Comparison of the construction of unmarked deletion mutations in Mycobacterium smegmatis, Mycobacterium bovis bacillus Calmette-Guérin, and Mycobacterium tuberculosis H37Rv by allelic exchange.

Until recently, genetic analysis of Mycobacterium tuberculosis, the causative agent of tuberculosis, was hindered by a lack of methods for gene disruptions and allelic exchange. Several groups have described different methods for disrupting genes marked with antibiotic resistance determinants in the slow-growing organisms Mycobacterium bovis bacillus Calmette-Guérin (BCG) and M. tuberculosis. In this study, we described the first report of using a mycobacterial suicidal plasmid bearing the counterselectable marker sacB for the allelic exchange of unmarked deletion mutations in the chromosomes

Mycobacterium abscessus group pulmonary disease.

PURPOSE OF REVIEW: Mycobacterium abscessus is the second most isolated nontuberculous mycobacterial (NTM) respiratory pathogen in United States. It is also among the most difficult to treat NTM respiratory pathogens. The purpose of this review is to highlight current opportunities and limitations in the management of M. abscessus pulmonary disease and ways to optimize therapy to avoid treatment choices that make management of these patients even more difficult. RECENT FINDINGS: The main themes of the manuscript emphasize recognition of M. abscessus drug resistance mechanisms and their implica

Clinical characteristics and antimicrobial susceptibility profiles of Mycobacterium abscessus and Mycobacterium massiliense pulmonary infection.

OBJECTIVES: Mycobacterium abscessus complex (MABC) is the most common rapidly growing Mycobacterium species in structural pulmonary diseases and can be life-threatening. This study aimed to assess the clinical characteristics and drug-susceptibility statuses of different M. abscessus (MAB) subspecies in the Zhejiang Province. METHODS: DNA sequencing was used to differentiate clinical MABC subspecies isolates. The Clinical and Laboratory Standards Institute guidelines were used to determine in vitro susceptibility of imipenem-relebactam (IMP-REL), omadacycline, and other conventional antibioti

Immunogenicity and vaccine potential of clinical isolate Mycobacterium kansasii strain against Mycobacterium tuberculosis infection.

Mycobacterium kansasii is a bacterium included in non-tuberculous mycobacteria (NTM) that can cause lung disease. It shares a significant number of antigens with Mycobacterium tuberculosis (Mtb), suggesting that it has the potential to be used as a tuberculosis (TB) vaccine. Therefore, we subcutaneously vaccinated mice with reference strain, M. kansasii-ATCC12478 [M. kansasii-American Type Culture Collection (ATCC)], and clinically isolated strain, M. kansasii-SM-1 to evaluate potential as a TB vaccine by comparing with bacille Calmette-Guerin (BCG) vaccine. Ten weeks after vaccination, we eva

Immunogenicity and cross-reactivity against Mycobacterium tuberculosis of proteoliposomes derived from Mycobacterium bovis BCG.

The only currently available vaccine against tuberculosis (TB) is Mycobacterium bovis Bacille Calmette-Guerin (BCG), which has inconsistent efficacy to protect against the disease in adults. M. tuberculosis (MTB) cell wall components have been implicated in the pathogenicity of TB and therefore have been a prime target for the identification and characterization of cell wall proteins with potential application in vaccine development. In this regard, proteoliposomes (PLs) derived from mycobacteria containing lipids and cell wall proteins could be potential vaccine candidates against TB. In the

Immunological evaluation of a novel Mycobacterium tuberculosis antigen, Rv3117, absent in Mycobacterium bovis BCG.

Tuberculosis (TB) remains a global infectious disease. To investigate the value of a novel Mycobacterium tuberculosis (M. tuberculosis) region of difference 5 (RD5)-encoded antigen, Rv3117, in the development of effective immuno-diagnostics and vaccines against TB, the immune responses to the antigen were examined in human subjects, as well as in C57BL/6 mice. The results showed that Rv3117 was able to evoke specific humoral and cellular immune responses. Consistent with the results from the RD1-encoded antigens, culture filtrate protein 10 kDa (CFP-10) and early secreted antigenic target 6 kD

Vaccine efficacy of Mycobacterium bovis BCG against Mycobacterium sp. infection in amberjack Seriola dumerili.

Mycobacteriosis, caused by the intracellular parasitism Mycobacterium sp., causes economic damages to aquaculture production in Japan, particularly in seriola fish production. Antibiotics are not effective against Mycobacterium sp. and so a potent vaccine is needed. We previously reported that BCG vaccine (Mycobacterium bovis BCG) induces adaptive immunity against Mycobacterium sp. in Japanese flounder, Paralichthys olivaceus. In a phylogenetic tree, the genes for a major antigen, the Ag85 complex, in Mycobacterium sp. TUMSAT-Msp001 are closely related to homologues in Mycobacterium ulcerans.

Mycobacterium bovis BCG vaccination as prophylaxis against Mycobacterium ulcerans osteomyelitis in Buruli ulcer disease.

Mycobacterium ulcerans disease, or Buruli ulcer (BU), causes significant morbidity in West Africa. Clinically, the disease presents in the skin as either nonulcerative or ulcerative forms and often invades bones either subjacent to the skin lesion (contiguous osteomyelitis) or remote from the skin lesion (metastatic osteomyelitis). Osteomyelitis represents a severe form of the disease that often requires numerous surgical interventions, even amputations. Surgery is accepted as the present definitive treatment for BU. In the absence of an effective drug treatment, the need for the development o

In Vitro Resistance against DNA Gyrase Inhibitor SPR719 in Mycobacterium avium and Mycobacterium abscessus.

