Mathematical model of excitation-contraction in a uterine smooth muscle cell.

Uterine contractility is generated by contractions of myometrial smooth muscle cells (SMCs) that compose most of the myometrial layer of the uterine wall. Calcium ion (Ca(2+)) entry into the cell can be initiated by depolarization of the cell membrane. The increase in the free Ca(2+) concentration within the cell initiates a chain of reactions, which lead to formation of cross bridges between actin and myosin filaments, and thereby the cell contracts. During contraction the SMC shortens while it exerts forces on neighboring cells. A mathematical model of myometrial SMC contraction has been dev

Frusemide inhibits angiotensin II-induced contraction on human vascular smooth muscle.

AIMS: Frusemide is widely used in the treatment of acute pulmonary oedema, chronic congestive heart failure and, to a lesser degree, in the treatment of hypertension. Evidence suggests that frusenuide exerts an anti-vasoconstrictor effect independent of its diuretic properties. Since angiotensin II is a highly potent vasoconstrictor involved in the pathophysiology of these diseases, we have investigated the effect of frusemide on the contraction elicited by angiotensin II on human internal mammary artery (IMA) and saphenous vein (SV). METHODS: Rings of IMA and SV were suspended for isometric

Microvascular responses to oxygen and muscle contraction in hypertensive Dahl rats.

We evaluated the possibility that increased arteriolar tone in salt-sensitive hypertension could be partially due to an altered vascular responsiveness to oxygen and/or tissue metabolites. The microvasculature of the superfused spinotrapezius muscle was studied with fluorescence microscopy in Dahl salt-sensitive rats fed low (0.45%) or high (7%) salt diets for 4 weeks. High salt intake produced hypertension (mean arterial pressure = 162 +/- 5 mm Hg vs 130 +/- 4 mm Hg for low salt rats, p < 0.05), and increased vascular tone at the level of the arcade arterioles but not in the smaller transvers

Mediation of anaphylaxis-induced jejunal circular smooth muscle contraction in rats.

Altered intestinal motility and diarrhea are features of food protein-induced intestinal anaphylaxis in the conscious rat. These experiments were performed to determine the mediator(s) responsible for jejunal circular smooth muscle contraction during this response. Hooded-Lister rats were sensitized by intraperitoneal injection of 10-micrograms egg albumin, and controls were sham-sensitized with saline. Fourteen days later the contractility of the circular muscle in jejunal segments (mucosa intact) was examined in standard tissue baths in response to antigen (Ag) or other agents. While control

Can muscle co-contraction protect knee ligaments after injury or repair?

A computer-based model of the knee was used to study forces in the cruciate ligaments induced by co-contraction of the extensor and flexor muscles, in the absence of external loads. Ligament forces are required whenever the components of the muscle forces parallel to the tibial plateau do not balance. When the extending effect of quadriceps exactly balances the flexing effect of hamstrings, the horizontal components of the two muscle forces also balance only at the critical flexion angle of 22 degrees. The calculations show that co-contraction of the quadriceps and hamstring muscles loads the

Defect in colonic smooth muscle contraction in patients with ulcerative colitis.

Patients with ulcerative colitis have decreased postprandial colonic contractions. The purpose of this study was to determine whether the smooth muscle from patients with ulcerative colitis responds abnormally in vitro to different stimuli. Circular colonic smooth muscle strips from patients with ulcerative colitis, acute diverticular disease, or adenocarcinoma were stretched to the optimal length and stimulated with electrical field stimulation (EFS), bethanechol, or increased concentrations of extracellular K+. The EFS-stimulated on-contraction was similar in each group, but the off-contract

Contraction of porcine ocular ciliary muscle by epidermal growth factor-urogastrone.

Epidermal growth factor-urogastrone (EGF) caused a concentration-dependent contractile response in porcine ocular ciliary muscle preparations. The half-maximal contraction was observed at 23 ng/ml EGF (4 nM). The contractile action of EGF, which was not abolished by the removal of extracellular calcium, was not affected by atropine, tetrodotoxin, phentolamine and indomethacin. In addition to causing contractions on its own, the contractile action of EGF was potentiated in the presence of prostaglandin E1 and vasopressin. Our data point to a potential role for EGF in the regulation of ciliary m

Effects of cricothyroid muscle contraction on laryngeal resistance and glottic area.

To determine the functional significance of the cricothyroid muscle (CT) in respiration, laryngeal resistance was measured in anesthetized dogs, along with electromyographic activity of the posterior cricoarytenoid muscle (PCA) and CT. In two dogs the larynx was videotaped simultaneously via a telescope. Increased CT activity was induced by airway occlusion or hypercapnia. Observations were carried out before and during cold blockade of the recurrent laryngeal nerve (RLN) or the nerve to the CT (external branch of the superior laryngeal nerve [Ext SLN]). Paralysis of the CT had no effect on la

Antigen-induced contraction of jejunal smooth muscle in the sensitized rat.

We examined contractility of longitudinal muscle strips of jejunum from control rats and rats infected 34-85 days previously with Trichinella spiralis. Antigen prepared from T. spiralis larvae contracted muscle from previously infected but not control rats. Contraction was specific for the sensitizing agent because antigen from Nippostrongylus brasiliensis did not induce contraction. Contraction was resistant to atropine or tetrodotoxin but was inhibited by the 5-hydroxytryptamine (5-HT) antagonist cyproheptadine and by 5-HT desensitization. Neither histamine antagonists, diphenhydramine (H1-r

Skeletal muscle contraction characteristics in vivo in malignant hyperthermia susceptible subjects.

