Caiman-biting mosquitoes and the natural vectors of Hepatozoon caimani in Brazil.

Mosquitoes that feed on crocodilians are poorly known, despite the potential role of these exothermic animals as reservoirs of arboviruses. In this article, we assessed the frequency, abundance, and temporal variation of caiman-biting mosquitoes as well as searched for the natural vectors of the blood parasite of caimans, Hepatozoon caimani, in the Pantanal area of central-western Brazil from captures conducted bimonthly from September 2006 to September 2007 and in February 2008. A total of 5,272 mosquitoes belonging to 10 species of five genera was caught on caimans. The most abundant species

Anopheles mosquitoes: not just flying malaria vectors... especially in the field.

The polymorphism of genes involved in the immunity of malaria vectors has been the subject of several recent studies with mosquitoes from natural populations. Most of the genes examined are known for their role against Plasmodium berghei and not necessarily for their role against Plasmodium falciparum. It seems, therefore, to be highly important not only to be cautious when linking natural selection with malaria epidemiology but also to consider the importance of other parasites and the environment on the mosquito genome.

Direct and indirect immunosuppression by a malaria parasite in its mosquito vector.

Malaria parasites develop as oocysts within the haemocoel of their mosquito vector during a period that is longer than the average lifespan of many of their vectors. How can they escape from the mosquito's immune responses during their long development? Whereas older oocysts might camouflage themselves by incorporating mosquito-derived proteins into their surface capsule, younger stages are susceptible to the mosquito's immune response and must rely on other methods of immune evasion. We show that the malaria parasite Plasmodium gallinaceum suppresses the encapsulation immune response of its m

Vector competence of Egyptian mosquitoes for Rift Valley fever virus.

Reintroduction of Rift Valley fever (RVF) into Egypt in 1993 raised concerns about the potential for Egyptian mosquitoes to transmit the virus. We evaluated the ability of Aedes caspius, Culex pipiens, Cx. antennatus, Cx. perexiguus, Cx. poicilipes, and Anopheles pharoensis collected in the Aswan area and Cx. pipiens collected in the Nile Delta to transmit RVF virus. All mosquito species tested were susceptible to RVF virus infection, with An. pharoensis and Ae. caspius being the most sensitive to infection. However, none of 12 An. pharoensis, including 10 with a disseminated infection, transm

A comparative assessment of mosquito trapping methods for vector surveillance in Scotland.

BACKGROUND: While mosquito vector-borne disease (VBD) risk remains low in Northern Europe, surveillance is needed to monitor potential vector species. However, most mosquito trapping methods have been evaluated only in areas of high mosquito density, with limited knowledge of their sensitivity in low-abundance areas. Here, we assessed the relative performance of four commercially available traps in an urban setting in Scotland, where mosquito density is low. METHODS: Two trap evaluation experiments were conducted in an urban marsh in Glasgow City during August 2023. In experiment 1, the Bioge

Experimental evidence rules out mosquitoes as vectors of Lyme disease.

BACKGROUND: Lyme disease, caused by Borrelia burgdorferi sensu lato (s.l.), is the most common vector-borne disease in the Northern Hemisphere, with Ixodes ticks as its primary vectors. However, many patients do not recall tick bites, fueling speculation about alternative transmission routes, particularly via mosquito bites. This belief is reinforced by studies reporting Borrelia presence in mosquitoes. This study evaluates whether three mosquito species can acquire, maintain, and transmit Borrelia spirochetes. METHODS: Mosquitoes (Aedes aegypti, Culex quinquefasciatus, and Culex pipiens biot

Chromobacterium biopesticide overcomes insecticide resistance in malaria vector mosquitoes.

Vector mosquito control is an integral part of malaria control. The global emergence of insecticide resistance in malaria-transmitting Anophelines has become an impediment and has created an urgent need for novel mosquito control approaches. Here, we show that a biopesticide derived from the soil-dwelling bacterium Chromobacterium sp. Panama (Csp_P) kills insecticide-resistant Anopheles mosquitoes, regardless of their resistance mechanisms. In addition, sublethal dose of Csp_P acts as a synergist to now used chemical insecticides across multiple classes. Moreover, Csp_P reduces host-seeking be

Wolbachia cifB induces cytoplasmic incompatibility in the malaria mosquito vector.

Wolbachia, a maternally inherited intracellular bacterial species, can manipulate host insect reproduction by cytoplasmic incompatibility (CI), which results in embryo lethality in crosses between infected males and uninfected females. CI is encoded by two prophage genes, cifA and cifB. Wolbachia, coupled with the sterile insect technique, has been used in field trials to control populations of the dengue vector Aedes albopictus, but CI-inducing strains are not known to infect the malaria vector Anopheles gambiae. Here we show that cifA and cifB can induce conditional sterility in the malaria

Vector competence of Virginia mosquitoes for Zika and Cache Valley viruses.

