Gas decomposition of C3F7CN/CO2 mixtures: a chemical kinetics modelling study.

C3F7CN mixtures are more promising environmentally friendly alternatives to SF6 for gas-insulated equipment. Decomposition of C3F7CN mixtures driven by partial discharges during operation can generate toxic by-products, degrade dielectric performance, and pose risks to maintenance personnel. We develop a physics-based chemical kinetic framework to quantify the time-resolved and cumulative concentrations of decomposition products under prolonged partial discharge activity in a C3F7CN/CO2 mixture. A two-region, zero-dimensional model couples Bolsig+ electron-energy kinetics with Cantera gas-phas

Predicting blood-brain barrier permeability of chemicals by machine learning modeling.

In vitro studies are widely used to measure chemical neurotoxicity or neuroactivity. To show similar activity in vivo, chemicals need to cross the blood-brain barrier (BBB), a membrane that controls chemical movement from the bloodstream to the brain. Machine learning (ML) models present a rapid, cost-effective alternative to experimental methods for predicting BBB permeability. We developed and validated a set of ML models to predict BBB permeability by employing various molecular descriptors and ML algorithms. Models were rigorously evaluated using 5-fold cross-validation and scaffold splitt

Chemical composition and evaluation of the antipsoriatic potential of Cistus sintenisii in mouse model.

Cistus species have been used in the treatment of various ailments, including skin disorders in the traditional medicine. The present study aimed to evaluate the antipsoriatic potential of Cistus sintenisii extracts, using an imiquimod-induced psoriasis mouse model. The most promising effects were observed in the highest concentrations of the extracts, which attenuated psoriatic alterations, such as hyperplasia of the epidermis and inflammation and improved epidermal structure. Furthermore, reduction of approximately 50% in the Psoriasis Area and Severity Index (PASI) score on day 6 indicated

An Experimental Model: Modifications of the Chemical Silicone Medical Characteristics for an Improved Biocompatibility.

OBJECTIVE: Despite its good biocompatibility and processability, the severe surface hydrophobicity of polydimethylsiloxane (PDMS) limits its biomedical applications. This study aimed to enhance the surface properties of PDMS by depositing gelatin onto it via polydopamine (PDA) mediation and to evaluate the resulting cell-material interactions. METHODS AND RESULTS: The 1% gelatin-modified PDA-PDMS (G-PDA-PDMS) group showed the highest cell viability at 48 h. Surface hydrophilicity was significantly improved, with the contact angle reduced to 21.97° ± 2.06 (p <0.01). XPS confirmed successful mo

Hybrid non-animal modeling: A mechanistic approach to predict chemical hepatotoxicity.

Developing mechanistic non-animal testing methods based on the adverse outcome pathway (AOP) framework must incorporate molecular and cellular key events associated with target toxicity. Using data from an in vitro assay and chemical structures, we aimed to create a hybrid model to predict hepatotoxicants. We first curated a reference dataset of 869 compounds for hepatotoxicity modeling. Then, we profiled them against PubChem for existing in vitro toxicity data. Of the 2560 resulting assays, we selected the mitochondrial membrane potential (MMP) assay, a high-throughput screening (HTS) tool th

Inhibition of 2-Oxoglutarate Dehydrogenase as a Chemical Model of Acute Hypobaric Hypoxia.

Both hypoxia and inhibition of 2-oxoglutarate dehydrogenase complex (OGDHC) are known to change cellular amino acid pools, but the quantitative comparison of the metabolic and physiological outcomes has not been done. We hypothesize that OGDHC inhibition models metabolic changes caused by hypoxia, as both perturb the respiratory chain function, limiting either the NADH (OGDHC inhibition) or oxygen (hypoxia) supply. In the current study, we quantify the changes in the amino acid metabolism after OGDHC inhibition in the highly sensitive to hypoxia cerebellum and compare them to the earlier chara

Modeling secondary organic aerosol formation from volatile chemical products.

Volatile chemical products (VCPs) are commonly-used consumer and industrial items that are an important source of anthropogenic emissions. Organic compounds from VCPs evaporate on atmospherically relevant time scales and include many species that are secondary organic aerosol (SOA) precursors. However, the chemistry leading to SOA, particularly that of intermediate volatility organic compounds (IVOCs), has not been fully represented in regional-scale models such as the Community Multiscale Air Quality (CMAQ) model, which tend to underpredict SOA concentrations in urban areas. Here we develop a

Perfluoroalkyl chemicals in neurological health and disease: Human concerns and animal models.

Perfluoroalkyl acids (PFAAs) are man-made organic pollutants that are found ubiquitously in the environment and may impact human health. Here, we review the published literature concerning PFAA impacts on neurobiological, neuroendocrine, and neurobehavioral outcomes. We find that there are many mechanisms through which PFAAs may enter the brain and interact with biochemical endpoints to impact neurological function. These results are supported by epidemiological evidence in humans and experimental evidence in animals that demonstrate numerous and varied PFAA impacts on the nervous system. Howe

A Chemical Biology Approach to Model Pontocerebellar Hypoplasia Type 1B (PCH1B).

