Acid-dependent dismutation of nitrogen oxides may be a critical source of nitric oxide in human macrophages.

The cytotoxic activity of the macrophage relies greatly on the secretion of a number of reactant intermediates, including superoxide (O(2)(-)), hydroxyl radical (OH(-)) and nitric oxide (NO). The latter, synthesized via cytokine-mediated induction of inducible NO-synthase (iNOS), is readily observed in murine macrophages. However, a poorly reproducible or minimal response to cytokine-stimulation in the human macrophage has questioned the presence or significance of this important pathway in man. Nevertheless, iNOS is present in other human phagocytic cells, e.g. neutrophils, while the NO metab

Inhibition of cathepsin K by nitric oxide donors: evidence for the formation of mixed disulfides and a sulfenic acid.

The cysteine protease cathepsin K is believed to play a key role in bone resorption as it has collagenolytic activity and is expressed predominantly and in high levels in bone resorbing osteoclast cells. The addition of nitric oxide (NO) and NO donors to osteoclasts in vitro results in a reduction of bone resorption, although the mechanism of this effect is not fully understood. The S-nitroso derivatives of glutathione (GSNO) and N-acetylpenicillamine (SNAP) and the non-thiol NO donors NOR-1 and NOR-3 all inhibited the activity of purified cathepsin K in a time- and concentration-dependent man

Oxidant stress, anti-oxidants, nitric oxide and essential fatty acids in peptic ulcer disease.

In patients with duodenal ulcer (DU), the plasma levels of nitrite and lipid peroxides, the anti-oxidant content of red cells and plasma phospholipid fatty acid analysis were performed both before and after healing of the ulcer following treatment with lansoprazole, a proton pump inhibitor. These results showed that during the phase of active DU, the concentrations of antioxidants (superoxide dismutase, SOD, catalase and glutathione peroxidase) in red cells were low where as those of lipid peroxides and nitric oxide were high. Of the fatty acids measured, the concentration of palmitic acid (16

Exogenous and endogenous nitric oxide eluting polylactic acid-based nanofibrous scaffolds for enhancing angiogenesis of diabetic wounds.

Delayed wound healing is a major complication that diabetic patients suffer from due to high microbial infection susceptibility, high diabetic wound alkalinity, a low lymphangiogenesis rate, and a high inflammation rate, resulting in severe gangrene. Hence, this study aims to develop a multifunctional adhesive nanofibrous patch to promote the wound healing process. Phenytoin, sildenafil citrate, and/or nitric oxide-eluting nanoparticles were incorporated separately within the polylactic acid nanofibrous layer. Polylactic acid was fabricated in the form of highly porous nanofibrous matrices tha

Nitrogen oxide reduction through absorbent solutions containing nitric acid and hydrogen peroxide in hollow fiber membrane modules.

Emissions of nitrogen oxides such as NO and NO2, which are commonly known as NOx, are threats to human existence and cause environmental problems. Mainly, two techniques have been developed to drastically reduce these emissions, which are dry and wet processes. The wet process has several advantages, major identifiable advantages are the adaptability to the flue gas, low operating temperatures and no poisoning and inactivation catalyst. Also, a mixture of hydrogen peroxide and nitric acid are used as absorbents solution for NOx reduction in the wet process. The advantages of using this mixture

Nitrogen Oxides and Nitric Acid Enable the Sustainable Hydroxylation and Nitrohydroxylation of Benzenes under Visible Light Irradiation.

A new type of waste recycling strategy is described in which nitrogen oxides or nitric acid are directly employed in photocatalyzed hydroxylations and nitrohydroxylations of benzenes. Through these transformations, otherwise costly denitrification can be combined with the synthesis of valuable compounds for various applications.

Evaluation of nitric and acetic acid resistance of cement mortars containing high-volume black rice husk ash.

