Twice daily topical administration of echothiophate for 2 weeks to the eyes of living cynomolgus monkeys produced profound subsensitivity of the accommodative response to pilocarpine and an approximately 50% decrease in the number of specific binding sites for 3H-quinuclidinyl benzilate (3H-QNB) in the ciliary muscle without a change in their affinity. When echothiophate treatment was discontinued, functional cholinergic sensitivity and the number of QNB binding sites both returned to normal over a similar 4-8 week period. Most animals had a modest overshoot of both functional sensitivity and

Muscarinic receptors involved in cholinergic neurotransmission were studied in isolated innervated guinea-pig tracheas using preganglionic (nerve) and postganglionic (field) stimulation, after blocking sympathetic effects with bethanidine (5 microM). Neostigmine (10 nM) significantly increased responses to nerve and field stimulation. The M1 antagonist pirenzepine (0.1-100 nM) selectively reduced tracheal responses to nerve stimulation in control and in neostigmine-treated tissues. The M2 antagonist gallamine (0.1-100 microM) significantly increased tracheal responses to nerve and field stimul

The effect of cholinergic agonists and antagonists on the central pattern generator of the pharyngeal muscles has been studied in third instar larvae of Drosophila. The pharyngeal muscles are a group of rhythmically active fibers involved in feeding. Bath application of the cholinergic agonists carbachol, muscarine, pilocarpine, and acetylcholine (ACh) to a semiintact preparation including the pharyngeal muscles and the central nervous system (CNS), initiated long-lasting endogenous-like bursting activity in the muscles. The muscarinic antagonists, atropine and scopolamine, blocked these respo

Like other neurotransmitter receptors, muscarinic acetylcholine receptors are subject to regulation by the state of receptor activation. Prolonged increases in the concentration of muscarinic agonists result in a decrease in receptor density and loss of receptor sensitivity, both in vivo and in vitro. On the other hand, when the receptor is deprived of acetylcholine for a long duration in vivo, the receptor becomes more sensitive in responding to muscarinic agonists. However, it has been more difficult to demonstrate increases in receptor concentration that accompany this supersensitive state.

The cholinergic modulation of histamine release and synthesis was studied in rat brain slices or synaptosomes labeled with L-[3H]histidine. Carbachol in increasing concentrations progressively reduced the K+-induced [3H]histamine release from cortical slices. Pirenzepine, a preferential M1-receptor antagonist, reversed the carbachol effect in an apparently competitive manner and with Ki values of 1-6 X 10(-8) M. 11-[(2-[(Diethylamino)methyl]-1-piperidinyl)acetyl]-5,11-dihydro-6H- pyrido[2,3-b][1,4]benzodiazepine-6-one (AF-DX 116), considered a preferential M2-receptor antagonist, reversed the

Although gallamine and a number of other compounds have been reported to slow the rate of dissociation of labeled ligands, especially [3H]N-methylscopolamine (NMS), from muscarinic receptors of heart and brain, there has been some dispute as to whether the dissociation of [3H]quinuclidinyl benzilate (QNB) is subject to such allosteric regulation. The present studies were intended to determine whether past discrepancies might be due to differences between tissues. We have found that gallamine modulates the dissociation of [3H]QNB from muscarinic receptors of the heart in a biphasic manner. Low

Rat thymocytes possess a single class of saturable, high affinity binding sites for muscarinic antagonists of the benzilate type such as [3H]3-quinuclidinyl benzilate ([3H]3-QNB). The average number of receptors per cell is 3000 and the equilibrium dissociation constant of [3H]3-QNB on intact cells is 7.5 nM. In the work reported here we found that perturbation of the thymocyte membrane by addition of phytohemagglutinin (4 micrograms/ml) caused a transient increase in muscarinic antagonist binding, and hydrocortisone (100 mg/kg s.c.) treatment of rats for 2 days prior to sacrifice increased th

To test the assumption that activation of the hypothalamic-pituitary-adrenocortical (HPA) axis is mediated by cholinergic neurons, the cortisol response to the administration of the muscarinic agonist RS 86 was investigated in 12 healthy volunteers. In addition, prolactin and growth hormone secretion patterns were assessed. No significant increase in plasma cortisol or growth hormone was observed after the administration of 1.5 and 3.0 mg RS 86, respectively. However, there was a slight increase in prolactin serum concentration after 3.0 mg RS 86. As RS 86, when used in the same dose range, ca

The binding parameters of muscarinic agonists and antagonists in rat central (brain, cerebellum and striatum) and peripheral (heart and lung) tissues were determined at 2 degrees and 37 degrees from competitive binding experiments with (3H)-quinuclidinylbenzylate (QNB). Muscarinic ligands binding affinities for cerebellum, heart and lung were tightly correlated. These tissues were also characterized by their low concentration of muscarinic receptors with high affinity for agonists. In contrast, brain and striatum contained a higher concentration of muscarinic receptors with a lower affinity fo

The binding characteristics of muscarinic receptors in rat salivary and lacrimal glands were studied by means of radioligand binding techniques. In competition experiments against [3H]N-methylscopolamine, classical muscarinic antagonists ipratropium bromide, N-methylscopolamine and N-methylatropine exhibited very similar KD values in all the glands and their binding behavior was well described by a one binding site model (nH congruent to 1). The novel cardioselective antimuscarinic compound, AF-DX 116, displayed an equally low affinity in all the tissues examined. Pirenzepine and dicyclomine,

Muscarinic receptors in the rabbit carotid body were studied utilizing the specific muscarinic antagonist. [3H]quinuclidinyl benzilate. Biochemical and autoradiographic assays of ligand binding showed that receptor numbers and distributions remained unchanged following degeneration of axons and terminals of the carotid sinus nerve. The data suggest that muscarinic receptors are associated with carotid body parenchymal cells and are absent on afferent nerve terminals.

