Field-realistic doses of the neonicotinoid acetamiprid impact natural soil arthropod community diversity and structure.

The neonicotinoid acetamiprid is used as a foliar insecticide spray, which results in direct exposure of a wide variety of soil organisms. Laboratory testing indicated that acetamiprid is toxic to the Collembola (springtails) species Folsomia candida, while Acari (mites) seem relatively insensitive to neonicotinoids. Since such opposing effects on different soil arthropods might imbalance natural arthropod communities, this study determined: (i) if field-realistic doses of acetamiprid affect the abundance and diversity in soil arthropod communities, and (ii) whether these potential effects are

Sex-specific transgenerational effects on murine thyroid gland imposed by ancestral exposure to neonicotinoid thiacloprid.

Neonicotinoids, a relatively new widely used class of insecticide is used in agriculture to control insect populations. We examined the capacity of ancestral exposure to the neonicotinoid thiacloprid (thia) to induce transgenerational effects on thyroid tissue. Pregnant outbred Swiss female mice were exposed to thia at embryonic days E6.5 to E15.5 using 0, 0.6, and 6 mg/kg/day doses. Thyroid paraffin sections were prepared for morphology analysis. We apply ELISA method to measure T4 and TSH levels, RT-qPCR for gene expression analysis, ChIP-qPCR techniques for sperm histone H3K4me3 analysis, a

Exposure to neonicotinoid pesticides induces physiological disorders and affects color performance and foraging behavior in goldfish.

We investigated the effects of neonicotinoid pesticides (NEOs) on the spontaneous swimming and foraging behavior, as well as the morphological and physiological changes of goldfish. Most fish reared in thiamethoxam (THM)-sprayed rice fields showed the scales easily peeled off, and increased ascites. Some individuals showed decreased bio-defense activity and low plasma Ca2+. Similar changes were found in the exposure test to THM (1.0 and 20.0 μg/L) and dinotefuran (1.2 and 23.5 μg/L). Next, the effects of a low concentration of THM (1.0 μg/L) on the spontaneous swimming and foraging behavior of

The chelation mechanism of neonicotinoid insecticides influencing cadmium transport and accumulation in rice at different growth stage.

The combined exposure of heavy metals and organic contaminates can influence the transport and accumulation of heavy metals within the soil-rice system. However, the underlying mechanisms of this process remain largely unknown. Herein, this study investigated the influence of three neonicotinoid insecticides (NIs), including imidacloprid (IMI), clothianidin (CLO), and thiamethoxam (THI), on the Cd transport and accumulation in rice (Oryza sativa) at different growth stages. Particular focus lied on their complex interaction and key genes expression involved in Cd transport. Results showed that

Concentrations of Neonicotinoid insecticides and their metabolites in multiple urine samples collected from pregnant women in Japan.

Exposure to low doses of neonicotinoid (NEO) insecticides during pregnancy may have a negative impact on the neurodevelopment of children. Biomarkers are frequently used to investigate the exposure and its health effects in environmental epidemiology. In the present study, we aimed to evaluate the utility of urinary biomarkers of NEO exposure by calculating intra-class correlation coefficients (ICCs). Urine samples were collected from 30 pregnant women who were non-smokers and living in a central part of Japan, between 12 and 22 weeks of gestation. We collected first morning voided (FMV) urine

Acute poisoning with acetamiprid, a type of neonicotinoid insecticide causing severe lactic acidosis: A case report.

Acetamiprid is a type of neonicotinoid that belongs to the new class of insecticides. It is fatally toxic to insects by acting as an agonist to nicotinic acetylcholine receptors. Cases with neonicotinoids including acetamiprid poisoning in humans were reported around the world. Only one case with acetamiprid poisoning associated with severe adverse effects has been described from Sri Lanka. Serious side effects will occur if it is consumed in excess amount. Here, we described a case of acetamiprid poisoning in a man who developed severe lactic acidosis, myocardial suppression, hyperglycemia, a

Experimental evolution supports the potential of neonicotinoid-pyrethroid combination for managing insecticide resistance in malaria vectors.

The introduction of neonicotinoids for managing insecticide resistance in mosquitoes is of high interest as they interact with a biochemical target not previously used in public health. In this concern, Bayer developed a combination of the neonicotinoid clothianidin and the pyrethroid deltamethrin (brand name Fludora Fusion) as a new vector control tool. Although this combination proved to be efficient against pyrethroid-resistant mosquitoes, its ability to prevent the selection of pyrethroid and neonicotinoid resistance alleles was not investigated. In this context, the objective of this work

Effects of Neonicotinoid Insecticides on Physiology and Reproductive Characteristics of Captive Female and Fawn White-tailed Deer.