This paper presents the performance of cement mortar containing black rice husk ash (BRHA) under nitric and acetic acid attacks. The BRHA, collected from an electrical generating power plant that uses rice husk as fuel, was ground using a grinding machine. The compressive strength loss, weight loss, and expansion of mortars under nitric and acetic acid attack were investigated. The test results of BRHA properties in accordance with the ASTM C 618 standard found that the optimal grinding time was 4 h as this achieved a Blaine fineness of 5370 cm(2)/g. For parametric study, BRHA were used as a P

Suberoylanilide hydroxamic acid radiosensitizes tumor hypoxic cells in vitro through the oxidation of nitroxyl to nitric oxide.

The pharmacological effects of hydroxamic acids are partially attributed to their ability to serve as HNO and/or NO donors under oxidative stress. Previously, it was concluded that oxidation of the histone deacetylase inhibitor suberoylanilide hydroxamic acid (SAHA) by the metmyoglobin/H2O2 reaction system releases NO, which was based on spin trapping of NO and accumulation of nitrite. Reinvestigation of this system demonstrates the accumulation of N2O, which is a marker of HNO formation, at similar rates under normoxia and anoxia. In addition, the yields of nitrite that accumulated in the abs

Gibberellic acid nitrite stimulates germination of two species of light-requiring seeds via the nitric oxide pathway.

We used two species of light-requiring seeds, Paulownia tomentosa, which have absolute light requirement (no germination in darkness), and Stellaria media seeds, which germinate in darkness to a certain extent because of presence of preformed active phytochrome, to obtain results strongly suggesting that gibberellic acid nitrite stimulates seed germination via its capability as a functional NO donor. Exogenous application of gibberellic acid nitrite stimulates gibberellin-insensitive Stellaria media seed germination in darkness as do a wide variety of NO donors. Pure gibberellic acid could rep

Effects of nitrogen dioxide exposure and ascorbic acid supplementation on exhaled nitric oxide in healthy human subjects.

BACKGROUND: Nitric oxide (NO) is detectable in the exhaled breath, is involved in airway defence and inflammation, and probably modulates bronchial smooth muscle tone. Given the sensitivity of nitrogen oxides to local redox conditions, we postulated that exposure to oxidant or antioxidant compounds could alter concentrations of NO in the exhaled breath (eNO). We assessed the effect of nitrogen dioxide (NO(2)) and ascorbic acid exposure on eNO in healthy human subjects. METHODS: Ten healthy subjects were randomised to undergo a 20 minute single blind exposure to NO(2) (1.5 parts per million) o

Interaction of nitric oxide donors and ascorbic acid on D-[3H] aspartate efflux from rat striatal slices.

There are conflicting reports in the literature concerning the neuroprotective effect of ascorbic acid on excitotoxic processes in which excessive glutamate release and nitric oxide are supposed to be major factors. To study the influence of ascorbate on the nitric oxide modulated glutamate release rat striatal slices, preloaded with the tritiated glutamate analog D-aspartate, were used. The high potassium-induced efflux of D-[3H]aspartate was concentration dependently stimulated by the nitric oxide donors sodium nitroprusside, S-nitroso-N-acetylpenicillamine (SNAP) or 5-amino 3-morpholinyl-1,

Effect of oxygen toxicity and nitric acid-induced lung damage on drug absorption from the rat lung.

To investigate the effect of pulmonary oxygen toxicity and nitric acid-induced lung damage on absorption of drugs from the lung, rats were either exposed continuously to approximately 100% oxygen or given an intratracheal injection of 1% nitric acid solution (0.15 ml), and rates of drug absorption from damaged and control lungs were compared after various times. To measure pulmonary absorption rates, 0.1 ml of drug solution (0.1-10 mM) was administered through a tight-fitting tracheal cannula to anesthetized animals, and, after various times, lungs were assayed for unabsorbed drug. Prostaglandins as negative regulators against lipopolysaccharide, lipoteichoic acid, and peptidoglycan-induced inducible nitric oxide synthase/nitric oxide production through reactive oxygen species-dependent heme oxygenase 1 expression in macrophages.

