Pipiserpin orchestrates mosquito reproduction through dual control of vitellogenin integrity and 20-hydroxyecdysone-directed vitellogenesis.

Mosquito-borne diseases persist as a critical global health burden, driving the demand for novel vector control approaches. Here, we identify and characterize Pipiserpin, a female-specific serine protease inhibitor (serpin) in Culex pipiens pallens, and elucidate its role in regulating reproductive fitness. Pipiserpin exhibits tissue-specific expression in the fat body and ovary, peaking post-blood meal, and features a conserved serpin fold with a reactive central loop critical for protease inhibition. RNA interference-mediated knockdown of Pipiserpin severely impaired ovarian follicular devel

The role of citizen science in mosquito-borne disease surveillance and control: A scoping review.

BACKGROUND: Mosquito-borne diseases (MBDs) pose substantial global health and economic burdens. Although conventional MBDs surveillance systems remain essential, they are often resource-intensive, uneven in coverage, and often insufficiently responsive to spatio-temporal variations in mosquito presence and risk. Citizen science, increasingly enabled by mobile and digital technologies, offers a scalable complement to expand surveillance reach and timeliness. However, existing reviews have not comprehensively integrated evidence across diverse dimensions of citizen science applied to MBDs survei

Advances in mosquito gut microbiota: Mechanisms of regulating pathogen transmission and applications in vector control.

Mosquito-borne diseases pose a threat to global public health, yet traditional control measures are constrained by insecticide resistance and vaccine development bottlenecks. The gut microbiota-mediated transmission-blocking strategy has emerged as a promising alternative. This review summarizes three symbiont mechanisms: direct pathogen inhibition, host immunomodulation, and metabolic niche competition. We highlight research progress and field efficacy of three technical routes: Wolbachia-based population replacement and suppression, paratransgenesis using engineered symbionts, and deployment

An Estimation of Potential Vector Control Effect of Gravid Mosquito Trapping in Fort Worth, Texas.

Entomological surveillance is an essential component for integrated vector management (IVM), the current best practice for West Nile virus (WNV) prevention and control. The significance of vector mosquito surveillance, however, is not always recognized by the public, which increases vulnerability of IVM programs to elimination or downsizing when virus activities are low, particularly during interepidemics of WNV. In order to increase public recognition, the unrecognized contribution of mosquito surveillance with gravid (egg-carrying) mosquito trapping to WNV vector control was estimated using

Modelling Aedes aegypti mosquito control via transgenic and sterile insect techniques: endemics and emerging outbreaks.

The invasion of pest insects often changes or destroys a native ecosystem, and can result in food shortages and disease endemics. Issues such as the environmental effects of chemical control methods, the economic burden of maintaining control strategies and the risk of pest resistance still remain, and mosquito-borne diseases such as malaria and dengue fever prevail in many countries, infecting over 100 million worldwide in 2010. One environmentally friendly method for mosquito control is the Sterile Insect Technique (SIT). This species-specific method of insect control relies on the mass rear

Mass treatment with ivermectin for filariasis control in Papua New Guinea: impact on mosquito survival.

Field studies were carried out to determine the impact of mass human treatment with ivermectin on the survival of anthropophagic mosquitoes of the Anopheles punctulatus complex (Diptera: Culicidae), the vectors of lymphatic filariasis and malaria in Papua New Guinea. In a village where mass treatment had been given, using 400 microg/kg ivermectin plus 6 mg/kg diethylcarbamazine citrate (DEC), we performed pre- and post-treatment collections of freshly blood-engorged mosquitoes from the same nine bedrooms. All blood-fed mosquitoes collected less than 4 days after mass treatment died within 9 da

Soda-baited RNAi yeast insecticides as effective Attractive Targeted Sugar Baits (ATSBs) for mosquito control.

BACKGROUND: Attractive Targeted Sugar Baits (ATSBs) are a promising, environmentally friendly approach for mosquito control, but the direct field application, scalability and long-term effectiveness of ATSBs across diverse mosquito species remain significant challenges. METHODOLOGY/PRINCIPAL FINDINGS: We assessed the efficacy of a genetically engineered RNA interference (RNAi) yeast strain (Sh.463) formulated in three sugar baits: soda (Coca-Cola), 10% sucrose, and a commercial mosquito bait (BaitStab), on Aedes, Anopheles and Culex mosquitoes. All RNAi yeast bait formulations induced signifi

Prospection of plant-based bio-insecticides for mosquito vector control in Tanzania: A comprehensive review.