Over the past decade, abnormalities have been documented in white-tailed deer (Odocoileus virginianus) in west-central Montana. Hypotheses proposed to explain these anomalies included contact with endocrine disrupting pesticides, such as imidacloprid. We evaluated the effects of imidacloprid experimentally at the South Dakota State University Wildlife and Fisheries Captive Facility where adult white-tailed deer females and their fawns were administered aqueous imidacloprid (an untreated control, 1,500 ng/L, 3,000 ng/L, and 15,000 ng/L). Water consumption, thyroid hormone function, behavioral r

Hummingbirds and bumble bees exposed to neonicotinoid and organophosphate insecticides in the Fraser Valley, British Columbia, Canada.

To measure exposure to neonicotinoid and other pesticides in avian pollinators, we made novel use of cloacal fluid and fecal pellets from rufous (Selasphorus rufus) and Anna's (Calypte anna) hummingbirds living near blueberry fields in the Fraser River Valley and Vancouver Island, British Columbia, Canada. To examine on-farm exposure to pesticides in invertebrate pollinators, we also collected bumble bees native to Canada (Bombus mixtus, Bombus flavifrons, and Bombus melanopygus), their pollen, and blueberry leaves and flowers from within conventionally sprayed and organic blueberry farms. Matrix-Dependent Photolysis of Neonicotinoid Insecticides: An Overlooked Source of Reactive Volatile Organic Compounds and Brown Carbon.

Neonicotinoid insecticides (NNs) generate matrix-dependent photoproducts with significant atmospheric implications. Through high-resolution mass spectrometry analysis of five NNs (nitenpyram, thiamethoxam, imidacloprid, clothianidin, and dinotefuran), we demonstrate that solid-phase photolysis produces 2.5 times more gaseous products (430 vs 170 compounds) than liquid-phase reactions. Solid-state transformations yield concerning chlorinated VOCs (13% of products) and 59 light-absorbing brown carbon (BrC) species, while aqueous-phase reactions favor oxygenated CHO compounds (72-100% abundance).

Texture and Neonicotinoid Exposure Shape Bacterial Assemblages and Functions in Agricultural Soils: Responses Over Prolonged Exposure.

The physical and chemical properties of soil fundamentally shape its microbial communities. In a controlled 28-day microcosm experiment, we assessed bacterial community responses to imidacloprid in three soils with differing textures and classifications: a loamy sand (11 g/100 g clay; red Luvisol), a sandy loam (16 g/100 g clay; red Luvisol) and a clay soil (56 g/100 g clay, Vertisol). Analyses included 16S rRNA gene amplicon sequencing, indicator species analysis, co-occurrence network analysis and PICRUSt2-based functional prediction. Imidacloprid exposure elicited soil-specific shifts in ba

Neonicotinoid seed treatments and early soybean planting: quantifying pest abundance, crop protection, and effects on yield.

Soybean (Glycine max (L.) Merr.) yields have increased with advances in plant breeding and management practices, including earlier planting. Early planting in temperate climates maximizes yield potential but may increase vulnerability to certain pests and environmental stressors. Neonicotinoid seed treatments, particularly thiamethoxam, are widely marketed to protect soybean seedlings, yet their effectiveness under typical early-season field conditions in Indiana remains unexplored. This 2-year (2023 to 2024) study was conducted at 3 sites in Indiana. Soybeans were planted during 4 discrete pe

Disrupting effects of neonicotinoids and their interaction with metals on thyroid hormone, an evidence of children in a rural area, South China.

Neonicotinoids exposure was found to induce thyroid dysfunction. However, there lack of direct evidence between neonicotinoids exposure and thyroid hormone (TH) disruption in population study, especially in children, which limits the understanding on their health hazard. To fill this knowledge gap, we conducted a cross-sectional study on children of a rural area in South China (n = 88), and analyzed urinary ten neonicotinoids (including metabolites), serum TH, thyroxine-binding globulin (TBG), and thyroid stimulating hormone (TSH) levels. Based on linear regression, generalized additive model,

Next-generation effects of fetal and lactational exposure to the neonicotinoid pesticide clothianidin on the immune system and gut microbiota.

