Our studies showed that the spin-labeled (SL) compounds (free stable nitroxyl radical derivatives) accumulate predominantly in pigment melanoma and some of them possess high antimelanoma activity (100% curability of tumor bearing animals) and lower general toxicity. Taking into account our results we describe a proposed mechanism of the spin-labeled accumulation in melanoma, their antimelanoma activity and lower toxicity.

We measured the carotid body volume of rats treated with chronic hypoxia alone and chronic hypoxia together with a single neonatal injection of N-ethyl-N-nitrosourea (10 mg/kg). All the animals so treated showed enlargement of their carotid bodies, but no carotid body chemodectomas occurred.

B16 melanoma (mouse melanoma) and C24 melanoma (human malignant melanoma) transplanted in mice were treated by a combined therapy of ACNU [1-(4-amino-2-methyl - 5 - pyrimidinyl) - methyl - 3 - (2 - chlorethyl) - 3 - nitrosourea hydrochloride] (10 mg/kg) and hyperthermia (43 degrees C, 30 min). In both types of melanoma, a marked synergistic effect of the combined therapy was noted. Particularly in C24 melanoma a reduction in the size of tumor was observed. Histopathologic findings revealed a strong degeneration such as destruction of the tumor structure and vacuolization of nuclei.

Formation and loss of methylated purines in DNA of various fetal and maternal tissues were measured up to 7 days following intravenous administration of N-[14C]methyl-N-nitrosourea to rats on the 21st day of gestation. G-banding analysis was carried out in 45 mice after exposure to the leukemogen butylnitrosourea (BNU). Of 27 animals without clinical signs of leukemia, 2 had chromosomal rearrangements (7%) while, of 18 leukemic mice, 5 showed abnormal karyotypes (28%). Twenty-four patients with far advanced malignant tumors, resistent to established chemotherapy,, were treated with the combination of MNU and Cyclophosphamide. The drugs were administered in six-day cycles sequentially. MNU in doses of 4 mg/kg body weight and Cyclophosphamide in doses of 8 mg/kg body weight were given. Results of treatment showed response (greater than 50% tumor regression) in 10 (42%) of the 24 treated patients. Seven remissions were complete and three partial.
