affinity with chitosan matrix, and worked as a perfect micron-filler to enhance drug encapsulation and retard drug migration. The structure of the microspheres was characterized by fourier transform infrared spectroscopy (FT-IR), scanning electron microscopy (SEM) and X
