Comparing Processed Nerve Allografts and Assessing Their Capacity to Retain and Release Nerve Growth Factor.

Peripheral nerve gap injuries continue to present a clinical challenge to today's surgeons. One method of surgical repair, implantation of acellular allografts, has been developed with the aim of bridging the gap with a cadaveric graft after removal of its cellular components, thereby accelerating axonal regeneration and eliminating the need for immunosuppression in recipient patients. Although decellularizing allografts reduces rates of graft rejection, the same chemical processing modifies the neural microenvironment, removing neutrotrophic factors and modifying the complex extracellular mat

Expression of nerve growth factor and its receptor in distracted tibial nerve after limb lengthening.

Despite many experimental and clinical studies conducted on distraction osteogenesis (DO) in the past decade, changes in the surrounding tissues that occur after the procedure remains poorly understood. To study the biochemical changes of recovery in nerve tissues upon DO-induced nerve injury, we prepared a rabbit model of tibia lengthening to observe the expression pattern of nerve growth factor (NGF) and low-affinity NGF receptor (p75NGFR) in the distracted tibial nerve. The distracted tibial nerve was harvested at various time points during the consolidation period of new bone formation and

The early effect of nerve growth factor in the management of serious optic nerve contusion.

OBJECTIVE: the aim was to investigate the early effect of nerve growth factor in the management of serious optic nerve contusion (ONC). METHODS: nine patients with unilateral serious optic nerve contusion (seven males and two females) were enrolled in this study. Corticosteroid, energy mixture, red sage root, vitamins B1 and B12 were used for all patients within three weeks after trauma. Five of the nine patients were injected intramuscularly with 30 microg/time of nerve growth factor solution (once a day for a total of 14 days). Each patient was subjected to pattern visual evoked potential (

Altered nerve growth factor level in the optic nerve of patients affected by multiple sclerosis.

In this study we measured with a highly sensitive two-site immunoenzymatic assay the levels of nerve growth factor (NGF) in human optic nerve affected by multiple sclerosis (MS). The result of this study showed an elevated increase of NGF in the MS optic nerves, as compared to the level of NGF found in the optic nerves non-affected by this demyelinating disorder. Moreover, the results showed that the optic nerves also expressed trkA NGF receptor and NGFmRNA, most likely by oligodendrocytes, implying that NGF is locally produced and suggesting that the presence of NGF might be regulated by an a

Effects of nerve growth factor on nerve regeneration through a vein graft across a gap.

The limited availability of donor sites for nerve grafts and their inherent associated morbidity continue to stimulate research toward finding suitable alternatives. In the following study, the effect of direct administration of nerve growth factor (NGF) into a nerve conduit across a gap was tested in a rat sciatic nerve model. A 1-cm segment of the right sciatic nerve in Sprague-Dawley rats was resected, and the gap was then bridged using one of three methods: group I (NGF-treated group, n = 12), a vein graft filled with NGF (100 ng in 0.3-ml phosphate buffered saline); group II (control grou

Primary sensory neuron survival following targeted administration of nerve growth factor to an injured nerve.

Nerve injuries induce neurochemical changes within primary sensory neurons, including expression of neuropeptides, and a loss of a substantial proportion of the neurons may possibly be caused by a lack of neurotrophic support. In the present study the role of nerve growth factor (NGF) in preventing these changes was investigated in monkeys by giving NGF peripherally through a fibronectin (Fn) conduit. A sensory nerve (superficial radial) was transected and a gap of 5 mm was bridged with either autologous sural nerve graft (SNG), Fn, or Fn impregnated with NGF (Fn-NGF). After four months the do

Expression of choline acetyltransferase and nerve growth factor receptor within hypoglossal motoneurons following nerve injury.

In the present study we employed light microscopic immunocytochemical techniques in order to investigate the temporal response of choline acetyltransferase (ChAT) and nerve growth factor receptor (NGFr) within hypoglossal motoneurons following unilateral transection or crushing of the XII nerve or after intraneural injections of ricin into the nerve. In control rats (i.e., sham operated) virtually all the motoneurons of the XII nucleus displayed intense immunolabeling for ChAT and were devoid of NGFr immunoreactivity. As early as 3 days post-operative the intensity and the number of ChAT-label

Levodopa induces synthesis of nerve growth factor and growth hormone in patients with Parkinson disease.

