Crystal structure of a mutant mineralocorticoid receptor responsible for hypertension.

The S810L mutation within the human mineralocorticoid receptor (MR S810L) induces severe hypertension and switches progesterone from antagonist to agonist. Here we report the crystal structures of the ligand-binding domain of MR S810L in complex with progesterone and deoxycorticosterone, an agonist of both wild-type and mutant MRs. These structures, the first for MR, identify the specific contacts created by Leu810 and clarify the mechanism of activation of MR S810L.

Role of N-terminal of metacaspase of Candida albicans in calcium binding.

Metacaspases are members of the CD clan and share structural similarities with mammalian caspases but possess unique features. This study delves into the Candida albicans metacaspase CaMCA-Ia, a type I metacaspase. CaMCA-Ia demonstrates Ca2+-dependent autoprocessing and presents a hydrophobic N-terminal, which differs from that of type II metacaspases. Truncated CaMCA-Ia (CaMCA-Ia-ΔN86), lacking 86 N-terminal amino acids, undergoes gradual self-processing and intermolecular processing. Calcium addition induces multistep processing, leading to maturation. Like mammalian caspases, CaMCA-Ia-ΔN86

Non-standard amino acid incorporation into thiol dioxygenases.

Thiol dioxygenases (TDOs) are non‑heme Fe(II)‑dependent enzymes that catalyze the O2-dependent oxidation of thiol substrates to their corresponding sulfinic acids. Six classes of TDOs have thus far been identified and two, cysteine dioxygenase (CDO) and cysteamine dioxygenase (ADO), are found in eukaryotes. All TDOs belong to the cupin superfamily of enzymes, which share a common β‑barrel fold and two cupin motifs: G(X)5HXH(X)3-6E(X)6G and G(X)5-7PXG(X)2H(X)3N. Crystal structures of TDOs revealed that these enzymes contain a relatively rare, neutral 3‑His iron‑binding facial triad. Despite thi

Structure-Guided Engineering of Xylitol 4-dehydrogenase Unlocks Efficient L-Xylulose Biosynthesis: Mechanistic Insights into Catalytic Enhancement.

L-Xylulose, a potent α-glucosidase inhibitor and indispensable chiral precursor for L-nucleoside drugs, is highly valuable. However, chemical synthesis often suffers from poor atom economy and tedious protection and deprotection steps of functional groups. Here, a green biocatalytic route was explored by retrieving five XDH4 genes and selecting AYXDH4 as the best candidate. Through homology modeling and site-directed mutagenesis targeting potential synergy sites in the α-helix, the double mutant T171A/S172A was generated, which exhibited 43.97% higher catalytic efficiency (kcat/Km) than the wi

[Rational design and molecular engineering of a chitosanase for preparing specific chito-oligosaccharides with degree of polymerization≥5].

To achieve efficient enzymatic preparation of specific high-degree-of-polymerization chito-oligosaccharides (degree of polymerization≥5, DP≥5), rational molecular engineering of chitosanase Csn46-Mut4 was performed in this study. Through site-directed mutagenesis coupled with DNS enzyme activity-HPLC product distribution screening, a superior mutant, C41A/T47A/V144A, was obtained. Under conditions of 55℃ and pH 5.5, the catalytic efficiency and thermostability of this enzyme were remarkably enhanced, reaching 2.56-fold and 1.67-fold those of Csn46-Mut4, respectively. Peak area normalization an

Substrate expansion of Geotrichum candidum alcohol dehydrogenase towards diaryl ketones by mutation.

Chiral diaryl alcohols, such as (4-chlorophenyl)(pyridin-2-yl)methanol, are important intermediates for pharmaceutical synthesis. However, using alcohol dehydrogenases (ADHs) in the asymmetric reduction of diaryl ketones to produce the corresponding alcohols is challenging due to steric hindrance in the substrate binding pockets of the enzymes. In this study, the steric hindrance of the ADH from Geotrichum candidum NBRC 4597 (G. candidum acetophenone reductase, GcAPRD) was eliminated by simultaneous site-directed mutagenesis of Phe56 (in the large pocket) and Trp288 (in the small pocket). As a

Fine-Tuning of Neuronal Ion Channels-Mapping of Residues Involved in Glucose Sensitivity of Recombinant Human Glycine Receptors.