Ten malignant hyperthermia susceptible (MHS) subjects were investigated, all of them positive in in vitro tests. As a control group 12 healthy subjects were investigated. The investigation was done in a blind fashion during strictly standardized conditions. Electromechanical delay of contraction and half contraction time to tetanus were faster (P less than 0.05) while half relaxation time was shorter in the MHS subjects (P less than 0.05). Skin and intramuscular temperature were significantly higher in the MHS subjects (P less than 0.05). This indicates that MHS subjects differ in various skel

Effects of cholinergic drugs on muscle contraction in Moniliformis moniliformis (Acanthocephala).

In whole Moniliformis moniliformis spontaneous muscle contractions were rhythmic; longitudinal contractions were measured with a force transducer. The cholinergic agonists levamisole and nicotine significantly increased muscle tension in whole worms; these contractions were tonic and were antagonised by the ganglionic blocker pentolinium and by piperazine. In addition, levamisole-induced contractions were inhibited by gallamine, hexamethonium, and norepinephrine. In worm segments, where drugs in solution were injected through the worms, acetylcholine (ACh) and nicotinic agonists were effective

Gradient in excitation-contraction coupling in canine gastric antral circular muscle.

Slow waves decay in amplitude as they propagate through the thickness of circular muscle of the canine antrum. Slow waves are the excitable events that initiate contractions in the antrum. Excitation-contraction coupling occurs if slow wave depolarizations surpass a 'mechanical threshold'. The amplitude of slow waves recorded from circular muscle cells near the submucosa was insufficient to reach the mechanical threshold previously determined for muscle near the myenteric plexus, suggesting that either submucosal cells are normally mechanically quiescent, or that contractions of submucosal cel

Mechanisms of substance P-induced contraction of rabbit airway smooth muscle.

The contractile effects of substance P (SP) were studied in isolated rabbit tracheal smooth muscle (TSM) segments in vitro. Noncumulative administration of SP produced dose-dependent increases in TSM tension. The mean (+/- SE) peak isometric tension (Tmax) with SP was 35.7 (+/- 6.2%) of the corresponding Tmax response to methacholine. The dose of agonist producing 50% of Tmax (ED50) was significantly lower for SP, averaging 1.8 (+/- 0.4) X 10(-7) M, vs. 1.7 (+/- 0.32) X 10(-6) M for methacholine. Isometric muscle contraction and the active state: an analog computer study.

From Sandow's excitation-contraction coupling hypothesis and reasonable assumptions I obtain the kinetics of the active state, (AS), and thence, via empirical equations for series elastic and contractile components for frog sartorius around 20 degrees C, the tension, P, and dP/dt vs. time. Assumptions: (a) Rate of Ca(+2) injection is proportional to the Ca gradient, and a permeability, which increases from zero to a limit as the membrane potential rises above a threshold. (b) Released Ca(+2) is bound by the "muscle machinery," M, and removed by a carrier pump. (c) The AS is proportional to the

Feedback contributions to excitation-contraction coupling in native functioning striated muscle.

Skeletal and cardiac muscle excitation-contraction coupling commences with Nav1.4/Nav1.5-mediated, surface and transverse (T-) tubular, action potential generation. This initiates feedforward, allosteric or Ca2+-mediated, T-sarcoplasmic reticular (SR) junctional, voltage sensor-Cav1.1/Cav1.2 and ryanodine receptor-RyR1/RyR2 interaction. We review recent structural, physiological and translational studies on possible feedback actions of the resulting SR Ca2+ release on Nav1.4/Nav1.5 function in native muscle. Finite-element modelling predicted potentially regulatory T-SR junctional [Ca2+]TSR do

Attenuated Rapid-Onset Vasodilation to Forearm Muscle Contraction in Black Men.

PURPOSE: Non-Hispanic Black individuals have a blunted ability to vasodilate at rest compared with other racial groups. Limited studies have investigated blood flow responses to exercise in Black individuals. Recently, our laboratory demonstrated that Black men exhibit attenuated increases in forearm vascular conductance (FVC) during steady-state rhythmic handgrip. The mechanisms for this remain unknown. Herein, we used single muscle contractions, a modality that allows for assessment of rapid-onset vasodilation (ROV) independent of major elevations in shear stress, tissue metabolism, and syst

Systemic IL-6 regulation of eccentric contraction-induced muscle protein synthesis.

Systemic cytokines and contractile activity are established regulators of muscle protein turnover. Paradoxically, the IL-6 cytokine family, which shares the ubiquitously expressed membrane gp130 receptor, has been implicated in skeletal muscle's response to both contractions and cancer-induced wasting. Although we have reported that tumor-derived cachectic factors could suppress stretch-induced protein synthesis in cultured myotubes, the ability of systemic cytokines to disrupt in vivo eccentric contraction-induced protein synthesis has not been established. Therefore, we examined whether syst

Aortic smooth muscle contains limiting amounts of myosin light chain kinase (MLCK) for myosin regulatory light chain (RLC) phosphorylation and contraction that predisposes to thoracic aortic disease in humans containing heterozygous loss-of-function mutations in MYLK. We tested the hypothesis that thoracic aortic smooth muscle contraction may also be susceptible to variations in the smooth muscle-specific isoform of the motor protein myosin where inactivation of one Myh11 allele or the presence of one Myh11 missense variant associated with an increased risk of human aortic disease may result i
