Oleanolic acid modulates neuronal regeneration through KLF5-mediated FGF13 mRNA degradation to alleviate Alzheimer's disease-like cognitive dysfunction.

Despite understanding the pathophysiology of Alzheimer's disease (AD), the mechanisms of neuronal regeneration mediated by oleanolic acid (OA) through m6A RNA methylation remain unexplored, forming the crux of this study. In a streptozotocin (STZ)-induced AD rat model, we administered OA and conducted behavioral tests to evaluate cognitive functions. We employed BrdU incorporation assays and immunofluorescence to investigate NSC proliferation, and Western blotting, chromatin immunoprecipitation (ChIP), RNA immunoprecipitation (RIP), and MeRIP-qPCR assays to analyze protein expression and RNA s

Ultrathin Oleanolic Acid/Perfluorocarbon-Ferrocene Nanosheets Trigger Reactive Species Storm to Reshape Tumor Microenvironment for Enhanced Chemodynamic Therapy.

Ferrocene derivatives possess a typical "sandwich" structure that facilitates the formation of two-dimensional nanosheets (2D NSs), while their ferrous ions can effectively mediate chemodynamic therapy (CDT). However, conventional bottom-up synthesis methods for nanosheets often involve complex processes and stringent preparation conditions, limiting their therapeutic applications. In this study, we developed a simple yet efficient premix-dropwise-stir synthetic strategy to fabricate ultrathin oleanolic acid/perfluorocarbon-ferrocene (O/F-Fc) 2D NSs with considerable antitumor properties. Hyaluronic acid-based reduction responsive nanoparticles for Improved anti-psoriasis effects of traditional Chinese herb monomer oleanolic acid via blocking YAP-AREG axis.

BACKGROUND: Oleanolic acid (OA), a pentacyclic triterpenoid abundantly present in various traditional Chinese herbs, exhibits promising anti-psoriatic potential owing to its broad pharmacological activities. Nevertheless, its clinical translation has been hindered by challenges including poor aqueous solubility, limited cutaneous permeation, and rapid systemic clearance, all of which compromise bioavailability when administered topically. To address these limitations, we designed and developed encapsulated and disulfide bonded OA-hyaluronic acid nanoprodrugs (OA-NPs@OA) for topical treatment o

Oleanolic acid alleviates ANIT-induced cholestatic liver injury by activating Fxr and Nrf2 pathways to ameliorate disordered bile acids homeostasis.

BACKGROUND: Cholestasis is a clinical syndrome with high incidence and few effective treatments. Oleanolic acid (OA) is a triterpenoid compound with anti-cholestatic effects. Studies using bile duct ligation or lithocholic acid modeling have shown that the alleviating effect of OA on cholerosis is related to the regulation of nuclear factor erythroid 2 related factor (Nrf2) or farnesoid X receptor (Fxr). PURPOSE: This study aims to investigate the underlying mechanism of OA against alpha-naphthylisothiocyanate (ANIT)-induced cholestatic liver injury based on Nrf2 and Fxr dual signaling pathwa

Oleanolic Acid Cubic Liquid Crystal Nanoparticle-Based Thermosensitive Gel Attenuates Depression Symptoms in Chronic Unpredictable Mild Stress Rats.

PURPOSE: Major depressive disorder (MDD) is a global health concern. Studies have demonstrated that oleanolic acid (OA) has a regulatory effect on MDD. However, OA is poorly soluble, has low oral bioavailability, and faces challenges in crossing the blood-brain barrier. In this study, building upon a previous formulation of OA cubic liquid crystal nanoparticles (OA-LCNP), we combined nanoparticles with a thermosensitive gel for nasal administration and investigated the pharmacological effects of OA-LCNP thermosensitive gel (OANG) on depression. This study aimed to evaluate the effects of OANG

An oleanolic acid derivative reduces denervation-induced muscle atrophy via activation of CNTF-mediated JAK2/STAT3 signaling pathway.

Denervation caused by sciatic nerve injury has brought great harm to the patients, especially denervation-induced muscle atrophy. The body stress produces a large number of Schwann cells when the sciatic nerve is injured, and the cells secrete some cytokines including ciliary neurotrophic factor (CNTF) that not only play a role in promoting the repair of sciatic nerve, but also maintain the normal physiological function of the muscles surrounding the damaged nerves. CNTF upregulates janus kinase 2 (JAK2) and signal transducers and activators of transcription 3 (STAT3) signals in myoblasts, and

The Multi-pathway Mechanism and Co-delivery System of Oleanolic Acid Derivatives in Reversing Chemotherapy Resistance in Breast Cancer.

