The use of nucleic acid as a drug substance for vaccines and other gene-based medicines continues to evolve. Here, we have used a technology originally developed for mRNA in vivo delivery to enhance the immunogenicity of DNA vaccines. We demonstrate that neutralizing antibodies produced in rabbits and nonhuman primates injected with lipid nanoparticle (LNP)-formulated Andes virus or Zika virus DNA vaccines are elevated over unformulated vaccine. Using a plasmid encoding an anti-poxvirus monoclonal antibody (as a reporter of protein expression), we showed that improved immunogenicity is likely

BACKGROUND: Nucleic acid-based vaccines have been studied for the past four decades, but the approval of the first messenger RNA (mRNA) vaccines during the COVID-19 pandemic opened renewed perspectives for the development of similar vaccines against different infectious diseases. Presently available mRNA vaccines are based on non-replicative mRNA, which contains modified nucleosides encased in lipid vesicles, allowing for entry into the host cell cytoplasm, and reducing inflammatory reactions. An alternative immunization strategy employs self-amplifying mRNA (samRNA) derived from alphaviruses,

Lipid nanoparticle (LNP)-based mRNA therapeutics, highlighted by the success of SARS-CoV-2 vaccines, face challenges due to inflammation caused by ionizable lipids. These ionizable lipids can activate the immune system, particularly when co-delivered with nucleic acids, leading to undesirable inflammatory responses. We introduce a novel class of anti-inflammatory ionizable lipids functionalized with hydroxychloroquine (HCQ), which suppresses both lipid-induced and nucleic acid-induced immune activation. These HCQ-functionalized LNPs (HL LNPs) exhibit reduced proinflammatory responses while mai

This review investigates biomaterial-based strategies for improved treatment of MPXV. We focus on emerging synthetic biomedical approaches to combating the virus. These include peptide nucleic acids, CRISPR-based systems, and small-molecule therapeutics. These methods work by targeting and blocking viral proteins and enzymes. Such synthetic platforms may help reduce viral transmission and minimize side effects. They also offer potential solutions to challenges such as viral resistance in humans. In addition, biomaterials contribute to the development of more stable and effective vaccines. Comb

Streptococcus suis serotype 2 (SS2) is a major zoonotic pathogen responsible for severe infections in both pigs and humans. Despite its clinical significance, no effective vaccine is currently available, and the influence of capsular polysaccharide (CPS2) structural features on host immunity remains unclear. Here, we identified a fast-growing SS2 strain from a panel of 40 clinical isolates and established a novel, reliable CPS2 purification method. This approach combines polyethylenimine-mediated nucleic acid precipitation with phenol-based protein denaturation, eliminating the need for column

Molecular biology is making a tremendous impact on the diagnosis and treatment of liver diseases. Methods such as the polymerase chain reaction are changing the way physicians diagnose and monitor patients with viral hepatitis. Assays based on recombinant protein antigens allow for detection of specific autoantibodies in diseases such as primary biliary cirrhosis. The diagnosis of inherited metabolic diseases, such as hemochromatosis and Wilson disease, is being revolutionized by discovery of the defective genes involved and the development of methods to rapidly sequence DNA and identify mutat

Glycoconjugates are central mediators of molecular recognition in biological systems, yet their inherent structural microheterogeneity has long impeded mechanistic insight and therapeutic translation. Recent advances in synthetic chemistry, chemoenzymatic approaches, and glycoengineering now enable access to structurally defined glycoconjugates, creating new opportunities for precision medicine. Here, we review key developments in glycoconjugate-based therapeutics over the past two years, including glycoconjugate vaccines, glycoengineered peptides and proteins with improved pharmacological pro

By targeting the membrane (M) proteins of monkeypox virus (MPXV) strain VEROE6, we analyzed its evolutionary hierarchy and predicted its dominant antigenic B-cell epitope to provide a theoretical basis for the development of MPXV epitope vaccines and related monoclonal antibodies. In this study, phylogenetic trees were constructed based on the nucleic acid sequences of MPXV and the amino acid sequences of M proteins. The 3D structure of the MPXV_VEROE6 M proteins was predicted with AlphaFold v2.0 and the dominant antigenic B-cell epitopes were comprehensively predicted by analyzing parameters

Although universal HBV vaccination has markedly reduced the prevalence of overt HBV infection in Saudi Arabia, Occult hepatitis B infection (OBI) remains an underrecognized clinical and public health concern, associated with HBV reactivation during immunosuppression and progression to hepatocellular carcinoma. This review summarizes current evidence on the epidemiology, molecular mechanisms, diagnostic challenges, and public health implications of OBI in Saudi Arabia and across the Middle East. Important mechanisms include persistence of covalently closed circular DNA, viral genetic mutations,