BACKGROUND: Polyneuropathy is observed in treated patients with Parkinson disease. Peripheral axonal degeneration is related to nerve growth factor metabolism. The causes for this axonal degeneration and the role of levodopa itself are not known. Levodopa may experimentally exert supportive effects on nerve growth factor synthesis and on growth hormone production, a hormonal compound with regenerative potential. OBJECTIVE: The objective of this study was to investigate the effects of soluble 250-mg levodopa/benserazide administration on plasma occurrence of levodopa, nerve growth factor, and

Increased Epidermal Nerve Growth Factor without Small-Fiber Neuropathy in Dermatomyositis.

Small-fiber neuropathy (SFN) is suggested to be involved in the pathogenesis of some types of autoimmune connective tissue diseases. SFN with a reduction in epidermal nerve fibers might affect sensory fibers and cause neuropathic symptoms, such as pruritus and pain, which are common in both dermatomyositis (DM) and cutaneous lupus erythematosus (CLE). Nerve growth factor (NGF) has been recognized as important in nociception by regulating epidermal nerve fiber density and sensitizing the peripheral nervous system. The present study aimed to investigate whether SFN was associated with the cutane

Brain-derived nerve growth factor in the cochlea - a reproducibility study.

OBJECTIVE: Brain-derived nerve growth factor (BDNF) plays an important role in cochlear development so it is plausible that it could restore hearing loss if delivered directly into the cochlea. We wished to confirm our previous report that a single intracochlear injection of brain-derived nerve growth factor (BDNF) was beneficial for hearing in guinea pigs. We wished to assess the reproducibility of our results and assess possible improved methods with a view to developing a clinical treatment for sensorineural hearing loss. METHODS: CDDP was used to create partial hearing loss in 25 guinea p

Brain targeting of nerve growth factor using poly(butyl cyanoacrylate) nanoparticles.

The nerve growth factor (NGF) is essential for the survival of both peripheral ganglion cells and central cholinergic neurons in the basal forebrain. The accelerated loss of central cholinergic neurons during Alzheimer's disease may be a determinant cause of dementia, and this observation may suggest a possible therapeutic benefit from treatment with NGF. In recent years, convincing data have been published involving neurotrophic factors for the modulation of dopaminergic transmission within the brain and concerning the ability of NGF to prevent the degeneration of dopaminergic neurons. In thi

Developmental profile of serum nerve growth factor levels in Rett complex.

Rett syndrome (RS) is a progressive neurodevelopmental disorder predominant in females, characterised by mental deficiency, stereotyped hand-washing and apraxia. Notwithstanding the recent identification of the MECP2 gene likely involved in the pathogenesis of SR, the neurobiological bases of this syndrome are still largely unknown. Converging evidence shows that the brain levels of nerve growth factor (NGF), a neurotrophin regulating the development and functioning of central cholinergic neurons, are decreased in RS girls. In this study, the serum levels of NGF were measured in classic RS, in

Oestrogen and nerve growth factor - neuroprotection and repair in Alzheimer's disease.

The neurogenetics and neuropathology of Alzheimer's disease (AD) are still largely unknown, even though recent work has clarified some genetic components in this common and devastating neurodegenerative disease. Most of the genetic mutations have been shown to be, at least in the early onset type of AD, related to the function of a large transmembrane protein, amyloid precursor protein (APP). This protein is cleaved into various smaller fragments that are either soluble or aggregating. It is thought that this processing of APP is inherently important for the initiation and progression of AD. R

Nerve growth factor stimulates production of neuropeptide Y in human lymphocytes.

The production of neuropeptide Y (NPY) in lymphocytes obtained from human tonsils was investigated using radioimmunoassay. While unstimulated lymphocytes did not produce detectable amounts of NPY, NPY synthesis was induced after cell activation. Our results show that the addition of nerve growth factor (NGF) to unstimulated lymphocytes has an effect similar to that of mitogens, both leading to production of NPY. The study of purified B and T cells confirmed that only activated cells are able to synthesize NPY. The stimulatory effect of NGF on NPY production is not a common characteristic of al
