Domain-specific mutations in unc-6/Netrin differentially affect dorsal-ventral axon pathfinding in Caenorhabditis elegans.

UNC-6/Netrin is a conserved regulator of dorsal-ventral axon and cell migrations. Here, we identified missense mutations in distinct UNC-6 domains and assessed their roles in dorsal VD/DD motor axon guidance and ventral anterior ventral microtubule (AVM) axon guidance. A missense mutation in a conserved residue of the laminin N-terminal (LN) domain (G289D) resulted in dorsal and ventral axon guidance defects similar to the unc-6 null. A distinct missense mutation in the LN domain (S120F) strongly perturbed ventral AVM axon guidance with minimal effects on dorsal VD/DD axon guidance. Mutations

miR-27a-3p Upregulation and Netrin-1 Deficiency Drive Inflammatory Responses and Nerve Degeneration in Patients With Diabetes.

BACKGROUND: Inflammation is increasingly recognized in the pathogenesis of diabetic neuropathy (DN). Circulating miR-27a-3p levels are closely related to inflammation and increased in patients with diabetic nephropathy and retinopathy. miR-27a-3p has a predicted binding site in the 3 UTR of Netrin-1 mRNA, an anti-inflammatory nerve growth factor whose levels correlate with small nerve fiber loss in patients with DN. METHODS: Eighty-two participants with (DN+) and without (DN-) small nerve fiber damage and 45 healthy controls underwent measurement of plasma miR-27a-3p, PBMC levels of NF-kB and

Antitumor efficacy achieved by targeting PEGylated nanomedicines to netrin-1 in the extracellular matrix or EphA2 on the cancer cell membrane.

Targeting nanocarriers to tumors to increase their therapeutic efficacy requires selection of appropriate cancer targets and may depend on the particular formulation under investigation. Here, we compare the effectiveness of targeting PEGylated liposomal doxorubicin (PLD) or glycosidic switch liposomes (GSL) formulated with a glucuronide prodrug of 9-aminocamptothecin to ephrin receptor A2 (EphA2) present on the membrane of cancer cells or netrin-1 present in the tumor extracellular matrix. Targeting PLD to EphA2 or netrin-1 with bispecific PEG engagers significantly decreased the IC50 value o

Association of serum Netrin-1, NSE, and S100β with brain injury severity and prognosis in patients with sepsis-associated encephalopathy.

Sepsis-associated encephalopathy (SAE) represents the most prevalent neurological complication of sepsis and is frequently linked to unfavorable patient outcomes. This study aimed to evaluate the prognostic significance of serum Netrin-1, neuron-specific enolase (NSE), and S100β levels in patients diagnosed with SAE. A retrospective analysis was performed on 120 SAE patients, measuring serum levels of Netrin-1, NSE, and S100β and correlating these with Acute Physiology and Chronic Health Evaluation II (APACHE-II) scores. Independent risk factors for short-term mortality were identified, and th

Pan-cancer analysis reveals netrin-1 receptors as potential tumor biomarkers and immune-related therapeutic targets.

The signal pathways mediated by axon guiding molecule netrin-1 (NTN1) are transduced via its several membrane-bound receptors that include deleted in colorectal cancer (DCC), UNC5 (A-D), neogenin 1 (Neo1), melanoma cell adhesion molecule (MCAM), and Down syndrome cell adhesion molecule (DSCAM). Most of these genes play a role in the occurrence and progression of some solid tumors. Sufficient systematic studies have not been performed on the expression characteristics of the role of NTN1 and its receptors in the context of pan-cancer. Based on data from 10,437 subjects with 33 types of solid tu

UNC5B Overexpression Alleviates Peripheral Neuropathic Pain by Stimulating Netrin-1-Dependent Autophagic Flux in Schwann Cells.

Lesions or diseases of the somatosensory system can cause neuropathic pain (NP). Schwann cell (SC) autophagy plays an important role in NP. Uncoordinated gene 5 homolog B (UNC5B), the canonical dependent receptor of netrin-1, is known to be exclusively expressed in SCs and involved in NP; however, the underlying mechanisms were unclear. A rat model of sciatic nerve chronic constriction injury (CCI) was used to induce peripheral neuropathic pain. Adeno-associated virus (AAV) overexpressing UNC5B was applied to the injured nerve, and an autophagy inhibitor, 3-mechyladenine (3-MA), was intraperit

Netrin-1 receptor UNC5C cleavage by active δ-secretase enhances neurodegeneration, promoting Alzheimer's disease pathologies.

Netrin-1, a family member of laminin-related secreted proteins, mediates axon guidance and cell migration during neural development. T835M mutation in netrin receptor UNC5C predisposes to the late-onset Alzheimer's disease (AD) and increases neuronal cell death. However, it remains unclear how this receptor is molecularly regulated in AD. Here, we show that δ-secretase selectively cleaves UNC5C and escalates its proapoptotic activity, facilitating neurodegeneration in AD. Netrin deficiency activates δ-secretase that specifically cuts UNC5C at N467 and N547 residues and enhances subsequent casp

Increased expression of Netrin-4 is associated with allodynia in a trigeminal neuropathic pain model rats by infraorbital nerve injury.

Neuropathic pain refers to pain caused by lesions or diseases of the somatosensory nervous system that is characteristically different from nociceptive pain. Moreover, neuropathic pain occurs in the maxillofacial region due to various factors and is treated using tricyclic antidepressants and nerve block therapy; however, some cases do not fully recover. Netrin is a secreted protein crucially involved in neural circuit formation during development, including cell migration, cell death, neurite formation, and synapse formation. Recent studies show Netrin-4 expressed in the dorsal horn of the sp

Netrin-1 Regulates Fibrocyte Accumulation in the Decellularized Fibrotic Sclerodermatous Lung Microenvironment and in Bleomycin-Induced Pulmonary Fibrosis.

