Biomimetic Dendrimer-Peptide Conjugates for Early Multi-Target Therapy of Alzheimer's Disease by Inflammatory Microenvironment Modulation.

Current therapeutic strategies for Alzheimer's disease (AD) treatments mainly focus on β-amyloid (Aβ) targeting. However, such therapeutic strategies have limited clinical outcomes due to the chronic and irreversible impairment of the nervous system in the late stage of AD. Recently, inflammatory responses, manifested in oxidative stress and glial cell activation, have been reported as hallmarks in the early stages of AD. Based on the crosstalk between inflammatory response and brain cells, a reactive oxygen species (ROS)-responsive dendrimer-peptide conjugate (APBP) is devised to target the A

A highly luminescent ZnS/CdSe/ZnS nanocrystals-tetrapeptide biolabeling agent.

Stable colloidal ZnS/CdSe/ZnS nanocrystals capped with hexadecylamine were transferred into water by encapsulation in a protective shell of an amphiphilic polymer. The properties of the products have been investigated by dynamic lighting scattering, absorption, and photoluminescence spectroscopy. These nanocrystals, when conjugated with tuftsin in aqueous suspension, formed an effective labeling reagent for macrophages or lymphocytes. The labeling of cells was demonstrated by confocal microscopy. Hybrid bio-inorganic nanoconjugates are potentially powerful fluorescent tracking tools in biologi

Thiol functionalised gold nanoparticles loaded with methotrexate for cancer treatment: From synthesis to in vitro studies on neuroblastoma cell lines.

HYPOTHESIS: Colloidal gold nanoparticles (AuNPs) functionalised with hydrophilic thiols can be used as drug delivery probes, thanks to their small size and hydrophilic character. AuNPs possess unique properties for their use in nanomedicine, especially in cancer treatment, as diagnostics and therapeutic tools. EXPERIMENTS: Thiol functionalised AuNPs were synthesised and loaded with methotrexate (MTX). Spectroscopic and morphostructural characterisations evidenced the stability of the colloids upon interaction with MTX. Solid state (GISAXS, GIWAXS, FESEM, TEM, FTIR-ATR, XPS) and dispersed phas

Nanocomposites: silver nanoparticles and bacteriocins obtained from lactic acid bacteria against multidrug-resistant Escherichia coli and Staphylococcus aureus.

Drug-resistant bacteria such as Escherichia coli and Staphylococcus aureus represent a global health problem that requires priority attention. Due to the current situation, there is an urgent need to develop new, more effective and safe antimicrobial agents. Biotechnological approaches can provide a possible alternative control through the production of new generation antimicrobial agents, such as silver nanoparticles (AgNPs) and bacteriocins. AgNPs stand out for their antimicrobial potential by employing several mechanisms of action that can act simultaneously on the target cell such as the p

Lipid-Conjugated Reduced Haloperidol in Association with Glucose-Based Nanospheres: A Strategy for Glioma Treatment.

Aggressive glioma exhibits a poor survival rate. Increased tumor aggression is linked to both tumor cells and tumor-associated macrophages (TAMs), which induce pro-aggression, invasion, and metastasis. Imperatively, for effective treatment, it is important to target both glioma cells and TAMs. Haloperidol, a neuropsychotic drug, avidly targets the sigma receptor (SR), which is expressed in higher levels in both the cell types. Herein, we present the development of a novel cationic lipid-conjugated reduced haloperidol (±RHPC8), which aims to mediate the SR-targeted antiglioma effect. Hypothetic

Biological activities of metallic nanostructures functionalized with hexadeca-substituted copper(II) and cobalt(II) phthalocyanines.

A novel phthalonitrile derivative (a) containing three functional groups (hexyl, aminated ester, phenoxy) was synthesized and subsequently cyclotetramerized in the presence of the corresponding metal chloride salts to obtain hexadeca-substituted metal {M = Cu(II) and Co(II)} phthalocyanines (b and c). The water-soluble phthalocyanines (d and e) were prepared by treating the newly synthesized complexes (b and c) with methyl iodide. Moreover, gold nanoparticles (1) and silver nanoparticles (2) were prepared, and their surfaces were modified with quaternary phthalocyanines (d and e). The biologic

Developing FRET-based UCNP@SiO2-AIE/AIE@DSPE-PEG2000 nanomaterials for lysosome-targeted NIR photodynamic cancer therapy.

