Oleic acid and docosahexaenoic acid cause an increase in the paracellular absorption of hydrophilic compounds in an experimental model of human absorptive enterocytes.

Surface active compounds present in food possibly have the ability to enhance the absorption of water soluble toxic agents. Therefore, we investigated whether fatty acids such as oleic acid and docosahexaenoic acid (DHA), both commonly present in food, negatively affect the integrity of tight junctions (TJ) in the intestinal epithelium and thereby increase the absorption of poorly absorbed hydrophilic substances. Caco-2 cells, which are derived from human absorptive enterocytes, were grown on permeable filters for 20-25 days. Differentiated cell monolayers were apically exposed for 90min to ma

Functional properties of ice cream from milk of dairy cows abomasally infused with increasing amounts of high-oleic sunflower fatty acids.

The main challenge to producing a nutritionally improved milk fat composition is to increase monounsaturated fatty acids (MUFA) and decrease saturated fatty acids (SFA) without detriment to dairy product characteristics. Our aim was to determine the optimal amount of intestinally available oleic acid (via abomasal infusion) to produce higher-MUFA milk fat with satisfactory functional characteristics for ice cream. One control and four increasing doses of free fatty acids (FA) from high oleic sunflower oil (HOSFA) were infused into the abomasa of four lactating dairy cows in a changeover design

Resolvin E1 ameliorates obstructive meibomian gland dysfuction in oleic acid induced mice model.

This study explores the effects of Resolvin E1 (RvE1), a natural agonist of peroxisome proliferator-activated receptor-γ (PPAR-γ), in a novel mouse model of obstructive Meibomian gland dysfunction (oMGD) induced by oleic acid. Oleic acid was applied at varying concentrations to the eyelid margin to identify the optimal dose for MGD induction. Morphological and pathological changes in Meibomian glands (MGs) were assessed using slit-lamp examination, histological staining (H&E, Oil Red O), and molecular analyses. Male BALB/c mice (6-8 weeks) were randomly assigned to RvE1 treatment or PBS contro

Oleic acid modification enables lysosomal escape of phthalocyanine photosensitizer for effective bladder cancer treatment.

Lysosomal sequestration, driven by acidic hydrolases, degrades and inactivates therapeutics, severely compromising their efficacy. While unsaturated fatty acid (UFA) modifications confer robust lysosomal escape capabilities to nanomedicines, their utility in small-molecule drug design remains largely underexplored. Capitalizing on the unique structural attributes of UFA side chains, we designed and synthesized three zinc phthalocyanine (ZnPc) conjugates featuring distinct linker architectures with oleylamine (OA): ZnPc-OA (linker-free), ZnPc-PEG2-OA (PEG linker), and ZnPc-butyl-OA (alkyl chain

Oleic acid fuels cisplatin-resistant ovarian cancer through FABP4-driven lipid uptake.

BACKGROUND: Ovarian cancer (OC) depends on lipids as fuel for metastasis and growth. We previously showed that cisplatin resistant (Pt-R) OC cells uptake higher amounts of fatty acids (FAs) compared to sensitive (Pt-S) cells, a process which facilitates cancer cell survival under cisplatin-induced oxidative stress. METHODS: Isogenic pairs of Pt-S and Pt-R OC cell lines were cultured in low serum conditions supplemented with either 50 μM oleic acid (OA, unsaturated) or 50 μM palmitic acid (PA, saturated) and used for viability assays, RNA-Sequencing, and cell cycle analysis. The effects of an

Hydrophobic Ion Pairing of Polymyxin B with Oleic Acid: A Dissipative Particle Dynamics Simulation Study.

Background: Hydrophobic ion pairing is a technique for reducing the hydrophilicity of charged molecules (drugs) by pairing them with oppositely charged hydrophobic counterions. This method is used to control the solubility of charged molecules in a solvent and is of particular importance in drug delivery. Methods: Dissipative particle dynamics simulations were performed to provide a microscopic understanding of hydrophobic ion pairing in polymyxin B (PMB) and oleate (OA) ions. Solvents and ions were explicitly included in the simulations. Results: We investigated the effects of relative concen

Oleic acid-PPARγ-FABP4 loop fuels cholangiocarcinoma colonization in lymph node metastases microenvironment.

BACKGROUND AND AIMS: Lymph node metastasis is a significant risk factor for patients with cholangiocarcinoma, but the mechanisms underlying cholangiocarcinoma colonization in the lymph node microenvironment remain unclear. We aimed to determine whether metabolic reprogramming fueled the adaptation and remodeling of cholangiocarcinoma cells to the lymph node microenvironment. APPROACH AND RESULTS: Here, we applied single-cell RNA sequencing of primary tumor lesions and paired lymph node metastases from patients with cholangiocarcinoma and revealed significantly reduced intertumor heterogeneity

Nitro-oleic acid reduces thoracic aortic aneurysm progression in a mouse model of Marfan syndrome.