OBJECTIVE: Fibrocytes are collagen-producing leukocytes that accumulate in patients with systemic sclerosis (SSc; scleroderma)-related interstitial lung disease (ILD) via unknown mechanisms that have been associated with altered expression of neuroimmune proteins. The extracellular matrix (ECM) influences cellular phenotypes. However, a relationship between the lung ECM and fibrocytes in SSc has not been explored. The aim of this study was to use a novel translational platform based on decellularized human lungs to determine whether the lung ECM of patients with scleroderma controls the develo

A guide to selecting high-performing antibodies for Netrin-1 (UniProt ID: O95631) for use in western blot and immunoprecipitation.

Netrin-1 is a secreted protein that regulates cell migration and survival, controlling axonal guidance during development, morphogenesis, apoptosis, angiogenesis, inflammation and cancer progression. Here we have characterized fourteen Netrin-1 commercial antibodies for western blot and immunoprecipitation using a standardized experimental protocol based on comparing read-outs in knockout cell lines and isogenic parental controls. These studies are part of a larger, collaborative initiative seeking to address antibody reproducibility issues by characterizing commercially available antibodies f

Netrin-3 rectifies diabetes-induced cognitive impairment by counteracting hippocampal oxidative stress and NLRP3 inflammasome activation.

Diabetes frequently causes substantial cognitive decline, yet available treatments offer minimal benefit. Netrin-3, a laminin-like protein, regulates nervous system development and inflammatory responses. However, its involvement in diabetes-associated cognitive impairment remains poorly understood. The escalating global diabetes burden, combined with the absence of disease-modifying therapies for cognitive complications, creates an urgent need to identify novel molecular targets. Our research evaluated whether Netrin-3 could counter cognitive deterioration using db/db mice as a type 2 diabete

Netrin-1 promotes liver regeneration possibly by facilitating vagal nerve repair after partial hepatectomy in mice.

Hepatic regeneration after hepatectomy is a great concern in clinical practice. Recently, the neuronal guidance protein netrin-1 has been reported to enhance regeneration after nerve injury. The goal of this study was to preliminarily investigate whether netrin-1 stimulates vagus nerve regeneration to promote liver regeneration after partial hepatectomy in mice. The expression of netrin-1 in murine remnant livers after partial hepatectomy (PHx) was evaluated in initial studies. C57BL/6 mice that received exogenous netrin-1 after PHx were used to examine liver regeneration. PHx was performed in

Homozygous Missense Variants in NTNG2, Encoding a Presynaptic Netrin-G2 Adhesion Protein, Lead to a Distinct Neurodevelopmental Disorder.

NTNG2 encodes netrin-G2, a membrane-anchored protein implicated in the molecular organization of neuronal circuitry and synaptic organization and diversification in vertebrates. In this study, through a combination of exome sequencing and autozygosity mapping, we have identified 16 individuals (from seven unrelated families) with ultra-rare homozygous missense variants in NTNG2; these individuals present with shared features of a neurodevelopmental disorder consisting of global developmental delay, severe to profound intellectual disability, muscle weakness and abnormal tone, autistic features

Synergistic Activity of Floor-Plate- and Ventricular-Zone-Derived Netrin-1 in Spinal Cord Commissural Axon Guidance.

In vertebrates, commissural axons extend ventrally toward the floor plate in the spinal cord and hindbrain. Netrin-1, secreted by floor plate cells, was proposed to attract commissural axons at a distance. However, recent genetic studies in mice have shown that netrin-1 is also produced by ventricular zone (VZ) progenitors and that in the hindbrain, it represents the main source of netrin-1 for commissural axons. Here, we show that genetically deleting netrin-1 either from the VZ or the floor plate does not prevent midline crossing in the spinal cord, although axon pathfinding and fasciculatio

Netrin G1: its downregulation in the nucleus accumbens of cocaine-conditioned mice and genetic association in human cocaine dependence.

Netrin G1 is a presynaptic ligand involved in axonal projection. Although molecular mechanisms underlying cocaine addiction are still poorly understood, Netrin G1 might have a role as a regulator of anxiety, fear and spatial memory, behavioural traits impaired in the context of cocaine exposure. In this study, the Netrin G1 (Ntng1) expression was investigated in the nucleus accumbens of mice primarily conditioned to cocaine using a place preference paradigm. A genetic association study was then conducted on 146 multiplex families of the Collaborative study on Genetics of Alcoholism, in which s

Netrin-1 Therapy Restores Partial Hindlimb Movement in a Rat Model of High-Severity Chronic Spinal Cord Injury.

Spinal trauma caused by contusion or compression is the leading cause of spinal cord injury (SCI) worldwide. These injuries frequently progress to a chronic phase, especially in cases of severe damage. This process results in permanent impairment, affecting both physiological functions and voluntary motor control below the lesion level. At cellular level, the formation of a glial scar, which delineates the cystic cavity, interrupts the connectivity between the central nervous system (CNS) and muscles, as well as the neural communication between the peripheral and CNSs. This process, combined w

Spatiotemporal changes in Netrin/Dscam1 signaling dictate axonal projection direction in Drosophila small ventral lateral clock neurons.

Axon projection is a spatial- and temporal-specific process in which the growth cone receives environmental signals guiding axons to their final destination. However, the mechanisms underlying changes in axonal projection direction without well-defined landmarks remain elusive. Here, we present evidence showcasing the dynamic nature of axonal projections in Drosophila's small ventral lateral clock neurons (s-LNvs). Our findings reveal that these axons undergo an initial vertical projection in the early larval stage, followed by a subsequent transition to a horizontal projection in the early-to
