Rescue of adult hippocampal neurogenesis in a mouse model of HIV neurologic disease.

The prevalence of central nervous system (CNS) neurologic dysfunction associated with human immunodeficiency virus (HIV) infection continues to increase, despite the use of antiretroviral therapy. Previous work has focused on the deleterious effects of HIV on mature neurons and on development of neuroprotective strategies, which have consistently failed to show a meaningful clinical benefit. It is now well established that new neurons are continuously generated in discrete regions in the adult mammalian brain, and accumulating evidence supports important roles for these neurons in specific cog

Neurologic, gastric, and opthalmologic pathologies in a murine model of mucolipidosis type IV.

Mucolipidosis type IV (MLIV) is an autosomal recessive lysosomal storage disorder caused by mutations in the MCOLN1 gene, which encodes the 65-kDa protein mucolipin-1. The most common clinical features of patients with MLIV include severe mental retardation, delayed motor milestones, ophthalmologic abnormalities, constitutive achlorhydria, and elevated plasma gastrin levels. Here, we describe the first murine model for MLIV, which accurately replicates the phenotype of patients with MLIV. The Mcoln1(-/-) mice present with numerous dense inclusion bodies in all cell types in brain and particula

Logistic model analysis of neurological findings in Minamata disease and the predicting index.

OBJECTIVE: To establish a statistical diagnostic method to identify patients with Minamata disease (MD) considering factors of aging and sex, we analyzed the neurological findings in MD patients, inhabitants in a methylmercury polluted (MP) area, and inhabitants in a non-MP area. MATERIALS AND METHODS: We compared the neurological findings in MD patients and inhabitants aged more than 40 years in the non-MP area. Based on the different frequencies of the neurological signs in the two groups, we devised the following formula to calculate the predicting index for MD: predicting index = 1/(1+e(-

Neurological tissue dissection techniques in mouse models for reproducible scientific results: Brain, spinal cord, cerebrospinal fluid, and sciatic nerve.

Biomedical research studies, specifically regarding human neurodegenerative diseases, are bound by ethical challenges, and have limited diagnostic and treatment options. Transgenic mouse models provide the opportunity to conduct feasible and practical research with the ability to precisely define the progression of neurodegenerative disease over a well-controlled dosage and timeline. The use of transgenic mouse models has been extensive and is critical to advancing research in many ways, including understanding brain morphology and general tissue changes caused by neurological diseases. Often,

Therapeutic effects of low-intensity transcranial ultrasound on neurological recovery in rat models of MCAO with varying injury severities.

BACKGROUND: Low-intensity transcranial ultrasound (LITUS) is a safe, non-invasive neuromodulation method shown to improve neurological function after ischemic stroke in middle cerebral artery occlusion (MCAO) rats models. Given that the degree of severity of ischemic damage may influence treatment outcomes, it is important to investigate the therapeutic performance of LITUS in various injury models to provide a comprehensive understanding to support its clinical application. METHODS: Sixty adult male SD rats were randomly divided into four groups: Sham (S), MCAO only (M), MCAO + high-intensit

Perspectives in the investigation of Cockayne syndrome group B neurological disease: the utility of patient-derived brain organoid models.

Cockayne syndrome (CS) group B (CSB), which results from mutations in the excision repair cross-complementation group 6 (ERCC6) genes, which produce CSB protein, is an autosomal recessive disease characterized by multiple progressive disorders including growth failure, microcephaly, skin photosensitivity, and premature aging. Clinical data show that brain atrophy, demyelination, and calcification are the main neurological manifestations of CS, which progress with time. Neuronal loss and calcification occur in various brain areas, particularly the cerebellum and basal ganglia, resulting in dysk

Molecular changes evoked by the beta-lactam antibiotic ceftriaxone across rodent models of substance use disorder and neurological disease.

Ceftriaxone is a beta-lactam antibiotic that increases the expression of the major glutamate transporter, GLT-1. As such, ceftriaxone ameliorates symptoms across multiple rodent models of neurological diseases and substance use disorders. However, the mechanism behind GLT-1 upregulation is unknown. The present review synthesizes this literature in order to identify commonalities in molecular changes. We find that ceftriaxone (200 mg/kg for at least two days) consistently restores GLT-1 expression in multiple rodent models of neurological disease, especially when GLT-1 is decreased in the disea

Insights into the Pathology of the α3 Na(+)/K(+)-ATPase Ion Pump in Neurological Disorders; Lessons from Animal Models.

The transmembrane Na(+)-/K(+) ATPase is located at the plasma membrane of all mammalian cells. The Na(+)-/K(+) ATPase utilizes energy from ATP hydrolysis to extrude three Na(+) cations and import two K(+) cations into the cell. The minimum constellation for an active Na(+)-/K(+) ATPase is one alpha (α) and one beta (β) subunit. Mammals express four α isoforms (α1-4), encoded by the ATP1A1-4 genes, respectively. The α1 isoform is ubiquitously expressed in the adult central nervous system (CNS) whereas α2 primarily is expressed in astrocytes and α3 in neurons. Na(+) and K(+) are the principal io

Efficacy of a Novel Dimeric Macrocyclic Gadolinium Chelate With High Relaxivity in Preclinical Models of Neurological and Oncological Pathologies.

BACKGROUND: Gadolinium-based contrast agents (GBCAs) are crucial for enhancing MRI contrast, especially in neurological and oncological imaging. Achieving high relaxivity is a primary focus in contrast media research. Recently, digadoglucitol, the first dimeric macrocyclic gadolinium complex, was introduced, offering significantly higher relaxivity compared with first-generation commercial macrocyclic GBCAs. PURPOSE: This study assesses the performance of digadoglucitol in preclinical models of neurological and oncological diseases. It explores whether digadoglucitol provides comparable diagn

Neuroimaging data repositories and AI-driven healthcare-Global aspirations vs. ethical considerations in machine learning models of neurological disease.