Photodynamic therapy (PDT) has emerged as a promising strategy for cancer treatment due to high spatiotemporal selectivity. However, most of the conventional photosensitizers (as the core component of PDT) are activated by visible light, which suffers from limited tissue penetration, thereby restricting the clinical applications of PDT. In this study, two carboxymethyl-functionalized aggregation-induced emission (AIE) compounds, (E)-2-(4-(bis(4-methoxyphenyl)amino)styryl)-5-carboxy-3-methylbenzo[d]thiazol-3-ium (MOTBAC) and (E)-5-carboxy-2-(2-(5-(4-(diphenylamino)phenyl)thiophen-2-yl)vinyl)-3-

Coaxial Synthesis of PEI-Based Nanocarriers of Encapsulated RNA-Therapeutics to Specifically Target Muscle Cells.

In this work, we performed a methodological comparative analysis to synthesize polyethyleneimine (PEI) nanoparticles using (i) conventional nanoprecipitation (NP), (ii) electrospraying (ES), and (iii) coaxial electrospraying (CA). The nanoparticles transported antisense oligonucleotides (ASOs), either encapsulated (CA nanocomplexes) or electrostatically bound externally (NP and ES nanocomplexes). After synthesis, the PEI/ASO nanoconjugates were functionalized with a muscle-specific RNA aptamer. Using this combinatorial formulation methodology, we obtained nanocomplexes that were further used a

Potentials and emerging trends in nanopharmacology.

Nanopharmacology is a relatively newer branch of pharmacology which investigates interaction of a nanomedicine with living systems at the nanoscale level. Modern medicine is increasingly concerned with various surface modified nanocarriers, such as dendrimers, nanoparticles, carbon based nanomaterials, polymer-drug nanoconjugates, etc., which have immense therapeutic potential by target specific drug delivery, using nanoscaffolding and nanocontainers, owing to the specific physical, chemical and biological properties of these moieties that is related to their nanoscale size range. Multi-scale modeling of folic acid-functionalized TiO2 nanoparticles for active targeting of tumor cells.

Strategies based on the active targeting of tumor cells are emerging as smart and efficient nanomedical procedures. Folic acid (FA) is a vitamin and a well-established tumor targeting agent because of its strong affinity for the folate receptor (FR), which is an overexpressed protein on the cell membranes of the tumor cells. FA can be successfully anchored to several nanocarriers, including inorganic nanoparticles (NPs) based on transition metal oxides. Among them, TiO2 is extremely interesting because of its excellent photoabsorption and photocatalytic properties, which can be exploited in ph

Antibacterial Efficacies of Nanostructured Aminoglycosides.

The widespread use of broad-spectrum aminoglycoside antibiotics is restricted from various clinical applications due to the emergence of bacterial resistance and the adverse effects such as ototoxicity and nephrotoxicity. The intensive applicability of nanoparticles in modern medicinal chemistry has gained the interest of researchers for modification of aminoglycosides as nanoconjugates either via covalent conjugation or physical interactions to alleviate their undesirable effects and bacterial resistance. In this context, various carbohydrates, polymers, lipids, silver, gold, and silica-attac

Clinically Approved Drugs against CNS Diseases as Potential Therapeutic Agents To Target Brain-Eating Amoebae.

Central nervous system (CNS) infections caused by free-living amoebae such as Acanthamoeba species and Naegleria fowleri are rare but fatal. A major challenge in the treatment against the infections caused by these amoebae is the discovery of novel compounds that can effectively cross the blood-brain barrier to penetrate the CNS. It is logical to test clinically approved drugs against CNS diseases for their potential antiamoebic effects since they are known for effective blood-brain barrier penetration and affect eukaryotic cell targets. The antiamoebic effects of clinically available drugs fo

Synthesis of Chitosan Nanoparticles Conjugated With Protoporphyrin IX and Vitamin B9 for Their Application in Photodynamic Therapy.