Although prostaglandins (PGs) were reported to exert proinflammatory and anti-inflammatory effects in macrophages, their action mechanisms remain unclear. The effects of PGs including PGJ2 (J2), Δ-PGJ2 (Δ), 15-deoxy-Δ PGJ2 (15d), PGE2 (E2), and PGF2α (F2α) on lipopolysaccharide (LPS)-, lipoteichoic acid (LTA)-, and peptidoglycan (PGN)-induced inducible nitric oxide (NO) synthase (iNOS)/NO production by RAW264.7 macrophages were investigated. First, we found that induction of cyclooxygenase 2 (COX-2) protein occurred at a time earlier than that of heme oxygenase 1 (HO-1) protein, and the additi

Nitric acid-modified biochar ameliorates saline-alkali soil and promotes cotton growth by regulating microbiomes and metabolites.

Biochar is extensively utilized for saline-alkali soil remediation; however, the effects of feedstock origin on its performance remain unclear. Moreover, there is a growing need to develop modified biochar with improved efficiency in saline-alkali soil. Therefore, cotton straw biochars derived from saline-alkali soil (SBC) and non-saline-alkali (NBC), along with their nitric acid-modified forms (HSBC and HNBC), were prepared and assessed via a cotton pot experiment. The results showed that the pH and total soluble salts (TSS) of SBC were significantly higher than those of NBC by 0.33 units and

Gibberellic acid (GA) alleviates Al toxicity in rice (Oryza sativa) through a nitric oxide (NO) dependent pathway.

Over 50% of global arable soils are acidic; acidic soil-induced aluminum (Al) phytotoxicity primarily inhibits root elongation, thereby reducing the plant's absorption of water and nutrients, which suppresses crop yield. Gibberellic acid (GA), a class of critical plant hormones, acts as a core regulator of plant development and growth mechanisms and contributes to the physiological adaptation of plants under stress conditions. In this study, we selected the rice variety Nipponbare (Nip) to investigate whether GA exerts an effect on alleviating Al toxicity and to explore the underlying mechanis

Successive nitric oxide and lipoic acid priming mitigates [ZnO]NPs toxicity in wheat: experimental and DFT insights.

The global food security is threatened by the accumulation of anthropogenic nanomaterials in the environment. We investigated the phytotoxic effects of elevated zinc oxide nanoparticles ([ZnO]NPs: 300-400 mg L⁻1) on wheat (Triticum aestivum L.) and evaluated a novel detoxification strategy using successive redox-priming. Wheat caryopses were primed with alpha-lipoic acid (LA) and sodium nitroprusside (SNP, a nitric oxide donor) either individually or in successive sequences (LA→SNP and SNP→LA). Exposure to [ZnO]NPs resulted in significant growth inhibition, reduction of biomass and degradation

Near-infrared light-triggered nitric oxide-releasing hyaluronic acid hydrogel for precision transdermal therapy of androgenic alopecia.

Progressive follicle and vascular atrophy, insufficient nutrient supply, and hormonal imbalance are direct causes of androgenic alopecia (AGA), limiting treatment options. Nitric oxide (NO) has demonstrated significant advantages in cell proliferation, vascular repair, and inflammation regulation. However, precise control of NO concentration and efficient utilization limit its clinical application for AGA treatment. To address this, we developed a near-infrared (NIR) light-triggered NO-releasing delivery system (Gel@L-Arg). The system uses Chlorin e6 (Ce6) grafted onto oxidized hyaluronic acid

Interplay of nitric oxide and indole-3-acetic acid signaling in regulating arsenate toxicity in tomato seedlings.

The problem of arsenic contamination is widespread, posing serious risks to both human health and crop productivity. Therefore, eco-friendly strategies are urgently needed to reduce metal stress-related yield losses and minimize its impacts on human health. Although the roles of nitric oxide (NO) and indole-3-acetic acid (IAA) in combating abiotic stresses, including metal toxicity, have been reported, the mechanisms underlying their interaction remain unclear. This study investigates whether IAA and NO function through shared or independent signaling pathways in regulating arsenate [As(V)] to