The aminobenzimidazole SPR719 targets DNA gyrase in Mycobacterium tuberculosis. The molecule acts as inhibitor of the enzyme's ATPase located on the Gyrase B subunit of the tetrameric Gyrase A2B2 protein. SPR719 is also active against non-tuberculous mycobacteria (NTM) and recently entered clinical development for lung disease caused by these bacteria. Resistance against SPR719 in NTM has not been characterized. Here, we determined spontaneous in vitro resistance frequencies in single step resistance development studies, MICs of resistant strains, and resistance associated DNA sequence polymor

Disseminated Mycobacterium gordonae and Mycobacterium mantenii infection with elevated anti-IFN-γ neutralizing autoantibodies.

A case of disseminated nontuberculous mycobacteria(l) (NTM) infection in a patient with positive neutralizing anti-interferon-γ (IFN-γ) autoantibodies involving bone, bronchus, systemic lymph nodes, and skin is reported. The causative NTMs were two different strains: Mycobacterium gordonae, which rarely causes true disease, and Mycobacterium mantenii, which is extremely rare. Anti-mycobacterial treatment successfully ameliorated all disseminated lesions. Although the concentration of anti-IFN-γ autoantibodies increased during the pre-treatment period, it gradually decreased after anti-mycobact

Characterization of novel Mycobacterium tuberculosis and Mycobacterium smegmatis mutants hypersusceptible to beta-lactam antibiotics.

Our laboratory previously constructed mutants of Mycobacterium tuberculosis and Mycobacterium smegmatis with deletions in the genes for their major beta-lactamases, BlaC and BlaS, respectively, and showed that the mutants have increased susceptibilities to most beta-lactam antibiotics, particularly the penicillins. However, there is still a basal level of resistance in the mutants to certain penicillins, and the susceptibilities of the mutants to some cephalosporin-based beta-lactams are essentially the same as those of the wild types. We hypothesized that characterizing additional mutants (de

Fatty acid-dependent cidality in Mycobacterium avium and Mycobacterium abscessus: intracellular acidification by itaconic acid.

Non-tuberculous mycobacterial (NTM) lung disease is now recognized as a chronic condition with considerable morbidity and mortality with incidence rising globally. This is concerning as these infections are often mis- or undiagnosed and currently available treatment regimens have lengthy (>1 year) treatment courses with variable to poor success rates. Identification of vulnerable targets and pathways in NTM pathogens is urgently needed to develop improved therapeutics. Itaconic acid (ITA), a macrophage-produced metabolite with immunomodulatory and antibacterial activity, inhibits growth of myc

Phenotypic and transcriptomic characterization of two newly described Mycobacterium species within the Mycobacterium gordonae complex.

INTRODUCTION: Non-tuberculous mycobacteria constitute a rapidly emerging class of opportunistic pathogens with escalating clinical significance globally. We recently characterized two novel species within the Mycobacterium gordonae complex-Mycobacterium camsae and Mycobacterium pumcae-both isolated from human cutaneous infections. METHODS: Comprehensive phenotypic and transcriptomic analyses were conducted to elucidate their pathobiological properties. Comparative analysis included M. gordonae (closest phylogenetic relative), M. marinum (slow-growing reference), and M. abscessus (rapid-growin

Aerobiology and Transmission of Mycobacterium tuberculosis.

Despite vast changes in human social structures through evolution-from small hunter-gatherer bands to densely populated megacities-Mycobacterium tuberculosis (Mtb) has persisted within human populations for millennia, underscoring the bacterium's exceptional transmissibility. This paper examines the aerobiological foundations of Mtb transmission, tracing the bacillus from expulsion in respiratory aerosols through environmental survival to deposition and infection in new hosts. It considers how host, mycobacterial, and environmental factors interact to influence transmission efficiency, highlig

Mycobacterium tuberculosis as a model system.

Over the past 30 years, the study of Mycobacterium tuberculosis has become a vibrant field that supports basic investigation into mycobacteria, wide-ranging studies of M. tuberculosis pathogenesis, and drug and therapeutic development. In this minireview, we highlight the multiple ways in which M. tuberculosis is a model. M. tuberculosis has emerged as a model for prokaryotic biology, revealing basic mechanisms of gene expression, genomic integrity, and mutagenesis that expand prokaryotic dogmas established in earlier model organisms. Next, we highlight the experimental systems used to model M

Comparative in silico analysis of vapBC3 toxin-antitoxin system in Mycobacterium Bovis and Mycobacterium tuberculosis.

Toxin-Antitoxin (TA) systems are highly conserved genetic elements in the Mycobacterium tuberculosis complex, playing a critical role in bacterial survival, stress response, and persistence. Small variations in sequence composition can lead to species-specific functional differences, potentially influencing host adaptation, antibiotic tolerance, and virulence strategies. In this study, a comparative analysis of TA systems in M. tuberculosis and M. bovis was conducted using BLAST NCBI, and SnapGene 5.3.1. A main mutation was identified at nucleotide 719 of the vapC3 gene in M. bovis isolates. W

Heterologous Substitution of Mycobacterium tuberculosis rRNA in Mycobacterium smegmatis and Its Impact on Antimicrobial Susceptibility.

Background: The global incidence of multidrug-resistant and extensively drug-resistant tuberculosis continues to rise. The ribosome serves as a target for multiple antimicrobials, making functional research on it hold great significance. Methods: Using homologous recombination combined with a multiple serine integrase-mediated site-specific recombination system, we replaced the two endogenous rRNA operons in Mycobacterium smegmatis MC2 155 with a single copy of the single rRNA operon from Mycobacterium tuberculosis H37Rv, constructing the M. smegmatis BRkoA strain. We assessed growth kinetics
