Cold-smoked salmon are ready-to-eat products that may support the growth of pathogenic Listeria monocytogenes during their long shelf-life. Consumption of such contaminated products can cause fatal listeriosis infections. Another challenge and potential risk associated with CS salmon is their high levels of sodium salt. Excess dietary intake is associated with serious health complications. In the present study, anti-listerial bacteriocin (nisin), P100 bacteriophages (Phageguard L, PGL) and fermentates (Verdad N6, P-NDV) were evaluated as commercial bio-preservation strategies for increased con

Quantitative modeling of combination therapy can describe the effects of each antibiotic against multiple bacterial populations. Our aim was to develop an efficient experimental and modeling strategy that evaluates different synergy mechanisms using a rapidly killing peptide antibiotic (nisin) combined with amikacin or linezolid as probe drugs. Serial viable counts over 48 h were obtained in time-kill experiments with all three antibiotics in monotherapy against a methicillin-resistant Staphylococcus aureus USA300 strain (inoculum, 10(8) CFU/ml). A sequential design (initial dosing of 8 or 32

The aim of this study was to analyze the resistance of Staphylococcus aureus isolates from bovine mastitis in the eastern part of Poland to a set of 20 antibiotics and three alternative agents: lysostaphin, nisin and polymyxin B. Eighty-six out of 123 examined isolates were susceptible to all 20 tested antibiotics (70%). The highest percentage of resistance was observed in the case of β-lactam antibiotics: amoxicillin (n=22, 17.9%), ampicillin (n=28, 22.8%), penicillin (n=29, 23.6%) and streptomycin (n=13; 10.6%). Twenty-five of the penicillin-resistant strains were found to carry the blaZ gen

The effects of acid, oxgall, and H(2)O(2) on susceptibilities to antibiotics and nisin were examined for 13 strains of bifidobacteria. Susceptibilities to ampicillin, cloxacillin, penicillin, vancomycin, kanamycin, neomycin, paramomycin, streptomycin, chloramphenicol, erythromycin, tetracycline, and nisin A were assayed by a microdilution broth method. Acid-, oxgall- and H(2)O(2)-stressed variants were produced and assayed. Exposure to a pH of 2.0 for 60 min reduced susceptibilities to cloxacillin and nisin A but increased susceptibilities to ampicillin, vancomycin, aminoglycosides, chloramphe

Nisin, a natural substance from Lactococcus lactis, displays a promising antibacterial ability against the gram-positive bacteria. However, it is susceptible to the external environment, i.e. temperature, pH, and food composition. In this study, a dual stabilization method, coaxial electrospinning, was applied to protect nisin in food packaging materials and the effect of nisin concentration on the properties of the nanofibers was investigated. The core-shell nanofibers with pullulan as a core layer and carboxymethyl chitosan (CMCS)/polyethylene oxide (PEO) as shell layer were prepared, and th

Lactococcus lactis, an internationally recognized safe species of Gram-positive bacteria, produces a variety of antimicrobial peptides. In this study, we examined the irpT gene of L. lactis N8 and its influence on the bacterial stress response. We used the Cre-loxP system to knock out the irpT gene from the L. lactis N8Δply10300 genome. The mutant strains L. lactis N8Δply10300 and L. lactis N8Δply10300ΔirpT showed higher nisin tolerance and nisin production than the wild-type strain L. lactis N8, and also obtained tolerance for multiple stressors. The biological characteristics of the irpT gen

The ability of Lactococcus lactis subsp. lactis CECT 539 and Pediococcus acidilactici NRRL B-5627 to produce bacteriocins on both diluted and concentrated whey was investigated in batch fermentations. Both strains produced the higher amounts of biomass and bacteriocin titres on diluted whey. Luedeking and Piret expression was able to model the production of nisin, which was produced as a primary metabolite on both culture media. However, the pediocin production could not be typified in any case due to the negligible growth of P. acidilactici. Although the whey supported the growth and bacterio

Epidemiological studies reveal significant associations between periodontitis and oral cancer. However, knowledge about the contribution of periodontal pathogens to oral cancer and potential regulatory mechanisms involved is limited. Previously, we showed that nisin, a bacteriocin and commonly used food preservative, reduced oral cancer tumorigenesis and extended the life expectancy in tumor-bearing mice. In addition, nisin has antimicrobial effects on key periodontal pathogens. Thus, the purpose of this study was to test the hypothesis that key periodontal pathogens (Porphyromonas gingivalis,

BACKGROUND: Bacteriocins are promising alternatives to antibiotics in poultry production, offering pathogen control with minimal disruption to gut microbiota and reduced risk of resistance dissemination. This study compared the impact of three bacteriocins-microcin J25, nisin Z, and pediocin PA-1-against bacitracin as a positive control, specifically evaluating their effects on gut microbiota composition and metabolic activity. The study utilized the PolyFermS continuous fermentation model to simulate chicken caecal conditions.
RESULTS: 16S rRNA sequencing revealed that bacitracin and nisin Z