Mosquito-borne diseases, including malaria, dengue, chikungunya, zika, and lymphatic filariasis, pose significant public health challenges in Tanzania. Current vector control strategies largely depend on synthetic insecticides, but the emergence of insecticide resistance, environmental persistence, and non-target toxicity necessitates alternative, eco-friendly solutions. Plant-based bioinsecticides are gaining attention due to their biodegradability, low mammalian toxicity, and complex phytochemical profiles that hinder resistance development. This systematic review synthesizes 41 studies cond

Integrating zooprophylaxis, insecticide-treated livestock and mosquito nets for malaria control: a One Health proposal.

Malaria transmission persists despite widespread use of insecticide-treated nets (ITNs) due to multiple factors, including outdoor and early-evening biting and insecticide resistance. In this context, zooprophylaxis, which strategically uses livestock to divert mosquitoes from humans, has shown mixed epidemiological effects. Conversely, treating livestock with topical or systemic insecticides (known as insecticide-treated livestock [ITL]) has been demonstrated to significantly reduce mosquito survival. Combining ITL and zooprophylaxis with ITNs could improve vector control for species that fee

Beyond the Mosquito: Social Determinants of Health and Global Strategies for Aedes-Borne Disease Control.

BACKGROUND: Aedes-borne diseases, including dengue, chikungunya and Zika, have emerged as a growing global health concern, particularly in tropical and subtropical regions. Their spread is not only driven by biological and environmental factors but also shaped by a wide range of social determinants of health (SDH). This systematic review synthesizes available evidence on SDH that shape the transmission, prevention and control of these diseases, as well as the strategies implemented internationally to address them. METHODS: We conducted five systematic reviews following PRISMA guidelines, each

Exploring mosquito abundance and Plasmodium infection through nested-PCR: implications for disease surveillance and control.

The Plasmodium is responsible for malaria which poses a major health threat, globally. This study is based on the estimation of the relative abundance of mosquitoes, and finding out the correlations of meteorological parameters (temperature, humidity and rainfall) with the abundance of mosquitoes. In addition, this study also focused on the use of nested PCR (species-specific nucleotide sequences of 18S rRNA genes) to explore the Plasmodium spp. in female Anopheles. In the current study, the percentage relative abundance of Culex mosquitoes was 57.65% and Anopheles 42.34% among the study areas

Implementation of an Intelligent Trap for Effective Monitoring and Control of the Aedes aegypti Mosquito.

Aedes aegypti is a mosquito species known for its role in transmitting dengue fever, a viral disease prevalent in tropical and subtropical regions. Recognizable by its white markings and preference for urban habitats, this mosquito breeds in standing water near human dwellings. A promising approach to combat the proliferation of mosquitoes is the use of smart traps, equipped with advanced technologies to attract, capture, and monitor them. The most significant results include 97% accuracy in detecting Aedes aegypti, 100% accuracy in identifying bees, and 90.1% accuracy in classifying butterfli

Environmental safety review of methoprene and bacterially-derived pesticides commonly used for sustained mosquito control.

Some pesticides are applied directly to aquatic systems to reduce numbers of mosquito larvae (larvicides) and thereby reduce transmission of pathogens that mosquitoes vector to humans and wildlife. Sustained, environmentally-safe control of larval mosquitoes is particularly needed for highly productive waters (e.g., catchment basins, water treatment facilities, septic systems), but also for other habitats to maintain control and reduce inspection costs. Common biorational pesticides include the insect juvenile hormone mimic methoprene and pesticides derived from the bacteria Bacillus thuringie

nanos gene control DNA mediates developmentally regulated transposition in the yellow fever mosquito Aedes aegypti.

Transposable elements (TEs) are proposed as a basis for developing drive systems to spread pathogen resistance genes through vector mosquito populations. The use of transcriptional and translational control DNA elements from genes expressed specifically in the insect germ line to mediate transposition offers possibilities for mitigating some of the concerns about transgene behavior in the target vector species and eliminating effects on nontarget organisms. Here, we describe the successful use of the promoter and untranslated regions from the nanos (nos) orthologous gene of the yellow fever mo