BACKGROUND: Vector-borne diseases are a major public health concern and cause significant morbidity and mortality. Zika virus (ZIKV) is the etiologic agent of a massive outbreak in the Americas that originated in Brazil in 2015 and shows a strong association with congenital ZIKV syndrome in newborns. Cache Valley virus (CVV) is a bunyavirus that causes mild to severe illness in humans and ruminants. In this study, we investigated the vector competence of Virginia mosquitoes for ZIKV and CVV to explore their abilities to contribute to potential outbreaks. METHODS: To determine vector competenc

Experimental population modification of the malaria vector mosquito, Anopheles stephensi.

Small laboratory cage trials of non-drive and gene-drive strains of the Asian malaria vector mosquito, Anopheles stephensi, were used to investigate release ratios and other strain properties for their impact on transgene spread during simulated population modification. We evaluated the effects of transgenes on survival, male contributions to next-generation populations, female reproductive success and the impact of accumulation of gene drive-resistant genomic target sites resulting from nonhomologous end-joining (NHEJ) mutagenesis during Cas9, guide RNA-mediated cleavage. Antiviral Immunity and Virus-Mediated Antagonism in Disease Vector Mosquitoes.

More than 100 pathogens, spanning multiple virus families, broadly termed 'arthropod-borne viruses (arboviruses)' have been associated with human and/or animal diseases. These viruses persist in nature through transmission cycles that involve alternating replication in susceptible vertebrate and invertebrate hosts. Collectively, these viruses are among the greatest burdens to global health, due to their widespread prevalence, and the severe morbidity and mortality they cause in human and animal hosts. Specific examples of mosquito-borne pathogens include Zika virus (ZIKV), West Nile virus (WNV

Vector Competence of American Mosquitoes for Three Strains of Zika Virus.

In 2015, Zika virus (ZIKV; Flaviviridae; Flavivirus) emerged in the Americas, causing millions of infections in dozens of countries. The rapid spread of the virus and the association with disease outcomes such as Guillain-Barré syndrome and microcephaly make understanding transmission dynamics essential. Currently, there are no reports of vector competence (VC) of American mosquitoes for ZIKV isolates from the Americas. Further, it is not clear whether ZIKV strains from other genetic lineages can be transmitted by American Aedes aegypti populations, and whether the scope of the current epidemi

How Zoophilic Japanese Encephalitis Vector Mosquitoes Feed on Humans.

Japanese encephalitis virus (JEV) is the most frequent cause of mosquito-borne encephalitis in Asian countries. Several culicine species are potential vectors. The primary JEV vectors feed mainly on cows (a dead-end host for JEV), pigs (an amplifying host), and, occasionally, humans (a dead-end host). It is essential to determine blood-feeding patterns to understand the transmission cycle of the disease. Here we review blood-feeding characteristics of the primary JEV vectors Culex tritaeniorhynchus, Culex vishnui, and Culex gelidus based on experimental works and field surveys conducted in Asi

Olfactory learning and memory in the disease vector mosquito Aedes aegypti.

Olfactory learning in blood-feeding insects, such as mosquitoes, could play an important role in host preference and disease transmission. However, standardised protocols allowing testing of their learning abilities are currently lacking, and how different olfactory stimuli are learned by these insects remains unknown. Using a Pavlovian conditioning paradigm, we trained individuals and groups of Aedes aegypti mosquitoes to associate an odorant conditioned stimulus (CS) with a blood-reinforced thermal stimulus (unconditioned stimulus; US). Results showed, first, that mosquitoes could learn the

Odorant receptor-mediated sperm activation in disease vector mosquitoes.

Insects, such as the malaria vector mosquito, Anopheles gambiae, depend upon chemoreceptors to respond to volatiles emitted from a range of environmental sources, most notably blood meal hosts and oviposition sites. A subset of peripheral signaling pathways involved in these insect chemosensory-dependent behaviors requires the activity of heteromeric odorant receptor (OR) ion channel complexes and ligands for numerous A. gambiae ORs (AgOrs) have been identified. Although AgOrs are expressed in nonhead appendages, studies characterizing potential AgOr function in nonolfactory tissues have not b

Eastern equine encephalitis virus in mosquitoes and their role as bridge vectors.

Eastern equine encephalitis virus (EEEV) is maintained in an enzootic cycle involving Culiseta melanura mosquitoes and avian hosts. Other mosquito species that feed opportunistically on mammals have been incriminated as bridge vectors to humans and horses. To evaluate the capacity of these mosquitoes to acquire, replicate, and potentially transmit EEEV, we estimated the infection prevalence and virus titers in mosquitoes collected in Connecticut, USA, by cell culture, plaque titration, and quantitative reverse transcription-PCR. Cs. melanura mosquitoes were the predominant source of EEEV (83 [

A system for studying vector competence of mosquitoes for Japanese encephalitis virus.

A method to infect mosquitoes with Japanese encephalitis virus (JEV) and to demonstrate virus transmission after an extrinsic incubation period is described. Using per oral feeding method infection rate as high as 90% could be achieved. Demonstration of transmission of the virus was achieved by allowing the infected mosquitoes to probe a suitable serum medium and testing the probed serum for virus. Both infection and transmission were demonstrated by using insect-bioassay.