Antigen transfer refers to the process of intercellular information exchange, where antigenic components including nucleic acids, antigen proteins/peptides and peptide-major histocompatibility complexes (p-MHCs) are transmitted from donor cells to recipient cells at the thymus, secondary lymphoid organs (SLOs), intestine, allergic sites, allografts, pathological lesions and vaccine injection sites via trogocytosis, gap junctions, tunnel nanotubes (TNTs), or extracellular vesicles (EVs). In the context of vaccine inoculation, antigen transfer is manipulated by the vaccine type and administratio

The Brighton Collaboration Benefit-Risk Assessment of VAccines by TechnolOgy (BRAVATO) Working Group has prepared standardized templates to describe the key considerations for the benefit-risk assessment of several vaccine platform technologies, including nucleic acid (RNA and DNA) vaccines. This paper uses the BRAVATO template to review the features of a vaccine employing a proprietary mRNA vaccine platform to develop Moderna COVID-19 Vaccine (mRNA-1273); a highly effective vaccine to prevent coronavirus disease 2019 (Covid-19) caused by severe acute respiratory syndrome coronavirus 2 (SARS-C

Coxsackievirus A6 (CVA6) has emerged as the dominant etiological agent of hand, foot and mouth disease worldwide. At present, neither specific antivirals nor effective vaccines are available, creating an urgent need for rapid, low-cost in vitro diagnostics that can interrupt transmission. This study developed an electrochemical biosensor based on a "screening-validation" strategy. It utilizes a MoS2/MWCNT/AgNPs three-dimensional highly conductive network as the substrate, tightly anchored with p(NIPAm-co-SBMA) microgel to form a "protective-conductive" synergistic interface. The capture probe

PURPOSE OF REVIEW: With improved malaria control, acute undifferentiated febrile illness studies in tropical regions reveal a startling proportion of rickettsial illnesses, especially scrub typhus, murine typhus, and spotted fever group rickettsioses. Laboratory diagnosis of these infections evolved little over the past 40 years, but combinations of technologies like PCR and loop-mediated isothermal amplification, with refined rapid diagnostic tests and/or ELISA, are promising for guidance for early antirickettsial treatment.

Infectious virus outbreaks pose a significant challenge to public healthcare systems. Early and accurate virus diagnosis is critical to prevent the spread of the virus, especially when no specific vaccine or effective medicine is available. In clinics, the most commonly used viral detection methods are molecular techniques that involve the measurement of nucleic acids or proteins biomarkers. However, most clinic-based methods require complex infrastructure and expensive equipment, which are not suitable for low-resource settings. Over the past years, smartphone-based point-of-care testing (POC

Vaccine development against SARS-CoV-2 has been highly successful in slowing down the COVID-19 pandemic. A wide spectrum of approaches including vaccines based on whole viruses, protein subunits and peptides, viral vectors, and nucleic acids has been developed in parallel. For all types of COVID-19 vaccines, good safety and efficacy have been obtained in both preclinical animal studies and in clinical trials in humans. Moreover, emergency use authorization has been granted for the major types of COVID-19 vaccines. Although high safety has been demonstrated, rare cases of severe adverse events

Lipid nanoparticles (LNPs) have become a key technology for delivering nucleic acids, playing a critical role in the success of mRNA vaccines and RNA-based therapeutics. Established manufacturing methods, including microfluidic mixing and impingement jet mixing, have exhibited excellent scalability, reproducibility, and clinical relevance, most notably during the development of vaccines for SARS-CoV-2. Nevertheless, these techniques are accompanied by challenges related to the handling of organic solvents (ethanol), scalability for personalized medicine, and environmental considerations. This

Vaccines are derived from a variety of sources including tissue extracts, bacterial cells, virus particles, recombinant mammalian, yeast and insect cell produced proteins and nucleic acids. The most common method of vaccine production is based on an initial fermentation process followed by purification. Production of vaccines is a complex process involving many different steps and processes. Selection of the appropriate purification method is critical to achieving desired purity of the final product. Clarification of vaccines is a critical step that strongly impacts product recovery and subseq

Cervical cancer and cervical intraepithelial neoplasia (CIN) are well-known outcomes of a human papillomavirus (HPV) infection. Viral oncogenes expressions like E6, E7, and, recently recognized E5, lead to HPV-related malignant progression. Although HPV prevention by powerful vaccines against most frequent and oncogenic genotypes is feasible, current treatment against cervical neoplasia is distant from an ideal one. In addition, late diagnosis is commonly associated with a poor prognosis. On top of that, radiotherapy, chemotherapy, or surgery are less effective in high-grade lesions. Areas cov

Glioma, particularly glioblastoma (GBM), remains one of the most lethal primary brain tumors, characterized by aggressive biological behavior, profound intratumoral heterogeneity, and a highly immunosuppressive tumor microenvironment. Despite encouraging advances in cancer immunotherapy, durable clinical benefit from glioma vaccines has remained limited, highlighting a substantial translational gap between early immunogenicity and long-term therapeutic efficacy. In this review, we provide an integrative overview of current glioma vaccine strategies, including peptide-based, dendritic cell-base