One of the main obstacles of Photodynamic Therapy (PDT) to damage and destroy abnormal cells is that most photosensitizers (Ps) have a highly hydrophobic nature with a tendency to aggregate in aqueous solutions and the non-specificity towards target cells. Nanotechnology proposes new tactics for the development of monomeric Ps nanotransporters and active targeting mole-cules with the use of biodegradable polymeric nanoparticles to improve the specificity towards target cells. The goal of this work was to optimize the synthesis of chitosan polymeric nanoparticles conjugated with protoporphyrin

Nanomaterials with a photothermal effect for antibacterial activities: an overview.

Nanomaterials and nanotechnologies have been expected to provide innovative platforms for addressing antibacterial challenges, with potential to even deal with bacterial infections involving drug-resistance. The current review summarizes recent progress over the last 3 years in the field of antibacterial nanomaterials with a photothermal conversion effect. We classify these photothermal nanomaterials into four functional categories: carbon-based nanoconjugates of graphene derivatives or carbon nanotubes, noble metal nanomaterials mainly from gold and silver, metallic compound nanocomposites su

Nanotechnology and nanotoxicology in retinopathy.

Nanoparticles are nanometer-scaled particles, and can be utilized in the form of nanocapsules, nanoconjugates, or nanoparticles themselves for the treatment of retinopathy, including angiogensis-related blindness, retinal degeneration, and uveitis. They are thought to improve the bioavailability in the retina and the permeability of therapeutic molecules across the barriers of the eye, such as the cornea, conjunctiva, and especially, blood-retinal barriers (BRBs). However, consisting of multiple neuronal cells, the retina can be the target of neuronal toxicity of nanoparticles, in common with

Targeted Nanodelivery of WGX50 and Curcumin via Gold Nanoparticles for Alzheimer's Therapy.

Alzheimer's disease (AD) is a progressive neurodegenerative disorder, posing a global health challenge. It affects millions of people, causing cognitive decline and a heavy burden on healthcare systems. Neuroinflammation is a key pathological feature of AD, often associated with the dysregulation of microRNAs such as hsa-miR-146a-5p. WGX50 (N-[2-(3,4-Dimethoxy-phenyl)-ethyl]-3-phenyl-acrylamide), a small molecule derived from Zanthoxylum bungeanum Maxim, has antioxidant and anti-inflammatory activities. While WGX50 demonstrates potent inhibition of neuroinflammation, its poor blood-brain barri

Biodistribution of tumor-targeted MXene-antibody conjugates.

The biodistribution profile is a critical determinant of the success of nanomaterial applications, governing both therapeutic efficacy and systemic safety in emerging photothermal therapy (PTT) agents. Despite the promise of PTT, the clinical translation of many nanomaterial-based photothermal agents is impeded by intrinsic toxicity and insufficient tumor specificity. Moreover, off-target biodistribution of photosensitizers after systemic administration can increase systemic exposure, reduce tumor delivery, and ultimately compromise therapeutic efficacy, underscoring the need for advanced mate

Novel dual-targeting of biofilm formation and pyocyanin production in clinical Pseudomonas aeruginosa isolates using glutamine-modified thiosemicarbazone-conjugated ZnO nanoparticles.

Pseudomonas aeruginosa is a highly adaptable opportunistic pathogen frequently associated with chronic and hard-to-treat infections, particularly in burn units and immunocompromised patients. Its intrinsic and acquired resistance to multiple antibiotics poses a major therapeutic challenge. While ZnO nanoparticles conjugated with thiosemicarbazone (TSC) have shown promise in general antimicrobial applications, their potential for simultaneously inhibiting biofilm formation and pyocyanin production-key virulence factors-in clinical P. aeruginosa strains remains unexplored. In this study, ZnO nan

The gold/ampicillin interface at the atomic scale.

In the fight against antibiotic resistance, gold nanoparticles (AuNP) with antibiotics grafted on their surfaces have been found to be potent agents. Ampicillin-conjugated AuNPs have been thus reported to overcome highly ampicillin-resistant bacteria. However, the structure at the atomic scale of these hybrid systems remains misunderstood. In this paper, the structure of the interface between an ampicillin molecule AMP and three flat gold facets Au(111), Au(110) and Au(100) has been investigated with numerical simulations (dispersion-corrected DFT).