Bacteriocins, antimicrobial peptides produced by bacteria, are expected to be novel antimicrobial agents. Previous studies demonstrated that several two-component regulatory systems (TCSs) linked to the expression of lantibiotic transporters are involved in resistance to lantibiotics in Staphylococcus aureus. In this study, we examined the susceptibility to four Class Ia bacteriocins (lantibiotics) among clinical isolates of S. aureus and sought to reveal the mechanisms underlying differences in susceptibility among strains. A direct assay was performed using nisin A, Pep5, nukacin ISK-1, and

According to the World Health Organization (WHO), multiple drug-resistant (MDR) bacterial infection is a top threat to human health. Since bacteria evolve to resist antibiotics faster than scientists can develop new classes of drugs, the development of new materials which can be used, not only for separation, but also for effective disinfection of drug resistant pathogens is urgent. Driven by this need, we report for the first time the development of a nisin antimicrobial peptide conjugated, three dimensional (3D) porous graphene oxide membrane for identification, effective separation, and com

With the increasing application of antimicrobial peptides (AMPs) to replace antibiotics in medicine, food and agriculture, it is highly desired to develop a fast, reliable, and convenient strategy for sensitive detection of AMPs. Herein, a novel electrochemical (EC) and electrochemiluminescence (ECL) dual-modality sensing platform was developed based on probe encapsulated liposomes and nanochannel array modified electrodes, which enables sensitive determination of nisin in food samples. The bifunctional probe with both EC and ECL signals, tris(2,2-bipyridyl) dichlororuthenium (II) (Ru(bpy)3 2+

: The effects of ultrasound (US), thermosonication (TS), ultrasound combined with nisin (USN), TS combined with nisin (TSN), and conventional thermal sterilization (CTS) treatments on the inactivation of microorganisms in grape juice were evaluated. TS, TSN, and CTS treatments provided the desirable bactericidal and enzyme inactivation, and nisin had a synergistic lethal effect on aerobic bacteria in grape juice while not having any obvious effect on the mold and yeast. Compared with CTS, the sensory characteristics of grape juice treated with TS and TSN are closer to that of fresh juice, its

In this research, the chemical composition and biological properties of Tunisian Myrtus communis (McEO) flowers were investigated. The antibacterial effect of McEO toward some bacteria was assessed, alone and in combination with nisin. The major components of McEO were α-pinene, 1,8-cineol, limonene, and linalool. McEO exhibited cytotoxicity toward HepG2 and MCF-7 cell lines. The microbiological data showed that Gram-positive bacteria were more susceptible to McEO. McEO had a bactericidal effect against L. monocytogenes. McEO is able to prevent lipid oxidation, microbial development at noncyto

We characterized the regulated activity of the lactococcal nisA promoter in strains of the gram-positive species Streptococcus pyogenes, Streptococcus agalactiae, Streptococcus pneumoniae, Enterococcus faecalis, and Bacillus subtilis. nisA promoter activity was dependent on the proteins NisR and NisK, which constitute a two-component signal transduction system that responds to the extracellular inducer nisin. The nisin sensitivity and inducer concentration required for maximal induction varied among the strains. Significant induction of the nisA promoter (10- to 60-fold induction) was obtained

Staphylococcus aureus is a bacterium that mainly colonizes the nasal cavity and skin. To colonize the host, it is necessary for S. aureus to resist many antibacterial factors derived from human and commensal bacteria. Among them are the bacteria-derived antimicrobial peptides (AMPs) called bacteriocins. It was reported that some two-component systems (TCSs), which are signal transduction systems specific to bacteria, are involved in the resistance to several bacteriocins in S. aureus. However, the TCS-mediated resistance is limited to relatively low concentrations of bacteriocins, while high c

Nanofibrous zein/PEG based membranes incorporated with natural antimicrobial compounds were fabricated by electrospinning method. Structural and thermal analysis of prepared nanofibers revealed that the applied processing technique did not significantly affect the structure of pristine zein polymer. Morphological characterization showed a higher degree of polydispersity in the fibers modified with eugenol, thymol, nisin, or their combinations, and an average fiber diameter in the range from 300 to 390 nm. Nanofibrous samples with eugenol and thymol prevented the growth of Escherichia coli and

Bacteriophages infecting dairy starter cultures are a serious threat in the dairy industry leading to failed or low-quality milk fermentations. The aim of this work was to assess if the presence of bacteriocins, either intentionally added or produced in situ during milk fermentation, increase the risk of fermentation failure in the event of phage contamination. Bacteriocin-phage interactions (BaPIs) were determined in vitro for nisin, a bacteriocin commonly used as a food biopreservative. Nisin increased the plaque size for the lactococcal phages P2 and bIL170. However, bIL170 burst size and l

The Lactococcus lactis subsp. lactis 194-K strain has been established to be able to produce two bacteriocins, one of which was identified as the known lantibiotic nisin A, and the other 194-D bacteriocin represents a polypeptide with a 2589-Da molecular mass and comprises 20 amino acid residues. Both bacteriocins were produced in varying proportions in all of the studied nutrient media, which support the growth of the producer. Depending on the cultivation medium, the nisin A content was 380- to 1123-fold lower in the 194-K stain culture fluid than that of the 194-D peptide. In comparision to
