First attempt of boron neutron capture therapy for adenoid cystic carcinoma liver metastasis: a case report and literature review.

This report details a 55-year-old male with liver metastases from Adenoid Cystic Carcinoma (ACC), enrolled in a study on Boron Neutron Capture Therapy (BNCT) utilizing Boronophenylalanine (BPA) as a carrier. The patient underwent two BNCT sessions. Within three months of the first BNCT, the tumor area remained stable, and pre-existing telangiectasia completely disappeared. No irreversible adverse effects of BNCT were observed. Despite ongoing dose exploration, this study highlights BNCT's therapeutic potential for ACC-related liver lesions.

Biomimetic Macrophage Cell Membrane-Based Nanoparticles for Effective Treatment of Glioblastoma Through Boron Neutron Capture Therapy Combined With Immunotherapy.

Glioblastoma multiforme (GBM) remains largely incurable due to the blood-brain barrier (BBB) and immunosuppressive microenvironment. While boron neutron capture therapy (BNCT) selectively eradicates tumor cells via 10B(n, α)-7Li reactions, its clinical potential in GBM is unrealized because of the suboptimal pharmacokinetics of conventional boron agents and their failure to achieve therapeutic intracranial concentrations. Here, we design macrophage membrane-coated PLGA nanoparticles (M@PLGA-10BN) that exploit innate biological transport mechanisms to overcome dual therapeutic barriers. The mac

Assessment of long-term risks of secondary cancer in paediatric patients with brain tumours after boron neutron capture therapy.

This study firstly explored the risks of secondary cancer in healthy organs of Chinese paediatric patients with brain tumours after boron neutron capture therapy (BNCT). Three neutron beam irradiation geometries (i.e. right lateral, top to bottom, posterior to anterior) were adopted in treating patients with brain tumours under the clinical environment of BNCT. The concerned organs in this study were those with high cancer morbidity in China (e.g. lung, liver and stomach). The equivalent doses for these organs were calculated using Monte Carlo and anthropomorphic paediatric phantoms with Chine

Boron neutron capture therapy and 18F-labelled borophenylalanine positron emission tomography: a critical and clinical overview of the literature.

Positron emission tomography (PET) is considered one of the most useful tool for molecular imaging both in clinical and preclinical research for in vivo assessing of biochemical and pharmacological processes. Boron neutron capture therapy (BNCT) is a biologically-targeted radiotherapy that can selectively hit the tumour cells, saving the surrounding normal tissue. Boron 10 ((10)B) is the isotope widely used for this purpose, and acts as killer for tumor cells, releasing highly reactive α and (7)Li-particles when it absorbs a thermal neutron. The basic requirements for a successful BNCT treatme

Current role of boron neutron capture therapy in head and neck cancer: insights from 4 years of clinical experience.

PURPOSE: Boron neutron capture therapy (BNCT) was approved in Japan in June 2020 as a curative option for unresectable, locally advanced, or recurrent head and neck cancers. Relevantly, its efficacy and safety in clinical practice should be regularly monitored, particularly regarding its optimal positioning. Therefore, to advance its clinical application, we assessed clinical data on BNCT for head and neck cancers. METHODS: We retrospectively analyzed 285 patients who received initial BNCT between June 2020 and May 2024 with available ≥1 month follow-up data. The primary endpoint was local co

From tumor imaging probes to boron neutron capture therapy: a strategy for designing targeted boron carriers for pancreatic Cancer.

Pancreatic cancer is characterized by late detection, early metastatic dissemination, therapy resistance, and a dense desmoplastic stroma that restricts drug access. Surgical resection with adjuvant chemotherapy confers only modest short-term benefit, but systemic toxicity, acquired chemoresistance, and postoperative relapse constrain long-term survival. Boron neutron capture therapy (BNCT) is a binary radiotherapy in which thermal neutron capture by 10B triggers the 10B(n, α)7Li reaction, releasing high-linear-energy-transfer alpha particles and lithium-7 nuclei. Their 5-9 μm range confines c

Development of a Theranostic Agent for Bone Metastases: Integration of Nuclear Medicine Imaging and Boron Neutron Capture Therapy (BNCT).