Breast cancer threatens the health of women worldwide. However, the use of chemotherapy for breast cancer is prone to generating side effects and drug resistance. Therefore, identifying natural compounds with anticancer activity is a better solution to the problem of drug resistance. Oleanolic acid (OA), a kind of pentacyclic triterpenoid, is widely studied and used in the field of oncology. It has biological activity against breast cancer and has few side effects on normal cells. OA can be used as a frame for chemical modification to synthesize new compounds for the development of new drugs.

A green extract rich in oleanolic acid obtained from Aglianico red grape pomace as a promising anti-browning agent.

Enzymatic browning has deleterious implications in food industry. In musts, especially in white ones, this phenomenon driven by polyphenol oxidases threatens wine quality and stability. Herein, an oleanolic acid (OA)-enriched extract from Aglianico red grape pomace obtained by the green solvent dimethylcarbonate was investigated as an alternative anti-browning agent. The extract inhibited tyrosinase with an IC₅₀ of 83.84 ± 7.42 μg/mL. When added to Fiano di Avellino musts (0.01-5.0 mg/mL), it reduced browning, as demonstrated by CIELAB colorimetry, with effective stabilization at values ≥0.08

Polymeric Nanocarriers and Polymer-Assisted Delivery Platforms for Oleanolic Acid: Design Strategies, Controlled Release, Translational Challenges, and Clinical Perspectives.

Oleanolic acid is a naturally occurring pentacyclic triterpenoid with broad preclinical promise in inflammation, oxidative stress, liver injury, metabolic disorders, cancer-related models, skin disease, and wound repair. Its further development, however, is constrained by poor aqueous solubility, low and variable bioavailability, limited barrier transport, crystallinity, and strong dependence of biological response on the formulation used. These properties make oleanolic acid a useful example of a hydrophobic natural compound whose pharmacological performance is inseparable from delivery desig

Fabrication of polysaccharide-coated oleanolic acid-curcumin-coassembled nanoparticles (OA/Cur NPs): Enhancement of colloidal stability and water solubility.

Natural terpenoid carriers, such as oleanolic acid (OA), can enhance the water solubility and stability of hydrophobic compounds such as curcumin (Cur). However, improving the colloidal stability of nanoparticle emulsions and resolving the redispersion problem of freeze-dried nanoparticle powders remain significant challenges. In this study, we fabricated coassembled oleanolic acid-curcumin nanoparticles (OA/Cur NPs) and applied a polysaccharide surface coating method to improve their colloidal stability and water solubility. The results showed that the optimal ratio of Cur/OA for preparing OA

Co-delivery of cisplatin and oleanolic acid by silica nanoparticles-enhanced apoptosis and reverse multidrug resistance in lung cancer.

Multidrug resistance (MDR) of chemotherapy is one of the significant concerns in cancer therapy. Here in our study, cisplatin (DDP) and oleanolic acid (OA) were co-loaded in mesoporous silica nanoparticles (Nsi) to construct DDP/OA-Nsi and solve the DDP-resistance in lung cancer therapy. The cytotoxicity and apoptosis assays demonstrated that in DDP-resistant A549/DDP cells, the cytotoxicity of DDP/OA-Nsi was significantly higher than that of free DDP or DDP single delivery system (DDP-Nsi). The intracellular drug accumulation study revealed that the intracellular DDP concentration in the DDP/

Oleanolic Acid Decreases IL-1β-Induced Activation of Fibroblast-Like Synoviocytes via the SIRT3-NF-κB Axis in Osteoarthritis.

Synovial inflammation is a major pathological feature of osteoarthritis (OA), which is a chronic degenerative joint disease. Fibroblast-like synoviocytes (FLS), localized in the synovial membrane, are specialized secretory cells. During OA synovitis, FLS produce chemokines and cytokines that stimulate chondrocytes to secrete inflammatory cytokines and activate matrix metalloproteinases (MMPs) in FLS. Recent studies have demonstrated that sirtuin 3 (SIRT3) performs as a key regulator in maintaining mitochondrial homeostasis in OA. This study aims at ascertaining whether SIRT3 is involved in OA

Self-assembled nanoparticles of oleanolic acid as a potential functional food ingredient with improved dispersibility and metabolic modulation activity.

Oleanolic acid (OA) is a plant-derived pentacyclic triterpenoid with anti-obesity and anti-inflammatory activities, but its poor solubility limits bioavailability. In this study, self-assembled OA nanoparticles (OA-NM) were prepared via an anti-solvent method to enhance dispersibility and efficacy. OA-NM exhibited uniform spherical morphology (∼228 nm), good stability, and supramolecular reorganization driven mainly by hydrophobic interactions and hydrogen bonding. Its cellular uptake occurred primarily via clathrin-mediated endocytosis. Compared with free OA, OA-NM showed lower cytotoxicity a