Mutations of EXOSC3 have been linked to the rare neurological disorder known as Pontocerebellar Hypoplasia type 1B (PCH1B). EXOSC3 is one of three putative RNA-binding structural cap proteins that guide RNA into the RNA exosome, the cellular machinery that degrades RNA. Using RNAcompete, we identified a G-rich RNA motif binding to EXOSC3. Surface plasmon resonance (SPR) and microscale thermophoresis (MST) indicated an affinity in the low micromolar range of EXOSC3 for long and short G-rich RNA sequences. Although several PCH1B-causing mutations in EXOSC3 did not engage a specific RNA motif as

Fluorescence Visualization of the Enteric Nervous Network in a Chemically Induced Aganglionosis Model.

Gastrointestinal motility disorders, severe variants in particular, remain a therapeutic challenge in pediatric surgery. Absence of enteric ganglion cells that originate from neural crest cells is a major cause of dysmotility. However, the limitations of currently available animal models of dysmotility continue to impede the development of new therapeutics. Indeed, the short lifespan and/or poor penetrance of existing genetic models of dysmotility prohibit the functional evaluation of promising approaches, such as stem cell replacement strategy. Here, we induced an aganglionosis model using to

The Behind-the-Knee test: an efficient model for evaluating mechanical and chemical irritation.

BACKGROUND/PURPOSE: The 'Behind-the-Knee' method (BTK test), using the popliteal fossa as a test site, evaluates both the inherent chemical irritation, and the potential for mechanical irritation of substrates and products. This approach eliminates some of the difficulties of in-use clinical test systems while still providing reliable results. In this publication, examples of the results of BTK tests on several materials are presented with direct comparisons, where possible, with results of in-use clinical testing conducted on the same materials. METHODS: In in-use clinical tests, volunteer p

Quantitative structure-activity relationship modeling of dermatomyositis activity of drug chemicals.

Dermatomyositis (DM) is an idiopathic inflammatory disorder consisting of skin and skeletal muscle involvement. Some drugs induce DM or dermatomyositis-like syndrome (DM-LS), the others provoke polymoysitis (PM) or cause elevation of serum levels of muscle enzymes (SE) or give muscle damage (M). The unexpected adverse reactions to drugs causing myositis are not a solved contemporary problem. The aim of this study was to determine the structural requirements of eliciting drug-induced DM as compared with drug induced PM. The Common Reactivity Pattern (COREPA) approach was used to describe the st

Biochemical reaction network modeling: predicting metabolism of organic chemical mixtures.

All organisms are exposed to multiple xenobiotics, through food, environmental contamination, and drugs. These xenobiotics often undergo biotransformation, a complex process that plays a critical role in xenobiotic elimination or bioactivation to toxic metabolites. Here we describe the results of a new computer-based simulation tool that predicts metabolites from exposure to multiple chemicals and interconnects their metabolic pathways, using four common drinking water pollutants (trichloroethylene, perchloroethylene, methylchloroform, and chloroform) as a test case. The simulation tool interc

An integrated model for the differentiation of chemical-induced allergic and irritant skin reactions.

Contact and photocontact allergic as well as irritant and photoirritant skin reactions represent a major problem in clinical dermatology and during the development of new pharmaceuticals. Furthermore, there is a lack of in vitro and in vivo assays that provide a clear differentiation between allergic and irritant skin reactions. Here, we describe an integrated model to differentiate between chemical-induced allergic and irritant skin reactions by measuring objective and easy-to-determine parameters within both skin and skin-draining lymph nodes. Dose-response studies with standard contact and

Effect of supplemental L-arginine in a chemical-induced model of colorectal cancer.

L-Arginine inhibits the development of spontaneous, transplantable solid tumors and chemically induced mammary tumors. The aim of the present study was to investigate the effect of l-arginine on chemically induced colorectal cancer in male Wistar rats. Colorectal cancer was induced in all animals by weekly subcutaneous injections of the colonic procarcinogen 1,2-dimethylhydrazine (DMH) at a dosage of 20 mg/kg body weight. Arginine was given in a 1% solution of drinking water. Group I was the DMH control; group II, arginine for 22 weeks; group III, arginine for the first 10 weeks only. Lymphocy

Drug-free treatment selection for chemical abusers: a diagnostic-based model.

The frequent treatment failures of patients with alcohol and drug-abuse disorders are often attributable to poor treatment choice. The absence from the literature of clinically relevant criteria for serving this population is noted. Four categories of patients are identified, and a diagnostic framework for the selection of drug-free treatments is proposed.

Hepatic lipid abnormalities in a chemical/viral mouse model for Reye's syndrome.

We have examined hepatic lipid profiles in a mouse model for Reye's Syndrome (RS) in which young animals are exposed to nontoxic doses of an industrial pesticide emulsifier and subsequently are infected with sublethal doses of mouse-adapted human Influenza B (Lee) virus (FluB). The purpose of this study was to determine whether liver lipid content was altered in the mice, the time course of any changes, and whether lipid changes were consistent with liver pathology. Neonatal mice exposed dermally to the emulsifier, Toximul MP8 (Tox), had significantly elevated levels of hepatic cholesterol, wi

Chemically induced congenital thymic dysgenesis in the rat: a model of the DiGeorge syndrome.

The DiGeorge syndrome, a variable complex of thymic aplasia, congenital heart disease, hypoparathyroidism, and anomalies of the face and neck, is thought to result from exposure to teratogenic agents. A group of congenital defects closely resembling this syndrome can be produced in newborn rats by the administration of the fat-soluble zinc chelating agent WIN 18,446, a bis-dichloroacetylamine. This drug, nontoxic to adult animals, is a powerful teratogen when administered to pregnant rats during days 9-12 of the 21-day gestation period. Our animal data suggest that the human syndrome results f