Prostatic specimens of adenomatous tissue (PA) were obtained from the lateral lobe of the prostate in patients with benign prostatic hyperplasia (BPH). Non-hyperplastic specimens of the outer prostatic tissue (PC) were taken from the dorsal part of the prostate in patients undergoing cystourethrectomy. Effects of alpha-adrenoceptor and muscarinic receptor active drugs were studied. Noradrenaline (NA) and the alpha 1-adrenoceptor agonist phenylephrine induced concentration-dependent contractions in PC and PA preparations. The alpha 2-adrenoceptor agonist clonidine was without effect on PC but c

The large B cells of bull-frog sympathetic ganglia are well known to be depolarized by slow synaptic transmission, muscarinic agonists, analogues of luteinizing hormone-releasing hormone (LHRH), and substance P. Voltage-clamp analysis shows that these actions result from two underlying mechanisms: inhibition of the M-current, a voltage-dependent potassium current; and in some cells, an inward current associated with an increase in conductance. The additional inward current appears as a voltage-insensitive change in the instantaneous conductance (i.e. apparent leak conductance). The additional

Because of the possibility that olivocochlear bundles may use acetylcholine as an inhibitory transmitter, we examined the cochlea of the guinea pig for cholinergic receptor binding sites. Binding of the potent muscarinic antagonist [3H]quinuclidinyl benzilate (3H-QNB) showed that the cochlea has sites with the pharmacological specificity of muscarinic cholinergic receptors. Specific 3H-QNB binding was saturated at 0.8 nM and was half-saturated at 0.03 nM concentrations. Total 3H-QNB binding was reduced 70% in the presence of saturating doses of acetylcholine, oxotremorine and atropine, and hal

Nicotinic and muscarinic sites on glomus cell membranes of cats, rabbits and mice were determined in carotid body slices. Cells were impaled under Nomarski optics. Resting potentials were 11.1-73.1 mV and input resistances were 11-250 M omega. Nicotine, pilocarpine or bethanechol depolarized glomus cells and their input resistance decreased. Curare or alpha-bungarotoxin reduced nicotine effects. Atropine had similar effects on the responses to pilocarpine or bethanechol.

A voltage-sensitive outward membrane current ('M') and a consequent change in conductance (delta G) appear with a slow time-constant, in principal neurons of rabbit superior cervical ganglion (SCG), only when membrane potentials (Vm) are depolarized to less than -60 mV. Effects of muscarine on the voltage-current curves indicate that, in this depolarized range of less than -60 mV, suppression of M-current could contribute a muscarinic depolarization accompanied by a decrease in G; but that, at all Vms tested (about -90 to -40 mV), there is an additional larger muscarinic depolarization with no

We describe the synthesis and characterization of [125I]3-quinuclidinyl-(3-iodo-4-hydroxy-benzilate), a binding probe for the muscarinic cholinergic receptor of high specific radioactivity. The binding isotherm of this 125I-labeled compound to a rat synaptic plasma membrane-enriched fraction consists of two components: a 'specific' component which is saturable and closely fits hyperbolic binding to a single class of sites as evaluated by Scatchard analysis (Kd = 1.5 to 3 nM), and a linear component which may be measured directly by preincubating membranes in 0.1 microM 3-quinuclidinyl-benzilat

Dexmedetomidine, an alpha(2)-adrenoceptor agonist, has been approved for clinical use, although the mechanism of dexmedetomidine action has not been fully elucidated. Several studies have shown that G protein-coupled receptors (GPCRs) are recognized as targets for anesthetics and analgesics. Therefore, it is of interest to determine whether dexmedetomidine affects the function of GPCRs other than the alpha(2)-adrenoceptor. We examined the effects of dexmedetomidine on M(1), M(3), 5-HT(2C), substance P, and orexin 1 receptors in Xenopus oocytes expressing individual receptors. In addition, we i

Previously, we have reported that the NMDA antagonist AP-5, injected unilaterally into the nucleus accumbens of mice, induces ipsilateral rotation in monoaminergically intact mice, but contralateral rotation in monoamine-depleted animals. In this paper we report that the muscarine antagonist methscopolamine, injected unilaterally into the nucleus accumbens, induced predominantly contralateral rotation in monoaminergically intact mice. In monoamine-depleted animals intra-accumbens methscopolamine induced only a weak stimulation of rotational behaviour (not significant), but the direction of the