AIMS: Marfan syndrome (MFS) is a connective tissue disorder caused by mutations in the Fibrillin-1 gene. It is associated with formation of thoracic aortic aneurysms that can potentially be a life-threatening condition due to aortic rupture or dissection. Excessive non-canonical transforming growth factor beta signalling, mediated by activation of extracellular signal-regulated kinases 1/2 (ERK1/2), as well as inducible nitric oxide synthase (NOS2)-dependent nitric oxide production, have been identified to drive aortic pathology in MFS through induction of elastin fragmentation and smooth musc

Activation of Liver X Receptor Attenuates Oleic Acid-Induced Acute Respiratory Distress Syndrome.

Liver X receptors (LXRs) were identified as receptors that sense oxidized cholesterol derivatives. LXRs are best known for their hepatic functions in regulating cholesterol metabolism and triglyceride synthesis, but whether and how LXRs play a role in the lung diseases is less understood. To study the function of LXRs in acute respiratory distress syndrome (ARDS), we applied the oleic acid (OA) model of ARDS to mice whose LXR was genetically or pharmacologically activated. The VP-LXRα knock-in (LXR-KI) mice, in which a constitutively activated LXRα (VP-LXRα) was inserted into the mouse LXRα lo

Effect of oleic acid ligand on photophysical, photoconductive and magnetic properties of monodisperse SnO2 quantum dots.

Oleic acid capped monodisperse SnO(2) quantum dots (QDs) of size 2.7 nm were synthesized by thermal decomposition and oxidation of Sn(II)(oleate) complex in high boiling nonpolar solvent octadecene using oleic acid as a capping agent and N-methylmorpholine N-oxide as an oxidizing agent. FTIR, DSC and TGA were employed to understand the growth of the oleic acid capped SnO(2) QDs through the decomposition of metal fatty acid complex. The surface defect-related luminescence properties of the QDs were demonstrated using steady-state and time-resolved spectroscopy. The oleic acid capping on the QD

Effects of Endogenous PPAR Agonist Nitro-Oleic Acid on Metabolic Syndrome in Obese Zucker Rats.

Nitroalkene derivatives of nitro-oleic acid (OA-NO(2)) are endogenous lipid products with novel signaling properties, particularly the activation of PPARs. The goal of this proposal was to examine the therapeutic potential of this OA-NO(2) in treatment of obesity and obesity-related conditions in obese Zucker rats. The animals were randomly divided to receive OA-NO(2), oleic acid (OA), both at 7.5 mug/kg/d, or vehicle ethanol via osmotic mini-pumps for 2 weeks. Following OA-NO(2) treatment, food intake was decreased as early as the first day and this effect appeared to persist throughout the e

Decreased fluid tolerance, accelerated transit, and abnormal motility of the human colon induced by oleic acid.

To determine whether the presence of unabsorbed fat in the colon altered colonic motility, intraluminal pressures were recorded in the terminal ileum and proximal colon, and serial 1-min gamma camera scans were obtained while test solutions labeled with diethylenetetramine pentaacetic acid chelate of indium 111 were infused into the middle portion of the ascending colon. Seven subjects received a control solution, and 6 subjects received an emulsion of oleic acid (4.3 g/100 ml). Oleic acid accelerated colonic transit; isotope accumulated in the rectosigmoid faster in the first 120 min (4343 +/

Albumin Nanoparticle-Based Delivery of Oxaliplatin-Oleic Acid Prodrug for Enhanced Breast Cancer Therapy.

Triple-negative breast cancer (TNBC) remains a therapeutic outlier, with limited targeted options and frequent relapse despite chemotherapy. While platinum therapy can benefit some TNBC cases, including BRCA1/2-mutant tumors, toxicity and limited tumor-selective exposure often restrict its impact. To address these barriers, we applied a lipid-metallodrug prodrug approach and synthesized an oxaliplatin-oleic acid (OXA-OA) conjugate that coupled OXA's cytotoxicity with OA-associated anticancer activity. The prodrug was encapsulated into genipin-crosslinked albumin nanoparticles (OXA-OA Alb NPs)

Obesity-induced oleic acid metabolic dysregulation may exacerbate osteoarthritis through the degradation of SOX9.

Obesity-related osteoarthritis (OA) is a degenerative joint disease accompanied by metabolic disorders. However, the specific metabolites and their molecular mechanisms remain unclear. Here, we establish a mouse model of obesity by feeding a high-fat diet (HFD) and find that HFD feeding promotes articular cartilage degeneration and accelerates OA progression. By using the untargeted metabolomics analysis, we identify oleic acid (OLA), a common monounsaturated fatty acid, as a significantly upregulated metabolite accumulating in articular cartilage. We further show that OLA disrupts extracellul