The inhibitory glycine receptor (GlyR) mediates synaptic inhibition in the spinal cord, brain stem, and other regions of the mammalian central nervous system. Glucose was shown to potentiate α1 GlyRs by interacting with K143. Here, additional amino acids involved in glucose modulation were identified using a structure-based approach of site-directed mutagenesis followed by whole-cell patch-clamp analysis. We identified two additional lysine residues in the α1 GlyR extracellular domain, K16 and K281, that were involved in glucose modulation. Mutation of either residue to alanine abolished gluco

The K428 residue from Thermus thermophilus SG0.5JP17-16 laccase plays the substantial role in substrate binding and oxidation.

Laccases are multi-copper oxidases (MCOs) that catalyze the oxidation of various organic and inorganic compounds with concomitant reduction of dioxygen to water, and are considered to be green catalysts. Previous studies revealed that in MCOs, the structure of substrate-binding pocket was closely related to the substrate specificity. The amino acids on the pocket-constructing loops were involved in the identification of substrate and affected the catalytic properties of the enzyme. In the Thermus thermophilus SG0.5JP17-16 laccase (lacTT), the Lys428 residue is on pocket-constructing loops, to

Efficient use and recycling of the micronutrient iodide in mammals.

Daily ingestion of iodide alone is not adequate to sustain production of the thyroid hormones, tri- and tetraiodothyronine. Proper maintenance of iodide in vivo also requires its active transport into the thyroid and its salvage from mono- and diiodotyrosine that are formed in excess during hormone biosynthesis. The enzyme iodotyrosine deiodinase responsible for this salvage is unusual in its ability to catalyze a reductive dehalogenation reaction dependent on a flavin cofactor, FMN. Initial characterization of this enzyme was limited by its membrane association, difficult purification and poo

Critical role of W60d in thrombin allostery.

Mutation of residue W60d of thrombin, located 17 A from the Na+ binding site, suppresses Na+ binding and the functional differences between the slow and fast forms. The molecular basis for the long-range effect of this mutation is provided by a conspicuous network of water molecules which connects the Na+ binding environment to the specificity sites S1 and S2 of the enzyme. The mutation appears to stabilize thrombin in a hybrid conformation that is overall similar to the slow form, but with the fibrinogen recognition site functioning as in the fast form. It also affects the switch in specifici

Significantly enhanced specific activity of Bacillus subtilis (2,3)-butanediol dehydrogenase through computer-aided refinement of its substrate-binding pocket.

(2,3)-Butanediol dehydrogenases (BDHs) are widely utilized for the stereoselective interconversion between α-hydroxy ketones and vicinal diols to produce various functional building blocks. In this study, to enhance the specific activity towards (R)-phenyl-1,2-ethanediol (1a) for 2-hydroxyacetophenone (1b), the substrate-binding pocket of a Bacillus subtilis BDH (BsBDHA) was refined through site-directed mutagenesis. Based on molecular docking simulations, 14 residues were identified and subjected to alanine scanning mutagenesis. After screening, two residues, His42 and Gly292, were singled ou

Prion propagation is controlled by discrete structural regions of PrP rather than overall stability.

Mutational analysis of the cellular prion protein (PrPC) has revealed various regions of the protein that modulate prion propagation. However, these approaches involve deletions, insertions, or replacements in the presence of the WT PrPC, which may mask the true phenotype. Here, site-directed alanine mutagenesis of the prion protein (PrP) was conducted to identify sites, particularly potential "surface patches" of PrPC required for prion propagation. Mutations were targeted to the helical, sheet, and loop regions of PrPC, or a combination thereof and the mutated proteins expressed in mouse neu

Enhancing catalytic efficiency of two microbial uricases making by directed evolution.