Neuroimaging data repositories are data-rich resources comprising brain imaging with clinical and biomarker data. The potential for such repositories to transform healthcare is tremendous, especially in their capacity to support machine learning (ML) and artificial intelligence (AI) tools. Current discussions about the generalizability of such tools in healthcare provoke concerns of risk of bias-ML models underperform in women and ethnic and racial minorities. The use of ML may exacerbate existing healthcare disparities or cause post-deployment harms. Do neuroimaging data repositories and thei

The Thyroid Hormone-target Gene Rhes a Novel Crossroad for Neurological and Psychiatric Disorders: New Insights from Animal Models.

Ras homolog enriched in striatum (Rhes) is predominantly expressed in the corpus striatum. Rhes mRNA is localized in virtually all dopamine D1 and D2 receptor-bearing medium-sized spiny neurons (MSNs), and cholinergic interneurons of striatum. Early studies in rodents showed that Rhes is developmentally regulated by thyroid hormone, as well as by dopamine innervation in adult rat, monkey and human brains. At cellular level, Rhes interferes with adenosine A2A- and dopamine D1 receptor-dependent cAMP/PKA pathway, upstream of the activation of the heterotrimeric G protein complex. Besides its inv

Neurological outcomes of animal models of uterine artery ligation and relevance to human intrauterine growth restriction: a systematic review.

AIM: This review explores the molecular, neurological, and behavioural outcomes in animal models of uterine artery ligation. We analyse the relevance of this type of model to the pathological and functional phenotypes that are consistent with cerebral palsy and its developmental comorbidities in humans. METHOD: A literature search of the PubMed database was conducted for research using the uterine artery ligation model published between 1990 and 2013. From the studies included, any relevant neuroanatomical and behavioural deficits were then summarized from each document and used for further a

Do Microplastics Have Neurological Implications in Relation to Schizophrenia Zebrafish Models? A Brain Immunohistochemistry, Neurotoxicity Assessment, and Oxidative Stress Analysis.

The effects of exposure to environmental pollutants on neurological processes are of increasing concern due to their potential to induce oxidative stress and neurotoxicity. Considering that many industries are currently using different types of plastics as raw materials, packaging, or distribution pipes, microplastics (MPs) have become one of the biggest threats to the environment and human health. These consequences have led to the need to raise the awareness regarding MPs negative neurological effects and implication in neuropsychiatric pathologies, such as schizophrenia. The study aims to u

Normative Modeling of Amygdala Subfields and Disease-Specific Vulnerabilities Across Neurological Disorders.

As an integrative center of the limbic system, the amygdala and its subfields participate in diverse neural functions and demonstrate heterogeneous structural changes in various neurological disorders. In this study, three-dimensional T1-weighted MRI data from 22,258 healthy participants were analyzed using normative modeling to establish normative references for the volumes of the amygdala and its subfields. These models elucidated the developmental and aging curves of the amygdala and identified distinct ages at peak volume across subfields, ranging from 24.6 to 43.7 years. Based on these re

Extra-Neurological Characterization of Seckel Syndrome-Model Mice Harboring CEP152 Variants.

Centrosomal protein 152 (CEP152) is a key regulator of centriole architecture and function, essential for proper cell division and polarity. Pathogenic variants in CEP152 cause Seckel syndrome (SCKL), a systemic disorder characterized by microcephalic primordial dwarfism. However, the mechanisms underlying its multi-organ manifestations remain poorly understood. To investigate this, we utilized two mouse models harboring patient-derived CEP152 variants, Cep152W105*/K897* and Cep152Q32P/Q32P. While our previous work focused on neurodevelopmental defects, here we systematically analyzed extra-ne

Human Myelin Spheres for in Vitro Oligodendrocyte Maturation, Myelination and Neurological Disease Modeling.

Demyelinating diseases, such as multiple sclerosis, damage the protective myelin sheaths of the central nervous system. The development of effective therapies has been hampered by the lack of models that accurately replicate human myelin biology. Here we present a novel method to generate human myelin spheres (MyS) by coculturing of hPSC-derived neuronal and oligodendrocyte precursor cells, to create myelinated neurons. Using multimodal analyses including confocal and (electron)microscopy, single-nuclei transcriptomics, lipidomics, and electrophysiology, we demonstrate myelination in MyS as ea

Improved survival and neurological outcomes with lysophosphatidylcholine supplementation in severe cardiac arrest model.

Cardiac arrest (CA) results in a loss of blood circulation, leading to whole-body ischemia-reperfusion injuries. A deficiency in plasma lysophosphatidylcholine (LPC) levels has been observed in both human patients and rat models and is implicated in organ dysfunction following CA. Building on previous findings from a mild injury model, this study explored the therapeutic potential of LPC supplementation in a severe 12-min rat CA model. The study also compared the effects of combining multiple LPC species with individual species to better determine the most effective strategy and enhance the tr

Artificial Intelligence in Neurology: Assessing the Diagnostic and Therapeutic Reasoning of Large Language Models.

Background Artificial intelligence (AI) and large language models (LLMs) are increasingly integrated into medicine, yet their role in clinical decision-making remains underexplored. Neurology provides an important testing ground for these systems because neurological diagnoses often involve complex, overlapping syndromes, subtle clinical distinctions, and time-sensitive decision-making that can significantly affect patient outcomes. Objective This study evaluated the diagnostic and therapeutic reasoning of three LLMs: ChatGPT 4.0, Google Gemini, and Claude 3.5 Sonnet, using a complex neurologi