Recently, the effects of exposure to clothianidin (CLO) on the thymus and gut microbiota have become clear, but no report has examined its next-generation impacts. Pregnant C57BL/6N mice were administered a no-observed-adverse-effect-level dose of CLO until weaning. We examined CLO's effects on the gut microbiota and immune organs of dams and their 3- and 10-week-old male offspring. CLO administration led to several alterations of the top 30 bacterial genera in the gut microbiota in dams and 3-week-old mice. Compared to controls, 10-week-old mice had more thymic Hassall's corpuscles, and both

The neonicotinoid thiacloprid leads to multiple defects during early embryogenesis of the South African clawed frog (Xenopuslaevis).

There is increasing concern about the health effects of pesticides that pollute natural waters. In particular, the use of neonicotinoids, such as thiacloprid (THD), is causing unease. THD is considered non-toxic to non-target vertebrates. Studies classify THD as carcinogenic, toxic to reproduction, and therefore harmful to the environment. A detailed study of possible THD effects during the amphibian embryogenesis is needed because leaching can introduce THD into aquatic environments. We incubated stage 2 embryos of the South African clawed frog in various THD concentrations (0.1-100 mg/L) at

Comparative study of the effect of solvents on the efficacy of neonicotinoid insecticides against malaria vector populations across Africa.

BACKGROUND: New insecticides with a novel mode of action such as neonicotinoids have recently been recommended for public health by WHO. Resistance monitoring of such novel insecticides requires a robust protocol to monitor the development of resistance in natural populations. In this study, we comparatively used three different solvents to assess the susceptibility of malaria vectors to neonicotinoids across Africa. METHODS: Mosquitoes were collected from May to July 2021 from three agricultural settings in Cameroon (Njombe-Penja, Nkolondom, and Mangoum), the Democratic Republic of Congo (Nd

Ca2+ imaging with two-photon microscopy to detect the disruption of brain function in mice administered neonicotinoid insecticides.

Neonicotinoid pesticides are a class of insecticides that reportedly have harmful effects on bees and dragonflies, causing a reduction in their numbers. Neonicotinoids act as neuroreceptor modulators, and some studies have reported their association with neurodevelopmental disorders. However, the precise effect of neonicotinoids on the central nervous system has not yet been identified. Herein, we conducted in vivo Ca2+ imaging using a two-photon microscope to detect the abnormal activity of neuronal circuits in the brain after neonicotinoid application. The oral administration of acetamiprid

Role of nicotinic acetylcholine receptor subunits in the mode of action of neonicotinoid, sulfoximine and spinosyn insecticides in Drosophila melanogaster.

Insecticides remain valuable tools for the control of insect pests that significantly impact human health and agriculture. A deeper understanding of insecticide targets is important in maintaining this control over pests. Our study systematically investigates the nicotinic acetylcholine receptor (nAChR) gene family, in order to identify the receptor subunits critical to the insect response to insecticides from three distinct chemical classes (neonicotinoids, spinosyns and sulfoximines). Applying the CRISPR/Cas9 gene editing technology in D. melanogaster, we were able to generate and maintain h

Evolution of tolerance to chlorpyrifos causes cross-tolerance to another organophosphate and a carbamate, but reduces tolerance to a neonicotinoid and a pharmaceutical.

Exposure to pesticides is a major stressor in freshwater ecosystems. While populations can evolve tolerance to pesticides and thereby ensure their persistence in contaminated environments, this may have important consequences for their sensitivity to other pollutants. Indeed, tolerance to one pollutant may both increase (as a cost of tolerance) or decrease (cross-tolerance) the sensitivity to other pollutants. Despite the increasing concern of pharmaceuticals in waterbodies, no patterns of pesticide-induced (cross-)tolerance have been studied. We conducted 48 h acute toxicity assays with a ran

Development of fluorescence polarization immunoassay for the rapid detection of 6-chloronicotinic acid: main metabolite of neonicotinoid insecticides.

A fluorescence polarization immunoassay (FPIA) for the quantitative determination of 6-chloronicotinic acid (6-CNA) using polyclonal antibody was developed. The 6-CNA-protein (bovine serum albumin and soybean trypsin inhibitor) conjugates and fluorescein-labeled 6-CNA derivative (tracer) were prepared and used as the immunogens and tracer, respectively. The synthesized tracer was purified by thin layer chromatography (TLC) and showed a good binding to antiserum (73/5) which was obtained from the immunized rabbit (No. 73) with 6-CNA-BSA conjugate. The detection limit (10% inhibition) of FPIA wa