Boron neutron capture therapy (BNCT) is one of the promising cancer treatment methods with 10B-labeled compounds and neutron irradiation. The 10B(n,α)7Li reaction produces a 7Li nucleus and an α-particle with high linear energy transfer, which are responsible for the therapeutic effects. We hypothesized that BNCT could effectively treat bone metastases by selectively accumulating 10B at metastatic lesion sites. In this study, we designed, synthesized, and evaluated [nat/67Ga]Ga-DOTA-K(ε-closo-dodecaborate)D11 ([nat/67Ga]6), a compound integrating closo-dodecaborate for BNCT, a [nat/67Ga]Ga-DOT

Tumor Eradication by Boron Neutron Capture Therapy with 10 B-enriched Hexagonal Boron Nitride Nanoparticles Grafted with Poly(Glycerol).

Boron neutron capture therapy (BNCT) has emerged as a treatment modality with high precision and efficacy of intractable tumors. At the core of effective tumor BNCT are 10 B carriers with facile preparation as well as advantageous pharmacokinetic and therapeutic profiles. Herein, the design and preparation of sub-10 nm 10 B-enriched hexagonal boron nitride nanoparticles grafted with poly(glycerol) (h-10 BN-PG), and their application to cancer treatment by BNCT are reported. By virtue of their small particle size and outstanding stealth property, h-10 BN-PG nanoparticles accumulate efficiently

Development of Monte Carlo based real-time treatment planning system with fast calculation algorithm for boron neutron capture therapy.

PURPOSE: We simulated the effect of patient displacement on organ doses in boron neutron capture therapy (BNCT). In addition, we developed a faster calculation algorithm (NCT high-speed) to simulate irradiation more efficiently. METHODS: We simulated dose evaluation for the standard irradiation position (reference position) using a head phantom. Cases were assumed where the patient body is shifted in lateral directions compared to the reference position, as well as in the direction away from the irradiation aperture. For three groups of neutron (thermal, epithermal, and fast), flux distributi

Assessment of soft error risks to cardiac implantable electronic devices for boron neutron capture therapy using field-programmable gate arrays.

PURPOSE: Boron neutron capture therapy (BNCT) is a promising cancer treatment; however, it poses the risk of soft errors in cardiac implantable electronic devices (CIEDs). This study aimed to assess the occurrence of soft errors in CIEDs during BNCT and to correlate these errors with the neutron flux. MATERIALS AND METHODS: A field-programmable gate array (FPGA) was used as a surrogate for CIED to measure the soft errors under thermal and epithermal neutron irradiation. The neutron flux at the FPGA position was assessed using gold wire activation, and the effective soft-error cross-section wa

Design, Synthesis, and Biological Evaluation of Boron-Containing Macrocyclic Polyamines and Their Zinc(II) Complexes for Boron Neutron Capture Therapy.

Boron neutron capture therapy (BNCT) is a binary therapeutic method for cancer treatment based on the use of a combination of a cancer-specific drug containing boron-10 (10B) and thermal neutron irradiation. For successful BNCT, 10B-containing molecules need to accumulate specifically in cancer cells, because destructive effect of the generated heavy particles is limited basically to boron-containing cells. Herein, we report on the design and synthesis of boron compounds that are functionalized with 9-, 12-, and 15-membered macrocyclic polyamines and their Zn2+ complexes. Their cytotoxicity, i

Exploring Targeted Delivery Systems for Boron Neutron Capture Therapy and Its Potential as a Promising Therapeutic Modality for Hepatocellular Carcinoma.

BACKGROUND: Boron neutron capture therapy (BNCT) is a precision binary radiotherapy. In this modality, thermal neutrons combine with 10B to induce a nuclear reaction that kills tumor cells. Its therapeutic efficacy depends on the targeted accumulation of boron delivery agents. BNCT has demonstrated clinical efficacy in treating head and neck cancers and recurrent gliomas. However, there is limited evidence regarding its application in hepatocellular carcinoma (HCC). This review systematically examines recent advances in novel boron carriers. It also assesses the potential of BNCT for treating

Salvage surgery following boron neutron capture therapy in a case of recurrent hypopharyngeal cancer after radiotherapy: Surgical and pathological insights.

Radiotherapy (RT) is a cornerstone management strategy for head and neck cancer (HNC); however, local recurrence often requires salvage surgery, which is associated with an elevated risk of complications, particularly in previously irradiated fields. Boron neutron capture therapy (BNCT) has broadened treatment options; however, the safety and feasibility of salvage surgery after BNCT remain unclear owing to the limited case number. Herein, we describe a rare case of hypopharyngeal squamous cell carcinoma initially treated with RT, followed by BNCT for local recurrence, and ultimately with succ