Uricase is a key enzyme in purine metabolism that catalyzes the oxidation of uric acid to allantoin, widely used in the treatment of hyperuricemia and gout. In this study, error-prone PCR and one high-throughput screening method were employed to generate uricase mutants with enhanced enzymatic activity from Aspergillus flavus and Candida utilis. After several rounds of mutation and selection, an A. flavus uricase mutant, af-UAM, with activity of 46.21 U/mg, and a C. utilis uricase mutant, cu-UAM, with activity of 31.43 U/mg, were obtained-representing the highest uricase activities reported up

Red-mediated recombineering of Salmonella enterica genomes.

The mutagenesis of enterobacterial genomes using phage λ Red recombinase functions is a rapid and versatile experimental tool. In addition to the rapid generation of deletions in the genome of Salmonella enterica, variations of the method allow site-directed mutagenesis, generation of reporter fusions, generation of chimeric genes, or transplantation of regulatory elements directly in the chromosome. We describe the application of these approaches with focus on practical aspects and critical steps.

A novel approach for the rapid mutagenesis and directed evolution of the structural genes of west nile virus.

Molecular clone technology has proven to be a powerful tool for investigating the life cycle of flaviviruses, their interactions with the host, and vaccine development. Despite the demonstrated utility of existing molecular clone strategies, the feasibility of employing these existing approaches in large-scale mutagenesis studies is limited by the technical challenges of manipulating relatively large molecular clone plasmids that can be quite unstable when propagated in bacteria. We have developed a novel strategy that provides an extremely rapid approach for the introduction of mutations into

16S rRNA mutations that confer tetracycline resistance in Helicobacter pylori decrease drug binding in Escherichia coli ribosomes.

Tetracycline resistance in clinical isolates of Helicobacter pylori has been associated with nucleotide substitutions at positions 965 to 967 in the 16S rRNA. We constructed mutants which had different sequences at 965 to 967 in the 16S rRNA gene present on a multicopy plasmid in Escherichia coli strain TA527, in which all seven rrn genes were deleted. The MICs for tetracycline of all mutants having single, double, or triple substitutions at the 965 to 967 region that were previously found in highly resistant H. pylori isolates were higher than that of the mutant exhibiting the wild-type seque

In vitro analysis of the replicative capacity and phenotypic susceptibility to integrase inhibitors of HIV-2 mutants with integrase insertions.

BACKGROUND: HIV-2 resistance to integrase strand-transfer inhibitors (INSTIs) is characterized by two main pathways: (i) mutations at codons 143, 148 and155; and (ii) amino acid insertion after integrase codon 231 (231ins). OBJECTIVES: To complete INSTI resistance data on HIV-2 by determining the viral replicative capacity and INSTI phenotypic susceptibility of integrase mutants obtained through site-directed mutagenesis. METHODS: Site-directed mutants (SDMs) were constructed and viral stocks produced. Viral replicative capacity was assessed by measuring HIV-2 viral load at days 3, 7 and 14.

Functional analysis of the carboxy-terminal domain of Bacillus anthracis protective antigen.

Protective antigen (PA) is the common receptor-binding component of the two anthrax toxins. We investigated the involvement of the PA carboxy-terminal domain in the interaction of the protein with cells. A deletion resulting in removal of the entire carboxy-terminal domain of PA (PA608) or part of an exposed loop of 19 amino acids (703 to 722) present within this domain was introduced into the pag gene. PA608 did not induce the lethal-factor (LF)-mediated cytotoxic effect on macrophages because it did not bind to the receptor. In contrast, PA711- and PA705-harboring lethal toxins (9- and 16-am

Biosynthesis of Crinipellin Diterpenes in Mushroom Marasmius fiardii PR-910.

Crinipellins are a distinctive family of 5/5/5/5 tetracyclic diterpenoids previously reported exclusively from mushrooms of the genus Crinipellis. Despite extensive synthetic studies, the biosynthetic machinery responsible for crinipellin formation has remained elusive. Here, we identify the crinipellin biosynthetic gene cluster (mfd) from the mushroom Marasmius fiardii PR-910, a member of the family Marasmiaceae to which Crinipellis also belongs, although M. fiardii PR-910 itself has not been previously reported to produce crinipellins.
